This study was carried out to investigate the effect of new foot-bath facility and detergent solution (sodium molylbdenate, citrate, potassium nitrate, tataric acid, sodium hypo-cholorite, and zinc sulfate) on claw health in lactating dairy cows. Minimal inhibition concentration (MIC) and minimal bactericidal concentration (MBC) of copper sulphate were 0.31% for E. coli and Bacillus isolated from cows claw. The MIC and MBC of new detergent for E. coli were 1.25% and 5%, respectively, however their respectively values for Bacillus were noticed 0.63% and 2.5%. Both 5E. coli and Bacillus populations in petri-dishes were significantly reduced (more than 95%) with the application of new detergent solution (5% or 16%). Locomotion score (LS 1-5; very good to severely bad) of lactating cows were significantly improved with in 30 days with the use of new detergent solution in foot bath. The LS2 (n=16), LS3 (n=16), and LS4 (n=7) were shown 100%, 43.8%, and 14.3% recovery rate within 30 days with the use of new detergent solution. However, LS5 (n=2) were not recovered to normal claw health and locomotion score within 30 days of new detergent application. Usage of new detergent solution for 60 days in a foot bath have shown 81.3%, 71.4% and 50.0% recovery rate in cows with LS3, LS4 and LS5, respectively. Abnormal claw incidence was reduced from 18.8% to 1.5% in overall herd (n=80) with the use of new detergent solution (16%) in a foot bath for 90 days. In conclusion, usage of 16% of our detergent solution for 60 days in a foot bath can significantly improve the cow claw health and thus mitigate the negative effects of abnormal claw on productivity of cows and dairy farm income.
An integrated wastewater treatment pond system is developed for treatment and recycling of excreta from dairy cattle. It is composed of three ponds in series. A pit with a capacity of $10m^3$, 2-day hydraulic residence time, and overflow velocity of $1.5m^3m^{-2}day^{-1}$ is located internally in primary pond. It is designed for efficient sludge sedimentation and effective methane fermentation. It receives $5m^3/day$ of diluted cattle excreta by the water used for clearing stalls. A submerged gays collector for the recovery of methane is installed on the top of the pit. The average BOD_5 concentration of influent is 398.7mg/l. That of the effluent from primary pond is 49.2mg/l. About 88% of BOD_5 are removed in primary pond. It is assumed that about 60% of the influent BOD_5 is removed in the pit and that almost all of the carbon of the removed BOD_5 in the pit is converted to methane and carbon dioxide. Methane fermentation of the pit is well established at $16^{\circ}C$. This phenomena results from temperature stability, complete anaerobic condition, and neutral pH of the pit. Gas from the collector is almost 90% methane, less than 9% nitrogen, and less than 1% carbon dioxide. Thus a purified methane is produced, which can be used as energy source.
Chemical heat pump system of 2-propanol/acetone/hydrogen is most suitable to the recovery of waste heat of power plant. various types of 5 wt% Pt-alumina catalysts were prepared for 2-propanol dehydrogenation using sol-gel method. The characteristics and the dehydrogenation reaction rate of each catalyst were investigated. Pt-alumina xerogel catalyst has excellent reaction rate and good durability in comparison with the existing alumina supported Pt catalysts. Pt-alumina aerogel catalyst had the highest reaction rate in all prepared catalysts, but sufficient aging time was necessary to maintain its reaction rate. A potential advantage of the aerogel catalyst is the fact that the high temperature heat treatment is not required. Without heat treatment or with low temperature heat treatment, the Pt-alumina aerogel catalyst has excellent reaction rate as well as durability and this gives us the economic advantage. Alumina xerogel supported Pt catalyst prepared by incipient wetness method showed good reaction rate, and had good mechanical strength. Blank alumina xerogel prepared by sol-gel method can be used for the support of metal catalysts.

The aim of this study was to investigate method validation for determination of sesamol, sesamin, and sesamolin in non-fermented sesame and fermented sesame by bioconversion. For validation, the specificity, linearity, precision, accuracy, limits of detection (LOD), and quantification (LOQ) of sesamol, sesamin, and sesamolin were measured by HPLC. Linearity tests showed that the coefficients of calibration correlation ($R^2$) for sesamol, sesamin, and sesamolin were 0.9999. Recovery rates of lignan contents in non-fermented and fermented sesame were high in the ranges of 100.27~115.10% and 98.43~114.90%, respectively. The inter-day and intra-day precisions of sesamin and sesamolin analyses for non-fermented and fermented sesame were 0.27~1.94% and 0.25~0.69%, respectively. The LOD and LOQ were $0.23{\sim}0.34{\mu}g/g$ and $0.70{\sim}1.03{\mu}g/g$, respectively. These results indicate that the validated method is appropriate for the determination of sesamol, sesamin, and sesamolin.
This study assesses of efficiency of oocyte recovery and in vitro development for during the non breeding season in goat. Thirty-four matured goats, maintained in a pen under natural day length and fed hay ad libitum, were pretreated with progestagen implanted CIDR for 10 days. Superovulation treatment of the goats received twice daily intramuscular injections of a total of 70 mg FSH for 3 days from Day 8 of CIDR. All the gonadotropin treated goats were injected with 10 mg $PGF_2{\alpha}$ on Day 8 and 400~600 IU hCG in the afternoon on Day 10. Oocytes were recovered by follicle aspiration or oviduct flushing at 35 to 40 h after hCG injection through mid-ventral incision. The in vivo matured oocytes were activated by ionomycin (5 min) and 6-DMAP (3.5~4 h). The activated oocytes were cultured in mSOF medium containing 0.8% BSA at $38.5^{\circ}C$ in an atmosphere of 5% $CO_2$, 5% $O_2$, 90% $N_2$ for 7~8 days. There was no significant difference in the mean number of CL and in vivo matured and follicular oocytes recovered. But, quality of I+II grade follicular oocytes was lower (p<0.05) in the prepubertal goat (25.0%) than the adults (52.4%). The same results were also observed in the cleavage and blastocyst rate of activated oocytes. The clavage and blastocyst rate from prepubertal derived oocytes were lower (p<0.05) in the prepubertal goat (54.5%, 23.3%) than the adult goat (86.8%, 46.6%). Considering overall these results, we suggest that maturation of donor goats is a major factor affecting recovered oocytes quality and in vitro development of activated goat oocytes. There was no significant difference in oocyte quality between seasonal treatments.

Background : The incidence of nontuberculous mycobacterium (NTM) infections in Korea is increasing. This retrospective study was performed to examine the recovery rate of NTM from respiratory specimens as well as the isolated NTM colony characteristics, and to assess the clinical significance of a NTM isolation. Methods : The results of the respiratory specimens requested for an acid-fast bacilli (AFB) examination during 2002 at Asan Medical Center, along with the patients clinical characteristics were analyzed. Results : A total 26,820 respiratory specimens were requested for the acid-fast bacilli (AFB) smear and culture during the study period. The proportion of M. tuberculosis and NTM isolation was 5.7% and 2.2%, respectively. Among the AFB smear and culture positive specimens, 12.2% were found to be NTM. The scotochromogen showing a low colony count < 20, which appeared to be contaminants, were isolated in 31.8% of the 584 NTM isolates. Excluding the low-colony scotochromogens, the M. avium-intracellulare complex was the most common NTM isolates (42.1%), and was also the most common causative organism for NTM pulmonary diseases. 8.4% (23/275) and 17.8% (49/275) of patients with NTM isolates met the American and British Thoracic Society diagnostic criteria for NTM pulmonary disease, respectively. Conclusion : In case of a positive AFB-smear or culture result, the possibility of NTM being a causative organism should always be considered, even in Korea, which has an intermediate incidence of tuberculosis.
Feather, generated in large quantities as a byproduct of commercial poultry processing, is almost pure keratin, which is not easily degradable by common professes. Four strains, SMMJ-2, FL-3, NO-4 and RM-12 were isolated from soil for production of extracellular keratinolytic protease. They were identified as Bacillus sp. based on their morphological and physiological characteristics. They shown high protease activity on 5.0% skim milk agar medium and produced a substrate like mucoid on keratin agar medium. Bacillus sp. SMMJ-2 had a faster production time for producing keratinolytic protease than other strains. This strain did not completely degrade whole chicken feather for five days in basal medium but completely degraded whole chicken feather when supplied with nitrogen source for 40hours in keratinolytic producing medium ($0.7%\;K_{2}HPO_{4},\;0.2%\;KH_{2}PO_{4},\;0.1%$ fructose, 1.2% whole chicken feather, $0.01%\;Na_{2}CO_3$, pH 7.0). When supplied with chicken feather as nitrogen source, keratinolytic protease activity was 89 units/ml/min. When soybean meal was used as nitrogen source, the keratinolytic protease production reached a maximum of 106 units/ml/min after 48 hours under $30^{\circ}C$, 180 agitation. To isolate the keratinolytic protease, the culture filtrate was precipitated with $(NH_4)_{2}SO_4$ and acetone. The recovery rate of keratinolytic protease was about 96% after treatment with 50% acetone. The enzyme was stable in the range of $30{\sim}50^{\circ}C$ and pH $6.0{\sim}12.0$.

This study has been carried out to elucidate the characteristics of the dissolution for Thermal Decomposed Solid Waste of uranium-bearing sludge (TDSW), the removal of impurities by an alkalization in a nitric acid dissolving solution of TDSW, and the selective removal (/recovery) of uranium by an acidification in an carbonate alkali solution, respectively. TDSW generated by thermal decomposition of U-bearing sludge which was produced in the uranium conversion plant operation, was stored in KAERI as a solid-powder type. It is found that the dissolution of TDSW is more effective in nitric acid dissolution than oxidative-dissolution with carbonate. At 1 M nitric acid solution, TDSW was undissolved about 30wt% as a solid residue, and uranium contained in TDSW was dissolved more than 99%. In order to the alkalization for the nitric acid dissolving solution of TDSW, carbonate alkalization is more effective with respect to remove the impurities. At the carbonate alkali solution controlled to about 9 of pH, Al, Ca, Fe and Zn co-dissolved with U in dissolution step was removed about $98{\pm}1%$. On the other hand, U could be recovered more than 99% by an acidification at pH about 3 in a carbonate alkali solution, which was nearly removed the impurities, adding 0.5M $H_2O_2$. It was found that uranium could be selectively recovered (/removed) from TDSW.

This study proposed the method of phosphate recovery from municipal wastewater by using ferrous iron waste, generated from the mechanical process in the steel industry. In the analysis of XRD, ferrous iron waste was composed of $Fe_3O_4$ (magnetite), practically with $Fe^{2+}$ and $Fe^{3+}$. It had inverse spinel structure. In order to identify the adsorption characteristic of phosphate on ferrous iron waste, isotherm adsorption test was designed. Experimental results were well analyzed by Freundlich and Langmuir isotherm theories. Empirical constants of all isotherms applied increased with alkalinity in the samples, ranging from 1.2 to 235 $CaCO_3/L$. In the regeneration test, empirical constants of Langmuir isotherm, i.e., $q_{max}$ (maximum adsorption capacity) and b (energy of adsorption) decreased as the frequency of regeneration was increased. Experiment was further performed to evaluate the performance of the treatment scheme of chemical precipitation by ferrous iron waste followed by biological aerated filter (BAF). The overall removal efficiency in the system increased up to 80% and 90% for total phosphate (TP) and soluble phosphate (SP), respectively, and the corresponding effluent concentrations were detected below 2 mg/L and 1 mg/L for TP and SP, respectively. However, short-circuit problem was still unsolved operational consideration in this system. The practical concept applied in this study will give potential benefits in achieving environmentally sound wastewater treatment as well as environmentally compatible waste disposal in terms of closed substance cycle waste management.
BACKGROUND: The analytical method was established for determination of fungicide chinomethionat in several animal commodities using gas chromatography (GC) coupled with electron capture detector (ECD). METHODS AND RESULTS: In order to verify the applicability, the method was optimized for determining chinomethonat in various livestock products including beef, pork, chicken, milk and egg. Chinomethionat residual was extracted using acetone/dichloromethane(9/1, v/v) with magnesium sulfate and sodium chloride (salting outassociated liquid-liquid extraction). The extract was diluted by direct partitioning into dichloromethane to remove polar co-extractives in the aqueous phase. The extract was finally purified with optimized silica gel 10 g. CONCLUSION: The method limit of quantitation (MLOQ) was 0.02 mg/kg, which was in accordance with the maximum residue level (MRL) of chinomathionate as 0.05 mg/kg in livestock product. Recovery tests were carried out at two levels of concentration (MLOQ, 10 MLOQ) and resulted in good recoveries (84.8~103.0%). Reproducibilities were obtained (Coefficient of variation <5.2%), and the linearity of calibration curves were reasonable (r2>0.995) in the range of 0.01-0.2 ㎍/mL. This established analytical method was fully validated and could be useful for quantification of chinomathionat in animal commodities as official analytical method.
