Revealing the Spatial Distribution of Inorganic Elements in Rice Grains

  • Femtosecond laser ablation (fs LA) was used in this study to identify pollution by heavy metals and the distribution of elemental nutrients at different rice milling ratios. Polished rice (degrees of milling of 3, 5, 7, 9, and 11) was collected from major Korean supermarkets and one sample thereof was selected. An internal quality control experiment was conducted using a rice flour certified reference material from the Korea Research Institute of Standards and Science (KRISS CRM) for the evaluation of the efficacy. To assess the effectiveness of the analysis method, the reliability was validated using a food analysis performance assessment scheme (FAPAS), with chili powder serving as an external quality control. The results of the analysis of the inorganic elements Ti, Ca, Al, Fe and Mn in white and brown rice with degrees of milling of 3, 5, 7, 9 and 11 using ICP-MS, ICP-OES and AAS revealed contents of 0.40, 49.2, 2.43, 5.36 and 10.3 mg/kg in white rice and 0.59, 78.0, 7.52, 11.0 and 18.5 mg/kg in brown rice, respectively. Among the elements, there were remarkable differences in the measured contents. By comparing the contents of the elements at different degrees of milling, Ti, Co, As, Ca, Al, Cu, Fe, and Mn were determined to be distributed on the surface of the rice grains, whereas the contents of Cd and Pb increased toward the center of the rice grains, and Si was evenly distributed. After the quantitative analysis of rice samples polished to different degrees of milling, Ca and Al, which were contained in large amounts, and Si were analyzed with specificity by fs LA. The results show that Ca and Al were distributed in the rice husk (protective covering of rice) and Si was distributed in all parts of the rice.

Analysis of Impact Characteristics of Bonded Dissimilar Materials for Center Pillar (센터필라 적용을 위한 이종 접합강의 충격 특성 해석에 관한 연구)

  • This study was carried out to analyze the dynamic characteristics of laser tailor-welded blanks (TWBs) made of dissimilar materials. The analysis was performed using Hyper Works 10.0 with Solver LS-DYNA v.971. 2D-Shell was used as the modeling element, and the number of elements and nodes was 35,641 and 36,561, respectively. The impact speed was 10 km/h. To analyze the impact characteristics according to the height of the weld line for the upper and lower parts of the center pillar, the length of the lower part was set as 300 and 400 mm. When the lower part was made of SPFC980 steel with a length of 300 mm, the deformation was the smallest and the absorbed energy of the impact force was the largest. On based the lower part of center pillar, the position of TWB shows the shorter and the better value. In other words, the performance depended on the proportion of the upper part made of high-strength SABC1470 steel. A lower part made of SPFH590 steel showed large deformation. In contrast, a lower part made of SPFC980 steel showed significantly lesser deformation. Therefore, the impact performance of a lower part made of SPFC980 steel with a length of 300 mm showed the best analysis result.

Comparison of CH4 Emission by Open-path and Closed Chamber Methods in the Paddy Rice Fields (벼논에서 open-path와 closed chamber 방법 간 메탄 배출량 비교)

  • The closed chamber method, which is one of the most commonly used method for measuring greenhouse gases produced in rice paddy fields, has limitations in measuring dynamic $CH_4$ flux with spatio-temporal constrains. In order to deal with the limitation of the closed chamber method, some studies based on open-path of eddy covariance method have been actively conducted recently. The aim of this study was to compare the $CH_4$ fluxes measured by open-path and closed chamber method in the paddy rice fields. The open-path, one of the gas ($CO_2$, $CH_4$ etc.) analysis methods, is technology where a laser beam is emitted from the source passes through the open cell, reflecting multiple times from the two mirrors, and then detecting. The $CH_4$ emission patterns by these two methods during rice cultivation season were similar, but the total $CH_4$ emission measured by open-path method were 31% less than of the amount measured by closed chamber. The reason for the difference in $CH_4$ emission was due to overestimation by closed chamber and underestimation by open-path. The closed chamber method can overestimate $CH_4$ emissions due to environmental changes caused by high temperature and light interruption by acrylic partition in chamber. On the other hand, the open-path method for eddy covariance can underestimate its emission because it assumes density fluctuations and horizontal homogeneous terrain negligible However, comparing $CH_4$ fluxes at the same sampling time (AM 10:30-11:00, 30-min fluxes) showed good agreements ($r^2=0.9064$). The open-path measurement technique is expected to be a good way to compensate for the disadvantage of the closed chamber method because it can monitor dynamic $CH_4$ fluctuation even if data loss is taken into account.

Improvement of Transfer Alignment Performance for Airborne EOTS (항공용 전자광학추적장비의 전달정렬 성능 개선)

  • An Electro-Optical Tracking System (EOTS) is an electric optical system with EO/IR cameras, laser sensors, and an IMU. The EOTS calculates coordinates of targets, using attitude and acceleration measured by the IMU. In particular for an armed aircraft, the performance of the weapon system depends on how quickly and accurately it acquires the target coordinates. The IMU should be operated after alignment is complete, to meet the coordinate accuracy required by the weapon system so the initial stabilization time of the IMU should be reduced, by quickly measuring the attitude and acceleration. Alignment is the process of determining the initial attitude by resolving the attitude error of the IMU, and the IMU of mission equipment such as an airborne EOTS, uses velocity matching based on the velocity from GPS/INS for aircraft navigation. In this paper, a method is presented to improve the transfer alignment performance of the airborne EOTS, by maneuvering aircraft and the mission equipment. First, the performance factor of the alignment was identified, as a heading error through the velocity matching model and simulation results. Then acceleration maneuvers and attitude changes were necessary, to correct the error. As a result of flight tests applied to an EOTS on a OOO aircraft system, the transfer alignment performance was improved as the duration time was decreased, by more than five times when the aircraft accelerated by more than 0.2g and the EOTS was moving until 6.7deg/s.

Differences in Structural Characteristics and Eu(III) Complexation for Molecular Size Fractionated Humic Acid (분자량별 분류에 따른 휴믹산의 구조적 특성 및 Eu(III)과의 착물 반응 특성 비교에 대한 연구)

  • A humic acid(HA, Aldrich Co) sample was subjected to ultrafiltration for molecular size fractionation and three fractions of different nominal size($F_1$: 1,000-10,000 daltons; $F_2$: 10,000-50,000 daltons; $F_3$: 100,000-300,000 daltons) were obtained. The structural characteristics of the size-fractionated HA were analyzed using their IR and solid state C-13 NMR spectral data, and the carboxylate group contents of the humic acids were determined using their pH titration data. The $^7F_0-{^5}D_0$ excitation spectra of Eu(III) complexes of the size-fractionated mgHA in aqueous solution were acquired($[Eu(III)]=1.0{\times}10^{-4}mol\;L^{-1}$, $(HA)=470-970mg\;L^{-1}$) at pH 5.0 using a pulsed tunable laser system, in which metal binding properties of the size-fractionated HA were elucidated and compared on another. Characterization of the IR and C-13 NMR spectral data indicated that the fraction($F_3$) with molecules of larger size were primarily aliphatic, while the fractions($F_1$, $F_2$) with smaller molecules of less than 50,000 daltons were predominantly aromatic. Titration data were consistent with an increase in the number of carboxylate groups per unit mass as molecular size became smaller. The $^7F_0-{^5}D_0$ excitation spectral data of Eu(III)-humate complexes showed that the peak maxima on these spectra were shifted toward lower energies with increasing molecular size of HA, indicating the higher degree of bindings of the Eu in the molecules of larger size. We also discussed the relationship of the lower energy shifts of the maximum peaks with increasing the molecular size of HA with the structural differences of the size-fractionated HA.

Mineral Composition and Grain Size Distribution of Fault Rock from Yangbuk-myeon, Gyeongju City, Korea (경주시 양북면 단층암의 광물 조성과 입도 분포 특징)

  • This paper is focused on mineral compositions, microstructures and distributional characters of remained grains in the fault rocks collected from a fault developed in Yongdang-ri, Yangbuk-myeon, Gyeongju City, Korea, using X-ray diffraction (XRD), optical microscope, laser grain size analysis and fractal dimension analysis methods. The exposed fault core zone is about 1.5 meter thick. On the average, the breccia zone is 1.2 meter and the gouge zone is 20cm thick, respectively. XRD results show that the breccia zone consists predominantly of rock-forming minerals including quartz and feldspar, but the gouge zone consists of abundant clay minerals such as chlorite, illite and kaolinite. Mineral vein, pyrite and altered minerals commonly observed in the fault rock support evidence of fault activity associated with hydrothermal alteration. Fractal dimensions based on box counting, image analysis and laser particle analysis suggest that mineral grains in the fault rock underwent fracturing process as well as abrasion that gave rise to diminution of grains during the fault activity. Fractal dimensions(D-values) calculated by three methods gradually increase from the breccia zone to the gouge zone which has commonly high D-values. There are no noticeable changes in D-values in the gouge zone with trend being constant. It means that the bulk-crushing process of mineral grains in the breccia zone was predominant, whereas abrasion of mineral grains in the gouge zone took place by continuous fault activity. Mineral compositions in the fault zone and peculiar trends in grain distribution indicate that multiple fault activity had a considerable influence on the evolution of fault zones, together with hydrothermal alteration. Meanwhile, fractal dimension values(D) in the fault rock should be used with caution because there is possibility that different values are unexpectedly obtained depending on the measurement methods available even in the same sample.

Physiological and Proteome Responses of Korean F1 maize (Zea mays L.) Hybrids to Water-deficit Stress during Tassel Initiation (옥수수 영양생장기 한발 스트레스에 의한 광합성의 생리적 반응 및 프로테옴 변화 분석)

  • Severe droughts in spring have occurred frequently in Korea in recent years, exerting a critical impact on corn yield. Therefore, it is necessary to find physiological and/or molecular indicators of the response to drought stress in maize plants. In this study, we investigated the effects of water-deficit stress on two Korean elite F1 maize hybrids, Ilmichal and Gwangpyeongok, by withholding water for 10 days at tassel initiation. The water deficit drastically reduced the relative leaf water content, leaf number, leaf area, and stem length, leading to dry matter reduction. Moreover, it reduced the SPAD values and stomatal conductance of leaves in drought-stressed plants of both hybrids. Importantly, the number of leaves and SPAD value were non-destructive and easy to investigate in response to water-deficit stress, suggesting that they may be useful indicators for screening drought-tolerant genetic resources. We detected more than 100 spots that were differentially accumulated under drought stress. Of these spots, a total of 21 protein spots (≥1.5-fold) from drought-exposed maize leaves were successfully analyzed by MALDI-TOF-TOF mass spectrometry. Functional annotation using Gene Ontology analysis revealed that most of the identified proteins were involved in carbohydrate metabolism, stress response fatty acid catabolism, photosynthesis, energy metabolism, and transport. The protein expression levels were increased in both Ilmichal and Gwangpyeongok, except for triosephosphate isomerase, fructose-bisphosphate aldolase, and an uncharacterized protein. The lactoylglutathione lyase delta (3,5)-delta (2,4)-dienoyl-CoA isomerase was overexpressed in Gwangpyeongok only. The results obtained from this study suggest that the drought-specific genes may be useful as molecular markers for screening drought-tolerant maize genotypes.

  • Double-layered spheres play an important role in controlling drug delivery for pharmaceutical application, because of the low initial burst compared with single-layered spheres and targetable delivery to specific organ. But it has drawback in loading drug and controlling size. In this study, we developed double-layered spheres using relatively simple oil-in-water (O/W) solvent evaporation method witw/without ultrasonication and investigated the size variation of the double-layered microspheres on the contents of poly(lactide- co-glycolide) (PLGA). Double - layered spheres were char-acterized by scanning elecron microscope (SEM), camscope, and confocal fluorescence laser microscope (CFLM). Double-layered spheres showed smooth surfaces and obvious difference between core and corona by SEM observation and camscope. We observed the fluorescent core in the double-walled spheres composed of FlTC-dextran and PLGA using CFLM. It was found that the core of the microsphere was dextran and the corona of the fabricate microsphere was PLGA. Also, the more PLGA concentration, the more the size of the fabricating double-layered sphere observed.

A study on the differentiation of MC3T3-E1 incubated on the layer-built silica/polycaprolactone non-woven fabric produced by electrospinning (전기방사법으로 제조된 실리카/폴리카프로락톤 적층형 부직포에 배양한 골아 세포의 중식, 분화에 관한 연구)

  • Silica is known as a promising osteoconductive material, and polycaprolactone is a bioactive and degradable material. The purpose of this study was to monitor the differentiation of MC3T3-E1 cells cultured on the layer-built silica/poly caprolactone non-woven fabric produced by electrospinning. Non-woven fabric (silica, polycaprolactone, PSP, SPS) was made by electrospinning and they were inserted in the 48 well cell culture plate. MC3T3-E1 cells were prepared by subculture. Cells were seeded to each well $1{\times}10^5$ concentration per well. Dulbecco's modified eagle medium with 10% FBS and 1% antibiotic-antimycotic solution was used. Confocal laser scanning microscope was taken 4 hours after incubation (95% air. 5% $CO_2$, $37^{\circ}C$). Cell proliferation was monitored by spectrophotometer on 1, 7, 14 days, and the morphology of the growing cells was observed by field emission scanning electron microscope. To monitor the differentiation of osteoblasts on the materials, MC3T3-E1 cells were incubated in 48 well culture plate after seeding with the density of $1{\times}10^5$ concentration. Then ELISA kit & EIA kit were used on to assess osteocalcin and osteopontin expression respectively. The other conditions were the same as above. MC3T3-E1 cells were proliferated well on all of the materials. There were no statistical differences among them. The osteopontin expression of silica, PSP, SPS was significantly higher than other groups on day 3 (p/0,05), but after that time, there were no statistically signigicant differences. The osteocalcin expression was significantly higher in silica and PSP than other groups on day 14. These findings show that PSP was as good as silica on the effect of osteoblast differentiation. The PSP non-woven fabric may have the possibility as bone graft materials.

Effect of Fusion Condition on In Vitro Development of Caprine Cloned Oocytes with Nuclear Transfer (재래산양에 있어서 핵이식란의 융합조건이 융합 및 체외발달에 미치는 영향)

  • This study was conducted to examine the effects of electric stimulation conditions on in vitro developmental ability of caprine embryos after somatic cell nuclear transfer. Recipient oocytes were surgically collected after superovulation by using CIDR and FSH, PMSG, hCG and estrous synchronization in Korean native goats. The caprine ear cells were cultured in vitro in serum-starvation condition (TCM-l99 + 0.5% FBS) for 3 to 5 days of cell confluence. The zona pellucida of in vivo and in vitro matured oocytes were partially drilled using laser system. Single somatic cell was individually transferred into the enucleated oocyte. The reconstructed oocytes were electrically fused with 0.3M mannitol. After the electofusion, embryos were activated by electric stimulation or Ionomycin + 6-DMAP. Nuclear transfer embryos were cultured in mSOF medium supplemented with 0.8% BSA 6∼7 days at 39 , 5% $CO_2$, 5% $O_2$, 90% $N_2$. The fusion rate of donor cells was 60.4% and 40.3 % in ear cell and fetal fibroblast, and cleavage rate were 40.6% and 48.2%, respectively. No significant difference was found in the fusion and cleavage rate in different donor cells. Nuclear transferred oocytes were fused by electric pulses of 1.30∼1.40, 2.30∼2.39 and 2.40∼2.46 ㎸/cm. There was no significant difference among different electric pulses in fusion rates (26.7, 34.8 and 43.8%). The cleavage rate was higher (p<0.05) in 1.30∼1.40 ㎸/cm (82.9%) than 2.30∼2.39 ㎸/cm (43.8%) and 2.40∼2.46 ㎸/cm. (51.8%). The fusion rates of recipient oocyte source were 1st (43.5% and 23.6%), 2nd (55.7% and 39.2%) and 3rd (66.1% and 52.8%) in in vivo and in vitro oocytes. However, fusion ratee were significantly higher (p<0.05) in in vivo than in vitro oocyte. The cleavage rate of fused oocytes from in vivo and in vitro sources were 52.6% and 54.4%, respectively. No significant difference was found in the cleavage rate according to the recipient oocyte source. These results suggest that factors such as field pulse of electric stimulation and oocyte source could affect in vitro developmental ability of nuclear transplanted caprine oocytes.
