Chlorella ellipsoidea 엽록체막과 틸라코이드막의당지질 대사에 미치는 식품보존제의 효과 (The effect of Antiseptics on the Galactolipid Metabolism of Chlorella ellipsoidea Chloroplast and Thylakoid Envelope)

  • Chlorella ellipsoidea의 엽록체막 및 틸라코이드 막의 당지질 생합성과 그의 지방산 조성에 식품 보전제가 미치는 영향을 분석하고자 potassium sorbate(0.3 mM, P.S) , sodium bensoate(0.4mM, S.b), calcium propionate(2 mM, C.P)를 처리한 배지에서 세포를 배양한 후 배양초와 배양 중간기에 세포를 수확하여 막을 분리하고, 이들 막을 구성하고 있는 당지질 중 MGDG, DGDG의 함량 및 그의 지방산 조성을 대조구와 비교 분석하였다. Total lipid와 MGDG, DGDG 함량은 대조구와 비교해 볼 때 처리구에서 현저하게 감소하였다. 염록체막의 BGDG를 구성하는 지방산은 대조구에서는 palmitoleic acid 15.55% , oleic acid 15.09%가 이용되었으며, P.S 처리구는 linolenic acid 14.36%, oleic acid 14.71%, S.B 처리구는 palmitoleic acid 18.26%, oleic acid 17.26%, C.P 처리구는 palmitoleic acid 16.31%, oleic acid 16.88%를 당지할 합성에 주로 이용하였다. DGDG 형성에 이용된 지방산은 대조구의 경우 linolenic acid 17.74%, oleic acid 15.75%, P.S처리구는 palmitoleic acid 15.97%, oleic acid 14.90%, S.B 처리구는 oleic acid 15.74%, palmitoleic acid 13.29% C.P 처리구에서는 oleic acid 14.52%, palmitoleic acid 14.03%가 당지질 합성에 도입되었다. 틸라코이ㅡ막에서의 당지질 중 MGDG 생합성에 이용되는 지방산은 대조구에서는 linolenic acid 가 14.76%, oleic acid가 12.90% 사용되었으며 P.S 처리구에서는 palmitic acid와 palmitoleic acid가 각각 13.00%씩 이용되었다. S.B 처리구는 palmitoleic acid 12.94%, oleic acid 12.44% , C.P 처리구는 oleic acid 13.27 %, palmitoleic acid 9.66%를 당지질 생합성에 이용하였다. DGDG 합성에 이용되는 지방산은 대조구의 경우 linolenic acid 18.01% , oleic acid 15.53%로 나타났으며, P.S 처리구는 linolenic acid 19.20%, linoleic acid 14.14%, S.B 처리구는 oleic acid 14.85%, palmitoleic acid 9.03%, C.P 처리구는 linolenic acid 12.66%, olei acid 13.90%를 주로 이용하였다.

Expressions of Tumor-Related Proteins and $TGF-{\beta}1$ in Colon Cancer

  • This study was designed to investigate the correlation between the expression rate of p53 and p21 proteins, c-erbB-2 oncoprotein and $TGF-{\beta}1$ and tumor prognostic factors in colon cancer including the tumor size, histological differentiation and Dukes' stage. The expression rate of p53 protein was 11.4% (4 cases) at well differentiation, 48.6% (17 cases) at moderately differentiation, and 17.1% (6 cases) at poorly differentiation. In other words, the poorer differentiation, the higher the expression rate of p53 protein (P<0.05). The expression rate of p21 protein was 17.1% (6 cases) at well differentiation, 40.0% (14 cases) at moderately differentiation, and 8.6% (3 cases) at poorly differentiation, indicating that, as the histological malignant degeneration progressed, the expression rate of p21 protein decreased distinctively (P<0.05). However, the correlation of the above mentioned proteins with tumor size and Dukes' stage was not recognized. The expression rate of c-cerbB-2 oncoprotein was 11.4% (4 cases) at well differentiation, 54.3% (19 cases) at moderately differentiation, and 17.1% (6 cases) at poorly differentiation, indicating that the poorer differentiation, the higher expression rate of c-erbB-2 oncoprotein (P<0.05). The expression rate of $TGF-{\beta}1$ was 17.1% (6 cases) at well differentiation, 48.6% (17 cases) at moderately differentiation, and 11.4% (4 cases) at poorly differentiation. As Dukes' stage progressed, the expression rate of $TGF-{\beta}1$ was 8.6% (3 cases) in stage A, 20.0% (7 cases) in stage B, 37.1 % (13 cases) in stage C, and 11.4% (4 cases) in stage D. There was a difference in expression rates between Dukes' stages (P<0.05). In 10 cases, p53 protein was positive while p21 protein was negative, and in 6 cases, p53 protein was negative whereas p21 was positive (P<0.05). Therefore, a statistically significant inverse correlation between the expression rate of p53 protein and that of p21 protein was observed. In conclusion, since there was a signigicant correlation between histological differentiation of colon cancer and the expressions of p53 and p21 proteins and c-erbB-2 oncoprotein, and between Dukes' stage and the expression of $TGF-{\beta}1$, it was conformed that the overexpression of p53 and p21 proteins, c-erbB-2 oncoprotein, and $TGF-{\beta}1$ is closely associated with the occurrence of colon cancer and its progress. Accordingly, this study may be greatly beneficial to the presumption of diagnosis, treatment and prognosis of colon cancer patients.

Reversion of Multidrug Resistance by SKI-II in SGC7901/DDP Cells and Exploration of Underlying Mechanisms

  • In order to investigate whether SKI-II could reverse drug resistance and its possible mechanisms, we treated SGC7901/DDP cells with SKI-II or SKI-II in combination with DDP. Then cell growth, apoptosis, micromorphological changes, and expression of SphK1, P-gp, NF-${\kappa}B$, Bcl-2 and Bax were assessed by MTT assay, flow cytometry, electron microscopy, immunocytochemistry and Western blot assay respectively. SGC7901/DDP cells were insensitive to cisplatin 2.5mg/L, but when pretreated with SKI-II, their proliferation was inhibited by cisplatin 2.5mg/L significantly, the inhibition rate increasing with time and dose. The apoptosis rate was also significantly elevated. Expression of SphK1 and P-gp was decreased significantly, Pearson correlation analysis showing significant correlation between the two (r=0.595, P<0.01). Expression of NF-${\kappa}B$ and Bcl-2 was decreased significantly,while that of Bax was increased, compared to the control group. There were significant correlations between SphK1 and NF-${\kappa}B$(r=0.723, P<0.01), NF-${\kappa}B$ and Bcl-2(r=0.768, P<0.01). All these data indicated that SKI-II could reverse drug resistance of SGC7901/DDP to cisplatin by down-regulating expression of P-gp and up-regulating apoptosis through down-regulation of SphK1. The increased apoptotic sensitivity of SGC7901/DDP to cisplatin was due to the decreasing proportion of Bcl-2/Bax via down-regulating NF-${\kappa}B$.

폐동맥폐쇄와 주대동맥폐동맥혈관부행지를 동반한 활로씨사징증 -다양한 외과적 치료접근의 비교- (Tetralogy of Fallot Associated with Pulmonary Atresia and Major Aortopulmonary Collateral Arteries -Comparison between the different surgical approaches-)

  • 배경: 폐동맥폐쇄와 주대동맥폐동맥혈관부행지를 동반한 활로씨사징증 환아들은 여러가지 이질적인 외과적인 원칙하에 치료되어 왔다. 본 연구에서는 상기 질환으로 본원에서 치료를 받은 환아들을 대상으로 그동안 시도하였던 5 가지 서로 다른 외과적인 치료전략을 중심으로 그 장단점을 비교하고자 하였다. 대상 및 방법: 1986년 1월부터 1998년 6월까지 본 질환을 가진 50명의 환아를 대상으로 최초 수술방법에 따라 일단계단일화술군(A-1, n=9), 단계적단일화술군(A-2, n=11), 우심실-폐동맥 연결술군(B-1, n=11), 우심실-폐동맥연결/단일화술 동시 시행군(B-2, n=11), 일단계 완전교정술군(C, n=8) 등 5군으로 분류하고 군별 형태학적 특성, 수술결과, 완전교정 가능성, 추적결과 등을 비교 분석하였다. 50명 환아들의 제 1차 수술당시 평균연령은 39$\pm$31 (1-177) 개월이었다. 결과: 각 군의 최초 수술 당시 평균 연령은 각각 57$\pm$18(A-1), 42$\pm$48(A-2), 18$\pm$14(B-1), 52$\pm$55(B-2), 32$\pm$34(C) 개월이었다 (p<0.05). 주대동맥폐동맥혁관부행지의 평균 숫자는 일단계 완전교정술 시행 군에서 가장 적었다 (A-1=4.3$\pm$1.0, A-2=4.5$\pm$1.3, B-1=4.1$\pm$1.9, B-2=4.1$\pm$1.6, C=3.4$\pm$1.8 : p<0.05). 직접 및 간접 주대동맥폐동맥혁관부행지 숫자의 비는 군간에 차이가 없었다 (A-1=91%/9%), A-2=78%/22%, B-1=80%/20%, B-2=80%/20%, C=81%/19% : p>0.05). 19명의 환아는 1개 이상의 의존성 주대동맥폐동맥혈관부행지를 가지고 있었다. 13명의 환아는 진폐동맥이 존재하지 않았으며 29명의 환아의 진폐동맥은 좌우폐동맥 연결이 있었다. 수술은 50명의 환아에 대하여 총 101 (1-4)회 시행되었으며 각 군의 최초 수술에 따른 수술사망율은 각각 33%(3/9, A-1), 18%(2/11, A-2), 0%(0/11, B-1), 36%(5/11, B-2), 13%(1/8, C)였다 (p<0.05). 군별 심실중격결손의 완전폐쇄까지를 포함한 완전교정술을 완료한 환아의 백분율은 각각 22%(2/9, A-1), 18%(2/11, A-2). 45%(5/11,B-1), 27%(4/11,B-2), 100%(8/8,C)(p<0.05)였다 (p<0.05). 사망원인은 저산소성 호흡부전 및 심부전(6), 뇌손상(2), 패혈증(4) 등이었고 술후 합병증으로 횡격막신경마비가 5례에서 발생하였다. 6례의 추적실패를 제외한 생존환자 31례는 평균 74$\pm$42 (3-145) 개월 외래추적되었으며 1년 및 5년 생존율이 각각 73%, 73%였다. 결론: 연구자등은 본 연구를 통하여, 본 질환에 대하여 제 1단계로 시행한 시술로 우심실-폐동맥 연결술이 가장 안전하고도 완전교정술 도달 확률이 높은 치료전략이라는 사실을 입증하였으며 주대동맥폐동맥혈관부행지의 크기나 숫자가 단일화하기 쉬운 형태학적 특징을 지닌 경우에는 조기에 일단계완전교정술을 시행하여 양호한 결과를 얻을 수 있다는 사실을 발견하였다. 반면 본 환아군 중 단일화술을 먼저 시도한 군에서는 비록 단계적인 단일화를 시도한 군에서 단일화술과 관계된 수술사망율이 약간 낮기는 하였으나 완전교정술까지 완료될 가능성에는 차이가 없었다. 그러나 이 경우 보다 정련된 적응 환자의 선택을 통한 단일화 우선전략의 시도와 장기 추적결과의 관찰이 요구된다.

65 nm CMOS 공정을 이용한 W-대역 전력증폭기 설계 (Design of a W-Band Power Amplifier Using 65 nm CMOS Technology)

  • 본 논문에서는 차량 충돌 방지 장거리 레이더(Long Range Radar: LRR)을 위한 77 GHz 전력증폭기를 65 nm CMOS 공정을 이용하여 설계하였다. 제안한 회로는 3단 차동 전력증폭기로 공통 소스 구조와 트랜스포머를 사용했다. 측정결과로 77 GHz에서 18.7 dB의 전압 이득과 13 GHz의 3 dB 대역폭을 얻었다. 측정된 최대 출력 전력은 10.2 dBm, 입력 $P_{1dB}$는 -12 dBm, 출력 $P_{1dB}$는 5.7 dBm이며, 측정된 최대 전력 효율은 7.2 %이다. 본 전력증폭기는 1.2 V의 공급전원으로부터 140.4 mW의 DC 전력을 소모한다.

비할로겐 M-P 난연제 제조 및 복합재료 응용 연구 (A Study on the Preparation of Halogen Free M-P Flame Retardant and Its Application to Composite Material)

  • In order to improve flame retardancy, the halogen free organic melamine phosphate(M-P) flame retardant was synthesized from melamine and phosphoric acid by the reaction of precipitation. The ignition test was carried out preparing hybrid flame retardant compound($H_bFRC$) consisting of organic M-P and inorganic Mg$(OH)_2$ as a flame retardant in the polyolefin resins. The flame retardancy and mechanical properties of flame retardant aluminum composite panel($H_bFRC$-ACP) were performed to investigate the possibility of the composite material, which was contained M-P, as a inner core for $H_bFRC$-ACP. For this study, the results of ignition test indicate that a char formation and drip suppressing effect, and combustion time reduced as the content of M-P increased. The limited oxygen index(LOI) values were measured 17.4vol% and 31.5vol% for LDPE only and $H_bFRC$-3(M-P content: 15wt%), respectively. And it was verified that the $H_bFRC$-3 was needed more oxygen quantity with the increase of M-P content when it combustion. Also, the results from thermogravimetric analysis were observed endothermic peak at $350^{\circ}C$ and $550^{\circ}C$, it was confirmed predominant thermal stability though the wide temperature range by the mixture of M-P and Mg$(OH)_2$. The LDPE-ACP (using only LDPE as a inner core), $35.13kW/m^2$ of heat release rate(HRR) and 13.43MJ/m2 of total heat release(THR) were measured while the $H_bFRC$-ACP, $10.44kW/m^2$ of HRR and 1.84MJ/m2 of THR were measured by results of cone calorimeter test. In case of $H_bFRC$-ACP, the average gas emission amount of CO and $CO_2$ could be decreased down to 25% and 20%, respectively, in comparison with LDPE-ACP. The mechanical properties such as tensile strength, bending strength and adhesion strength of $H_bFRC$-ACP were revealed slightly high values $54N/mm^2$, $152N/mm^2$ and 120N/25mm, respectively, compared with LDPE-ACP. It was confirmed that flame retardancy was improved with the synergy effect because of char formation by M-P and hydrolysis by Mg$(OH)_2$. The result of this study suggest that $H_bFRC$ can be applied for an adequate halogen free flame retardant composite material as a inner core for ACP.

The Combined Effects of Ginkgo Biloba Extracts and Aspirin on Viability of SK-N-MC, Neuroblastoma Cell Line in Hypoxia and Reperfusion Condition

  • Objective: The purpose of this study is to investigate the combined effects of ginkgo biloba extract, ginkgolide A and B and aspirin on SK-N-MC, human neuroblastoma cell viability and mRNA expression of growth associated protein43 (GAP43), Microtubule-associated protein 2 (MAP2), B-cell lymphoma2 (Bcl2) and protein53 (p53) gene in hypoxia and reperfusion condition. Methods: SK-N-MC cells were cultured with Dulbecco's Modified Eagle's Medium (DMEM) media in $37^{\circ}C$, 5% $CO_2$ incubator. The cells were cultured for 8 hours in non-glucose media and hypoxic condition and for 12 hours in normal media and $O_2$ concentration. Cell survival rate was measured with Cell Counting Kit-8 (CCK-8) reagent assay. Reverse transcriptase polymerase chain reaction (RT-PCR) was used to estimate mRNA levels of GAP43, MAP2, Bcl2, and p53 genes. Results: The ginkgolide A and B increased viable cell number decreased in hypoxic and reperfused condition. The co-treatment of ginkgolide B with aspirin also increased the number of viable cells, however, there was no additive effect. Although there was no increase of mRNA expression of GAP43, MAP2, and Bcl2 in SK-N-MC cells with individual treatment of ginkgolide A, B or aspirin in hypoxic and reperfused condition, the co-treatment of ginkgolide A or B with aspirin significantly increased GAP43 and Bcl2 mRNA levels. In MAP2, only the co-treatment of ginkgolide A and aspirin showed increasing effect. The mRNA expression of p53 had no change in all treating conditions. Conclusion: This study suggests that the combined treatments of Ginkgo biloba extracts and aspirin increase the regeneration of neuroblastoma cells injured by hypoxia and reperfusion.

Artemisia fukudo essential oil attenuates LPS-induced inflammation by suppressing NF-${\kappa}B$ and MAPK activation in RAW 264.7 cells

  • In the present study, the chemical constituents of Artemisia fukudo essential oil (AFE) were investigated using GC-MS. The major constituents were ${\alpha}$-thujone (40.28%), ${\beta}$-thujone (12.69%), camphor (6.95%) and caryophyllene (6.01%). We also examined the effects of AFE on the production of nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin-IL-$1{\beta}$ (IL-$1{\beta}$), and IL-6 in lipopolysaccharide (LPS)-activated RAW 264.7 cells. Western blotting and RT-PCR analyses indicated that AFE has potent dose-dependent inhibitory effects on pro-inflammatory cytokines and mediators. We investigated the mechanism by which AFE inhibits NO and $PGE_2$ by examining the level of nuclear factor-${\kappa}B$ (NF-${\kappa}B$: p50 and p65) activation within the mitogen-activated protein kinase (MAPK: ERK, JNK and p38) pathway, which is an inflammation induced signal pathway in RAW 264.7 cells. AFE inhibited LPS-induced ERK, JNK and p38 phosphorylation. Furthermore, AFE inhibited the LPS-induced phosphorylation and degradation of $I{\kappa}B-{\alpha}$, which is required for the nuclear translocations of the p50 and p65 NF-${\kappa}B$ subunits in RAW 264.7 cells. Our results suggest that AFE might exert an anti-inflammatory effect by inhibiting the expression of pro-inflammatory cytokines. Such an effect is mediated by a blocking of NF-${\kappa}B$ activation which consequently inhibits the generation of inflammatory mediators in RAW 264.7 cells. AFE may be useful for treating inflammatory diseases.

Cloning and Expression of the Gene Encoding Glucose Permease of the Phosphotransferase System from Brevibacterium flavum in Escherichia coli

  • A Brevibacterium flavum gene coding for glucose permease of the phosphoenolpyruvate-dependent phosphotransferase system (PTS) was cloned by complementing the Escherichia coli ZSCl13 mutations affecting a ptsG gene with the B. flavum genomic library. From the E. coli clone grown as red colony on a MacConkey plate supplemented with glucose as an additional carbon source, a recombinant plasmid was isolated and named pBFT93. The plasmid pBFT93 was identified as carrying a 3.6-kb fragment of B. flavum chromosomal DNA which enables the E. coli transformant to use glucose or man nose as a sole carbon source in an M9 minimal medium. The non-metabolizable sugar analogues, 2-deoxy-D-glucose (2-DG) and methyl-$\alpha$-D-glucopyranoside (MeGlc) affected the growth of ZSCl13 cells carrying the plasmid pBFT93 on minimal medium supplemented with non-PTS carbohydrate, glycerol, as a sole cabon source, while the analogues did not repress the growth of ZSCl13 cells without pBFT93. It was also found that both $2-deoxy-D-[U-^{14}C]glucose{\;}and{\;}methyl-{\alpha}-D-[U-^{14}C]glucopyranoside$ could be effectively transported into ZSCl13 cells transformed with plasmid pBFT93. Several in vivo complementation studies suggested that the B. flavum DNA in pBFT93 encodes a glucose permease specific for glucose and mannose.

생후 6개월 미만의 영아에서 백일해에 대한 적극적인 감시활동: 2011-2013년 단일기관 연구 (Active Surveillance of Pertussis in Infants Under 6 Months of Age: A Single Center Experience from 2011 to 2013)

  • 목적: 본 연구에서는 6개월 미만의 영아를 대상으로 백일해의 기초적인 역학조사를 확인하고자 단일 기관에서 실시하였다. 방법: 2011년 10월부터 2013년 4월까지 분당 차병원에 내원한 진찰 당시 호흡기계 증상을 보여서 입원한 생후 6개월 미만의 영아들을 대상으로 비인두 흡입물로 B. pertussis PCR 또는 배양검사를 시행하였다. 또한 respiratory syncytial virus, adenovirus, metapneumo-virus, rhinovirus, parainfluenza virus, influenza A, B virus 포함한 호흡기 바이러스들에 대한 multiplex RT-PCR 검사를 시행 후 분석하였다. 결과: 대상 환아 수는 총 79례였으며 환아 연령 중앙값은 2개월이었고 대부분 진단은 급성 세기관지염(60%)이거나 폐렴(28%)이었다. B. pertussis 이 확인된 경우는 13례(16%)였으며 환아 연령 중앙값은 2개월이었다. B. pertussis 이 확인된 13례 중 7례(53%)에서 respiratory syncytial virus가, 1례(7%)에서 influenza A virus가 확인되었다. B. pertussis 이 확인된 13증례 중 DTaP 접종력 없는 증례는 6례(46%)이었고 DTaP 백신 1회 접종한 증례는 6례(46%)이었고 DTaP 백신 2회 접종한 증례는 1례(8%)이었다. 결론: 호흡기계 증상들을 보이는 입원한 생후 6개월 미만의 영아의 원인 병원체 중 B. pertussis 이 16%를 차지하였다. 6개월 미만의 영아들에 대한 향후 전국적인 다 기관이 포함된 역학적 감시 활동이 필요할 것으로 사료된다.
