Antibacterial Activity of Edible Plant against Pathogenic Bacteria 1. Antibacterial Activity of Clove against Staphylococcus aureus (식용식물의 식중독세균에 대한 항균작용 1. Staphylococcus aureus에 대한 Clove의 항균작용)

  • The antibacterial activity of clove (Eugenia caryophyllata Thumb) in culture troth against S. aureus was tested at 35, 5, -20, 50, 55 and 60$^{\circ}C$. Tryptic soy broth(TSB) containing 0∼0.5%(w/v) of clove was inoculated with 105∼107 CFU/ml of S. aureus and incubated at each temperature. The growth of S. aureus occured at 0.1% clove after a prolonged lag period while viable cells of S. aureus decreased more than 2 log cycles at 0.3 and 0.4% clove during 12 hours storage at 35$^{\circ}C$. During 32 days of refrigerated storage at 5$^{\circ}C$, survivors of S. aureus were decreased with the progress of time and increasing clove concentration. At the presence of 0.2, 0.3 and 0.4% clove, bacterial cells were dead after 32, 20 and 16 days of refrigerated storage, respectively. During 32 days of frozen storage at -20$^{\circ}C$, survivors of S. aureus were decreased less than 0.5 log cycle at 0% clove. At the presence of 0.1∼0.4% clove, survivors were decreased 2.5∼3.0 log cycles after 1 day and then decreased 0.4∼0.8 log cycles through the frozen storage. There were small changes in populations of S. aureus in TSB between different concentrations of clove during frozen storage. The D-values of S. aureus at clove 0, 0,2, 0.4% were 28.53, 15.14, 8.9 min at 50$^{\circ}C$, 18.43, 10.32, 6.74 min at 55$^{\circ}C$ and 12.78, 9.88, 5.72 min at 60$^{\circ}C$, respectively. The D-values for S. aureus were decreased with the increasing temperature and clove concentration.

  • Purpose: The purpose of this study was to investigate the quality characteristics and antioxidant activity on consequent storage based on the temperature of new cultivar dew-drop pine mushroom (Lentinula edodes GNA01). Methods: Dewdrop Pine Mushroom were prepared under different storeage temperature (4, 10, $20^{\circ}C$). Results: Weight loss and hardness showed the least reduction rate when it was stored at $4^{\circ}C$ (p<0.05). The color value changed to dark brown at all storage temperatures during the storage period. When it was stored at $4^{\circ}C$, it maintained its initial color for a longer period than at different storage temperatures. With respect to the change in its viable cell count, the cells proliferated to less than 3.0 log CFU/g up to Ed: Please review the change. The earlier part was difficult to understand 2.83 log CFU/g until 15 days of storage at $4^{\circ}C$. On measuring the antioxidant activity of this mushroom, the polyphenol content was maintained without a large change until 9 days of storage at $4^{\circ}C$. The electron-donating action maintained high antioxidant activity, accounting for 81.99% until 12 days of storage from 83.08% during the initial storage at $4^{\circ}C$. When it was stored at $4^{\circ}C$, the sensory characteristics received the highest score among all items, such as appeaance, color, flavor and general preference, etc. Concolusion: In summary, new Cultivar Dewdrop Pine Mushroom (Lentinula edodes GNA01) can maintain its commercial value until the 12th day of $4^{\circ}C$ storage.

  • The experiment was canied out to investigate the effects of cutting frequencies (3, 4 and 5 cuts per year) in relation to fertilization of mineral nitrogen on crude protein yield and the contents of crude protein and acid detergent fibre in orchardgms(Dacty1is glomerata L.). Tne results are as follows : 1. The content of crude protein increased according to the increase in cutting frequency and showed the highest in the 1st cut in all treatments. At 4 and 5 cutting frequencies per year, the content of crude protein increased in a large extent according to the increased fertilization of mineral nitrogen. However at 3 cutting frequency, the content of cmde protein appeared higher for no application of mineral nitrogen than for the treatments with fertilization. 2. 'Ihe content of acid detergent fibre(ADF) was considerably reduced at 4 and 5 cutting frequencies and in all treatments, the lowest were recorded in the 1st cut or in the last cut. At 3, 4 and 5 cutting frequencies, the highest ADF were shown in the 2nd cut, 3rd cut and 3rd cut respectively. However, the general trend was that the content of ADF was greatly reduced according to increased level of mineral nitrogen fertilization. 3. The yeild of cmde protein was the highest at 5 cutting frequency(l.2 ton DMIha) and it indicated that the yield of cmde protein increased in accordance to the increase in fertilization of mineral nitrogen and cutting frequency. 4. Transdermal Delivery of Ketoprofen and the Effect of Electroosmosis (케토프로펜의 경피전달 및 전기삼투압의 영향)

  • We investigated some important factors which affect the transdermal flux of ketoprofen, a nonsteroidal anti-inflammatory agent, as a first step to provide some basic knowledge for the development of a iontophoretic transdermal patch system. Factors such as current density, polarity, buffer (HEPES) and electrolyte concentration and pH were studied using hairless mouse skin. The effect of poly(L-lysin), which is known to affect the electro-osmotic flow through skin, on flux was also studied. Passive flux was about $20\;{\mu}g/cm^2hr$ at pH 4.0, but was negligible at pH 7.4 where all ketoprofen molecules dissolved are ionized (ketoprofen pKa=5.94). At pH 4.0, application of anodal current increased the flux further above the passive level, however anodal flux at pH 7.4 was much smaller than passive flux at pH 4.0. The application of cathodal current at pH 4.0 increased the average flux to $30-40\;{\mu}g/cm^2hr$, depending on the current density applied. At pH 7.4, cathodal flux was only about $5\;{\mu}g/cm^2hr$. Decrease in buffer and electrolyte concentration increased this cathodal flux about 10 fold. However decrease in HEPES buffer concentration 100 fold did not affect the flux. Anodal flux of acetaminophen was much larger than cathodal flux, indicating that electroosmotic flow can be playing an important role in the flux. Poly(L-lysin) increased the cathodal flux at pH 7.4. These results provide some important insights into the mechanism of transdermal flux of ketoprofen and the role of electroosmotic flow.

Comparative Study of the Dissolution Profiles of a Commercial Theophylline Product after Storage

  • The purpose of this work was to study the effect of storage time and temperature on the in vitro release kinetics of a commercial sustained-release dosage form of theophylline, at different pHs of the dissolution medium. The formulation was stored at $35^{\circ}C$ for 16 months and at $45^{\circ}C$ for 8 months, with a relative humidity of 60%. The in vitro release tests were performed at pHs 2, 4, 6 and 7.4. The mean values of the transport coefficient n, were close to 0.5 in all the conditions tested, which indicates that the transport system is not modified after storage of the formulation at $35^{\circ}C$ and $45^{\circ}C$. The mean values of the dissolution rate constant ranged from 0.036 to 0.043 $min^{-n}$, under all the conditions tested. Significant differences (${\alpha}=0.05$) were found between pHs 2, 4 and 6, 7.4 for all the model-independent parameters studied. When the formulation was kept at $35^{\circ}C$ for 16 months, the mean percentage of drug dissolved at 8 hours was 25.61% (pHs 2, 4) and, 36.12% (pHs 6, 7.4), representing a 26% and 24% reduction, respectively. Simitar results were obtained after storing the formulation at $45^{\circ}C$ for 8 months, corresponding to 33.3% (pHs 2, 4) and, 22.5% (pHs 6, 7.4) diminution, respectively. The values of the similarity factory $f_2$, obtained were lower than 50, which indicates the lack of similarity among the dissolution profiles, after storing the formulation under the experimental Conditions tested.

  • Seasonal variations of the contents of some chemical constituents of the estuary water at two definite stations of the laver bed in Nack Dong River have been determined over one tidal cycle in spring tide from Nov. 1962 to Oct. 1963. The ranges of annual variations of the contents at station 1 and station 2 are as follows: water temp. $2.2-30.8^{\circ}C$, $3.3-28.0^{\circ}C$; pH 7.8-8.5, 7.9-8.4; chlorosity 0.025-19.66 g/l, 4.31-19.56 g/l; magnesium 0.00355-1.565 g/l, -1.524 g/l; calcium 0.00557-0.482 g/l, - -0.590 g/l; saturation % of dissolved oxygen 71.8-123.2%, 88.2-113.8%; silicate-Si 8.00-125.5 ${\mu}$g-at./l, 6.70-100.5 ${\mu}$g-at./l; phosphate-P 0.12-1.47 ${\mu}$g-at./l, 0.11-1.09 ${\mu}$g-at./l; ammonia-N 4.88-25.45 ${\mu}$g-at./l, 4.12-17.58 ${\mu}$g-at./l; nitrite-N 0.07-0.75 ${\mu}$g-at./l, 0.08-0.58 ${\mu}$g-at./l; nitrate-N 2.11-6.89 ${\mu}$g-at./l, 1.85-7.43 ${\mu}$g-at./l each. The annual tidal variations of the constituents at station 1 are more remarkable than of station 2. The chlorosity, magnesium and calcium contents are decreased nearing the slack after ebb, and increased abruptly then one hour after the slack. The contents of the other constituents are varied according to the chlorosity variety. The values of pH, chlorosity, magnesium and calcium contents are lower in summer than winter, while the difference of seasonal variations of the % saturation of dissolved oxygen is not remarkable. The phosphate-P and total nitrogen contents have a tendency of increasing within a definite range, while the silicate-Si increase proportionally, to the increasing of mixing percentage of fresh water. The average values of Si/P and N/P are several times greater than of the normal in sea water. The chemical composition considered from the value of Mg/Cl or Ca/Cl of estuarine water varies according to the variety of chlorosity, even at the high chlorosity of 19 g/l.

Quality Changes and Shelf-life of Cut Cabbage Kimchi under Various Storage Temperatures and Packing Materials (저장온도 및 포장재에 따른 절단배추 김치의 품질변화 및 Shelf-life)

  • To study the shelf-life of cut cabbage kimchi, we examined the several quality characteristics of cabbage kimchi under various storage temperatures and packing materials. The pH of kimchi extracts were decreased to 4.0 in the condition of storage at 8$^{\circ}C$ during 6 days, at 4$^{\circ}C$ during 18days, 0$^{\circ}C$ during 42 days. The increase patterns of the organic acid were reverse to the changes of pH under the all conditions. The contents of reducing sugar were continuously decreased at 8$^{\circ}C$ , but increased gradually at 0$^{\circ}C$ and 4$^{\circ}C$ storage conditions. The contents of vitamine C were decreased at the early storage but increased to optimum ripening stage of pH 4.2-4.4 and after decreased continuously. The total bacterial cell counts of cabbage kimchi were increased suddenly at the early storage and after decreased gradually. And at the higher storage temperature, the more lactic acid bacteria were. On the results of sensory evaluation of cabbage kimchi at marketable shelf-life, sour taste and sour smell were significant under all experimental conditions by Duncan's multiple range test. On the above all results, we concluded that the marketable shelf-life of cut cabbage kimchi were 42 days, 19 days, 6 days at 0$^{\circ}C$, 4$^{\circ}C$, 8$^{\circ}C$ storage condition respectively.

  • Objectives : The aim of this study was to examine the effects of Acupuncture at the GV20 and GV22 on normal human beings using power spectrum analysis. Methods : Electroencephalogram(EEG) power spectrum exhibits site-specific and state-related differences in various frequency bands. 8 channels Background Electroencephalogram (EEG) was carried out in 30 subjects(24 females and 4 males). Results : In ${\delta}$(theta) band, the power values decreased significantly at the 8-channel average value(p=0.03) and especially at T3(p=0.02), T4(p=0.001) and P3(p=0.03). In ${\alpha}$(alpha) band, the power values have no significant changes. In ${\beta}$(beta)band, the power values increased significantly at the 8-channel average value (p=0.02) and especially at T4(p=0.003), P3 (p= 0.03) and P4(0.02). In ${\beta}/{\delta}$(beta/theta) ratio, the value increased significantly at the 8-channel average value(p=0.002) and especially at Fp2(p=0.05), F4(p=0.007), T3(0.012), T4(0.005), P3 (0.007) and P4(0.03) Conclusions : Through this data, we conclude that acupuncture at the GV20 and GV22 on normal human beings could have possibility to awake the cerebral cortex by the functional mechanism.

Vertical Variation of Total Bacterial number in Daechung Reservoir (대청호에서 세균의 수직적 분포에 관한 연구)

  • To define the ecological roles of bacterial community in Daechung Reservoir. Vertical variation of total bacterial numbers were estimated at MAN site for one year with DAPI staining. The bacterial numbers were counted, environmental factors were monitored at the surface, -5m, -10m, -15m and -25 m of MAN sited and tried to explain the vertical bacterial numbers with environmental factors. The bacterial number was $1.6-1.7.0 x10^6$ cells/ml at the surface, $2.3-11.0x10^6$ cells/ml at 5 m depth, $1.2-1.4.0x10^6$ cells/ml at 10 m depth, $1.4-15.0x10^6$ cells/ml at 15 m depth and $1.4-1.3.0x10^6$ cells/ml at 25 m depth. The Mean bacterial number at the surface was more that those at any other depth. The explanation of the vertical total bacterial numbers with environmental factors were suggested.

Physiological Regulation of an Alkaline-Resistant Laccase Produced by Perenniporia tephropora and Efficiency in Biotreatment of Pulp Mill Effluent

  • Regulation of alkaline-resistant laccase from Perenniporia tephropora KU-Alk4 was proved to be controlled by several factors. One important factor was the initial pH, which drove the fungus to produce different kinds of ligninolytic enzymes. P. tephropora KU-Alk4 could grow at pH 4.5, 7.0, and 8.0. The fungus produced laccase and MnP at pH 7.0, but only laccase at pH 8.0. The specific activity of laccase in the pH 8.0 culture was higher than that in the pH 7.0 culture. At pH 8.0, glucose was the best carbon source for laccase production but growth was better with lactose. Low concentrations of glucose at 0.1% to 1.0% enhanced laccase production, while concentrations over 1% gave contradictory results. Veratryl alcohol induced the production of laccase. A trace concentration of copper ions was required for laccase production. Biomass increased with an increasing rate of aeration of shaking flasks from 100 to 140 rpm; however, shaking at over 120 rpm decreased laccase quantity. Highest amount of laccase produced by KU-Alk4, 360 U/mL, was at pH 8.0 with 1% glucose and 0.2 mM copper sulfate, unshaken for the first 3 days, followed by addition of 0.85 mM veratryl alcohol and shaking at 120 rpm. The crude enzyme was significantly stable in alkaline pH 8.0~10.0 for 24 hr. After treating the pulp mill effluent with the KU-Alk4 system for 3 days, pH decreased from 9.6 to 6.8, with reduction of color and chemical oxygen demand at 83.2% and 81%, respectively. Laccase was detectable during the biotreatment process.
