종양 표지 인자로서 혈장 Transforming Growth Factor-β1에 대한 연구 (Study of plasma transforming growth factor-β1 level as a useful tumor marker in various cancers)

  • Background : Many investigators have found transforming growth factor-${\beta}1$ (TGF-${\beta}1$) to be elevated in tumors. Changes in responsiveness to TGF-${\beta}1$ have been linked to malignant transformation, tumor progression and tumor regression. Many malignant cell lines of epithelial or hematopoietic origin are refractory to the antiproliferative effects of TGF-${\beta}1$. However, a little is known about the association of TGF-${\beta}1$ with progression of malignant tumor. Methods : In this study, we measured the plasma level of TGF-${\beta}1$ in various cancer patients and evaluated the utility of plasma TGF-${\beta}1$ as a possible tumor marker. Plasma TGF-${\beta}1$ levels were measured using enzyme-linked immunosorbent assay in cancer patients and normal controls. Carcinoembryonic antigen (CEA) and alpha-fetoprotein (AFP) as tumor marker were compared with TGF-${\beta}1$ in the aspects of sensitivity and specificity. Results : The mean of plasma TGF-${\beta}1$ levels was $1.219{\pm}0.834ng/ml$ in normal controls, $5.491{\pm}3.598ng/ml$ in breast cancer, $12.670{\pm}10.386ng/ml$ in lung cancer, $5.747{\pm}3.228ng/ml$ in hepatocellular carcinoma and $10.854{\pm}7.996ng/ml$ in cervical cancer. In comparison with CEA and AFP, TGF-${\beta}1$ is more sensitive. Conclusion : We conclude that the high levels of TGF-${\beta}1$ are common in the plasma of cancer patients. These results suggest that the plasma TGF-${\beta}1$ level can be a potent tumor marker in various cancer patients.

  • Alkalophilic Bacillus sp. HJ-12 producing $\beta $-1, 4-D-arabinogalactanase was isolated from soil in the alkalic condition, pH 10.0. $\beta $-1, 4-D-arabinogalactanase was maximaly produced in the medium consisting of 2% soybean arabinogalactan (SAG), 0.5% yeast extract, 0.5% polypeptone, 0.5% NaCl, 0.1% K$_{2}$HPO$_{4}$, 0.02% MgSO$_{4}$$\cdot $7H$_{2}$O, 0.1% Na$_{2}$CO$_{3}$ under the aerobic condition (pH 8.2). $\beta $-1, 4-D-arabinogalactanase is inducible enzyme so that its activity has been increased 10 fold in the SAG medium than in the glucose medium. Through the ammonium sulfate precipitation, DEAE- Sephadex A-50 ion chromatography, and Sephadex G-75 gel chromatography procedures, this enzyme was purified with a single protein of 11% vield and 110 fold's purity. $\beta $-1, 4-D-arabinogalactanase is endo type enzyme producing ollgosaccharide from SAG.

베타-1,3-글루칸 합성효소 저해제의 스크리닝을 위한 효소원 제조법 (Preparation of Enzyme Source for Screening of Enzyme Inhibitor of $\beta$-1,3-glucan Synthase)

  • Assay conditions for screening of $\beta$-1,3-glucan synthase inhibitor were evaluated. Cells in the beginning of mid-log phase showed the highest activity of the $\beta$-1,3-glucan synthase. Cells permeabilized with 1% digitonin treatment could be used as a good crude enzyme source for convenient screening of the $\beta$-1,3-glucan synthase inhibitors. Calcofluor white (0.125% in final) and papulacandin B (25 $\mu$g/ml) inhibit 90% and more than 50% of the $\beta$-1,3-glucan synthase activity, respectively. Cells grown at 37$\circ$C showed higher enzyme activity than those of 25$\circ$C. Catalytic factor of the $\beta$-1,3-glucan synthase was solubilized from particulated membrane preparations, holoenzyme, by extracting with 0.00938% CHAPS.

  • Transforming growth $factor-{\beta}(TGF-{\beta})$ 신호의 매개자 역할을 하는 Smad 계열 단백질은 발생과정에 중요한 역할을 한다고 알려져 있다. Estrogen receptor(ER)와 구조적으로 유사한 estrogen receptor-related receptor(ERR)은 포유동물에서 후기 배발생기에 외배엽 형성과 관련이 되어 있는 고아핵수용체이다. 본 연구에서는 해양무척추동물의 초기발생과정과 계절번식기 동안에 Smad3와 ERR의 유전자 발현이 발생과정과 성숙에 어떠한 연관성을 갖고 있는지 알아보기 위하여, 동해안 연안에 주로 서식하는 극피동물문 둥근성게과 둥근성게(Strongylocentrotus nudus)를 재료로 하여 계절별 및 배발생 과정중에 Smad3와 $ERR{\beta}$ like 1의 mRNA 농도를 real-time PCR 방법으로 조사하였다. Smad3 mRNA는 샘플링을 시작한 2004년 2월의 생식소와 비교하면 4월부터 그 농도가 증가하기 시작하여 6월까지 증가하였으며, 산란기인 8월에 감소하였다가 10월부터 12월까지 높은 수준을 유지하였다. $ERR{\beta}$ like 1 mRNA는 6월까지 낮은 수준이었으나, 산란기인 8월에 급증한 후 다시 감소하였다. 수정란부터 초기 유생기까지 발생과정을 분석한 결과, Smad3 mRNA는 8세포기 및 16세포기에 높은 발현이 관측되었다. 한편, $ERR{\beta}$ like 1 mRNA는 포배기, 낭배기, 초기 유생기에 현저하게 높은 발현패턴을 보였다. 이상의 결과로부터 둥근성게의 산란기 및 발생배의 발생후기에 $ERR{\beta}$ like 1이 중요한 역할을 담당할 것으로 추정되며, 초기 난할시기에는 Smad3의 관련성이 시사되었다.

Hydrogen Bonds in GlcNAc( β1,3)Gal( β)OMe in DMSO Studied by NMR Spectroscopy and Molecular Dynamics Simulations

  • Hydrogen bond is an important factor in the structures of carbohydrates. Because of great strength, short range, and strong angular dependence, hydrogen bonding is an important factor stabilizing the structure of carbohydrate. In this study, conformational properties and the hydrogen bonds in GlcNAc( ${\beta}$1,3)Gal(${\beta}$)OMe in DMSO are investigated through NMR spectroscopy and molecular dynamics simulation. Lowest energy structure in the adiabatic energy map was utilized as an initial structure for the molecular dynamics simulations in DMSO. NOEs, temperature coefficients, SIMPLE NMR data, and molecular dynamics simulations proved that there is a strong intramolecular hydrogen bond between O7' and HO3' in GlcNAc( ${\beta}$1,3)Gal(${\beta}$)OMe in DMSO. In aqueous solution, water molecule makes intermolecular hydrogen bonds with the disaccharides and there was no intramolecular hydrogen bonds in water. Since DMSO molecule is too big to be inserted deep into GlcNAc(${\beta}$1,3)Gal(${\beta}$)OMe, DMSO can not make strong intermolecular hydrogen bonding with carbohydrate and increases the ability of O7' in GlcNAc(${\beta}$1,3)Gal(${\beta}$)OMe to participate in intramolecular hydrogen bonding. Molecular dynamics simulation in conjunction with NMR experiments proves to be efficient way to investigate the intramolecular hydrogen bonding existed in carbohydrate.

인장시험에 의한 낚시의 변형과 파단하중 (The Deformation and Breaking Load of the Fishing Hook by the Tensile Test)

  • 연근해 어업용 낚시 6가지형에 대하여 만능실험기로 인장속도를 290mm/min와 780mm/min로 하여 실험한 결과는 다음과 같다. 1) 낚시의 직경은 d, 미늘 기부에서 허리굽이까지의 거리를 w라 하면 낚시의 파단하중은 $B={\alpha}\;wd^2+\beta$ (단, B는 kg, w와 d는 mm)로 나타내며, 인장연도가 290mm/min 일 때의 $\alpha,\;\beta$는 둥근형 낚시 $SR_{p}-F_{1}B,\;SR_{p}-F_{1}S$$\alpha_{s}=0.5,\;\beta_{s}=1.6$, 모난형 낚시 $SA_{a}-F_{1}S,\;SA_{a}-F_{1}S_k,\;LA_{a}-F_{1}S_{k}$$\alpha_{s}=1.1,\;\beta_{s}=2.0$긴 낚시 $LA_{p}-F_{1}S$$\alpha_{s}=0.4,\;\beta_{s}=1.5$정도이다. 2) 인장속도가 780mm/min 일 때 윗식에서 $\alpha,\;\beta$는 둥근형 낚시는 $\alpha_{f}=0.4,\;\beta_{f}=1.4$모난형 낚시는 $\alpha_{f}=1.0$ 일본산은 $\beta_{f}=0.9$, 국산은 $\beta_{f}=-2.5$ 긴 낚시는 $\alpha_{=}0.6,\;\beta_{f}=0.4$ 정도이다. 3) 낚시의 파단변형률은 $40\sim60\%$ 정도이며, 인장속도가 빠를 때의 변형률은 느릴 때에의 $95\%$ 정도이나, 빠를 때의 파단하중은 느릴 때의 $80\%$ 정도이다. 4) 낚시가 파단되는 부위는 인장속도에 관계없이 거의 모두 허리굽이 부분이며 꼭지 기부에서 파단점까지의 거리는 낚시 길이와 거의 같다.

  • 점박이송사리, Rivulus marmoratus에서 기원된 16개의 ${\beta}-actin$ 유전자의 염기서열 분석 결과 1,764~1,769 bp 범위를 가지는 ${\beta}-actin$ 유전자를 분리하였다. 이는 기존에 보고된 점박이송사리 ${\beta}-actin$ 유전자와 exon 1 및 intron 2지역에서 다소의 차이를 보여 우리는 이를 점박이송사리 ${\beta}-actin$ 2 유전자라 명명하였다. Multiple alignment를 이용하여 유전자 서로간의 차이를 비교한 결과, 점박이송사리 ${\beta}-actin$ 유전자는 variation을 볼 수 있었다. 따라서 본 연구에서는 점박이송사리에 있어 ${\beta}-actin$ 유전자의 종내 변이 (intraspecific variation)를 확인하였다.

  • [ ($1{\to}3$) ], ($1{\to}4$)-${\beta}$-D-glucans(${\beta}-glucans$) are a major component of the cell walls of grasses as a component of the cereal endosperm and aleurone cell walls. Although ${\beta}-glucans$ exist in all cereals, their concentration is highest in oats and barley. Genetic and environmental differences are found in total ${\beta}-glucan$ content. Both oats and barley ${\beta}-glucans$ have cholesterol-lowering effects. This suggests possible use as food additives. Structural characterization of ${\beta}-glucan$ is important because structure can influence physical and physiological properties. In this review, ${\beta}-glucans$ of barley and oats are discussed in details including structure, chemical and physical properties, and nutritional implications. The use of barley and oat products as well as ${\beta}-glucan$ as a food additive continues to increase. This can provide an additional market for barley and oats, thus increasing the value of the crops.
