Potential and Significance of Ammonium Production from Helicobacter pylori

  • Glutamine and urea, abundant in body fluids or plasma, yield net ammonium ions upon hydrolysis by ${\gamma}-glutamyl$ transpeptidase (${\gamma}-GTP$) and urease, respectively, and these two enzymes are largely produced from Helicobacter pylori. To investigate bacterial potential of ammonium production, we first quantified those in whole-cell systems and found that the relative ratio of their amounts varied greatly, especially with pH values and the cell's aging. During the H. pylori cultivation, the ratio appeared to be inversely proportional to each other, showing a progressive increase of the ${\gamma}-GTP$ with decreasing of the urease. Under the urease-defective conditions due to low pH or coccoids, the bacterial cells still possessed a considerable amount of ${\gamma}-GTP$, which was found exclusively in the external compartment, therefore, the cell's ammonium production was found to be solely dependent upon glutamine, and the external ammonium concentration was constant without any contribution of urea concentration. Such ammonium constancy would definitely have an adverse effect on the host, because of its absolute requirement for vacuolar degeneration by H. pylori VacA, maximized at approximately 10 mM $NH_4Cl$. It was also found that, by using the metal-saturated membrane vesicles, ammonium ions were likely to be involved in the pH-dependent cation-flux across the H. pylori membrane, where the role of ${\gamma}-GTP$ in ammonium homeostasis around cells was suggested, especially under the hostile conditions against H. pylori.

경주마(競走馬)의 혈액화학성상(血液化學性狀)에 관한 연구: I. 혈청효소(血淸酵素) 활성치(活性値) (Studies on the Blood Chemical Properties in Racehorses: I. Serum Enzyme Activities)

  • The present study was carried out to investigate the serum enzyme activities of racehorses. The enzymes investigated were aspartate and alanine aminotransferase (GOT, GPT), ${\gamma}$-glutamyl transpeptidase(${\gamma}$-GTP), Lactic dehydrogenase(LDH), creatine phosphokinase(CPK), and alkaline phosphatase(ALP). Animals used were 30 healthy racehorses(♀17, ♂13) average weighing 435kg and were from 2 to 8 years of age. LDH activity was progressively decreased with age and next in the order of GOT and CPK activities. GOT and ${\gamma}$-GPT activities were not changed with age but ALP activity tended to be decreased wish age. Activities of GOT, GPT, ${\gamma}$-GTP and ALP were higher in female than in male. There was no difference in CPK activity by sex, and no difference was found out among breeds.

Activity of Some Hepatic Enzymes in Schistosomiasis and Concomitant Alteration of Arylsulfatase B

  • The levels of arylsulfatases A and B, $\alpha$-amylase, aspartate transcarbamylase, and $\gamma$-glutamyl transpeptidase were investigated during the infection of mice with schistosoma mansoni. This infection caused a significant (p<0.001) increase in the activity of hepatic arylsulfatase B (ASB), aspartate transcarbamylases and $\gamma$-glutamyl transpeptidase. A non-significant difference occurred for $\alpha$-amylase (p<0.3) and arylsulfatase A (p>0.5) when compared to the control. The specific activity of hepatic ASB was progressively increased with the progression of the Schistosoma-infection. Moreover, the kinetic studies of hepatic ASB in Schistosoma-infection showed that a slight decrease in the value of $K_m$ and about a 40% increase in $V_{max}$ when compared to the control. In addition, the pH optimum of hepatic ASB was altered from 6 to 7 as a result of schistosomiasis. These observations suggest that there are schistosomiasis-associated changes of the catalytic and kinetic properties of hepatic ASB.

Residual Level, Histology, and Blood Biochemistry of Tebuconazole: A Repeated Dose 28-Day Oral Toxicity Study in Pigs

  • In this study, we investigated the residual properties of tebuconazole-treated pigs. Twenty pigs were treated with different concentrations (0.25, 1.25, 2.5, 12.5, and 25 mg/kg bw/d) of tebuconazole for 28 d. Blood biochemistry, histology, and residual levels were analyzed using the VetTest analyzer, Masson's trichrome staining kit, and liquid chromatography-mass spectrometry, respectively. The final body weights were not significantly different between the control and treatment groups. Alkaline phosphatase, blood urea nitrogen, cholesterol, and gamma-glutamyl transpeptidase levels were significantly different from those of the control after exposure for 14 d. However, alanine aminotransferase levels showed changes only after exposure to pesticides for 28 d. The biochemical parameters were separated during the experimental period (14 d versus 28 d) by principal component analysis. Based on variable importance plots, blood urea nitrogen/creatinine ratio, blood urea nitrogen, glucose, and gamma-glutamyl transpeptidase are candidate biomarkers for tebuconazole exposure. The residual levels were observed at T4 (12.5 mg/kg bw/d) and T5 (25 mg/kg bw/d) in the liver and fat tissues, respectively. Fibrosis increased in the liver, kidney, and fat tissues, depending on the tebuconazole concentration. In conclusion, the residue limits of tebuconazole and the physiological changes caused by dietary tebuconazole in pigs provide important information for establishing maximum residue limits of pork and pork products.

Toxic Effects of Polygalae Radix on Rat Kidney

  • The renal toxicity of the extract of Polygalae Radix was investigated in rats. Rats were treated with 3.5 mg/Kg of the extract, i.p., for 7 days. Changes in consumatory behavior, 24 hour-urine and the activities of urinary enzymes were determined during the administration of the extract. Significant decrease in body weight and food consumption and increase in 24 hour-urine volume were observed during the administration. However, the quantity of total creatinine in urine was decreased significantly. Those indicate that subacute treatment with the extract might induce diuresis and the ditiresis might be due to the decrease in water reabsorption. In the activities of urinary enzymes, the activities of alanine aminopejotidase (AAP) and gamma-glutamyl transpeptidase (GGT) were increased 4.3 and 3.5 times and then returned to the control. The activity of N-acetyl-${\beta}$-D-glucosaminidase (NAG) was increased 7.2 times and then decreased slowly. But, it was significantly higher than that of the control evea after the last administration. The activity of factate dehydrogenase (LDH) was increased continuozlsly during the treatment. It showed 32 times higher than the control. These results suggested that the extract of Polygalae Radix had toxic effect on kidney. Furthermore, the result suggested that the subacute administration of the extract induced resistance against the toxicity of Polygalae Radix.

알코올성 간독성에 미치는 누에배설물의 영향 (Effect of Silkworm (Bombyx mori) Excrement Powder on the Alcoholic Hepatotoxicit in Rats)

  • 알코올 급여 흰쥐에서 알코올성 간독성에 대한 누에배설물(silkworm excrement powder)의 영향을 검토하기 위하여 반합성 식이에 누에배설물을 3% (w/w) 수준으로 첨가하여 30일간 급여한 후 혈중 알코올 및 지질 농도, 간 기능 지표 효소 활성 및 간 조직 검사를 실시하였다. 임상생화학적으로 중요한 간 기능 지표 효소인 alanine aminotransferase (ALT), aspartate aminotransferase (AST), $\gamma$-glutamyl transpeptidase ($\gamma$-GTP) 및 lactate dehydrogenase (LDH) 활성이 알코올 대조군에서 증가하였다. 또한 혈중 알코올 농도도 알코올 섭취에 의해 증가하였다. 그러나 누에배설물 투여에 의해 혈중 ALT 및 LDH 활성은 현저하게 감소하였다. 혈중 중성지질, 콜레스테롤 농도는 알코올 대조군에 비해 누에배설물 투여에 의해 현저히 감소하였다. 또한 간 조직 검사에서 알코올 대조군에서 많은 지방적이 나타나 지방간이 확인되었으나 누에배설물 투여에 의해서는 지방적의 크기와 수가 많이 줄어드는 결과를 얻었다. 이상의 결과에서 누에배설물에 의한 알코올-유발 간독성의 개선효과는 간 조직의 임상생화학적 지표 효소의 활성 감소에 기인하는 것으로 나타났다.

열다한소탕(熱多寒少湯)이 간기능에 미치는 영향 (The Effects of Constant Use of Yeoldahanso-tang on Liver Function)

  • 1. Objectives The purpose of this study was to evaluate whether use of Yeoldahanso-tang(熱多寒少湯) may injure the liver function. We clinically studied the change of liver function rest in patients who were admitted in Dongseo Oriental Medical Hospital. 2. Methods We analyzed rhe serum alkaline phosphatase(AIP), aspartate aminotransferase(AST), alanine aminotransferase(ALT), ${\gamma}$-glutamyl transpeptidase(${\gamma}$-GT), total bilirubin of 25 patients from 1st. July. 2004 to 15th. October. 2007 admitted in Dongseo Oriental Medical Hospital. Liver function test were done on admission and before discharge. 3. Results For most patients, the values of AIP, AST, ALT, ${\gamma}$-GT and total bilirubin were within normal range or decreased. 4. Conclusions This study suggests that Yeoldahanso-tang(熱多寒少湯) does not injure liver function of human.

항암제 내성 L1210세포의 Glutathione 대사 관련효소 유전자의 발현 양상 (Gene Expression of Enzymes Related to Glutathione Metabolism in Anticancer Drug-resistant L1210 Sublines)

  • 생쥐의 백혈병세포 L1210과 항암제에 대하여 내성이 유도된 L1210AdR, L1210VcR과 L1210Cis에서 glutathione의 농도와 glutathione의 합성 조절에 관여하는 ${\gamma}$-glutamylcysteine synthetase(GCS)와 ${\gamma}$-glutamyl transpeptidase (GGT), 세포 이물질을 축합하는데 촉매하는 glutathione S-transferase(GST)의 효소 활성도와 유전자의 발현 여부를 관찰하였다. 세포내 glutathione농도(${\mu}M/mg$ protein)는 L1210이 $0.41{\pm}0.003$, L1210AdR가 $0.73{\pm}0.006$, L1210VcR은 $1.16{\pm}0.060$, L1210Cis가 $2.19{\pm}0.282$으로 모세포에 비하여 내성세포에서 통계적으로 유의한 증가를 관찰하였다. Buthionine sulfoxamine(BSO)를 1 ${\mu}M$농도로 첨가하여 12시간 배양한 세포들에서의 glutathione농도는 L1210이 88%, L1210AdR가 85%, L1210VcR이 89%, 그리고 L1210Cis는 79%의 감소를 보였다. GCS의 활성도(nM/mg protein/min)는 L1210이 104인데 비하여 L1210AdR가 128, L1210VcR는 227, 및 L1210Cis는 212로 증가하였다. GGT의 활성도(nM/mg protein/min)는 L1210이 $2.15{\pm}0.531$이었고, L1210AdR은 $2.80{\pm}0.498$, L1210VcR은 $2.42{\pm}0.389$, 그리고 L1210Cis는 $2.98{\pm}0.623$으로 내성인 세포들에서 증가하였으며 L1210AdR과 L1210Cis에서 유의하였다. GST활성도(nM/mg protein/min)는 L1210이 $16.70{\pm}4.798$이었고, L1210AdR은 $14.51{\pm}3.402$, L1210VcR은 $19.52{\pm}4.255$, L1210Cis $17.77{\pm}4.495$로 L1210VcR과 L1210Cis가 약간의 증가를 보였으며, L1210AdR은 오히려 감소를 보였다. DNA의 slot blot에서 GCS, GGT, GST 유전자의 모세포와 내성세포간에 별다른 차이를 보이지 않았다. Northern hybridization에서 GCS는 약 4.5kb 크기의 band, GST-${\pi}$는 약 1.05kb 크기의 band를 보였으며 내성세포 모두에서 발현 증가가 관찰되었다. GGT의 경우 크기가 다른 6개의 band가 보였으며 특히 11.5 kb크기의 band에서 L1210AdR과 L1 210VcR의 발현이 증가하였으며, L1210VcR에서는 L1210과 다른 내성세포에서 보이는 1.95kb크기의 band가 보이지 않고 2.2kb 크기의 다른 band가 관찰되었다. 이상에서 L1210AdR과 L1210VcR의 내성에는 mdr1 유전자가 관여하고, L1210Cis의 내성에는 특히 glutathione이 중요하다. GCS, GGT 및 GST등의 활성도 및 유전자의 발현도 내성세포들에서 증가하였으며 이중 GCS는 내성세포내의 glutathione 합성에 가장 중요한 조절인자라 할 수 있다.

직장인에서 비만과 혈액지표와의 상관성에 관한 연구 (A Study for the Correlations between Obesity and Blood Parameters at the Worksite)

  • Obesity, a state of having excessive body fat enough to attack one's health, is most blamed for causing chronical degenerative illness such as hypertension, diabetes mellitus as well as arteriosclerosis. According to many studies, the program for the weight control was the most demanded at the worksites. This study was designed to investigate the correlation between the degree of obesity and blood parameters of the employees at the worksites. The results of periodic medical examination for 571 employees(male: 418, female: 153) were analyzed; Age, BMI, systolic blood pressure(SBP), diastolic blood pressure(DBP), hemoglobin(Hb), fasting blood sugar(FBS), total-cholesterol(TC), serum glutamic oxaloacetic transaminase (sGOT), serum glutamic pyruvic transaminase(sGPT) and $gamma$-glutamyl transpeptidase($gamma$-GTP). Pearson correlation coefficient was used to find out the correlation between BMI and each blood parameters. In the case of male, 27.3% of the subjects were over-weight. The subject's age and their blood levels of uric acid, FBS, sGOT and sGPT did not show any significant difference regardless of BMI's, while SBP, DBP, Hb, TC and $gamma$-GTP levels of the overweight group were significantly higher than those of normal group. In the case of female, 9.2% of the subjects were over-weight. This study shows that the implementation of weight control program is required at the worksites

Effect of Zinc-enriched Yeast FF-10 Strain on the Alcoholic Hepatotoxicity in Alcohol Feeding Rats

  • The possible protective effects of highly zinc-containing yeast Saccharomyces cerevisiae, FF-10 strain, isolated from tropical fruit rambutan on acute alcoholic liver injury in rats were evaluated. Zinc concentration in this strain was 30.6mg%. The activities of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), and $\gamma$-glutamyl transpeptidase ($\gamma$-GTP) were highly increased when alcohol was treated, relative to the normal rats. Also, a highly significant increase in the blood alcohol and acetaldehyde levels by alcohol treatment was observed. Administration of FF-10 strain markedly prevented alcohol-induced elevation of the activities of serum ALT, AST, and $\gamma$-GTP, and the levels of blood alcohol and acetaldehyde, and these reduced levels reached to that of normal rats. As compared with alcohol treated control rats, the FF-10 strain supplementation showed highly decreased the triglyceride concentration in serum. Alcohol treatment induced the marked accumulation of small lipid droplets, hepatocytes necrosis, and inflammation, but FF-10 strain administration attenuated to alcohol-induced accumulation of small lipid droplets and hepatocyte necrosis in the liver. Therefore, the current finding suggests that zinc-enriched yeast FF-10 strain isolated from tropical fruit rambutan may have protective effect against alcohol-induced hepatotoxicity.
