Evaluation of Butyltin Compounds and its Distribution Among Seawater, Sediment and Biota from the Kwangyang Bay (광양만내 유기주석화합물의 해수, 퇴적물, 생물중 농도 및 분포 상관관계)

  • Seawater, sediment and biota in the Kwangyang Bay were analyzed by gas chromatography/quartz furnace atomic absorption spectroscopy (GC-QFAAS) to investigate concentrations and distribution pattern of butyltin compounds (TBT, DBT, MBT) during February, April and July, 1996, Marine biota analyzed were Tapes japcnicus and Crassostrea gigas. The concentrations of tributyltin (TBT) in seawater were in the range of ND-15.7 ng/L for the surface and ND-68.5 ng/L for the bottom. The highest concentration of TBT in seawater was detected in April for the both, surface and bottom water. The maximum value of $TBT_{(bottom)}/TBT_{(surface)}$, 3.6 in April showed the increased input of TBT from the surface water in April compared to February (2.1) and July (0.9). The concentrations of TBT in the sediment were in the range of ND-8.5 ng/g dry wt. The highest concentration of TBT in the sediment was measured in July, This result seems to attributed to the removal of TBT from water column via sorption onto particulate matters to the relatively undisturbed underlying sediment and increased input of TBT by increased fluxes of detritus of marine plankton after spring bloom, in July. The mean values of partitioning coefficient ($K_d$) of TBT between seawater and sediment were $3.0\times10^3$(February), $7.4\times10^3$(April) and $9.4\times10^3$(July). The concentrations of TBT in biosamples were in the range of ND-93.30 ng/g dru wt. (T. japonicus) and ND-138.53 ng/g dry wt. (C. gigas). The seasonal variation of TBT contents in biota was remarkable. The $K_d$ (biological concentration factor) was $7-41\times10^3$ for T. japonicus. and $5-34\times10^3$ for C. gigas. The measured TBT concentrations in seawater in the study area was sufficient to cause the imposex of shellfish and to retard the growth of aquatic organisms including oyster upon chronic exposure.

AN EXPERIMENTAL STUDY ON THE TENSILE STRENGTH OF POSTERIOR RESIN-BASED COMPOSITES (구치부 복합레진의 인장강도에 관한 실험적 연구)

  • The purpose of this study was to evaluate the tensile strength of light-cured restorative posterior resin-based composites. Five commercially available light-cured composites(Denfil : DF, P60 : PS, Unifil S : US, Z100 : ZH, Z250 : ZT) were used. Rectangular tension test specimens were fabricated in a teflon mold giving 5mm in gauge length and 2mm in thickness. Specimens were subjected to the 5,000 thermal cycles between $5^{\circ}C$ and $55^{\circ}C$ and the immersion time in each bath was 15 second per cycle. Tensile testing was carried out with Instron at a crosshead speed of 0.5mm/min and fractured surface were observed with scanning electron microscope. The obtained results were summarized as follows; 1. The tensile strength of PS was highest. PS was significantly higher than DF, US and ZH(p<0.05) but in the case of ZT was similar to PS(p>0.05). 2. The tensile strength DF was lowest. DF was significantly lower than PS, US, ZH and ZT(p<0.05). 3. The tensile strength of US and ZH were significantly lower than PS and ZT(p<0.05). but were significantly higher than DF(p<0.05). The tensile strength of US and ZH were similar(p>0.05).

Evaluation of Odors and Odorous Compounds from Liquid Animal Manure Treated with Different Methods and Their Application to Soils (액상 가축분뇨의 처리 및 토양환원에 따른 악취 및 악취물질의 평가)

  • To comply with stricter regulations provoked by increasing odor nuisance, it is imperative to practice effective odor control for sustainable livestock production. This study was conducted to assess odor and odorous compounds emitted from liquid animal manure with different treatment methods such as Fresh Manure(without treatment, FM), Anaerobic Digestion(AD) and Thermophilic Aerobic Digestion(TAD) and their application to soil. Air samples were collected at the headspace of liquid manure, upland and paddy soil, and analyzed for odor intensity and offensiveness using an olfactometry; odor concentration index using odor analyser; nitrogen-containing compound such as ammonia(NH3) using fluorescence method; and sulfur containing compounds such as hydrogen sulfide(H2S), methyl mercaptan(MeSH), dimethyl sulfide(DMS) and dimethyl disulfide(DMDS) using gas chromatography-pulsed flame photometric detector, respectively. Odor intensity, offensiveness and concentration index from TAD liquid manure was statistically lower than those from FM and AD(p<0.01). Mean concentrations of H2S, MeSH, DMS, DMDS and NH3 were 65.93ppb, 18.55ppb, 5.26ppb, 0.33ppb and 10.57ppm for liquid manure with AD; and 5.15ppb, 0.97ppb, 0.80ppb, 0.56ppb and 1.34ppm for liquid manure with TAD, respectively. More than 60% of malodorous compounds related to nitrogen and sulfur were removed by heterotrophic microorganisms during TAD treatment. When liquid manure was applied onto upland and paddy soil, NH3 removal efficiencies ranged from 51 to 94% and 22 to 91% for AD and TAD liquid manure, respectively. The above results show that liquid manure with TAD is superior to AD and FM with respect to the odor reduction and odor problem caused by land applied liquid manure is directly related to the degree of odor generated by the manure treatment method.

Evaluation of shear-bond strength between different self-adhesive resin cements with phosphate monomer and zirconia ceramic before and after thermocycling (인산염계 기능성 단량체가 첨가된 수종의 자가 접착 레진시멘트와 지르코니아 세라믹 사이 열순환 전후 전단결합강도 비교)

  • Purpose: This study compared shear bond strengths of five self-adhesive cements with phosphate monomer to zirconium oxide ceramic with and without airborn particle abrasion. Materials and methods: One hundred zirconia samples were air-abraded ($50{\mu}mAl_2O_3$). One hundred composite resin cylinders were fabricated. Composite cylinders were bonded to the zirconia samples with either Permacem 2.0 (P), $Clearfil^{TM}$ SA Luting (C), $Multilink^{(R)}$ Speed (M), $RelyX^{TM}$ U200 Automix (R), G-Cem $LinkAce^{TM}$ (G). All bonded specimens were stored in distilled water ($37^{\circ}C$) for 24 h and half of them were additionally aged by thermocycling ($5^{\circ}C$, $55^{\circ}C$, 5,000 times). The bonded specimens were loaded in shear force until fracture (1 mm/min) by using Universal Testing Machine (Model 4201, Instron Co, Canton, MA, USA). The failure sites were inspected under field-emission scanning electron microscopy. The data was analyzed with ANOVA, Tukey HSD post-hoc test and paired samples t-test ($\alpha$=.05). Results: Before and after thermocycling, $Multilink^{(R)}$ Speed (M) revealed higher shear-bond strength than the other cements. G-Cem $LinkAce^{TM}$ (G) showed significantly lower bond strengths after thermocycling than before treatment (P<.05), but the other groups were not significantly different (P>.05). Conclusion: Most self-adhesive cements with phosphate monomer showed high shear bond strength with zirconia ceramic and weren't influenced by thermocycling, so they seem to valuable to zirconia ceramic bonding.

The Study on the Methylmercury Analysis and the Monitoring of Total Mercury and Methylmercury in Fish (어류 중 메틸수은 분석법 확립 및 모니터링)

  • Procedure for analysis of methylmercury in fish was developed, involving addition of HCl, extraction with toluene, and clean-up using L-cystein solution. Obtained extract is analyzed by gas chromatography with electron capture detector using Ulbon HR-Thermon-Hg column. Detection limit and recovery of the method were 0.005mg/kg (expressed as Hg), 98-107 (103%), respectively. Total mercury and methylmercury concentrations in 175 commercial fish samples ranged from [mean-max (mean), unit: mg/kg]: 0.014-1.200 (0.270) and 0.006-0.901 (0.168) in tuna-fish, 0.020-0.934 (0.323) and 0.012-0.553 (0.149) in martin-fish, 0.082-0.782 (0.391) and 0.040-0.436(0.201) in shark, 0,023-0.031 (0.026) and 0,013-0.018 (0.015) in salmon, 0.098-0.193 (0.133) and 0.031-0.015(0.090) in tilefish, and 0,031-0.214 (0.089) and 0.016-0.093 (0.042) in canned tuna respectively. No sample of analyzed fish exceeded 1.0mg/kg wet wt., limit for methylmercury established by Codex. In all species examined, estimated weekly intake was lower than Provisional Tolerable Weekly Intake recommended by the JECFA (the Joint FAO/WHO Expert Committee on Food Additives).

Antioxidant Activity and Functional Component Analysis of Korean Mountain Ginseng's Different Sections (장뇌삼 부위별 기능성 성분분석과 항산화활성)

  • This study was conducted to investigate antioxidant activity and functional component analysis in different sections of Korean mountain ginseng (seed, leaf, stem and root). The major free amino acids were arginine, proline, $\gamma-amino-n-butyric$ acid, alanine and aspartic acid, and proline showed the highest content 22.98 mg/g in 80% MeOH extract of seed. Contents of $\gamma-amino-n-butyric$ acid and alanine were the highest in 80% MeOH extract of leaf, with 26.04 mg/g and 13.07 mg/g. Aspartic acid showed the highest content 23.42 mg/g in 80% MeOH extract of leaf. The major fatty acids were identified as palmitic acid, oleic acid and linoleic acid by GC. Content of total phenolic compounds were 737 mg% in 80% EtOH extract of seed, and 560 mg% in 80% MeOH extract of seed. The highest contents of ferulic acid and salicylic acid were $875{\mu}g/g\;and\;78{\mu}g/g$ in 80% MeOH extract of leaf, and $\rho-coumaric$ acid was $181{\mu}g/g$ in 80% MeOH extract of stem, respectively. The highest contents of ginsenoside-Rg1, Re, Rd and Rc were $14.5{\mu}g/g,\;48.78{\mu}g/g,\;27.57{\mu}g/g\;and\;4.87{\mu}g/g$ in 80% MeOH extract of leaf, respectively. Antioxidant activities by DPPH were 83.82% in water extract of leaf, 89.74% in 80% EtOH extract of leaf and 88.37% in 80% MeOH extract of leaf, and 92.81% in BHA (200 ppm). These results suggest that Korean mountain ginseng is very important as functional food material.

EFFECT OF ACID-TREATMENT ON DENTIN BONDING (산 처리가 상아질 접착에 미치는 영향)

  • The purpose of this study was to evaluate the effect of various acid treatments on dentin bonding. Freshly extracted human teeth were uprightly embedded in self curing acrylic resin, and their occlusal surfaces were grinded to expose flat dentin surfaces. The specimens were divided into 4 groups. Specimens of one group were not treated so as to be a control and those of the other three groups were threated with 10% polyacrylic acid, 10% phosphoric acid, and 10-3 solution(10% citric acid/3% ferric chloride) respectively. Primer, bonding resin and composite resin were applied over the treated dentin surfaces sequentially. All specimens were stored in $37^{\circ}C$ distilled water for 24 hours, then the tensile bond strength was measured and the treated dentin surfaces and fracured dentin surfaces were examined under a scanning electron microscope. The results were as follows: Bond strengths of acid-treated groups were higher than those of the untreated group. In the acid-treated groups, bond strength was found to be the highest in the 10-3 solution group followed by the 10% phosphoric acid group and the 10% polyacrylic acid group(P<0.01). On SEM examination of dentin surfaces, the untreated dentin surface showed a remaining smear layer and closed dentinal tubules. Dentin surfaces treated with 10 % polyacrylic acid showed a clean dentin surface without the smear layer, but showed remaining smear plugs in dentinal tubules. A dentin surface treated with 10% phosphoric acid or 10-3 solution showed open dentinal tubules without the smear layer or smear plugs. On SEM observation of the fractured dentin-resin interface, the untreated group showed that failure occurred in the smear layer. The group treated with 10% polyacrylic acid showed no resin tag remained in the dentinal tubules, but resin tags in the dentinal tubules were observed in the group treated with the 10% phosphoric acid or the 10-3 solution. On the failure mode examination, the higher the bond strength of the group, the higher the frequency of cohesive failure. The coefficient between bond strength and cohesive failure rate was 0.71.

Distribution and Behavior of Soil CO2 in Pohang area: Baseline Survey and Preliminary Interpretation in a Candidate Geological CO2 Storage Site (포항 지역 토양 CO2의 분포 및 거동 특성 연구: CO2 지중저장 부지 자연 배경 조사 및 예비 해석)

  • Distribution and behavior of baseline soil CO2 were investigated in a candidate geologic CO2 storage site in Pohang, with measuring CO2 concentrations and carbon isotopes in the vadose zone as well as CO2 fluxes and concentrations through ground surface. This investigation aimed to assess the baseline CO2 levels and to build the CO2 monitoring system before injecting CO2. The gas in the vadose zone was collected using a peristaltic pump from the depth of 60 cm below ground surface, and stored at gas bags. Then the gas components (CO2, O2, N2, CH4) and δ13CCO2 were analyzed using GC and CRDS (cavity ringdown spectroscopy) respectively in laboratory. CO2 fluxes and CO2 concentrations through ground surface were measured using Li-COR in field. In result, the median of the CO2 concentrations in the vadose zone was about 3,000 ppm, and the δ13CCO2 were in the wide range between −36.9‰ and −10.6‰. The results imply that the fate of CO2 in the vadose zone was affected by soil property and vegetations. CO2 in sandy or loamy soils originated from the respiration of microorganisms and the decomposition of C3 plants. In gravel areas, the CO2 concentrations decreased while the δ13CCO2 increased because of the mixing with the atmospheric gas. In addition, the relation between O2 and CO2, N2, and the relation between N2/O2 and CO2 implied that the gases in the vadose zone dissolved in the infiltrating precipitation or the soil moisture. The median CO2 flux through ground surface was 2.9 g/m2/d which is lower than the reported soil CO2 fluxes in areas with temperate climates. CO2 fluxes measured in sandy and loamy soil areas were higher (median 5.2 g/m2/d) than those in gravel areas (2.6 g/m2/d). The relationships between CO2 fluxes and concentrations suggested that the transport of CO2 from the vadose zone to ground surface was dominated by diffusion in the study area. In gravel areas, the mixing with atmospheric gases was significant. Based on this study result, a soil monitoring procedure has been established for a candidate geologic CO2 storage site. Also, this study result provides ideas for innovating soil monitoring technologies.

Comparative Study on the Composition of Floral Volatile Components in the Flowering Stages of Robinia pseudoacacia L. (아까시나무(Robinia pseudoacacia L.) 꽃의 개화 단계별 향기성분 조성 비교)

  • Floral scent emitted from many plants is the critical factors for pollinator attraction and defense for adaptation in environments. The fragrance components of flowers are different in composition by geographical origins, climate factors and the development stages of flowers. In the present study, we investigated the volatile-floral compounds in flowers of Robinia pseudoacacia L. and defined the chemical contribution for flowering periods. The volatile compounds analysis was performed by gas chromatography with mass selective detector after solid phase microextraction (SPME). We reported different compositional features of fragrance compounds according to flowering periods. The abundant compounds identified in stage 1 were ${\alpha}$-pinene (66.80%) and ${\beta}$-pinene (26.53%). Those of the stage 2 were (Z)-${\beta}$-ocimene (37.57%), ${\alpha}$-pinene (15.16%), benzaldehyde (16.63%), linalool (12.13%). The volatiles of stage 3 comprised an abundance of (Z)-${\beta}$-ocimene (64.94%), ${\alpha}$-pinene (9.84%), linalool (8.92%), benzaldehyde (1.71%). Leaf volatiles were distinct from those in the reproductive plant parts by their high relative amount of (E)-${\beta}$-ocimene (23.50%) and (Z)-3-Hexenyl acetate (27.87%). Differences in flower scents of the different stages and leaves are discussed in light of biochemical constraints on volatile chemical synthesis and of the role of flower scent in evolutionary ecology of R. pseudoacacia.

Monitoring of Pesticide Residues Concerned in Stream Water (전국 하천수 중 잔류우려 농약 실태조사)

  • BACKGROUND: This study was carried out to investigate pesticide residues from fifty streams in Korea. Water samples were collected at two times. Thee first sampling was performed from april to may, which was the season for start of pesticide application and the second sampling event was from august to september, which was a period for spraying pesticides multiple times. METHODS AND RESULTS: The 136 pesticide residues were analyzed by LC-MS/MS and GC/ECD. As a result, eleven of the pesticide residues were detected at the first sampling. Twenty eight of the pesticide residues were detected at the second sampling. Seven pesticides were frequently detected from more than 10 water samples. Ecological risk assessment (ERA) was carried out by using residual and toxicological data. Four scenarios were applied for the ERA. Scenario 1 and 2 were performed using LC50 values and mean and maximum concentrations. Scenarios 3 and 4 were conducted by NOEC values and mean and maximum concentrations. CONCLUSION: Frequently detected pesticide residues tended to coincide with the period of preventing pathogen and pest at paddy rice. As a result of ERA, five pesticides (butachlor, carbendazim, carbofuran, chlorantranilprole, and oxadiazon) were assessed to be risks at scenario 4. However, only oxadiazon was assessed to be a risk at scenario 3 for the first sampling. Oxadiazon was not assessed to be a risk at the second sampling. It seems to be temporary phenomenon at the first sampling, because usage of herbicides such as oxadiazon increased from April to march for preventing weeds at paddy fields. However, this study suggested that five pesticides which were assessed to be risks need to be monitored continuously for the residues.
