장용성 연질 캡슐 오메가3 지방산의 흡수율 평가 (Absorption Evaluation of Enteric Coated Capsules Containing Omega 3 Fatty Acids)

  • 본 연구에서는 혈액 내에서의 오메가-3 지방산의 함량 변화를 측정하여 연질 캡슐과 장용성 캡슐의 차이를 검토하였다. Fish oil 중의 EPA와 DHA의 총합의 함량이 62.87 g/100 g으로 대부분을 차지하고 있다. 연질 캡슐의 경우, 제 1시험액(위액)에서는 내부의 지방이 70% 용출된 반면, 장용성의 경우 10% 정도만이 용출되었다. 제 2액(소장액)에서도 6시간까지 50% 정도 용출되어 90분에 50%가 용출된 일반 연질 캡슐과 대조적이었다. 기존에 사용하였던 연질 캡슐의 EPA 및 DHA는 흡수 초기에 장용성 캡슐에 비하여 다소 높은 비율을 보인 반면, 8시간 이후 장용성 캡슐 섭취한 경우 EPA와 DHA의 혈액내 함량이 비율이 증가하는 경향을 보였다. 경구 투여 48시간 후 지방산 조성은 장용성 캡슐에서는 mono-, poly-unsaturated fatty acid 혈액 내의 함량이 다소 높은 경향을 보였으며, saturated fatty acid 함량 역시 다소 높은 경향을 보였다. EPA와 DHA의 함량은 혈액 내에서 각 지방 분획에서의 함량 차이를 정확하게 측정하기 힘들었다. 이상의 결과에 의하면 장용성 캡슐은 위장관을 통과하여 장에서 용출되는 특성 때문에 지속적인 효과를 기대할 수 있을 것으로 추정된다.

  • Omega-3 polyunsaturated fatty acids (${\omega}$-3 PUFAs) are essential nutrients for human beings and their potential roles against cancer development and progression have become of wide concern recently. Some studies have suggested that perioperative supplementation with omega-3 fatty acids may have beneficial effects in gastrointestinal cancer patients undergoing surgery, while other researchers reported contrary results. This paper reviews recent research to establish therapeutic effects as well as possible underlying mechanisms of ${\omega}$-3 PUFA actions, and to help explain possible reasons for inconsistent results from different institutions.

  • Essentiality was proposed in the field of lipid by Burr and Burr in 1929. When rats were raised on the fat-free diet, their growth retarded and their skin and tails showed the characteristic deficient symptoms, which were relieved by the addition of ${\omega}6(n-6)$ polyunsaturated fatty acids as linoleic(LA) and arachidonic(AA) acids to the basal diet. LA is dehydrogenated to ${\gamma}-linolenic$ acid(GLNA) by ${\Delta}6$ desaturase, then GLNA is 2 carbon chain elongated by elongase to $dihomo-{\gamma}-linolenic$ acid(DGLNA), which is desaturated by ${\Delta}5$ desaturase to AA. These acids are called LA family or ${\omega}6(n-6)$ polyunsaturated fatty acids(PUFA). ${\alpha}-Linolenic$ acid(ALNA) is converted through the series of desaturation and elongation steps to docosahexaenic acid(DHA) via eicosapentaenoic acid(EPA). These acids belong to ALNA family or ${\omega}3(n-3)$PUFA. Human who consume large amounts of EPA and DHA, which are present in fatty fish and fish oils, have increased levels of these two fatty acids in their plasma and tissue lipids at the expense of LA and AA. Alternately, vegetarians, whose intake of LA in high, have more elevated levels of LA and AA and lower levels of EPA and DHA in plasma lipids and in cell membranes than omnivores. AA and EPA are metabolized to substances called eicosanoids. Those derived form AA are known as prostanocids(prostaglandins and prostacyclins) of the 2-types and leukotrienes of the 4-series, whereas those derived from EPA are known as prostanoids of the 3-types and leukotrienes of the 5-series. DGLNA is a precursor of the 1-types of prostaglandins. The metabolites of AA and EPA have competitive functions. Ingestion of EPA from fish or fish oil replaces AA from membrane phospholipids in practically all cells. So this leads to a more physiological state characterized by the production of proatanoids and leukotrienes that have antithrombic, antichemotactic, antivasoconstrictive and antiinflammatory properties. It is evident that ${\omega}3$ fatty acids can affect a number of chronic diseases through eicosanoids alone.

Chemometric Approach to Fatty Acid Profiles in Soybean Cultivars by Principal Component Analysis (PCA)

  • The purpose of this study was to investigate the fatty acid profiles in 18 soybean cultivars grown in Korea. A total of eleven fatty acids were identified in the sample set, which was comprised of myristic (C14:0), palmitic (C16:0), palmitoleic (C16:1, ${\omega}7$), stearic (C18:0), oleic (C18:1, ${\omega}9$), linoleic (C18:2, ${\omega}6$), linolenic (C18:3, ${\omega}3$), arachidic (C20:0), gondoic (C20:1, ${\omega}9$), behenic (C22:0), and lignoceric (C24:0) acids by gas-liquid chromatography with flame ionization detector (GC-FID). Based on their color, yellow-, black-, brown-, and green-colored cultivars were denoted. Correlation coefficients (r) between the nine major fatty acids identified (two trace fatty acids, myristic and palmitoleic, were not included in the study) were generated and revealed an inverse association between oleic and linoleic acids (r=-0.94, p<0.05), while stearic acid was positively correlated to arachidic acid (r=0.72, p<0.05). Principal component analysis (PCA) of the fatty acid data yielded four significant principal components (PCs; i.e., eigenvalues>1), which together account for 81.49% of the total variance in the data set; with PC1 contributing 28.16% of the total. Eigen analysis of the correlation matrix loadings of the four significant PCs revealed that PC1 was mainly contributed to by oleic, linoleic, and gondoic acids, PC2 by stearic, linolenic and arachidic acids, PC3 by behenic and lignoceric acids, and PC4 by palmitic acid. The score plots generated between PC1-PC2 and PC3-PC4 segregated soybean cultivars based on fatty acid composition.

Effects of Omega-3 Fatty Acid Supplementation on Skeletal Muscle Mass and Strength in Adults: A Systematic Review

  • Previous studies have suggested that omega-3 polyunsaturated fatty acids, predominantly eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), have several health benefits. However, their effect on changes in skeletal muscle mass and strength has not been established, owing to differences in study designs. This systematic review aimed to investigate the recent evidence regarding the role of dietary EPA and DHA in muscle mass changes and their association with muscle strength. Databases including PubMed and Google Scholar were searched for randomized controlled trials and single-arm interventions that investigated the effects of omega-3 fatty acids on skeletal muscle mass, strength, and body composition in adults aged 18 years and older. A total of 18,521 studies were retrieved from the databases and manual searches; 21 studies were quality assessed, and the findings were summarized. Studies were categorized into 3 main categories according to the type of omega-3 fatty acid supplementation: pure compounds such as oil tablets, formulated forms with protein, leucine, and vitamin D, and ingredients added to enteral nutrition support products. Overall, the majority of the study results appeared to indicate that omega-3 fatty acids are beneficial for muscle health. However, meta-analysis was not conducted because of the heterogeneity of the study participants, evaluation method of muscle indices, and intervention periods among the studies. High-quality studies are required to validate our conclusions. However, this systematic review of the effects of EPA and DHA on skeletal muscle and body composition provides evidence that can be applied in both clinical and industrial settings.

Protective Effect of Omega-3 of Polyunsaturated Fatty Acid Docosahexaenoic Acid on the Organic Mercury-Induced Cytotoxicity in Cultured NIH3T3 Fibroblasts

  • To clerify the protective effect of omega-3 of polyunsaturated fatty acid docosahexaenoic acid (DHA) on the cytotoxicity induced by organic mercury in cultured NIH3T3 fibroblasts. The measurement of cell viability on ogranic mercury wad done by XTT assay after NIH3T3 fibroblasts were cultured with various concentrations of methyl mercuric chloride (MMC). And also, the effect of DHA on the MMC-mediated cytotoxicity was examined by cell viability, and antioxidant effect of DHA was also assessed by superoxide dismutase (SOD)-like activity and the lipid peroxidation activity in cultured NIH3T3 fibroblasts. In this study, MMC decreased cell viability and $XTT_{50}$ value was determined at $50{\mu}M$ of MMC in these culture. In the effect of DHA against the cytotoxicity induced by MMC, DHA significantly increased the cell viability damaged by MMC in cultured NIH3T3 fibroblasts. And also, DHA showed the antioxidant effect by showing the increase of SOD-like activity and the decrease of lipid peroxidation activity. From these results, it is suggested that organic mercury such as MMC has highly toxic effect on cultured NIH3T3 fibroblasts, and also, omega-3 of polyunsaturated fatty acid, DHA showed the protection on MMC-induced cytotoxicity and antioxidant effect.

입본특이적(立本特異的) 방법(方法)과 $^{13}C-NMR$ 기법(技法)에 의한 잣기름의 트리아실 글리세롤의 구성지방산(構成脂肪酸)의 분포(分布)에 관한 연구(硏究) (Studies on the Fatty Acid Distribution in the Position of Triacylglycerols from the Seed of Pinus Koraiensis by Stereo-specific Analysis and $^{13}C-NMR$ Techniques)

  • All the triacylglycerols including the molecular species having ${\Delta}^5$-unsaturated fatty acids from the seeds of Pinus Koraiensis, were split into a mixture of diacylglycerols by a Grignard reagent prepared with allyl bromide without arousing acyl chains of a glycerol moiety to migration, and were also easily partially hydrolyzed to diacylglycerols by pancreatic lipase. (S)-(+)-(1-naphthyl)ethyl urethane(NEU) derivatives of the diacylglycerol mixture derived from the triacylglycerols were fractionated into sn-1, 3-, sn-1, 2- and sn-2, 3-DG-NEU by silica-HPLC and the fatty acid composition of these fractions was analysed. $C_{18:1{\omega}9}$ is distributed evenly in the three positions of TG with $C_{18:2{\omega}6}$ mainly located in sn-2 position, while ${\Delta}^5$-unsaturated fatty acids such as ${\Delta}^{5.9}-C_{18:2}$, ${\Delta}^{5.9.12}-C_{18:3}$ and ${\Delta}^{5.11.14}-C_{20:3}$ are exclusively present in the sn-3 position. These results could be confirmed by $^{13}C$-NMR spectroscopy : the signals at $^{\delta}$173.231 ppm and $^{\delta}$172.811 ppm of the carbonyl carbon of acyl moieties indicate the presence of saturated acids and/or $C_{18:1{\omega}9}$ (oleic acid) in the ${\alpha}({\alpha}')$- or ${\beta}$- positions, and $C_{18:2{\omega}6}$ including $C_{18:1{\omega}9}$ in the ${\beta}$-position, respectively. In addition, the resonance at $^{\delta}$173.044 ppm suggested a location of ${\Delta}^5$-unsaturated fatty acid moiety in the ${\alpha}({\alpha}')$-position.

화학적 발암과정에서 식이의 ${\omega}6/{$\omega}3$비율이 쥐간의 전암성병변 및 지질과산화물 형성에 미치는 영향 (Dietary ${\omega}6/{$\omega}3$ ratios on the preneoplastic lesions and lipid peroxidation in diethylnitrosamine initiated rat hepatocarcinogenesis)

  • To study the effect of dietary $\omega 6/\omega 3$ fatty acid ratios on the preneoplastic lesions and lipid peroxidation in rat hepatocellular chemical carcinogenesis, placental glutathione S-transferase(GST-P) positive foci area and numbers, glucose 6-phosphatase(G6Pase) activity, thiobarbituric acid reactive substances (TBARS) were measured. Male Sprague-Dawley rats were fed 5 different diets-low $\omega 6/\omega 3$ ratio with fish oil (Low-F), low $\omega 6/\omega 3$ ratio with perilia oil(Low-P), moderate ratio with perilia oil(Moderate), blend of 10 different commercial fats and oils(High-BL) and high $\omega 6/\omega 3$ ratio(High)-for 8 weeks. Hepatocarcinogenesis was induced by modified Ito model. The area of GST-P positive loci was the lowest in Moderate group and in ascending order of Low-F < Low-P < High-BL < High. But statistically, only Moderate and High groups were significantly different. The number of GST-P positive foci showed the same trend as foci area. The activities of G6Pase, membrane stability marker, were increased as $\omega 6/\omega 3$ ratio decreased. Lipid peroxidation values (TBARS) were the lowest in Low-F group and it is significantly different from Moderate, High-BL and High groups. When dietary $\omega 6/\omega 3$ ratio was moderate(4.06), hepatocarcinogenesis was suppressed compared with high or low $\omega 6/\omega 3$ ratios. Blend fat, commonly consumed among Koreans, did not show any suppressive effect on carcinogenesis because of high ratio(6.7). These results suggest that dietary $\omega 6/\omega 3$ ratio influences hepatocellular chemical carcinogenesis. It is recommended that appropriate $\omega 6/\omega 3$ ratio should be around 4.0. and we recommend to use more $\omega 3$ fatty acid in food preparation to reduce the risk of hepatocarcinogenesis.

우리나라 일부 초등학교 아동의 육류 및 생선 섭취수준에 따른 지방산 섭취양상과 혈청 지방산조성의 비교연구 (Dietary Fatty Acid Pattern and Serum Fatty Acid Composition of Korean Elementary School Children)

  • The purpose of this study was to investigate the pattern of individual fatty acid intake and to compare total serum lipid levels and total serum fatty acid composition of elementary school children in Seoul by fish and meat intakes. The subjects were divided into High-Fish / Low-Meat(HFLM, n=49) and Low-Fish/High-Meat(LFHM, n=47) group. The total energy from fat of HFLM and LFHM group was 22.0 and 30.0% respectively. The average intake of cholesterol was 237-287mg in HFLM group and 358-387mg in LFHM group. The average P/M/S ratio for all subjects was 1.1-1.4/1.1 - 1.2/1.0, which was very similar to desired ratio. The $\omega$6/$\omega$3 ratio of dietary fatty acids of HFLM and LFHM group were 8.0-10.9 and 18.9-29.3 respectively, which in case of LFHM group, was fat beyond the suggested range, 4/1 - 10.1. Serum lipid levels of all subjects fell within the normal range and there was no significant difference in serum lipid levels between HFLM and LFHM groups, except for HDL-cholesterol in upper grades(4th-6th grades). In lower grades(1st-3rd grades), serum PUFA composition of HFLM (40.8%) group was significantly higher than that of LFHM (38.3%) group (p<0.05) and SFA composition of HFLM(33.8%) was significantly lower than that of LFHM group, (p<0.001) between them. The $\omega$6/$\omega$3 ratio of serum fatty acid tended to be higher in LFHM than in HFLM group but there was no significant difference between two groups. This study suggests that the dietary fat pattern of growing children could affect the change in total serum fatty acid composition though it did not influence serum lipid levels significantly. Energy and EPA intakes were negatively correlated with HDL-cholesterol levels(p<0.05). Serum fatty acid compositions were more closely correlated with serum triglyceride(TG) levels than serum cholesterol levels. Especially, total SFA (p<0.001) and individual SFA, such as C14:0(p<0.001) and C16:0(p<0.001)compositions were positively correlated with serum TG levels, but stearic acid(p<0.01) was negatively correlated with it.

미역과 파래의 지방산 조성 (Fatty acid Composition of Miyeok(undaria pinnatifida) and Pare (Enteromorpha compressa))

  • The contents of total lipids from Korean marine benthic algae Miyeok(Undaria pinnatifida) and Pare(Enteromorpha compressa)were 1.8% and 0.7% on the dry basis, respectively. They were fractionated by silicic acid column chromatography and identified by gae liquid chromatography. The ratios of neutral lipids, glycolipids and phospholipids in total lipids were 3.5 : 74.1 : 22.4 in Miyeok and 33.1 : 48.4 : 18.5 in Pare. Total fatty acids in Miyeok and Pare were composed of 28.5 and 33.2% of saturates, 9.8 and 10.8% of monoenes, 61.7 and 56.0% of polyenes, respectively and the polyunsaturated acid was the most predominent component. $\omega$-3 polyunsaturated fatty acids were C18:4(15.9 and 17.1%) and C20:5(10.6 and 6.0%).
