The protective effect of xBrassicoraphanus (BR) on liver inhury was evaluated in the rats with liver injury induced by i.p. injection of D-galactosamine (GalN) following 2 week oral treatment of ethanol extract of xBrassicoraphanus (EBR). EBR (200 mg/kg) significantly suppressed the levels of ALT, AST, SDH, ${\gamma}-GT$, ALP, LDH and lipid peroxidation compared with GalN treated control, while EBR at 100 mg/kg significantly suppressed AST and ${\gamma}-GT$. Similarly, EBR at 200 mg/kg significantly attenuated the levels of Phase I enzymes such as XO, AO, AH and AD as well as significantly increased the levels of Phase II enzymes such as SOD, catalase and GSH-Px in the GalN treated rats. Taken together, these results indicate that the ethanol extract of xBrassicoraphanus may have a hepatoprotective effect against GalN induced liver injury, suggesting the ethanol extract of xBrassicoraphanus can be applied as hepatoprotective functional food. However, its mechanism should be further studied in molecular and cellular view points.
Xenobiotics and their reactive intermediates bind to cellular macromolecules and/or generate oxidative stress. which provoke deleterious effects on the cell function. Induction of xenobiotic-biotrans-forming enzymes and antioxidant molecules is an important defense mechanism against such insults. A group of genes involved in the defense mechanism. e.g. genes encoding glutathione S-transferases. NAD(P)H: quinone oxidoreductase, UDP-glucuronosyltransferase (UDP-GT) and ${\gamma}$-glutamylcysteine synthetase (GGCS). have a common regulatory sequence, Antioxidant or Electrophile Responsive Element (ARE/EpRE). Recently. Nrf2. discovered as a homologue of erythroid transcription factor p45 NF-E2, was shown to bind ARE/EpRE and induce the expression of these defense genes. Mice that lack Nrf2 show low basal levels of expression and/or impaired induction of these genes. which makes the animals highly sensitive to xenobiotic toxicity. Indeed. we show here that nrf2-deficient mice had a higher mortality than did the wild-type mice when exposed to acetaminophen (APAP). Detailed analyses of APAP hepatotoxicity in the nrf2 knockout mice indicate that a large amount of reactive APAP metabolites was generated in the livers due to the impaired basal expression of two detoxifying enzyme genes, UDP-GT (Ugt1a6) and GGCS. while the cytochrome P450 content was unchanged. Thus. the studies using the nrf2 knockout mice clearly demonstrate significance of the expression of Nrf2-regulated enzymes in protection against xenobiotic toxicity.
이 연구는 5가지 물질로 구성된 혼합물의 항산화 및 모발 성장 촉진 효과를 확인하기 위해 수행되었다. 발효 5종 혼합물은 적양파, 늙은 호박, 검은콩, 솔잎, 검은깨, 발효 2종 혼합물은 적양파, 늙은 호박으로 구성되어있다. 항산화 효과는 DPPH 라디칼 소거활성 및 총 플라보노이드 함량으로 측정하였다. 발효 5종 혼합물은 발효 2종 혼합물보다 높은 DPPH 라디칼 소거활성과 플라보노이드 함량을 보였다. 모발 성장 효과는 생체 내 실험을 통해 검증되었다. 알칼리성 인산가수 분해효소(ALP), ${\gamma}$-글루타밀트랜스펩티다아제(${\gamma}-GT$) 및 육안적 관찰을 통해 측정하였다. 실험동물은 샘플에 따라 물(NOR), 판시딜(P-CON), 발효 5종 혼합물(5NC), 발효 2종 혼합물(2NC) 총 4군으로 나눴다. 샘플은 일정한 시간에 하루 1번 4주 동안 경구투여하였다. 5NC군에서 ALP, ${\gamma}-GT$ 활성 모두 2NC에 비해 증가하였다. 육안적 관찰에서 P-CON군은 털이 가장 길고 면도 전의 상태와 유사하였다. 5NC군은 P-CON군과 유사한 수준으로 털이 자랐다. 모든 결과에서 발효 5종 혼합물은 발효 2종 혼합물 보다 높은 항산화 효과와 모발 성장 촉진 효과를 보였다. 결론적으로 발효 5종 혼합물은 유용한 모발 성장 촉진제의 원료로 사용할 수 있음을 확인하였다.
Objectives : This study was undertaken to verify the effects of Massa Medicata Fermentata (MMF) on nonalcoholic fatty liver disease (NAFLD) using high fat diet-fed male mice. Methods : Fifty four male C57BL/6N mice (age matched) were used for all experiments. Nine standard chow diet-fed mice were used as normal group and forty five high fat diet-fed obese mice were randomly divided into 5 groups: control, atorvastatin-10mg/kg, MMF(1)-62.5mg/kg, MMF(2)-125mg/kg and MMF(3)-250mg/kg. After all groups were treated with several kinds of diets for 8 weeks, we measured body weight gain, adipose tissue weights, plasma lipid and glucose metabolism, visceral organ weights, histological analysis for liver on the mice. Results : MMF-treated mice had lower body weight gain compared with controls. Among MMF-treated mice, the effect was magnified in MMF(2). MMF(3)-treated mice had lower blood plasma total cholesterol (TC) and glucose level compared with controls. MMF decreased hepatic lipid accumulation, liver fibrosis and liver inflammation of mice compared with controls. The effects was maximized in MMF(2) and atorvastatin. Blood plasma aspartate aminotransferase (AST), alanine aminotransferase (ALT), ${\gamma}$-glutamyltransferase (${\gamma}$-GT) concentrations tends to be decreased by MMF compared with controls. Blood plasma AST, ALT, ${\gamma}$-GT concentrations and organ weights were not changed by MMF, indicating that all three kinds of MMF do not show any hepatotoxicity. Conclusions : These results suggest that MMF improves NAFLD by reducing body weight gain, hepatic lipid accumulation, liver fibrosis, liver inflammation.
The aim of the present investigation was to evaluate the effect of A ferruginea extract on Dalton's lymphoma ascites (DLA) induced tumours in BALB/c mice. Experimental animals received A ferruginea extract (10 mg/kg.b.wt) intraperitoneally for 14 consecutive days after DLA tumor challenge. Treatment with extract significantly increased the life span, total white blood cell (WBC) count and haemoglobin (Hb) content and decreased the level of serum aspartate transaminase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP), gamma glutamyl transferase (${\gamma}$-GT) and nitric oxide (NO) in DLA bearing ascites tumor models. In addition, administration of extract significantly decreased the tumour volume and body weight in a DLA bearing solid tumor model. The levels of pro-inflammatory cytokines such as tumor necrosis factor-alpha (TNF-${\alpha}$), interleukin-1 beta (IL-$1{\beta}$), interleukin-6 (IL-6) and granulocyte monocyte-colony stimulating factor (GM-CSF), as well as pro-angiogenic growth factors such as vascular endothelial growth factor (VEGF) and inducible nitric oxide synthase (iNOS) were elevated in solid tumour controls, but significantly reduced by A ferruginea administration. On the other hand, the extract stimulated the production of interleukin-2 (IL-2) and interferon-gamma (IFN-${\gamma}$) in animals with DLA induced solid tumours. Increase in $CD4^+$ T-cell population suggested strong immunostimulant activity for this extract. GC/MS and LC/MS analysis showed quinone, quinoline, imidazolidine, pyrrolidine, cyclopentenone, thiazole, pyrazole, catechin and coumarin derivatives as major compounds present in the A ferruginea methanolic extract. Thus, the outcome of the present study suggests that A ferruginea extract has immunomodulatory and tumor inhibitory activities and has the potential to be developed as a natural anticancer agent.
작물 초기생육에 대한 저선량 방사선의 효과를 보고자 벼, 콩, 들깨 종자에 저선량 $^{\gamma}$선을 조사하여 발아율과 초기 생육을 관찰하였다. 저선량 $^{\gamma}$선에 의한 작물의 생육초기 효과는 한정되었으나 작물과 생육특성에 따라 적정 선량은 다르게 나타났다. 벼종자의 발아율은 2Gy조사구에서, 유묘초장과 생체중은 0.5Gy에서 가장 높았으며,대두종자의 발아율과 유모생육은 4Gy에서 가장 양호하였다. 들깨종자의 저선량 $^{\gamma}$선 조사에 의해 생육이 다소 불량하였으나 발아율은 2Gy에서, 유묘초장과 생체중은 1Gy에서 증가효과를 나타내었다.
Hepatoprotective action of alcoholic extract of Bacopa monniera (BME) was evaluated on Dgalactosamine (D-GalN) induced rat liver toxicity. Bacopa monniera extract reduced the elevated serum enzyme activities of ALT, AST, ALP, LDH, ${\gamma}-GT$ and the formation of hepatic malondialdehyde induced by D-GalN. The alcholic extract of Bacopa monniera also significantly restored the decreased levels of glutathione and the decreased activities of glutathione peroxidase, glutathione reductase, superoxide dismutase, catalase and glucose-6-phosphatase. Therefore these results suggest that Bacopa monniera has hepatoprotective effect against D-GalN induced hepatotoxicity.
We treated a 53 year-old male patient. He had alcoholic liver cirrhosis and diabetes. The main symptoms were fatigue, ascites, edema, anorexia, indigestion, constipation, and abdominal pain. We diagnosed him as 'the retention of pathogenic damp-heat in the liver and gall bladder’ and adminstered Saenggangeonbi-tang three times a day. We observed clinical symptoms, liver function test(AST, ALT, ${\gamma}-GT$, ALP, total bilirubin), nutritional state(total protein, albumin) and capillary blood glucose. After treatment, all the main symptoms disappeared and the patient's nutritional state has improved. Child's grade using the improvement index in liver cirrhosis has changed from B to A. Although the very mechanism of Saenggangeonbi-tang was not proven, we could suggest that Saenggangeonbi-tang is not toxic to the liver and has a good effect on alcoholic liver cirrhosis with diabetes mellitus.
Cheung-Gan-Haeju tang has been used on 3 cases of alcoholic liver disease patients complicated with ascites, clinical symptom(fatigue. jaundice, urine dark, indigestion, anorexia. ascites etc), liver function (AST, ALT, ${\gamma}$-GT, ALP, total bilirubin), and index of nutritional state (total protein, albumin, cholesterol) were improved after the adminstration. Although the exact mechanism involved in the effects of Cheung-Gan-haeju tang on these disease is still unknown, it is possibly suspected that Cheung-Gan-Haeju tang is non-toxic to liver and has beneficial effects on treating alcoholic liver disease complicated ascites. Further reports with many case, however, will be needed.
목적: 다른 기관들에 비해 간에서는 상대적으로 FDG 섭취 양상이 매우 불규칙하며, 악성 병변이 없는 간에서 비정상적으로 FDG 섭취가 증가하기도 한다. 하지만, 이제까지 이러한 다양한 간의 FDG 섭취를 간의 다른 양성 질환과 연관시킨 연구가 없어, 저자들은 이 연구에서 간에서의 FDG 섭취 양상과 지방간, 간 기능 검사, 지질 프로필을 포함하는 다양한 임상 요인과의 관계를 밝히고자 하였다. 대상 및 방법: 건강검진 목적으로 $^{18}F-FDG$ PET/CT를 시행한 188명 (남/여: 120/68, 평균나이: $50{\pm}9$세)을 대상으로 분석하였다. 간의 FDG 섭취에 영향을 줄 수 있는 당뇨 및 내당증 장애 환자, 심한 간질환이나 오랜 약물 복용의 기왕력이 있는 환자는 연구대상에서 제외하였다. PET/CT의 횡단면에서 간의 5번 분절에 해당하는 부위에 관심영역을 설정하고 이 부위의 평균 SUV를 구하여, 지방간이 있는 군과 없는 군 사이의 차이를 분석하였다. 또한 혈중 간 기능(ALT, AST), 알코올성 간질환(${\gamma}-GT$), 총 콜레스테롤, 중성지방 수치와 간의 FDC 섭취와의 관계를 알아보았다. 지방간 환자들의 체질량지수, 음주력을 조사하여 이 인자들이 간의 FDG 섭취에 미치는 영향도 분석하였다 결과 지방간이 있는 군의 평균 SUV가 지방간이 없는 군보다 통계적으로 유의하게 높았다(p<0.05). 혈중 총 콜레스테롤과 중성지방 수치는 간의 FDG 섭취와 유의한 상관관계를 보인 반면, 다른 요인들은 상관관계를 보이지 않았다. 또한, 지방간 환자에서 음주나 체질량 지수의 위험군과 비위험군 사이에 간의 FDG 섭취 정도는 차이는 보이지 않았다. 다변량 분석 결과, 지방간의 유무와 혈중 중성지방 수치가 간의 평균 SUV에 영향을 주는 유의한 독립 인자였다. 결론: 결론적으로 간의 FDG 섭취 정도는 지방간의 유무에 영향을 받으며, 중성지방 수치와 유의한 상관관계를 보였다.
