Studies on Stream Ecosystem in the Bukhan River using Stable Isotopes (안정동위원소를 이용한 북한강 하천생태계 연구)

  • This study examined the epiphytes using stable isotopes. Separated epiphytes should represent the water quality. In study areas, where agricultural paddies were prevailing, biomass of epiphytes can increase nitrogen isotopes(${\delta}^{15}N$) and as a result decrease in carbon isotopes(${\delta}^{13}C$). Naerin stream and Inbuk upper areas showed higher nitrogen isotopes(${\delta}^{15}N$) than in Hangye and Buk stream. Also, stable carbon isotopes(${\delta}^{13}C$) in Naerin stream and Inbuk streams tended to increase than in Hangye and Buk stream. This can indicate the epiphytes can be a good indicator for water quality pollution.

Food Sources of the Ascidian Styela clava Cultured in Suspension in Jindong Bay of Korea as Determined by C and N Stable Isotopes (탄소 및 질소안정동위원소 조성에 의한 남해안 진동만 양식 미더덕의 먹이원 평가)

  • To examine the trophic ecology of the ascidian Styela clava in an aquaculture system of Korea, stable carbon and nitrogen isotopes were analyzed monthly in S. clava, coarse ($>20{\mu}m$, CPOM) and fine particulate organic matters ($0.7<<20{\mu}m$, FPOM). CPOM (means: $-18.5{\pm}1.2$‰, $9.3{\pm}0.7$‰) were significantly higher ${\delta}^{13}C$ and ${\delta}^{15}N$ values than those ($-20.5{\pm}1.5$‰, $8.4{\pm}0.5$‰) of FPOM. S. clava had mean ${\delta}^{13}C$ and ${\delta}^{15}N$ values of $-18.9({\pm}1.7)$‰ and $11.6({\pm}0.7)$‰, respectively. S. clava were more similar to seasonal variations in ${\delta}^{13}C$ and ${\delta}^{15}N$ values of FPOM than those of CPOM, suggesting that they rely largely on the FPOM as a dietary source. In addition, our results displayed that the relative importance between CPOM and FPOM as dietary source for the ascidians can be changed according to the availability of each component in ambient environment, probably reflecting their feeding plasticity due to non-selective feeding irrespective of particle size. Finally, our results suggest that dynamics of pico- and nano-size plankton (i.e., FPOM) as an available nutritional source to S. clava should be effectively assessed to maintain and manage their sustainable aquaculture production.

  • In this study, inter-laboratory comparison was done using elemental analyzer-isotope ratio mass spectrometers (EA-IRMSs) to determine carbon and nitrogen contents as well as stable carbon and nitrogen isotopic compositions (${\delta}^{13}C$ and ${\delta}^{15}N$) of five environmental samples containing lake and marine sediments, higher plant leaves, and fish muscle, and one organic analytical standard (Protein (Casein) Standard OAS). Five national laboratories participated in this comparison study, and each laboratory analyzed all five samples and the analytical standard. Results showed that variations in total organic carbon (TOC) and total nitrogen (TN) contents as well as ${\delta}^{13}C_{TOC}$ and ${\delta}^{15}N_{TN}$ values among the laboratories were large compared to the analytical uncertainties. The results highlighted the inhomogeneity of the test samples and thus, the need to select suitable standard reference materials for future inter-laboratory studies. Further inter-laboratory comparison exercises could promote good measurement practices in the acquisition of stable carbon and nitrogen isotopic composition data.

  • The nitrogen isotope value in both ammonium and nitrate ion were determined at 9 stations during both June and August 2016, in order to understand the origin of DIN at the Han river. ${\delta}^{15}N-NO_3$ and ${\delta}^{15}N-NH_4$ values in 8 stations (CP, SB, MHC, P4, SJ, SBC, P2, SC) were no significant variation. However ${\delta}^{15}N-NO_3$ and ${\delta}^{15}N-NH_4$ values in KK (Kyeongan stream) showed significant different in comparison with 8 stations, with an apparent increase of nitrogen isotope values. These results indicate that antropogenic nitrogen source influence on KK station. Also the ${\delta}^{13}C$ and ${\delta}^{15}N$ isotope ratio of phytoplankton (Diatom and Cyanobacteria) in KK (Kyeongan stream) showed heavier values, compared to other study stations. These results indicate that nitrogen isotope value in phytoplankton effects by different nitrogen source in study sites. These results suggest that the analysis of stable isotope ratios is a simple but useful tool for the identification of dissolved inorganic nitrogen origin in aquatic environments.

Food Habits of the Glass eel Anguilla japonica in the West Coast Estuaries of Korean Peninsula Determined by Using C and N Stable Isotopes (안정동위원소를 이용한 서해연안 실뱀장어의 먹이 습성)

  • Glass eels (Anguilla japonica) are caught in the west coast of Korea on their migratory route from the breeding grounds in the Mariana Trench along the North Equatorial Current and the Kuroshio Current. To identify the food source of natural glass eels, we analyzed the stable C and N isotopes of glass eels caught in April 2012 and investigated possible food sources in the survey area. In particular, with respect to the stable C and N isotopes of particulate organic matter, we extended the surveying area to the northern parts of East China Sea as well as the west coast of Korea. The stable C and N isotope ratios of the glass eels caught in the west coast were found to be $-20.7{\pm}0.1$‰ and $5.0{\pm}0.2$‰, respectively. The stable C and N isotope ratios of the particulate organic matter in the west coast of Korea, in which the glass eels are assumed to eat the particulate organic matter as food source, were estimated to be $-24.0{\pm}0.3$‰ and $2.8{\pm}0.4$‰, respectively. Similar data were obtained from the northern part of the East China Sea, $-24.5{\pm}0.5$‰ and $0.8{\pm}0.3$‰. The stable isotope ratios showed values differing from the stepwise increasing rates up the food web in natural aquatic ecosystem, showing that particulate organic matter in the west coast of Korea and East China Sea was not served as the glass eels food source. This result suggested that the glass eels caught in the west coast might not assimilate nutrition from the marine environment during long migration.

Planting Conditions of Korean Cannabis Derived from Stable Isotope Ratio & Tetrahydrocannabinol Contents (안정동위원소 비율 및 테트라하이드로칸나비놀 함량 분석을 통한 한국산 대마의 재배 환경 추론)

  • Stable isotope ratio of carbon and nitrogen ($\delta^{13}C$ & $\delta^{15}N$), and $\Delta^{9}$-tetrahydrocannabinol (THC) contents were measured on 37 Korean cannabis and 10 commercial grade marijuana seized in Korea. Factors influencing on the measured values and their variations were investigated. $\delta^{13}C$ value of cannabis is specified mainly by water availability. Korean cannabis showed relatively low $\delta^{13}C$ values ranging -33.29$\sim$-27.01% (mean=-31.01%), which reflect geographic conditions of Korea where is rainy, especially during summer. $\delta^{15}N$ values, which reflect individual planting conditions, were relatively high up to -0.5$\sim$18.0% (mean=6.44%). It reflects characteristics of Korean cannabis growing wild in forest or cultivated in fertile soil. Tetrahydrocannabinol is the major hallucinogenic compound of cannabis. Ethanol extracts of cannabis leaves were derivatized by N,O-bis(trimethylsilyl)trifluoroacetamide (BSTFA), and the derivatives were analyzed by GC-MS in selected ion monitoring (SIM) mode. THC contents of Korean cannabis ranged 0.11$\sim$4.34% (mean=1.47%), which were relatively low compared with commercial grade marijuana.

The Origin of Organic Matters Utilized by Soft Bottom Macrozoobenthos in Tongyeong (안정동위원소 분석을 이용한 통영 연질조하대 대형저서동물의 유기물 기원 추적)

  • To elucidate the organic matter sources in soft-bottom macrozoobenthos at Tongyeong, Korea, macrozoobenthos and their potential food sources were sampled in May 2004, and their C and N stable isotope compositions were analyzed. In total we collected 128 macrozoobenthic species, which ranged from 465 to 3,775 individuals/$m^2$(average 2,108 ind.$m^2$) in density and from 47.8 to 539.9 gwwt/$m^2$(average 366.0 gwwt/$m^2$) in biomass. Cluster and multi-dimensional scaling analyses indicated that the macrozoobenthic community was divided into two distinct groups. Coastal inner stations, where commercial fish cages were established, were dominated by Amphioplus ancistrotus, Scoletoma longifoila and Tharyx sp., whereas open sea stations were dominated by Chaetozone spinosa, Scoletoma longifolia and Capitella capitata. ${\delta}^{13}C$ values of sedimentary organic matter showed a distinct gradient in the range of -18.4 to $-15.2\%_{\circ}$, with a declining trend from the coastal inner stations to open sea stations. This probably reflects the settling rate of organic wastes such as feces and pellets from fish cages near the coastal inner stations. The macrozoobenthos showed a broad ${\delta}^{13}C$ range from -19.5 to $-8.9\%_{\circ}$ at the coastal inner stations, and a relatively narrow range from -21.3 to $-12.9\%_{\circ}$ at the open sea stations. ${\delta}^{13}C$ values of macrozoobenthos paralleled those of sedimentary organic matter, Our isotope results suggested that macrozoobenthos near the coastal inner stations used organic wastes derived from fish cages along with phytoplankton, whereas the macrozoobenthos near the open sea stations used organic wasters derived mainly from phytoplankton.

Geochemical Studies of $CO_2$-rich Waters in Chojeong area II. Isotope Study (초정지역 탄산수의 지화학적 연구 II. 동위원소)

  • The $CO_2$-rich waters in the Chojeong area are characterized by low pH (5.0~5.8), high $CO_2$pressure (about 1 atm) and high amounts of total dissolved iou (up to 989 mg/L) and chemically belong to Ca-HC $O_3$type. The oxygen. deuterium and tritium isotope data indicate that the mixing process occurred between $CO_2$-rich water and surface water and/or shallow groundwaters and also suggest that the $CO_2$-rich water has been derived from meteoric waters. According to $\delta$$^{13}$ C values (-8.6~-5.3$\textperthousand$). the $CO_2$ in the water is attributed from deep seated $CO_2$gas. The high dissolved carbon (-14.4~-6.8$\textperthousand$. $\delta$$^{13}$ C) in groundwater of the granitic terrain might be affected by $CO_2$-rich water, whereas the dissolved carbon (-17.9~-15.2$\textperthousand$. $\delta$$^{13}$ C) in groundwater of the metamorphic terrain is likely controlled by soil $CO_2$ and from the reaction with calcite in phyllite. Sulfur isotope data (+3.5~+11.3$\textperthousand$,$\delta$$^{34}$ $S_{SO4}$) also support the mixing process between $CO_2$-rich water and shallow groundwater. Strontium isotopic ratio ($^{87}$ Sr/$^{86}$ Sr) indicates that the $CO_2$-rich water (0.7138~0.7156) is not related to vein calcite (0.7184) of Buak mine or calcite (0.7281~0.7346) in phyllite. By nitrogen isotope ($\delta$$^{15}$ $N_{NO3}$) the sources of nitrogen (up to 55.0 mg/L, N $O_3$) in the $CO_2$-rich water are identified as fertilizer and animal manure. It also indicates the possibility of denitrification during the circulation of nitrogen in the Chojeong area. The possible evolution model of the $CO_2$-rich water based on the hydrochemical and environmental isotopic data was proposed in this study. The $CO_2$-rich waters from the Chojeong area were primarily derived from the reaction with granite by supply of deep seated $CO_2$. and then the $CO_2$-rich water was mixed and diluted with the local groundwater.ter.

Identification of pollutant sources using water quality and stable isotope ratios of inflow tributaries in the lower reaches of the Han-River

  • Despite the expansion of sewage treatment facilities to reduce pollutants in the tributaries of the Han River, water pollution accidents such as fish deaths continue to frequently occur. The purpose of this study was to identify the pollutant sources using water quality and stable isotope ratio (${\delta}^{15}N$, ${\delta}^{13}C$, ${\delta}^{15}N-NH_4$, ${\delta}^{15}N-NO_3$) analysis results in the three inflow tributaries (Gulpocheon (GP), Anyangcheon (AY) and Sincheon (SC)) of the Han River. Water quality was analyzed in June and October from 2013 to 2017, and the results showed that the concentrations of nutrients, such as T-N, $NO_3-N$, and T-P, were increased at GP4, AY3, SC3, and SC4, which lie downstream of sewage treatment facilities. The results of ${\delta}^{15}N$ for June 2017 indicated that the source of nitrogen was sewage or livestock excreta at GP4 and SC4, and organic fertilizers at AY3 and SC3. ${\delta}^{15}N-NO_3$ results suggested that the source of nitrogen was related to organic sewage, livestock or manure at GP4, AY3 and SC4. Therefore, GP4 and SC4 were more influenced by effluent from sewage treatment facilities than by their tributaries, AY3 and SC3 were considered to be influenced more by their tributary than effluent from sewage treatment facilities. With the results of this study, the source of contamination (sewage treatment facility effluent) of river inflow downstream of Han River could be confirmed using water quality and stable isotope ratio.
