찰성 및 메성 쌀보리 ${\beta}-Glucan$ Gum 특성 비교 (Characteristics of ${\beta}-Glucan$ Gums from Normal and Waxy Hull-less Barleys)

  • 국내산 쌀보리 품종중 ${\beta}-glucan$ 함량이 유사한$(4.6{\sim}5.0%)$ 메성 쌀보리 (무등쌀보리)와 찰성 쌀보리(찰쌀 보리) 각각 1품종씩을 선별한 후 이들로부터 ${\beta}-glucan$ gum 물질을 추출하여 물리화학적 특성을 비교하였다. Crude ${\beta}-glucan$ gum의 평균 수율은 찰성 쌀보리에서 높았으며 ${\beta}-glucan$ 함량은 $57{\sim}64%$였다. 보리 ${\beta}-glucan$ gum의 점도는 찰성과 메성에서 유사하였고 의가소성의 유동특성을 보였으며 ${\beta}-glucan$ 농도가 증가할수록 그 점도도 증가하였다. 쌀보리가루의 아밀로 그래프에 의한 호화특성은 메성 쌀보리가 찰성 쌀보리에 비해 최고점도가 높은 반면 내부효소 불활성화처리에 의해서는 찰성 쌀보리의 최고점도가 더 높게 나타났다. 쌀보리로부터 분리한 전분과 ${\beta}-glucan$ gum을 혼합하여 호화특성을 살펴본 결과 ${\beta}-glucan$은 쌀보리 전분의 점도를 증가시켰으며 그 효과는 메성 쌀보리 전분에서 더 큰 것으로 확인되었다.

보리겨 $\beta$-glucan의 추출 및 이화학적 특성 (Extraction and Physicochemical Characterization of Barley Bran $\beta$-glucan)

  • Bran(1∼5번)의 총 평균 $\beta$-glucan 함량은 5.52%로서 가장 높은 회수율 (48.5%)을 보인 추출 조건은 pH 10 및 45$^{\circ}C$, 다음은 pH 7 및 45$^{\circ}C$로 나타났다. 보리겨의 gum 물질은 배유 부분의 gum 물질과는 달리 상당량의 전분, 단백질이 혼입되어 있었고 정제된 $\beta$-glucan 도 단백질 및 전분 함량이 감소되긴 하였으나 호분층 안쪽의 bran 1,2,3,4에서도 $\beta$-glucan 함량이 78.2∼85.8%를 나타내어 세포벽의 peptide, arabinoxylan 등과 강하게 결합됨을 알 수 있었다. $\beta$-glucan을 이루는 구성당은 glucose가 주종이며, 그 외 arabinose, xylose, mannose, galactose였으며, 분자량은 보리겨에 따라 차이가 있으나 bran 1∼3의 경우 4.09 x $10^{5}$ 에서 4.41 x $10^{5}$ 정도의 분자량이 90% 이상을 차지하였으며, 다당 고유의 특성인 분자량의 분산성을 나타내었다. 한편 $\beta$-glucan의 구조 역시 보리겨에 따라 다소 차이가 있으나 $\beta$-(1\$\longrightarrow$3) 결합, $\beta$-(1\$\longrightarrow$4) 결합이 대부분을 차지하였고, $\beta$-glucan용액의 유동특성은 농도가 증가함에 따라 유동지수는 감소하여 의가소성이 증가하는 경향을 나타내었다.

  • Oat gum, a soluble dietary fiber, was extracted from rolled oat and the content of $\beta$-glucan and the cholesterol adsorption activity of oat gum were examined. The content of oat gum extracted from rolled oat was 5.15% and the amount of $\beta$-glucan in the oat gum was 29.48%. Most of the cholesterol adsorption of oat gum occurred within 10 hours in the dialysis cell and the cholesterol adsorption activity of oat gum was between 12 to 25%.

Physicochemical and Sensorial Properties of Probiotic Yogurt as Affected by Additions of Different Types of Hydrocolloid

  • The main attributes of yogurt that affect consumer satisfaction are taste, consistency, and a firm texture. This study evaluates the influence of xanthan gum, barley beta-glucan, and guar gum in concentrations of 0.05%, 0.1%, 0.2%, and 0.3% on probiotic yogurt. The set-type yogurt samples were prepared by using raw cow's milk. The statistical analysis showed that none of these gum additions had any marked effect on pH, titratable acidity, total solids content, and probiotic bacteria counts of yogurt samples. Evaluations for syneresis and water-holding capacity (WHC) in the yogurt samples were affected by the type and concentration of the stabilizer. Yogurts treated with 0.1% xanthan gum and 0.3% beta-glucan recorded the highest WHC and the least syneresis. The largest amount of gel firmness was recorded in yogurt samples treated with 0.2% xanthan gum and 0.3% beta-glucan. Yogurt samples treated with 0.1% xanthan gum and 0.3% beta-glucan were considered acceptable by trained panelists and gained the highest scores in sensory evaluations. The correlation coefficient between the amount of syneresis, WHC and stiffness of texture was significant compared to scores for sensory evaluation (p<0.01). Results for effects of guar gum on the tested parameters were contrary to the results expected from a gum. According to this study, the use of xanthan gum and beta-glucan are highly recommended for low-fat yogurt production.

  • A soluble dietary fiber, $\beta$-glucan, contained in oat and barley has nutritional benefits such as hypocholesterolemic effects and influences blood glucose regulation. The contents of $\beta$-glucan in both cereals range from 3 to 7% with the exception of a certain barley genotype which contains up to 16% $\beta$-glucan. $\beta$-Glucan is distributed mainly in the cell walls of endosperm and the distal (bran) portion of kernel. Various procedures have been developed for increasing the extraction yield of $\beta$-glucan. Oat gum prepared with weak alkali extractionand alcohol proecipitation following protein removal usually contains 80% $\beta$-glucan.The most commonly used method for $\beta$-glucan quantitiation is an enzymatic procedure combining lichenase plus $\beta$-glucosidase followed by measuring the amount of glucos released by glucose oxidase-peroxidase treatment. The increase in foam-and emulsion-stabilizing capacity of $\beta$-glucan is due to the increase in viscosity of the aqueous phase. Therefore, $\beta$-glucan shows great potentials as a thickener and a stabilizer.

Characterization and Functional Properties of an Oat Gum Extracted from a Drought Harvested Oat (Avena sativa)

  • An oat gum was extracted from whole seeds of a drought harvested oat (Avena sativa). Oat gum presented a ${\beta}-glucan$ content of 65%(w/w) and an intrinsic viscosity of 141 mL/g. Gelling capability of oat gum at different concentrations was investigated. Gel hardness increased from 0.08 to 0.25 N as the oat gum concentration changed from 5 to 10%(w/v). Whippability, foam stability, emulsion stability, and reduced viscosity of oat gum at different pH were also investigated. Oat gum whippability was maximum at pH 7 (146%), while the higher foam and emulsion stability values were found at pH 9 (88 and 96%, respectively). The gum reduced viscosity increased from 715 to 958 mL/g as the pH changed from 7 to 9. Oat gum shows great potential as a gel forming, thickening, and stabilizing agent.

  • 맥주맥(麥灌麥) 장려품종의 원맥(原麥)과 맥아(麥芽)에서 수용성(水溶性) ${\beta}-glucan$함량(含量)은 각각(各各) $1.4{\sim}4.0%$$0.3{\sim}0.6%$범위(範圍)였다. 원맥(原麥)의 ${\beta}-glucan$함량(含豊)은 품종(品種), 재배지역(載培地域) 및 질소질비료(窒素質肥料) 수준(水準)의 영향(影響)을 많이 받는 것으로 나타났다. 원맥(原麥)의 ${\beta}-glucan$함량(含量)은 점도(粘度)와 고도(高度)의 정(正)의 관(關)을 보였으며 , 단백질함량(蛋白質含量)과 total gum함량(會量)에 대(對)하여는 각각(各各) 부(負)의 상관(相關)을 나타내었다. 맥아(麥芽)의 ${\beta}-glucanase$활성(活性)은 점도감소(粘度減少)의 상대시간(相對時間)으로 표시(標示)할 때 $11.0{\sim}20.0$초(秒)였으며, 품종(品種), 재배지역(栽培地域) 및 질소비료(室素肥料) 수준(水準)의 영향(影響)을 많이 받았으며, 제맥아중(製麥芽中) ${\beta}-glucanase$활성(活性)은 제6일(第6日)째 이후(以後)부터 최고수준(最高水準)을 유지하였다. 맥아(麥芽) ${\beta}-glucanase$활성(活性)은 맥아(麥芽)의 잔존(殘存) ${\beta}-glucan$함량(含量)과 부상관(負相關)을 추출수량(抽出收量)과는 정상관(正相關)을 보였다. 맥아(麥芽)의 단백질함량(蛋白質含豊)은 추출수량(抽出收量)과 부상관(負相關)을, 효소력가(酵素力價)와 정상관(正相關)을 보였다.

  • Bacillus licheniformis WL-12의 carboxymethyl cellulase (cellulase) 유전자를 함유한 대장균 균체 파쇄상등액으로부터 DEAE-Sepharose와 Q-Sepharose 컬럼 크로마토그래피를 통해 cellulase를 정제하였다. 정제된 효소의 비활성은 163 U/mg이었으며, SDS-PAGE에 의해 측정된 분자량은 약 49.5 kDa으로 나타났다. pH 5.5와 $55^{\circ}C$에서 최대 반응활성을 보였으며, SDS (5mM)에 의해서는 cellulase의 활성이 완전히 저해되었고 $Cu^{2+}$5mM)에 의해서는 약간 증진되었다. 정제된 cellulase는 CMC, konjac, barley $\beta$-glucan과 lichenan을 가수분해하였으나 xylan, locust bean gum 및 p-nitrophenyl-$\beta$-glucopyranoside를 분해하지 못하였다. Cellooligosaccharides를 정제된 WL-12 cellulase로 분해하였을 때 cellobiose와 cellotriose가 주된 최종 반응산물로 관찰되었으며 cellobiose보다는 중합도가 큰 cellotriose, cellotetrasoe와 cellopentaose는 분해하였으나 cellobiose는 분해하지 못하는 것으로 확인되었다.

Cloning of a Bacillus subtilis WL-7 Mannanase Gene and Characterization of the Gene Product

  • A gene encoding the mannanase of Bacillus subtilis WL-7, which had been isolated from Korean soybean paste, was cloned into Escherichia coli, and the gene product was purified from the culture filtrate of the recombinant E. coli. This mannanase gene, designated manA, consisted of 1,086 nucleotides, encoding a polypeptide of 362 amino acid residues. The deduced amino acid sequence was highly homologous to those of mannanases belonging to the glycosyl hydrolase family 26. The molecular mass of the purified mannanase was 38 kDa as estimated by SDS-PAGE. The enzyme had a pH optimum at 6.0 and a temperature optimum at $55^{\circ}C$. The enzyme was active on locust bean gum, konjak, guar gum, and lichenan, while it did not exhibit activity towards yeast mannan, laminarin, carboxymethylcellulose, $\beta$­glucan, xylan, and para-nitrophenyl-$\beta$-mannopyranoside.

  • Sparassis latifolia is a fungus abundant in β-glucan and amino acids and is highly valued as a medicinal mushroom. Among amino acids, γ-aminobutyric acid (GABA) is a free amino acid and has biological effects, such as increase/decrease of hypertension, improvement of cerebral blood flow, and prevention of dementia. In this study, biological elicitors were used to increase bioactive substances as a biofortification method. Sodium alginate extracted from seaweed (Sargassum horneri, Sargassum fulvellum, Sargassum fusiforme) were used as the elicitor. The levels of β-glucan and GABA in the mycelium and fruiting body grown by adding the elicitor to the medium were investigated. Addition of sodium alginate positively affected GABA production and negatively affected the β-glucan production in these fungi. Sodium alginates extracted from S. fulvellum induced the highest increase in GABA in the mycelium and fruiting bodies. Moreover, we investigated the effects of the extracts from mycelium and fruiting bodies on dendrite development in primary cortical neurons. We found that the extract from the fruiting bodies of sodium alginate treated fungi with increased levels of GABA inhibited the dendrite outgrowth of excitatory neurons, but not inhibitory neurons.
