Study on analysis of components and artificial cultural practice on several culture media of Paecilomyces japonica (눈꽃동충하초 배지별 인공재배법과 성분분석에 관한연구)

  • This experiment was carried out to find the method for mass production by artifical cultivition and to analyze the components of Paecilomyces japonica according to several media. Time of inoculation of the Paecilomyces japonica using silkworm was on first day of five molting and infection rate was 72.0%. Optium medium for mass production of the Paecilomyces japonica was known effective for increasing dry weight and fruitbody at brown rice 80g plus pupa powder 20g. Dry weight of Paecilomyces japonica using fungus of silkworm was 1.2g including pupa and length of fruitbody was appeared 3.0cm to 3.5cm. Content of $\beta$ - glucan was very high as 40.5% at inoculation on the first day of the five molting while 16.4% at brown rice, 20.7% at pupa, 23.1% at brown rice plus pupa powder, and 28.7% at pine sawdust plus wheat bran. Mycelium was poor and pinkly conidiospore was formed on media of centipede, maggot and powder of silkworm.

Artificial Dermis Composed of Gelatin, Hyaluronic Acid and (1\longrightarrow3),(1\longrightarrow6)-$\beta$-Glucan

  • Porous scaffolds composed of gelatin and polysaccharides such as hyaluronic acid and $\beta$-glucan were prepared by using the freeze-drying method after cross-linking with l-ethyl-(3-3-dimethylaminopropyl) carbodiimide hydrochloride (EDC). The scaffold had an inter-connected pore structure with the sufficient pore size for use as a support for the growth of fibroblasts. Results for the contact angle and cell attachment confirmed that high gelatin content in a mixture was suitable for cellular attachment and distribution in two- or three-dimensional fibroblast cultures. However, the addition of polysaccharides aroused the synergistic effects of morphologic and mechanical property of gelatin-based scaffolds. To prepare the artificial dermis for the wound dressing to mimic the normal human dermal skin, fibroblasts were isolated from a child's foreskin, and cultured in gelatin-based scaffolds. An in vivo study showed that the artificial dermis containing the fibroblasts enhanced the wound healing rate and re-epithelialization of a full-thickness skin defect rather than the acellular scaffold after one week.

Quality Properties of Oak Mushroom (Lentinus edodes) Based on Extraction Conditions and Enzyme Treatment (표고버섯(Lentinus edodes)의 추출조건 및 효소처리에 따른 품질 특성)

  • The quality properties of oak mushroom were investigated using extraction conditions and enzyme treatments. The physiochemical properties were excellent at the extraction temperatures of $20^{\circ}C\;and\;40^{\circ}C$. The quality was increased as the extraction time increased but was best at the extraction time of 10 hr. The physiochemical properties such as soluble solid, reducing sugar and crude protein contents were best at the solvent ratio of 1:100 (w/v) so that it was set at 1:100. Thus, enzyme treatment was done at $50^{\circ}C$ for 2 hr with the solvent ratio of 1:100. The result showed that the best quality was shown using 0.2% protease and 0.2% cellulase. With the enzyme treatment, the essential amino acid contents increased by two folds but no difference was shown in $\beta-glucan$ content.

Physico-chemical characteristics and antioxidant activities in oyster mushroom (Pleurotus ostreatus) cultivated with liquid spawn (액체종균으로 배양된 느타리버섯(Pleurotus ostreatus)의 이화학적 특성 및 항산화 활성)

  • The physicochemical characteristics of oyster mushrooms (Pleurotus ostreatus) cultivated using liquid spawn (MLS) were compared with those of commercial mushrooms cultivated using solid spawn. The color intensity of the two types of mushrooms showed no remarkable difference. The hardness of the MLS-cultivated mushrooms was significantly higher, but their moisture content (86.80%) was significantly lower than that of the commercial mushrooms. Mineral contents in MLS-cultivated mushrooms (421.17 mg/100 g) were significantly higher than those in the commercial mushrooms (333.26-362.78 mg/100 g); in particular, the potassium (K) content was the most abundant in the former. The amino acid content in the MLS-cultivated mushrooms (4,695.22 mg/100 g) was about 1.4-2.0 times that in the commercial mushrooms. The essential amino acid contents and sum of aspartic acid and glutamic acid were higher in the MLS-cultivated mushrooms than in the commercial mushrooms. The ${\beta}-glucan$ content in the MLS-cultivated mushrooms was 1.1-2.3 times higher than that in the commercial mushrooms. The total phenol and flavonoid contents and the DPPH and ABTS radical-scavenging activities of the MLS-cultivated mushrooms were significantly higher than those of the commercial mushrooms; however, the reducing power showed an opposite trend. Therefore, MLS-cultivated mushrooms contained higher amounts of valuable components and higher antioxidant activities than commercial mushrooms.

Comparison of physiological activity of medicinal mushrooms produced in Korea and China using different extraction solvents (한국 및 중국산 약용버섯류의 추출용매에 따른 생리활성 성분 비교)

  • In the present study, physiological activities of hot water extracts and 70% ethanol extracts of three types of medicinal mushrooms (Hericium erinaceus, Cordyceps militaris, and Ganoderma lingzhi) produced in Korea and China were investigated. Both the hot-water and 70% ethanol extracts of H. erinaceus and C. militaris from Korea displayed the highest 2,2-diphenyl-1-picrylhydrazy radical scavenging activities. Nitrite scavenging activities of hotwater extracts of G. lingzhi from Korea and China (41% and 39%, respectively) were higher than the activities of 70% ethanol extracts. Total polyphenol contents of hot water extracts of H. erinaceus and C. militaris from Korea were higher than those of 70% ethanol extracts. The ethanol extracts of G. lingzhi from Korea displayed the highest total polyphenol content. C. militaris from Korea displayed the highest ${\beta}-glucan$ level (45.11%). ${\beta}-glucan$ content of H. erinaceus from China (30.87%) was higher than H. erinaceus from Korea (16.94%). The findings indicate that healthy ingredients can be maximally extracted using the optimal solvents for each mushroom. These results will be useful in understanding the difference in physiological activities between the solvents used for the extraction of medicinal mushrooms from Korea and China.

Impact of Korean Malting Barley Varieties on Malt Quality

  • Barley has been used for the production of malt in the brewing industry. Malting is the process of preparing barley through partial germination. Malt extract is the most important quality parameter for malt quality. The grain and malt quality parameters of ten Korean malting barley varieties were studied. Malts was prepared using Phoeix automated micro malting system(Phoenix Bio, Australia). Quality analysis of Barley and malt was determined according to European brewery convention(EBC, 1998) and American society of brewing chemists(ASBC, 1997) method. And the hordeins of barley and malt were extracted with 50% isopropyl alcohol(IPA, 2-propanol) of 1% dithiothreitol(DTT). The analysis of hordeins was carried out by ultra-performance liquid chromatography(UPLC). The mean values of 1000-grains weight, assortment rate, protein content, starch content, beta-glucan content, husk rate, germination energy, germination capacity and water sensitivity of grain were 45.8g, 86.8%, 11.9%, 58.0%, 3.8%, 14.0%, 96.2%, 97.2%, 10.0%, respectively. The mean values of protein content, friability, diastatic power, extract, soluble protein, Kolbach index, beta-glucan of malt and wort were 11.3%, 87.6%, 201WK(Windish Kolbach), 79.3%, 4.6%, 41%, 85mg/L, respectively. UPLC analysis of grain and malt hordeins revealed that the amount of hordeins significantly degraded during malting. Also, we could successfully be used to compare hordein polypeptide patterns with malt quality.

Characteristics of Polysaccharide Isolated from the Fruit Body and Cultured Mycelia of Phellinus linteus IY001 (Phellinus linteus IY001의 자실체와 균사체 배양물로부터 분리한 다당류의 물리화학적 특성 비교)

  • This study was conducted to investigate the characteristics of polysaccharides isolated from the fruit body and cultured mycelia of Phellinus linteus IY001. All fractions were extracted by hot water, followed by ethanol precipitation (F-THE and M-HE) or ultrafiltration (M-HU) (F-TH, F-THE; fruit body, M-HE, M-HU; cultured mycelia). Among these fractions, F-TH fraction was obtained at the highest yields of 6.83% and yield of F-THE was at the level 2.79%. The carbohydrates of these fractions was found to be a heteroglucan composed of glucose, galactose, mannose, fructose, ribose and xylose by analysis of gas chromatography. The total carbohydrate contents of M-HE and M-HU fractions were 99.2%, and 86.0% respectively. The glucose content of M-HE, M-HU and F-THE ranged from 54 to 84.8% of the total monosaccharide. Amino acid pattern showed that all fractions contained a large amount of aspartic acid, glycine, glutamic acid, alanine. Serine and threonine were found to be involved in the linkage, O-linked type. These fractions, except F. TH, contained polysaccharides with the molecular weights of 12 kD and showed the characteristics of IR absorption for ${\beta}-glucosides$ at $890\;cm^{-1}$.

Physicochemical characteristics and antioxidant activities of Lentinula edodes cultivated with liquid spawn according to harvest cycle (액체종균에 의한 표고의 수확 주기에 따른 이화학적 특성 및 항산화 활성)

  • The physicochemical characteristics and antioxidant activities of Lentinula edodes cultivated with liquid spawn were compared according to the harvest cycle (1-3 cycles). There was no significant difference in moisture content. The crude lipid and crude protein contents tended to decrease according to harvest cycle. The content of β-glucan was the highest in mushrooms after one cycle. The β-glucan content in cycles 2 and 3 was 68.5% and 62.3% that of the content after cycle 1, respectively. Mineral content in the pileus and stipe was similar. The mineral content was highest in the pileus after two cycles. Composition and contents of free amino acids tended to decrease according to the harvest cycle. Contents of essential amino acids were higher in mushroom after one cycle. Total phenol content was highest in the pileus (31.96 mg/100 g) and the stipe (21.10 mg/100 g) after one cycle and tended to decrease with subsequent cycles. The flavonoid content was 11.13 mg/100 g after one cycle and significantly decreased according to the harvest cycle. Antioxidant activities were also highest after one cycle and significantly decreased according to the harvest cycle. When compared to L. edodes cultured for one cycle, L. edodes cultivated with liquid spawn for three cycles was considered to have similar marketability in terms of quality characteristics.

Selection of suitable conditions of mycelial growth and materials of bag cultivation in Sparassis crispa (꽃송이버섯의 액체배양조건 및 봉지재배 적합배지 재료선발)

  • Sparassis crispa(Cauliflower mushroom) was an edible mushroom that shows remarkably high contents of ${\beta}$-glucan compared to other edible mushroom. In this study, we aimed to select suitable conditions and materials for mycelial growth and fruit body production. The longer saccharification time of barely result in higher sugar content for eight hours. The optimal sugar content of media for mycelial growth was showed at level of 6 $^{\circ}Brix$. Optimum temperature and pH for mycelial growth in liquid spawn were $25^{\circ}C$ and pH 5.0~6.0, respectively. In case of sawdust was used Larix kaempferi as main material, the fruit body yield and cultivation period of supplemented with 10% wheat flour and 20% corn flour were highest and fastest, respectively.

Change of Physicochemical Characteristics and Functional Components in the Cereals of Saengsik, Uncooked Food by Washing with Electrolyzed Water (전해수 처리한 생식 원료 곡류의 이화학적 특성 및 기능성 성분의 변화)

  • We investigated changes in the physicochemical characteristics and functional components of the Saengsik cereal grains-milled rice, brown rice, glutinous rice, and barley- by washing with electrolyzed water. There were no changes in the composition of all cereal grains that was dried with hot-air and freeze-dried after being washed with electrolyzed water. The L and a values were not affected by drying in milled rice, brown rice, glutinous rice, or barley, but the b value increased in milled rice and glutinous rice-this increase was greater with hot-air drying than with freeze-drying. The dietary fiber content of the all cereals increased with hot-air drying. The niacin content decreased in all cereals; the extent of this change was greater with hot-air drying than with freeze-drying. The ${\beta}-glucan$ content of barley was higher in samples that underwent freeze-drying than in those treated by hot-air drying, but was not affected by washing with electrolyzed water. Thus, other than a change in color for milled and glutinous rice, no changes in the physicochemical characteristics and functional components these products were observed with freeze-drying. Data indicate that the electrolyzed water washing could be effectively used as pasteurization step in the uncooked cereal grains.
