농업생태계와 산림생태계 숲 유형에 따른 대형나방 군집 다양성 연구 (The study on diversity of macromoth community in two different forest types from agro and forest ecosystems)

  • 이 연구의 목적은 농업생태계에 존재하는 작고 파편화된 숲과 인근 산림생태계 내 연속된 숲에 서식하는 대형나방의 종 다양성과 종구성의 차이를 알아보는데 있다. 전남 서남부 8개 지역에서 2018과 2019년 6월과 8월 자외선 등 트랩을 이용하여 대형나방을 채집, 분석한 결과 두 숲 유형 간 뚜렷한 차이를 보였으며 두 그룹 간 종 수, 개체수, 종다양성 지수(H') 모두 산림생태계 숲이 농업생태계 숲보다 더 높게 나타났다. 유사도지수를 이용하여 나방 군집간 차이를 알아본 결과 농업생태계 숲에서 베타다양성이 높음을 확인하였다. 또한 종 구성의 변화율(Turnover)과 내포성(Nestedness)을 따로 나누어 분석한 결과 산림생태계는 종 구성 변화율이 큰 반면 농업생태계는 내포성이 비교적 크게 나타나 서식지 유형에 따라 나방 군집 구성의 차이가 다른 것을 확인할 수 있었다.

Differences in Temporal Variation of Ground Beetle Assemblages (Coleoptera: Carabidae) between Two Well-Preserved Areas in Mt. Sobaeksan National Park

  • Understanding how future climate conditions will be impact on the biodiversity and species composition is important, because biodiversity becomes more important in environment assessment. To understand the biological changes including diversity and species composition over time (temporal variation within a year), the species diversity and composition of ground beetles were investigated in two well-preserved areas in the Sobaeksan National Park using pitfall traps. In addition, relationships between ground beetles and environmental variables were studied by considering temporal variation. We collected 2,146 ground beetle specimens representing 45 species, and individual-based rarefaction curves indicated that similar species richness was found between Geumseon Valley (GV) and Namcheon Valley (NV). The Bray-Curtis matrix comparisons between study sites were characterized by similar ground beetles sample heterogeneity, while temporal variations in abundance, species richness, and ${\beta}-diversity$ of ground beetles showed rather difference over time according to location of study sites. In GV site, minimum temperature was selected as the best predictor for abundance, species richness, and ${\beta}-diversity$ of ground beetles, while those relationships in NV site were more complicated. In conclusion, our study suggests that understanding the different response of ground beetles to climatic variables related to local habitat conditions is important to predict the effect of climate change on biological communities.

Penicillium Diversity from Intertidal Zone in Korea

  • Penicillium species are commonly isolated from various outdoor and indoor environments, including marine environments such as sponges, algae and sand. Penicillium is especially important because numerous bioactive compounds have been isolated. Penicillium was the most common species in intertidal zone in Korea, however the diversity and ecological roles of Penicillium in intertidal zone are not clarified. We explored diversity and ecological roles of marine-derived Penicillium from tidal flat and sea sand in Korea. The diversity of marine-derived Penicillium from Korea was investigated using both culture-dependent and culture-independent approach by ${\beta}$-tubulin sequence. In addition, we evaluated optimal temperature, halo-tolerance, and enzyme activity of Penicillium strains, such as extracellular alginase, endoglucanase, ${\beta}$-glucosidase, and protease. For culture-dependent approach, a total of 182 strains of 62 Penicillium species were isolated, with 53 species being identified. The most common species was Penicillium oxalicum, followed by P. crustosum, P. brasilianum, P. koreense, and P. griseofulvum. Species richness and composition were not significantly different by season, substrates, and seaside. For culture-independent approach using Illumina sequencing, 73 OTUSs were detected. The most frequently observed species was P. antarcticum, followed by P. koreense, P. crustosum, and P. brevicompactum. Diversity of Penicillium was higher during winter season than during summer season and in western sea than in southern sea, respectively. Community structure was significantly different by season and sea side. 52 species were detected by both methods. Unique species were isolated from each of methods - 10 from culture methods and 21 from Illumina sequencing. Furthermore, salinity adaption of the Penicillium varied depending on species. Many Penicillium species showed endoglucanase, ${\beta}$-glucosidase, and protease activity. Some species including P. paneum and P. javanicum degraded the polycyclic aromatic hydrocarbons. Thus, our results demonstrate that intertidal zone in Korea harbors diverse Penicillium community and marine-derived Penicillium play important ecological roles as decomposers of organic material in marine environments.

Genetic Diversity of Plasmodium vivax in Clinical Isolates from Southern Thailand using PvMSP1, PvMSP3 (PvMSP3α, PvMSP3β) Genes and Eight Microsatellite Markers

  • Plasmodium vivax is usually considered morbidity in endemic areas of Asia, Central and South America, and some part of Africa. In Thailand, previous studies indicated the genetic diversity of P. vivax in malaria-endemic regions such as the western part of Thailand bordering with Myanmar. The objective of the study is to investigate the genetic diversity of P. vivax circulating in Southern Thailand by using 3 antigenic markers and 8 microsatellite markers. Dried blood spots were collected from Chumphon, Phang Nga, Ranong and, Surat Thani provinces of Thailand. By PCR, 3 distinct sizes of $PvMSP3{\alpha}$, 2 sizes of $PvMSP3{\beta}$ and 2 sizes of PvMSP1 F2 were detected based on the length of PCR products, respectively. PCR/RFLP analyses of these antigen genes revealed high levels of genetic diversity. The genotyping of 8 microsatellite loci showed high genetic diversity as indicated by high alleles per locus and high expected heterozygosity ($H_E$). The genotyping markers also showed multiple-clones of infection. Mixed genotypes were detected in 4.8% of $PvMSP3{\alpha}$, 29.1% in $PvMSP3{\beta}$ and 55.3% of microsatellite markers. These results showed that there was high genetic diversity of P. vivax isolated from Southern Thailand, indicating that the genetic diversity of P. vivax in this region was comparable to those observed other areas of Thailand.

국립공원 내 자생하는 고산 및 아고산식물의 분포 특성과 다양성 (Distribution Characteristics and Diversity of Alpine and Subalpine Plants Growing Naturally in National Parks)

  • Alpine and subalpine plants are isolated and distributed in mountaintop areas at high altitudes and, are among the species most vulnerable to global warming. If the rate of warming continues at its current rate, it is highly likely that this species will be unable to find new refugia and will be the last to go extinct in their current habitats. Therefore, research on the distribution and diversity of alpine and subalpine plants is urgently needed and is important from the perspective of biodiversity conservation. Therefore, this study focused on the distribution of alpine and subalpine plants native to national parks. Alpine and subalpine plants distributed across the 12 national parks accounted for approximately half (47.78%) of all alpine plants in Korea. The average relative frequency of occurrence was 0.23, plant similarity between national parks was 37.19%, and beta diversity was 0.63. The number of species was positively correlated with the latitude, elevation, and area of the national park. Thus the geographical distributions of alpine and subalpine plants haves distinct spatial specificities and physical environmental differences.

Molecular Characterization of TEM-type $\beta$-Lactamases Identified in Cold-Seep Sediments of Edison Seamount (South of Lihir Island, Papua New Guinea)

  • To determine the prevalence and genotypes of $\beta$-lactamases among clones of a metagenomic library from the cold-seep sediments of Edison seamount (10,000 years old), we performed pulse-field gel electrophoresis, antibiotic susceptibility testing, pI determination, and DNA sequencing analysis. Among the 8,823 clones of the library, thirty clones produced $\beta$-lactamases and had high levels of genetic diversity. Consistent with minimum inhibitory concentration patterns, we found that five ($167\%$) of thirty clones produced an extended-spectrum $\beta$-lactamase. 837- and 259-bp fragments specific to bla$_{TEM}$ genes were amplified, as determined by banding patterns of PCR amplification with designed primers. TEM­1 was the most prevalent $\beta$-lactamase and conferred resistance to ampicillin, piperacillin, and cephalothin. TEM-116 had a spectrum that was extended to ceftazidime, cefotaxime, and aztreonam. The resistance levels conferred by the pre-antibiotic era alleles of TEM-type $\beta$-lactamases were essentially the same as the resistance levels conferred by the TEM-type alleles which had been isolated from clinically resistant strains of bacteria of the antibiotic era. Our first report on TEM-type $\beta$-lactamases of the pre-antibiotic era indicates that TEM-type $\beta$-lactamases paint a picture in which most of the diversity of the enzymes may not be the result of recent evolution, but that of ancient evolution.

Comparison of Microbial Diversity of Korean Commercial Makgeolli Showing High ${\beta}$-Glucan Content and High Antihypertensive Activity, Respectively

  • We measured physiological functionalities, including antihypertensive angiotensin I-converting enzyme inhibitory activity and immun-stimulating ${\beta}$-glucan content for sixty kinds of Makgeolli that is commercially available from the market. As a result, we selected R-12 commercial raw Makgeolli, with a high content of immuno-stimulating ${\beta}$-glucan, and R-14 commercial raw Makgeolli, exhibiting high antihypertensive activity. Due to the similarities in their overall physicochemical properties and raw materials used for fermentation, we compared the microbial flora in order to investigate the reason for the differences in their functionalities. Nested PCR and denaturing gradient gel electrophoresis for yeasts and bacteria were performed for analysis of microbial diversity of two different kinds of Makgeolli (i.e., R-12, R-14), which showed immuno-stimulating ${\beta}$-glucan content and exhibited a very high level of antihypertensive activity, respectively. Analysis of the 18S rDNA amplicon revealed a major presence of the yeast strain Pichia burtonii in every Makgeolli sample. Analysis of the 16S rDNA amplicon revealed a predominance of lactic acid bacteria, and the most frequent lactic acid bacteria were Lactobacillus ingluviei, L. fermentum, and L. harbinensis, and Lactobacillus sp. Among these, L. harbinensis was detected only in R-12 and L. ingluviei was found only in R-14. Different functionalities from the individual commercially available Makgeolli may be attributed to actions of different microbial flora during fermentation.

고성군 화당리 연안에서 식물플랑크톤의 계절 및 지점별 조성 변화 (Spatial and Temporal Variability of Phytoplankton at Hwadang-ri, Goseng-gun)

  • 본 연구는 2013년 고성군 화당리에 있는 7개 지점에 대한 식물 플랑크톤의 공간적 분포, 계절적 분포, 표층과 심층의 깊이에 따른 빈도에 대해 기술한 것이다. 화당리에서 식물 플랑크톤 군집은 3강 60분류군으로 다양하였다. 규조강(Bacillariophyceae)은 41분류군으로 가장 높은 다양성을 나타내었으며 그 다음으로는 와편모강(Dinophyceae)으로 16분류군이었고, 황금색조식물강(Cryptophyceae)이 2분류군, 유글레나식물강(Eugenophyceae)이 1분류 군이었다. 표층은 비교적 높은 밀도와 풍부도를 유지하고 있었다. 그런데 Shannon-Weaver의 다양도 지수는 1월을 제외하고는 표층보다 저층에서 더 높았다. 또 균등도 지수도 1월을 제외하고는 표층보다 저층에서 더 높았다. 전체 군집에 대해 ${\beta}$-다양도는 낮거나(7개 정점의 공간적 표층은 1.125, 저층은 1.481) 보통(7개 정점의 시간적 표층은 1.725, 저층은 1.347)이었다. 계절에 따라서는 식물 플랑크톤의 군집 간에 분류학적 동질성이 있었다. 깊이에 대해서도 역시 동질성을 나타내었다. 그러나 풍부도의 분포와 생체량은 동-서 방향 구배가 유의한 차이를 나타내었다.

Analysis of Fungicide Sensitivity and Genetic Diversity among Colletotrichum Species in Sweet Persimmon

  • Anthracnose, caused by Colletotrichum gloeosporioides (C. gloeosporioides; Teleomorph: Glomerella cingulata), is the most destructive disease that affects sweet persimmon production worldwide. However, the biology, ecology, and genetic variations of C. gloeosporioides remain largely unknown. Therefore, in this study, the development of fungicide resistance and genetic diversity among an anthracnose pathogen population with different geographical origins and the exposure of this population to different cultivation strategies were investigated. A total of 150 pathogen isolates were tested in fungicide sensitivity assays. Five of the tested fungicides suppressed mycelial pathogen growth effectively. However, there were significant differences in the sensitivities exhibited by the pathogen isolates examined. Interestingly, the isolates obtained from practical management orchards versus organic cultivation orchards showed no differences in sensitivity to the same fungicide. PCR-restriction fragment length polymorphism (RFLP) analyses were performed to detect internal transcribed spacer regions and the ${\beta}$-tubulin and glutamine synthetase genes of the pathogens examined. Both the glutamine synthetase and ${\beta}$-tubulin genes contained a complex set of polymorphisms. Based on these results, the pathogens isolated from organic cultivation orchards were found to have more diversity than the isolates obtained from the practical management orchards.

Fungal Diversity and Enzyme Activity Associated with the Macroalgae, Agarum clathratum

  • Agarum clathratum, a brown macroalgae species, has recently become a serious environmental problem on the coasts of Korea. In an effort to solve this problem, fungal diversity associated with decaying A. clathratum was investigated and related ${\beta}$-glucosidase and endoglucanase activities were described. A total of 233 fungal strains were isolated from A. clathratum at 15 sites and identified 89 species based on morphology and a multigene analysis using the internal transcribed spacer region (ITS) and protein-coding genes including actin (act), ${\beta}$-tubulin (benA), calmodulin (CaM), and translation elongation factor (tef1). Acremonium, Corollospora, and Penicillium were the dominant genera, and Acremonium fuci and Corollospora gracilis were the dominant species. Fifty-one species exhibited cellulase activity, with A. fuci, Alfaria terrestris, Hypoxylon perforatum, P. madriti, and Pleosporales sp. Five showing the highest enzyme activities. Further enzyme quantification confirmed that these species had higher cellulase activity than P. crysogenum, a fungal species described in previous studies. This study lays the groundwork for bioremediation using fungi to remove decaying seaweed from populated areas and provides important background for potential industrial applications of environmentally friendly processes.
