Effect of Temperature on the Extraction of β-Glucan from Different Jeju Barley Varieties

  • The effect of different temperatures (45, 55, 65, and $75^{\circ}C$) on the extraction of ${\beta}$-glucan and the properties of extracted ${\beta}$-glucan were investigated with four different varieties of barley. Jeju naked barley, blue barley, beer barley, and black barley contained 6.85, 5.13, 3.58, and 4.16% of ${\beta}$-glucan, respectively. ${\beta}$-glucan in barley was extracted in the range of 64.88 to 93.84% depending on the extraction temperature and barley variety. The ${\beta}$-glucans in Jeju naked barley, Jeju blue barley, and black barley were optimally extracted at $65^{\circ}C$ for 3 h and Jeju beer barley at $75^{\circ}C$. The extracted ${\beta}$-glucan resolubilized to 43.48-81.73% and the ratio of ${\beta}(1{\rightarrow}3)$ to ${\beta}(1{\rightarrow}4)$ linkage was in the range of 1:3.8-5.8. These results suggest that purification and properties of ${\beta}$-glucan depend not only on the water extraction temperature, but also on the barley variety.

대식세포주에서 베타-글루칸에 의한 염증성 사이토카인의 발현 (Expression of Inflammatory Cytokines by Beta-glucan in Macrophage Cell Line)

  • Immune system can protect host attacking from a variety of microorganism and virus through innate and adaptive immunities. The innate immune system can be activated by recognition of conserved carbohydrates on the cell surface of pathogen resulting in protection, immunity regulation and inflammation. Immunostimulating and anti-tumor ${\beta}$-glucan, major cell wall component of many fungi, could be recognized as pathogen associated molecular pattern (PAMP) by C-type lectin such as pathogen recognition receptor (PRR) of host innate immunity cells. In spite of many studies of basidiomycetes ${\beta}$-glucan on immunostimulation, little is known about the precise mechanism as molecular-level. Among C-type lectins, dectin-1 was cloned and reported as a ${\beta}$-glucan receptor. In this report, we demonstrated induction of cytokine gene transcription by Ganoderma lucidum ${\beta}$-glucan in the absence or presence of lipopolysaccharide (LPS) by RT-PCR analysis. The expression of murine dectin-1 (MD-1) on RAW264.7 macrophage by RT-PCR showing both the full length, 757 bp $(MD-1{\alpha})$ and alternative spliced form, 620 bp $(MD-1{\beta})$. Both $MD-1{\alpha}$ and $MD-1{\beta}$ mRNAs were induced by ${\beta $-glucan both in the absence and presence of LPS. To explore expression of inflammatory cytokines by ${\beta}$-glucan, RAW264.7 cells were treated with ${\beta}$-glucan for 12 hours. As a result, the expressions of IL-1 IL-6, IL-l0 and $TNF-{\alpha}$ were increased by ${\beta}$-glucan treatment in a dose-dependent fashion. From these results, ${\beta}$-glucan induced transcriptions of dectin-1 and immune activating cytokine genes, indicating induction of immune allertness by expressing dectin-1 and secreting inflammatory cytokines.

  • Nude mouse의 세균 감염 피부에서 Aureobasidium 유래 ${\beta}$-glucan이 창상 치유에 미치는 영향을 평가하기 위해 본 실험을 실시하였다. 실험 결과, ${\beta}$-glucan은 S. aureus, S. pyogenes 및 P. aeruginosa 에 대해 살균 작용을 나타내지는 않았으나 대조군에 비해 감염 창상 크기를 현저히 감소시켰다. 따라서 ${\beta}$-glucan을 새로운 창상 치유 촉진제로서 고려해볼 수 있을 것이다.

Beta glucan: Heath benefits of optimal ingredients via the mixture ratio of rice bran and Sarcodon aspratus

  • Beta-glucan is a polymerized polysaccharide on beta-1,3 chemical bonds. It has the main pharmacological action of various anticancer mushrooms and is colorless and odorless. In addition, it enhances phagocytosis of macrophages against mycobacterium tuberculosism and increases resistance of host against food poisoning bacteria. Owing to these advantages, beta-glucan was used in various health supplement foods such as immune boosters, hypotensive agent and hypoglycemic agent. Our study was aimed to investigate the effective components of beta-glucan, which was optimized via the contents of Sarcodon aspratus and rice bran. First, the mixture ratio condition of Sarcodon aspratus and rice bran were respectively 10:0, 7:3, 5:5, 3:7, and 0:10. The raw materials were fabricated using hot water extraction method. Distilled water of 100 mL was added to the raw material and extracted. After the extraction processed, the content of effective components was analyzed. The absorbance of beta-glucan was measured using a beta-glucan kit (Megazyme, (1-3) (1-4) BETA-D-GLUCAN ASSAY KIT). The absorbance analysis was repeated three times for accurate analysis. After the extracts were lyophilized, the yields of extracted raw materials according to the mixture ratio conditions of Sarcodon aspratus and rice bran (10:0, 7:3, 5:5, 3:7, and 0:10) were respectively 33.1%, 34.5%, 35.3%, 30.7%, and 24.3%. The absorbance was 1.52, 1.47, 1.50, 1.79, and 1.56, respectively. As a result, the optimum ratio of beta-glucan is 3:7 at Sarcodon aspratus and rice bran. This study suggested that the optimal amount of beta-glucan could be used as a health supplement foods and food additive such as immune boosters, hypotensive agent and hypoglycemic agent. However, the new condition (temperature, time) of hot water extraction to maximize the content of beta-glucan could be considered, could be necessary to compare with the existing extracts.

Physicochemical Properties of β-Glucan from Acid Hydrolyzed Barley

  • This study was performed to investigate changes in the content and purity, as well as physical characteristics of ${\beta}$-glucan extracted from acid hydrolyzed whole grain barleys. Waxy and non-waxy barleys (Hordeum vulgare) were hydrolyzed with different concentrations of HCl (0.1~0.5 N) for 1 h. As the HCl concentration increased, the contents of total and soluble ${\beta}$-glucan from acid hydrolyzed barley decreased. However the ratio of soluble/total ${\beta}$-glucan content and purities of ${\beta}$-glucan significantly increased. The ratio of ${\beta}-(1{\rightarrow}4)/{\beta}-(1{\rightarrow}3)$ linkages, molecular weight, and viscosity of soluble ${\beta}$-glucan of raw barleys were 2.28~2.52, $6.0{\sim}7.0{\times}10^5g/mol$, and 12.8~32.8 centipoise (cP). Those of isolated soluble ${\beta}$-glucan were significantly decreased to 2.05~2.15, $6.6{\sim}7.8{\times}10^3g/mol$, and 3.6~4.2 cP, respectively, with increasing acid concentration. The re-solubility of raw barley ${\beta}$-glucan was about 50%, but increased to 97% with increasing acid concentration. Acid hydrolysis was shown to be an effective method to produce ${\beta}$-glucan with high ratio of soluble ${\beta}$-glucan content, purity, water solubility, and low viscosity.

표고 톱밥재배에서 배지조성과 버섯발생 온도에 따른 β-glucan 함량 비교 (Comparison of β-glucan Contents of Lentinula edodes Cultivated on Sawdust according to Medium Composition and Fruiting Temperature)

  • 표고는 항암활성과 면역활성을 촉진하는 렌티난이라는 ${\beta}$-glucan을 가진 식용버섯이다. 본 연구에서는 표고 톱밥재배에서 배지의 영양원의 종류와 버섯 발생온도에 따른 표고조직의 ${\beta}$-glucan 함량의 변화를 메가자임법으로 조사하였다. 배지 중량이 감소하면서 버섯생산량은 증가하였지만, 이 생산량은 중량감소보다 영양원 종류에 더 크게 영향을 받았다. 콩비지 + $CaCl_2$ 처리구(50.4%)를 제외한 표고의 ${\beta}$-glucan 함량은 39.5~42.1% 범위였다. 자실체 발생 온도에 따른 ${\beta}$-glucan 함량은 발생온도 증가에 따라 반비례 관계가 나타났고, 저온 발생조건에서 42.4%였다.

β-Glucan 추출에 미치는 온도, 용매 농도 및 pH의 영향 (Effect of Temperature, Solvent Concentration, and pH on the β-Glucan Extraction)

  • 추출조건에 따른 귀리겨 ${\beta}$-glucan의 추출조건을 확립하기 위하여 추출온도($40{\sim}60^{\circ}C$), 추출용매의 에탄올 농도(0~20%) 및 pH(5~9)를 변수로 중심합성계획법으로 설계하여 ${\beta}$-glucan 추출특성을 조사하였다. 추출시간의 증가에 따라 ${\beta}$-glucan의 추출속도는 추출초기에 급격히 증가하였으나, 이후에는 완만하였다. 총괄물질전달계수는 $3.363{\sim}8.552{\times}10^{-6}cm/min$ 범위로 낮은 추출온도에서는 추출용매의 농도가 낮을수록, 높은 추출온도에서는 추출용매의 농도가 높을수록 증가하였지만 pH에 의한 영향은 적은 것으로 나타났다. 추출용매의 pH는 중성일 때보다 산성이나 알칼리성일 때 ${\beta}$-glucan의 추출량이 증가하였다. 추출조건에 따라서 ${\beta}$-glucan 수용액의 점도는 유의적인 차이를 보였으며, 총괄물질전달계수가 높을수록 crude ${\beta}$-glucan 수용액의 점도가 높게 나타났으며, 점도, 추출율 및 총괄물질전달계수의 상관관계를 분석한 결과, 각각의 요인들이 높은 양의 상관관계(p<0.01)를 나타내었다.

보리와 귀리의 ${\beta}-Glucans$ 및 가공에 의한 용해성의 변화 (${\beta}-Glucans$ in Barley and Oats and Their Changes in Solubility by Processing)

  • 한국산 보리 및 귀리품종의 수용성, 불용성 및 총 ${\beta}-glucan$ 함량을 분석하여 ${\beta}-glucan$의 용해성을 조사하였다. 보리원맥의 총 ${\beta}-glucan$ 함량은 $3.3{\sim}5.6%$ 범위였으며 $65{\sim}70%$ 정맥수율로 도정하여 껍질 및 강층을 제거한 정맥의 ${\beta}-glucan$ 함량은 $3.5{\sim}7.1%$로 증가하였다. 보리원맥의 수용성 ${\beta}-glucan$ 함량은 $1.4{\sim}3.3%$의 분포였으며 총 ${\beta}-glucan$에 대한 수용성 ${\beta}-glucan$의 백분을로 나타낸 용해성(% solubility)은 $43{\sim}61%$ 범위로 총량의 약 반가량이 수용성인 것으로 나타났다. 보리정맥에 있어서는 총 ${\beta}-glucan$과 불용성 ${\beta}-glucan$ 함량은 증가한 반면 수용성 ${\beta}-glucan$은 약간 감소하는 경향을 보여 용해성이 $35{\sim}55%$로 원맥보다 다소 낮았다. 귀리는 총 ${\beta}-glucan$ 함량이 겉귀리에서 $3.1{\sim}4.0%$, groats에서 $4.0{\sim}4.8%$였으며 불용성 ${\beta}-glucan$ 함량이 $0.5{\sim}0.7%$로 보리에서 보다 훨씬 낮아 추출되어 나오는 수용성 ${\beta}-glucan$은 약 84%로 매우 높게 나타났다. 보리와 귀리는 추출초기에 빠른 속도로 ${\beta}-glucan$이 추출되었으며 귀리가 보리에 비해 추출이 보다 급격히 이루어졌는데 $2{\sim}3$시간 후에는 대부분의 수용성 ${\beta}-glucan$이 추출되었다 추출온도가 $23{\sim}45^{\circ}C$로 증가함에 따라 수용성 추출량이 증가하였으나 $65^{\circ}C$에서는 보리의 경우 추출량이 떨어졌다. 보리 및 귀리는 증자에 의한 가열처리에 의해 불용성 ${\beta}-glucan$ 함량이 증가하여 ${\beta}-glucan$의 용해성이 떨어진 반면 보리의 볶음처리에서는 ${\beta}-glucan$이 가용화됨에 따라 용해성이 증가하는 것으로 나타났다.

Ginseniside Rh2와 베타 글루칸의 암세포 전이억제효과 (Antimetastatic and Antitumor Effect of Ginsenoside Rh2 and ${\beta}-glucan$ in Mice)

  • We have investigated the antimetastatic and antitumor effects of Ginsenoside Rh2 and ${\beta}-glucan$ unsing an experimental metastatic mouse model intravenously injected with B 16 melanoma F 10 cells. Animal groups are divided into six groups according to the dosage of drug administration and the kind of drugs. The groups are control, ${\beta}-glucan$ with 50, 100 and 200 mg/kg, Geinsenoside Rh2 50 mg/kg, and ${\beta}-glucan$ 50 mg/kg + Ginsenoside Rh2 50 mg/kg. Oral administration of various concentration of ${\beta}-glucan$( 50, 100, and 200 mg/kg) were reduced the lung- metastatics induced by metastatic B16 melanoma F 10 cells injection with a dose dependent manner in the syngenic mice. At same dosage group, Ginsenoside Rh2 (50 mg/kg) has more antimetastatic effect than the ${\beta}-glucan$(50 mg/kg). The highest antimetastatic effects was observed in the ${\beta}-glucan$ 50 mg/kg + Ginsenoside Rh2 50 mg/kg group and has a similar tendency in the anti-tumor effects, including decrease of the average tumor weight and increase of the average survival rate. There are no differences of the average tumor weights were apparent in the ${\beta}-glucan$ groups, however there were little decrease of the average tumor weight in Ginsenoside 50 mg/kg group and ${\beta}-glucan$ 50 mg/kg + Ginsenoside Rh2 50 mg/kg group than that of the control group. The rate of average survival rate in the ${\beta}-glucan$ 50 mg/kg + Ginsenoside Rh2 50 mg/kg group, ${\beta}-glucan$ 200 mg/kg, ${\beta}-glucan$ 100 mg/kg and ${\beta}-glucan$ 50 mg/kg, and Ginsenoside 50 mg/kg groups were highly in order. These data suggest that antimetastatic and antitumor effect of combination of Ginsenodide Rh2 and ${\beta}-glucan$ be the highest in this study.

메성 및 찰성보리 ${\beta}-Glucan$의 리올로지 특성 (Rheological Properties of ${\beta}-Glucan$ Isolated from Non-waxy and Waxy Barley)

  • ${\beta}-Glucan$의 점증제 및 겔화제로서의 이용성을 규명하기 위하여 메성 및 찰성보리 ${\beta}-glucan$ 용액의 리을로지 특성을 비교하였다. 메성 및 찰성보리 ${\beta}-glucan$ 용액 모두 의가소생 유체 특성을 나타내었으며, Power law mdel에 잘 적용되었다. Arrhenius 식에 의해 구한 층밀림 속도 $700s^{-1}$에서의 겉보기 점도의 활성화에 너지는 농도가 1%에서 4%로 .증가함에 따라 메성 ${\beta}-glucan$의 경우 20.01에서 16.78kj/mol로, 찰성 ${\beta}-glucan$의 경우 지수의 경우 27.18에서 17.82kj/mol로 감소하였으며, 지수함수식에 의해 구한 농도의존성 상수인 A는 온도가 $20^{circ}C$에서 $65^{circ}C$로 증가함에 따라 메성 ${\beta}-glucan$의 경우 $0.37%^{-1}$에서 $0.42%^{-1}$로, 찰성 ${\beta}-glucan$의 경우 $0.49%^{-1}$에서 $0.57%^{-1}$로 증가하였다. 찰성보리 ${\beta}-glucan$의 고유점도는 2.38dL/g으로 메성보리 ${\beta}-glucan$의 1.60dL/g보다 높게 나타났으며, 비점도와 환원농도의 관계에서 구한 critical coil overlap parameter($C^*[{\eta}]$)를 경계로 기울기가 달라져 $C[{\eta}] 영역에서는 메성 및 찰성보리 ${\beta}-glucan$ 간에 차이가 나지 않았지만 $C[{\eta}]>C^*[{\eta}]$ 영역에서는 각각 2.907과 3.757을 나타내었다. 동적점탄성 측청 결과 진동수 변환중 cross-over가 일어나고 이후의 진동수 영역에서는 저장탄성율이 손실탄성율보다 높은 값을 나타내었으며, 24시간 경과 후 겔을 형성하여 겔화제로서의 이용 가능성을 보여주었다.
