  • In this paper, two weight conditions are introduced and the multiple weighted strong and weak characterizations of the multilinear fractional new maximal operator 𝓜ϕ,β are established. Meanwhile, we introduce the ${\mathcal{S}}_{({\vec{p}},q),{\beta}}({\varphi})$ and $B_{({\vec{p}},q),{\beta}}({\varphi})$ conditions and obtain the characterization of two-weighted inequalities for 𝓜ϕ,β. Finally, the relationships of the conditions ${\mathcal{S}}_{({\vec{p}},q),{\beta}}({\varphi}),\,{\mathcal{A}}_{({\vec{p}},q),{\beta}}({\varphi})$ and $B_{({\vec{p}},q),{\beta}}({\varphi})$ and the characterization of the one-weight $A_{({\vec{p}},q),{\beta}}({\varphi})$ are given.

  • Milk from dairy cows has long provided a high quality source of protein and selected micronutrients as calcuim to most populations. Recently, a relationship between disease risk and consumption of specific bovine ${\beta}$-casein fraction either A1 or A2 genetic variants has identified. Populations, which consume milk contain high containing high levels of ${\beta}$-casein A2 variants, have a lower incidence of cardiovascular disease and type 1 diabetes. Furthermore, consumption of milk with the A2 variants may be associated with less severe symptoms of autism and schizophrenia. The mechanism of action focuses on ${\beta}$-casein A1 and related forms preferentially that are able to produce a bioactive opioid peptide, ${\beta}$-casomorphin-7(${\beta}$-CM-7) during digestion. Infants may absorb ${\beta}$-CM-7 due to an immature gastrointestinal tract. Adult, on the other hand, appear to reap the biological activity locally on the intestinal brush boarder. ${\beta}$-CM-7 can potentially affect numerous opioid receptors in the nervous, endocrine, and immune system. Whether there is a definite health benefit to milk containing the A2 genetic variant is unknown and requires further investigation.

  • In order to express recombinant porcine TGF-${\beta}1$ protein in a baculovirus expression system the entire TGF-${\beta}1$gene containing extra amino acids at the N terminus was cloned into pFBa and pFBb of the Bac-To-$Bac^{TM}$ baculovirus expression system. One of the clones contained 106 extra amino acids and was designated pFBa-106 TGF-${\beta}1$, and the other had 28 extra amino acids and was designated pFBb-28 TGF-${\beta}1$. The orientation of the gene was identified with restriction enzyme mapping and PCR with internal TGF-${\beta}1$ primers. Sf-9 cells were infected at a m.o.i. of 10 by the recombinant viruses generated from the two expected sizes of 55 kD and 46.4kD. these precursor forms of TGF-${\beta}1$ with a polyclonal antibody against human TGF-${\beta}1$. No mature form of TGF-${\beta}1$ protein was detected on SDS gels and an immunoblot indicated that TGF-${\beta}1$ precursor is not properlu processed in insect cells.

Characterization of a New Antidementia $\beta$-Secretase Inhibitory Peptide from Rubus coreanus

  • In order to develop a potent antidementia $\beta$-secretase inhibitor from phytochemicals, $\beta$-secretase inhibitory activities of extracts from many medicinal plants and herbs were determined. Water extracts from Rubus coreanus showed the highest $\beta$-secretase inhibitory activity of 84.5%. After purification of the $\beta$-secretase inhibitor from R. coreanus using systematic solvent extraction, ultrafiltration, Sephadex G-10 column chromatography, and reverse-phase high performance liquid chromatography (HPLC), a purified $\beta$-secretase inhibitor with $IC_{50}$ inhibitory activity of $6.3{\times}10^3\;ng/mL$ ($1.56{\times}10^{-6}\;M)$ was obtained with a 0.08% solid yield. The molecular mass of the purified $\beta$-secretase inhibitor was estimated to be 576 Da by liquid chromatography-mass spectrometry (LC-MS) and $\beta$-secretase inhibitor also is a new tetrapeptide with the sequence Gly-Trp-Trp-Glu. The purified $\beta$-secretase inhibitory peptide inhibited $\beta$-secretase non-competitively and also show less inhibition on trypsin, however no inhibition on other proteases such as $\alpha$-secretase, chymotrypsin, and elastase.

Cholesterol Removal from Homogenized Milk with Crosslinked β-cyclodextrin by Adipic Acid

  • The present study was carried out to develop crosslinking of $\beta$-cyclodextrin ($\beta$-CD) using adipic acid, and to determine the optimum conditions of different factors ($\beta$-CD concentration, mixing temperature, mixing time and mixing speed) on cholesterol reduction from homogenized milk. Crosslinked $\beta$-CD was prepared with adipic acid. When the milk was treated with different conditions, the cholesterol removal rate was in the range of 92.1 to 93.1% with 1% $\beta$-CD addition, which were not significantly different among treatments. After cholesterol removal from milk, the used crosslinked $\beta$-CD was washed for cholesterol dissociation and reused. For recycling study, the cholesterol removal rate in the first trial was 92.5%, which was mostly same as that using new crosslinked $\beta$-CD. With repeated ten time trials using same sample, 81.4% of cholesterol was removed from milk. Therefore, the present study indicated that the optimum conditions for cholesterol removal using crosslinked $\beta$-CD were 1% $\beta$-CD addition and 10 min mixing with 400 rpm speed at 5$^{\circ}C$ with over 90% cholesterol removal. In addition, crosslinked $\beta$-CD made by adipic acid resulted in the effective recycling efficiency.

Effects of Dietary ${\beta}$-Cyclodextrin on Plasma Lipid and Tissue Cholesterol Content in Swine

  • This study examined the effects of dietary ${\beta}$-cyclodextrin (${\beta}CD$) on the cholesterol of blood and tissues of swine. Thirty six male castrated swine ($Landrace{\times}Yolkshire{\times}Duroc$) weighing 50 kg were randomly assigned to one of four dietary groups until their weight reached 110 kg. The groups were: basal diet without ${\beta}CD$ (control) and basal diets containing 1.5%, 3.0%, or 5.0% ${\beta}CD$. Diets and water were offered ad libitum. No significant difference was found between treatments in terms of feeding performance measured by daily intake, daily weight gain, and feed efficiency. Addition of ${\beta}CD$ to the diets significantly reduced total lipid, triglyceride and total cholesterol levels in swine blood, particularly in the group receiving 5.0% ${\beta}CD$, which showed decreases (p<0.05) of 21.9%, 55.6% and 27.7%, respectively. Cholesterol levels in back fat, loin, belly and ham portions of swine fed ${\beta}CD$ significantly differed (p<0.05) from controls, especially in the 5.0% ${\beta}CD$-fed group, with reductions of 26.0%, 27.5%, 17.9% and 18.3%, respectively. These results suggested that the addition of ${\beta}CD$ to the diet of swine could reduce their body cholesterol by decreasing the migration of cholesterol through the blood.

Cholesterol Removal from Lard with Crosslinked ${\beta}$-Cyclodextrin

  • The present study was carried out to determine the optimum conditions of different factors (ratio of lard to water, ${\beta}$-CD concentrations, mixing temperature, mixing time and mixing speed) on cholesterol reduction from lard by using crosslinked ${\beta}$-CD. Crosslinked ${\beta}$-CD was prepared with adipic acid. When the lard was treated under different conditions, the range of cholesterol removal was 91.2 to 93.0% with 5% crosslinked ${\beta}$-CD, which was not significantly different among treatments. In a recycling study, cholesterol removal with crosslinked ${\beta}$-CD in the first trial was 92.1%, which was similar to that with new crosslinked ${\beta}$-CD. In up to eight time trials, over 90% of cholesterol removal was found. The present study indicated that the optimum conditions for cholesterol removal using crosslinked ${\beta}$-CD were a 1:3 ratio of lard to water, 5% crosslinked ${\beta}$-CD concentration, $40^{\circ}C$ mixing temperature, 1 h mixing time and 150 rpm mixing speed. In addition, crosslinked ${\beta}$-CD made by adipic acid resulted in an effective recycling efficiency.

Fine Tuning and Cross-talking of TGF-β Signal by Inhibitory Smads

  • Transforming Growth Factor (TGF)-$\beta$ family, including TGF-$\beta$, bone morphorgenic protein (BMP), and activn, plays an important role in essential cellular functions such as proliferation, differentiation, apoptosis, tissue remodeling, angiognesis, immune responses, and cell adhesions. TGF-$\beta$ predominantly transmits the signals through serine/threonine receptor kinases and cytoplasmic proteins called Smads. Since the discovery of TGF-$\beta$ in the early 1980s, the dysregulation of TGF-$\beta$/Smad signaling has been implicated in the pathogenesis of human diseases. Among signal transducers in TGF-$\beta$/Smad signaling, inhibitory Smads (I-Smads), Smad6 and Smad7, act as major negative regulators forming autoinhibitory feedback loops and mediate the cross-talking with other signaling pathways. Expressions of I-Smads are mainly regulated on the transcriptional levels and post-translational protein degradations and their intracellular levels are tightly controlled to maintain the homeostatic balances. However, abnormal levels of I-Smads in the pathological conditions elicit the altered TGF-$\beta$ signaling in cells, eventually causing TGF-$\beta$-related human diseases. Thus, exploring the molecular mechanisms about the regulations of I-Smads may provide the therapeutic clues for human diseases induced by the abnormal TGF-$\beta$ signaling.

Secretion of Bovine $\beta$-Casein by Saccharomyces cerevisiae

  • Yeast expression plasmids containing an appropriate leader sequence and bovine $\beta$-casein cDNA were constructed to produce $\beta$-casein for the study of its functional characteristics. Two kinds of expression systems for $\beta$-casein were constructed using pCGYl444 as a precursor plasmid. This plasmid is a yeast-E. coli shuttle vector which contains the chelatin promoter. The plasmid pISB202 contains the invertase leader sequence and $\beta$-casein gene. The plasmid pDEB303 contains the original bovine $\beta$-casein leader sequence gene. These two plasmids were introduced into S. cerevisiae AB116 which is a strain deficient in the major yeast proteinases. Each clone was grown in minimal media for 24 h before induction by $CuSO_4$. The cells were thus grown under expression conditions. Both strains harbouring pISB202 and pDEB303 expressed bovine $\beta$-casein. The $\beta$-casein was detected using immunochemical staining after western blot. Secretion of $\beta$-casein was detected in the culture broth. The estimated amount of secreted $\beta$-casein was approximately 50 ${\MU}g$/l.

Immunostimulatory Effects of ${\beta}$-glucan Purified from Paenibacillus polymyxa JB115 on Mouse Splenocytes

  • We investigated the effects of ${\beta}$-glucan purified from Paenibacillus polymyxa JB115 on the viability and proliferation of splenocytes. Splenocytes play a critical role in host immunity. MTT assays and trypan blue exclusion tests revealed that ${\beta}$-glucan significantly promoted the viability and proliferation of splenocytes over a range of concentrations. However, there was no specific subset change. ${\beta}$-glucan protected splenocytes from cytokine withdrawal-induced spontaneous cell death. For further mechanistic studies, ELISA assay revealed that ${\beta}$-glucan enhanced the expression of anti-apoptotic molecules and interleukin 7 (IL-7), a cytokine critical for lymphocyte survival. We also investigated the IL-2 dependency of ${\beta}$-glucan-treated splenocytes to determine if treated cells could still undergo clonal expansion. In flow cytometric analysis, ${\beta}$-glucan induced increased levels of the activation marker CD25 on the surface of splenocytes and ${\beta}$-glucan-treated splenocytes showed higher proliferation rates in response to IL-2 treatment. This study demonstrates that ${\beta}$-glucan can enhance the survival of splenocytes and provides valuable information to broaden the use of ${\beta}$-glucan in research fields.
