Inhibitory effects of the extract of Viscum album on the proliferation of human tumor cell lines

  • A bioassay-guided fractionation of the whole extract of Viscum album (a parasitic plant : Loranthaceae) led to the isolation of two triterpenoidal components, oleanolic acid (1), ${\beta}$-amyrin acetate (2), homoflavoyadorinin B (3) as well as large quantity of free fatty acid mixtures as active ingredients of the extract responsible for the antitumoral property. The EtOAc soluble part and BuOH soluble part of the extract demonstrated a significant inhibition on the proliferation of cultured human tumor cells such as A549 (non small cell lung), SK-OV-3 (ovary), SK-MEL-2 (melanoma), XF498 (central nerve system) and HCT-15 (colon) in vitro, whereas the remaining water soluble part exhibited a poor inhibition.   • Sterol compositions of three solanaceous plant seed oils i.e., tobacco(Nicotiana tabacum L.), datura (Datura stramonium L.) and Chinese lycium (Lycium chinense Mill.) were analyzed by thin layer chromatography, gas liquid chromatography and gas liquid chromatography-mass spectrometry. Cholesterol, cholest-7-enol, campesterol, 24-methylenecholesterol, stigmasterol, sitosterol, and 28-isofucosterot were identified in 4-des-methylsterol fraction. 31-Norlanost-8-enol, $4{\alpha}$-methylcholest-8-enol, lophenol, 31-norlanosterol, obtusifoliol, 31-norcycloartenol, cyloeucalenol, gramisterol (24-methylenelophenol), and citrostadienol in 4-monomethylsterol fraction, and lanost-8-enol, cycloartanol, lanosterol, ${\beta}$-amyrin, 24-methylenelanost-8-enol, cycloartenol, lupeol and 24-methylenecycloartanol in 4,4-dimethylsterol fraction were identified respectively.

  • Isoprenoids represent the most diverse group of metabolites, which are functionally and structurally identified in plant organism to date. Ginsenosides, glycosylated triterpenes, are considered to be the major pharmaceutically active ingredient of ginseng. Its backbones, categorized as protopanaxadiol (PPD), protopanaxatriol (PPT), and oleanane saponin, are synthesized via the isoprenoid pathway by cyclization of 2,3-oxidosqualene mediated with dammarenediol synthase or beta-amyrin synthase. The rate-limiting 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGR), which is the first committed step enzyme catalyzes the cytoplasmic mevalonate (MVA) pathway for isoprenoid biosynthesis. DXP reductoisomerese (DXR), yields 2-C-methyl-D-erythritol 4-phosphate (MEP), is partly involved in isoprenoid biosynthesis via plastid. Squalene synthase and squalene epoxidase are involved right before the cyclization step. The triterpene backbone then undergoes various modifications, such as oxidation, substitution, and glycosylation. Here we will discuss general biosynthesis pathway for the production of ginsenoside and its modification based on their subcellular biological functions.

Antiulcerogenic effects of Gymnosporia rothiana

  • Gymnosporia rothiana (walp) Lawson (celastraceae), commonly known as Maytenus rothiana, is used in Indian folk medicine as an antiulcerogenic agent. However, there have been no scientific reports regarding its antiulcer activity. Therefore, this study was intended to evaluate the antiulcer property of petroleum ether, chloroform, and methanolic extract of leaves of Gymnosporia rothiana at different dose levels in ethanol induced and indomethacin induced gastric ulcer models. It was observed that oral administration of all the extract of Gymnosporia rothiana produces significant reduction in ulcer lesion index as well as increase in volume and pH of gastric content in both experimental models, being petroleum ether extract the most effective at dose of 250 mg/kg; it significantly reduced gastric lesion index (70.06%), in comparison to omeprazole (71.20%) and methanolic extract at a dose of 500 mg/kg (67.22%). Increased gastric mucosal defense mechanism by petroleum ether extract is probably due to its high levels of terpenoids like $\beta$ amyrin, lupeol acetate. The present results clearly shows antiulcer effect of Gymnosporia rothiana against various irritants has been mainly due to cytoprotective effect mediated through prostaglandin and partly due to free radical scavenging activity.

Effects of Precursor and Thidiazuron on Triterpene Saponin Biosynthesis in Centella asiatica (L.) Urban

  • Plants have been known to accumulate a very diverse range of triterpene saponins. We have investigated the regulation of saponin biosynthesis in higher plants using Centella asiatica (L.) Urban as a model plant. Effects of a feeding precursor on asiaticoside production from leaves and on the level of two-type OSCs mRNA were investigated. As a feeding precursor, squalene negatively affected the levels of CYS and bAS mRNA, but it also decreased the production of asiaticoside from whole plants. Plant hormones regulate secondary metabolism, and in plant tissue cultures they could affect both culture growth and secondary metabolite production. Although enhancement of asiaticoside production from whole plant cultures by addition of TDZ (thidiazuron) has been reported, the positive effect of TDZ on the levels of OSCs transcripts was not observed.

Comprehensive Expression Analysis of Triterpenoid Biosynthesis Genes Using Pac-Bio Sequencing and rnaSPAdes assembly in Codonopsis lanceolata

  • Codonopsis lanceolata (C. lanceolata) has been widely used in East Asia as a traditional medicine to treat various diseases such as bronchitis, convulsions, cough, obesity, and hepatitis. C. lanceolata belonging to Campanulaceae contains bioactive compounds such as polyphenols, saponins, and steroids. However, despite the pharmacological significance of C. lanceolata, the genetic information of this plant is limited and there are few studies of its transcriptome. In this study, we constructed a unigene set of C. lanceolata using Pac-Bio sequencing. Furthermore, the reads generated from Pac-bio and Illumina sequencing were mixed and assembled using rnaSPAdes. All genes involved in the triterpenoid pathway, a major bioactive compounds of C. lanceolata, were searched from the two unigene sets and the expression profiles of these genes were analyzed. The results showed that lupeol, beta-amyrin, and dammarenediol synthesis genes were activated in the leaves and roots of C. lanceolata. In particular, the expression of genes related to lupeol synthesis was relatively high, suggesting that the main triterpenoid of C. lanceolata is lupeol. Transcriptome studies related to lupeol synthesis in C. lanceolata have been rarely reported. Lupeol has been reported to have pharmacological effects such as anti-inflammatory, anti-cancer, and anti-bacterial. This study suggests the importance of C. lanceolata as a lupeol producing plant.

Chemical Variability of Leaf Cuticular Waxes According to Leaf Position in Tea Tree

  • Cuticular waxes on tea (Camellia sinensis L.) loaves consisted mainly of alkanes, fatty acids, primary alcohols, triterpenes, and a group of unknown compounds, dominated by primary alcohols and triterpenes. Tea tree accessions used in this study were M-1, M-2, Sakimidori, and Yabukita. For all accessions, the alkane, fatty acid, and primary alcohol constituents consisted of a homologues series, and the major constituents of primary alcohol class were the C28 and C30 homologues. Triterpenes consisted of friedelin, $\beta-amyrin$, and three unidentified ones and friedelin was the most abundant. Leaf area and the total amounts of cuticular waxes per leaf increased with lower leaf position from the apical bud in Yabukita variety. With different leaf position, total wax amount per unit leaf area on the youngest leaves of P1 (the uppermost leaf position) showed the largest amount $(12.80{\mu}g/cm^2)$, and on mature loaves of P2 to P6 ranged from 7.08 to $7.77{\mu}g/cm^2$, and then on the oldest loaves of P7 (the lowest leaf position) remained at an increased level $(17.53{\mu}g/cm^2)$. During leaf development (lower leaf position), the amount of primary alcohols decreased from P1 to P6 and increased at P7, whereas that of triterpenes increased from P1 to P7. The percentage of each wax class in the total wax amount occurred a decrease in primary alcohol and an increase in triterpene, with leaf age.

The EtOAc soluble fraction and BuOH soluble fraction of the extract demonstrated a significant inhibition on the proliferation of cultured human tumor cells such as A549 (non small cell lung), SK-OV-3 (ovary), SK-MEL-2 (melanoma), XF498 (central nerve system), and HCT-15 (colon) in vitro, whereas the remaining water soluble fraction exhibited a poor inhibition. The intensive phytochemical investigation of the EtOAc soluble fraction and BuOH soluble fraction of the extract indicated that the oleanolic acid (1) and large amounts of free fatty acid mixtures might be attributed to the in vitro antitumoral property of the whole extract of Viscum album var. coloratum.

Identification of Korean Ginseng Cytochrome P450 gene and Its Characterization by Transformation System (고려인삼 유래 Cytochrome P450 유전자의 동정 및 형질전환에 의한 특성검정)

  • Triterpenoid saponins were synthesized in Panax ginseng C.A. Meyer via the isoprenoid pathway by cyclization of 2,3-oxidosqualene to give primarily oleanane (beta-amyrin) or dammarane triterpenoid skeletons. The triterpenoids are backbone and undergoes various modifications (oxidation, substitution and glycosylation), mediated by cytochrome P450 (CYP)-dependent monooxygenases, glycosyltransferase and other enzymes. This is likely to be due in part to the complexity of the molecules and the lack of pathway intermediates for biochemical studies. A cDNA clone encoding a putative CYP gene was isolated from flower bud of ginseng and transformed into the plant(Nicotiana tabacum cv. Xanthi) and confirmed by PCR analysis. The CYP gene (PgCYP) contained an open reading frame(ORF) encoding mature protein of 500 amino acids. The expression of PgCYP were investigated in transgenic tobacco by reverse transcriptase-polymerase chain reaction (RT-PCR).

The Effect of Haliotidis Concha on the Growth and Ginsenoside Biosynthesis of Korean Ginseng Hairy Root (인삼 모상근의 생장과 Ginsenoside 생합성에 미치는 석결명의 영향)

  • In order to investigate the effects of elicitors on the growth and ginsenoside biosynthesis of ginseng hairy roots, we treated Panax ginseng hairy root with various concentrations of Haliotidis concha according to different time course. Haliotidis concha supplement increased the biomass and ginsenoside accumulation at 10 mg/L concentration. The growth rate of hairy root under a lighter concentration was greater than hairy root treated with a denser concentration. The highest content and productivity of ginsenosides appeared at 2 weeks after the treatment of 10 mg/L Haliotidis concha. The gene expression of squalene synthase, squalene epoxidase, dammarenediol synthase, cycloartenol synthase, $\beta$-amyrin synthase in hairy roots of ginseng were examined by RT-PCR. The Haliotidis concha treatment resulted in the obvious accumulation of the mRNA of triterpene biosynthesis in Panax ginseng hairy root as compared with the control. In this study, Haliotidis concha acts as a kind of elicitor for the production of ginsenosides.
