한약재 찌꺼기로 제조한 한방보드의 휘발성 성분의 방출 특성 (Emission Properties of Volatile Compounds from Medicine Herb Residues Board)

  • 한약재 찌꺼기로 제조한 한방보드로부터 방출되는 향기성분을 Tenax 튜브에 흡착시킨 후 TD GC/MS에 의해 동정하고, 온도에 따른 방출특성을 검토하였다. 한방보드로부터 방출되는 휘발성 물질로는 88종이 동정되었다. 온도별로는 $25^{\circ}C$에서 44종, $35^{\circ}C$에서는 55종, $45^{\circ}C$에서는 65종이 검출되었으며, 온도가 상승함에 따라 화합물의 수와 전체피크 면적 모두 증가하였으며, 측정한 모든 온도조건에서 검출된 향기성분은 34종이었다. $25^{\circ}C$에서 검출된 향기성분의 93%, $35^{\circ}C$에서는 92%, $45^{\circ}C$에서는 90%가 Hydrocarbon류의 화합물이었으며, 그 외 Ether류가 약 4%, Ketone, Aldehyde, Acid류의 물질이 소량 검출되었다. Hydrocarbons에는 17종, 11%가 monoterpenes이며, 80%, 39종이 sesquiterpenes이었다. monoterpenes에는 limonene이 가장 많이 검출되었으며, sesquiterpene류의 주요 향기 물질로는 ${\alpha}-curcumene$, zingiberene, ${\beta}-elemene$, ${\beta}-selinene$, ${\alpha}-amorphene$, ${\alpha}-copaene$이었다. 특히 ether화합물인 anethole (3.26%)이 모든 온도에서 비교적 많이 검출되었다. 이상의 결과에서 한약재 찌꺼기로 제조한 한방보드에는 다양한 종류의 향기성분이 다량 포함되어 있으며, 향기성분을 활용한 다양한 제품의 제조에 활용할 수 있을 것으로 생각된다.

  • 한국특산 섬오갈피나무 및 지리산오갈피나무의 잎과 줄기에서 추출한 주요 정유성분으로, 섬오갈피나무의 경우에는 ${\delta}-3-carene$ (31.34%), l-limonene (17.01%), ${\beta}-elemene$ (4.53%), trans-p-menth-2-ene-1,8-diol (3.13%), 1,8-cineole (4.73%), 1-dodecen-3-yne (2.64%), (Z)-3,7-dimethyl-1,3,6-octatriene (3.21%), 지리산오갈피나무의 경우에는 ${delta}-3-carene$ (14.78%), limonene (7.24%), t-ocimene (7.22%), ${\alpha}-terpinolene$ (8.76%), ${gamma}-elemene$ (4.32%), ${\beta}-selinene$ (7.72%), veridifloral (3.25%), dodecane (2.44%) 등의 성분함량이 높음을 알 수 있었다. 특히 ${\delta}-3-carene$는 섬오갈피나무 및 지리산오갈피나무에서 가장 높은 함량을 나타내었다.

수리취와 참취 엽의 휘발성 향기성분 비교 (Comparison of Volatile Aroma Compounds between Synurus deltoides and Aster scaber Leaves)

  • This study was investigated to compare the volatile aroma compounds of Synurus deltoides and Aster scaber. The volatile aroma compounds from Synurus deltoides and Aster scaber were extracted by soild-phase microextraction (SPME) methods. S. deltoides had 97 volatile aroma compounds such as including 5-acetyl-1,2-dihydro acenaphtylene (14.63%), ${\beta}$-cubebene (9.31%), caryophyllene (8.97%), ${\beta}$-chamigrene (7.14%), ${\beta}$-selinene (2.71), ${\alpha}$-farnesene (2.47%), ${\alpha}$-bergamotene (2.26%), ${\beta}$-elemene (1.94%), etc. A. scaber had 84 volatile aroma compounds such as (+)-epi-bicyclosesquiphellandrene (10.38%), terpinolen (10.09%), caryophyllene (6.04%), 8-isopropenyl-1,5-dimethyl-1,5-cyclodeca diene (5.42%), ${\alpha}$-himachalene (5.04%), ${\beta}$-thujene (4.37%), ${\beta}$-pinene (4.28%), ${\beta}$-cubebene (3.99%), etc. Conclusively, the main common volatile aroma compounds in S. deltoides and A. scaber leaves were 19 volatile aroma compounds such as caryophyllene, terpinolen, ${\beta}$-cubebene. But the composition and amount of volatile aroma compounds were very different between the two species.

Sesquiterpene Derivatives Isolated from Cyperus rotundus L. Inhibit Inflammatory Signaling Mediated by NF-${\kappa}B$

  • The immune system is finely balanced by the activities of pro-inflammatory and anti-inflammatory mediators or cytokines. Unregulated activities of these mediators can lead to the development of various inflammatory diseases. A variety of safe and effective anti-inflammatory agents are available with many more drugs under development. Of the natural compounds, the sesquiterpenes (nootkatone, ${\alpha}$-cyperone, valencene and ${\alpha}$-selinene) isolated from C. rotundus L. have received much attention because of their potential antiinflammatory effects. However, limited studies have been reported regarding the influence of sesquiterpene structure on anti-inflammatory activity. In the present study, the anti-inflammatory potential of four structurally divergent sesquiterpenes was evaluated in lipopolysaccaride (LPS)-stimulated RAW 264.7 cells, murine macrophages. Among the four sesquiterpenes, ${\alpha}$-cyperone and nootkatone, showed stronger anti-inflammatory and a potent NF-${\kappa}B$ inhibitory effect on LPS-stimulated RAW 264.7 cells. Molecular analysis revealed that various inflammatory enzymes (iNOS and COX-2) were reduced significantly and this correlated with downregulation of the NF-${\kappa}B$ signaling pathway. Additionally, electrophoretic mobility shift assays (EMSA) elucidated that nootkatone and ${\alpha}$-cyperone dramatically suppressed LPS-induced NF-${\kappa}B$-DNA binding activity using 32Plabeled NF-${\kappa}B$ probe. Hence, our data suggest that ${\alpha}$-cyperone and nootkatone are potential therapeutic agents for inflammatory diseases.

감귤 주스 착즙박을 이용하여 재배된 버섯균사체의 용매추출에 의한 휘발성 성분 (Solvent Extracted Volatile Components of Mushroom Mycelia Cultivated with Citrus Juice Processing Wastes)

  • 감귤 부위별 및 감귤 주스 가공 후 폐기되는 착즙박을 배지로 하여 재배된 간, 표고, 새송이, 산호침, 참부채 및 영지 버섯) 균사체의 GC/MS에 의한 휘발성 물질을 분석하였다. 감귤 분말에서 휘발성 물질은 29종류이었는데, ${\delta}-elemene$, ${\beta}-elemene$, ${\beta}-caryophyllene$, caryophyllene, ${\gamma}-elemene$, germacrene-D 및 ${\delta}-cadinene$은 감귤류의 essential oil 중의 성분들이었으나 8-hydroxy-linalool, ${\beta}-selinene$, tetradecanoic acid 및 pentadecanal은 essential oil 중의 성분들이 변화한 것이었다. 이외에도 tangeretin, nobiletin, 및 stigmasterol이 검출되었다. 버섯 균사체 분말에서 검출된 휘발성 성분들은 18종이었으며, ${\beta}-elemene$, germacrene-D 및 ${\delta}-cadinene$은 버섯이나 균사체에서 보고되지 않은 것으로 보아 균사체 배지로 사용한 착즙박에서 유래한 것으로 생각된다. caryophyllene, hexadecanoic acid, decanoic acid 및 tetradecanoic acid는 기 보고된 버섯이나 균사체의 성분들이었으며, Limonene-1,2-epoxide, ethyllinoleate, 1,2-benzenedicarboxylic acid, dicyclohexyl ester 및 ($3{\beta}$,24S)-stigmast-5-en-3-ol (${\gamma}-sitosterol$)은 감귤이나 버섯 균사체에서 보고되지 않은 성분들이 검출되었다.

야산 재배 더덕의 휘발성 향기성분에 관한 연구 (The Volatile Flavor Components of Fresh Codonopsis lanceolata cultivated on a wild hill)

  • 갓 채취한 야산재배 더덕의 휘발성 향기성분을 Head space sampling 방법으로 추출하고 GC/MS로 분석한 결과 terpene 및 terpene alcohol류 28종, hydrocarbon류 34종, alcohol류 31종, aldehyde 및 ketone류 13종, ester류 25종, acid류 6종, ether류 10종 등 총 167 종의 휘발성 향기성분을 확인하였다. 크로마토그램 상에서 차지하는 면적 비율이 큰 10대 주요 휘발성 성분은 dl-limonene(10.2%), ${\alpha}$-guaiene(9.0%), 2,2,6-trimethyl-octane(8.6%), hexadecane(8.0%), isolongifolan-8-ol(4.2%), 2,4,4-trimethyl-1,3-pentanediol diisobutyrate(4.1%), ${\beta}$-selinene(3.9%), 2,2,3-trimethylnonane(3.6%), 3-methyl-5-propyl-nonane(3.1%), 및 ledene(3.1%) 등으로 주로 20분 이내와 40${\sim}$60분 범위에서 나타났으며, 전체의 58%를 차지하였다. 식품의 향기는 기계적 분석 결과만으로 설명하기 어려우므로 전문 조향사들의 도움을 받아 관능평가를 실시하였으며, 그 결과 신선 더덕의 향기 특성으로 풋냄새, 흙냄새, 우유발효 냄새 및 장뇌냄새 등이 묘사되었다. 이러한 향기 특성을 GC/MS에서 확인된 20대 주요 휘발성 성분들과 관련시켜 보면, dl-limonene은 풋냄새 성분이며, ${\alpha}$-guaiene과 isolongifolan-8-ol은 흙냄새를, 우유발효취를 내는 성분들에는 2,2,6-trimethyloctane, hexadecane, 3-methyl-5-propylnonane, 2,2,3-trimethylnonane 등이 속하였다. 이 외에 과일향을 내는 2,4,4-trimethyl-1,3-pentanediol diisobutyrate와 쓴냄새 성분인 ${\beta}$-selinene, ledene, isoledene, ${\delta}$-cadinene 및 나무 냄새를 갖는 ${\alpha}$-humulene, dihydro-${\beta}$-agarofuran 등도 식품의 향기와 관련이 있는 것들이다. 이에 반해 풋냄새와 관련 있는 성분들 즉, 1-hexanol, 2-methylhexan-3-ol, 3-hexen-1-ol, cis-3-hexenyl butyrate, ethylhexanol, hexyl acetate, trans-2-hexen-1-ol이나 camphor, longiborneol, menthol 등 장뇌냄새의 원인 물질은 함유량이 상대적으로 적은데도 불구하고 전문조향사들은 신선 더덕의 특징적인 향기로 풋냄새와 장뇌취를 선정하였다. 따라서 유통업자를 중심으로 향기 특성 및 강도를 판정할 수 있는 훈련이이루어진다면 향기는 더덕의 신선도를 판정하는 지표로 활용될 수 있다고 생각된다.

전통 황칠 도료 개발에 관한 연구 (II) - 황칠나무 수피 추출물 및 삼출액의 화학적 조성과 도료용 오일특성 - (Studies on the Technical Development of Traditional Korean Golden Varnish (Hwangchil)(II) - Chemical Composition and Coatings-oil Characteristics of the Exudates and Bark-extractives of Hwangchil-namu(Dendrogapax morbifera Lev.) -)

  • In order to reconstruct the traditional technology of Korean golden varnish(Hwangchil) made from the exudates of D. morbifera, this study in the second step was carried out to analyze the chemical composition and oil properties of the exudates for coatings and bark extractives made from the trees of D. morbifera more than 20 years old grown in Wando and Jejudo islands in the southwestern part of Korean peninsula according to the physiological seasons. The results obtained are as follows: 1. The exudates appeared to be a phytoalexin because it exudated after several days in tapping only in summer season. 2. Both the exudates and bark-extracts with acetone contained 50~60% unsaponifiables and 30~50% saponifiables 3. A major component of essential oil in bark-extractives was ${\beta}$-cubebene and reached to 60~80%, while that of exudates included only 34% ${\beta}$-cubebene and some other compounds such as ${\beta}$-cadinene of 12%, aromadendrene of 9%, ${\beta}$-selinene of 9%, CAS030021-74-0 of 10%, etc. in GC-MS spectrometer. 4. A mjor component of fatty oil fraction in bark-extractives was linoleic acid and reached to more than 60%, while the exudates had mostly other components such as terpenes and phenolics instead of fatty acids accordint to GC-MS spectrometer. 5. Iodine value of samples after oil refining had 214mg/g in the exudates and more than than 150mg/g in bark-extractives, so the latter belonged to a drying oil.. 6. Therefore, the exudates from D. morbifera for traditional Korean golden varnish seems to have beeen used to a good varnish because it has some specific compounds different from its bark-extractives or general varnishes.

다년생 초본류의 향기성분 분석 (Analysis of Essential Oil from Perennial Herbaceous Plants)

  • The chemical composition of essential oil from the perennial herbaceous plants (Houttuynia cordata, Filipendula glaberrima, Peucedanum japonicum, and Ainsliaea acerifolia) was determined by GC/MS spectrometric analysis with the aid of NBS, Wiley Library and RI indice searches. The major constituents identified were $\alpha$-phellandrene (18.97%), $\gamma$-terpinene (12.32%), decanal (8.72%), 1-decanol (10.92%), decanoic acid (12.12%), and 2-undecanone (12.32%) from H. cordata, farnesol (2.83%), l-$\alpha$-terpineol (2.72%), benzenmethanol (2.03%), (Z)-3-hexen-1-ol (4.32%), and T-muurolol (2.07%) from F. glaberrima, $\alpha$-phellandrene (14.25%), endobornyl acetate (3.84%), heptanal (47.52%), octanal (2.65%), (E,E)-2,4-decadienal (2.75%), and octanoic acid (4.52%) from P. japonicum, and geyrene (9.74%), $\beta$-cubebene (11.15%), berkheyaradulen (22.32%), $\beta$-elemene (6.21%), (-)-A-selinene (4.85%), benzaldehyde (4.52%), and benzenacetaldehyde (3.40%) from A. acerifolia.

참나물의 휘발성 향기성분 분석 (Analysis of Volatile Flavor Components of Pimpinella brachycarpa)

  • Volatile flavor components of fresh, shady air dried, and presteamed shady air dried Chamnamul (Pimpinella brachycarpa) were collected by simultaneous steam distillation-extraction method, and essential oils were analyzed by gas chromatography-mass spectrometry (GC/MS). Twenty five, 17 and 23 volatile flavor components were identified in essential oils extracted from the fresh, shady air dried, and presteamed shady air dried Chamnamul samples, respectively; however, the kinds of individual components and its percent content of the total volatiles were varied depending on samples. The principal components of Chamnamul were isobutanal, trans caryophyllene, trans ${\beta}$-farnesene, and ${\alpha}$-selinene. Terpenoid compounds reached 44.11%, 33.91% and 72.63% respectively in fresh, shady air dried, and presteamed shady air dried Chamnamul.

Antibacterial and phytochemical properties of Aphanamixis polystachya essential oil

  • Now a day's rise of new antibiotic resistant bacterial strains is a global threat. Ethnic people of India have been employing Aphanamixis polystachya (Wall.) R. Parker wood extract in healing cancerous wounds. The aim of this study was to evaluate the antimicrobial activity and to identify the medicinally potent chemicals in the essential oil extract of A. polystachya. The antibacterial properties of various organic extracts were evaluated against a range of bacteria (gram-positive and gram-negative bacteria) based on the disc diffusion method and GC-MS based analysis for finding active oil extract components. All extracts of A. polystachya leaves showed potential antibacterial activity, notably ethyl acetate, while petroleum ether extracts revealed highly sensitive activity against all tested bacteria (zones of inhibition ranging from 8.83 to 11.23 mm). In addition, the petroleum ether extract had the lowest MIC value (32 to $256{\mu}g/mL$) against E. coli, S. lutea, X. campestris, and B. subtilis bacteria. The major compounds detected in oil [${\beta}$-elemene (16.04 %), ${\beta}$-eudesmol (12.78 %), ${\beta}$-caryophyllene (19.37 %), ${\beta}$-selinene (11.32 %), elemol (5.76 %), and ${\alpha}$-humulene (5.68 %)] are expected to be responsible for the potent antimicrobial activity. The results of this study offer valuable insights into the potent role of A. polystachya essential oil extract in pharmaceutical and antibiotic research.
