Characteristics of mushroom Phellinus baumii extracts with enzyme pretreatment (효소 전처리에 의한 상황버섯 β-glucan 추출물의 특성)

  • This study was conducted to establish optimized ${\beta}-glucan$ extraction method through enzymatic hydrolysis from Phellinus baumii and investigate ${\beta}-glucan$ contents and physicochemical properties. The optimal condition was obtained with the enzyme concentration of 0.66% (v/v), reaction time of 6.08 h ($R^2=0.9245$) and the ${\beta}-glucan$ contents from the Phellinus baumii extracts under the optimized condition was 1.9594 g/100 g. ${\beta}-Glucan$ yield (0.76-16.40%) of enzyme beta-glucan extract (EBE) was three fold higher than that of non-enzyme beta-glucan extract (NEBE). ${\beta}-Glucan$ purity (11.15-59.05%) of non-enzyme beta-glucan (NEB) and that of enzyme beta-glucan (EB) were higher than that of NEBE and that of EBE. ${\beta}-Glucan$ purity of EB (59.05%) and ${\beta}-glucan$ contents of EB (3.38 g/100 g) showed higher than those of others. Total sugar contents (0.61-1.17 mg/mL) showed that NEB and EB were higher than that of NEBE and EBE, EB had the highest total sugar content as 1.17 mg/mL, respectively. Protein contents (0.44-11.73 mg/mL) of NEBE and that of EBE were higher than that of NEB, that of EB. In FT-IR spectrum, the band at $890cm^{-1}$ of microcapsule was attributed to a ${\beta}-1,3-glucan$. The toxicities of ${\beta}-glucan$ from Phellinus baumii in both melanoma cell lines was determined using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazoli um bromide assay and ${\beta}-glucan$ from Phellinus baumii has no toxicity until $30{\mu}g/mL$. The effects of ${\beta}-glucan$ from Phellinus baumii on inhibition of cancer cell proliferation were detected by using a wound healing assay. The effect of NEB and EB were higher than NEBE and EBE, especially $30{\mu}g/mL$ of EB had the highest in both melanoma cell lines.

Effects of Malt Modification on ${\beta}$-Glucan Solubility and Beer Viscosity (보리의 발아정도가 맥아의 ${\beta}$-glucan 용해성 및 맥주의 점도에 미치는 영향)

  • Two barley malt samples were selected at two different stages of germination, a well-modified malt germinated for 96 hr and a poorly-modified malt for 60 hr, and were analyzed for total, insoluble, and soluble ${\beta}$-glucan contents. The total ${\beta}$-glucan content in raw barley was 3.96%, and the content was reduced during malting. The total ${\beta}$-glucan contents of the poorly- and well-modified malts were 1.02% and 0.18%, respectively. After 4 days of germination, approximately 95% of the ${\beta}$-glucan present in the barley was degraded. A significantly higher proportion of water-soluble ${\beta}$-glucan was found in the well-modified malt, suggesting that ${\beta}$-glucan solubility was dependent on cell wall modifications in the malt (${\beta}$-glucan breakdown). The proportion of water-soluble ${\beta}$-glucan was also affected by the extraction temperature. The two differently modified malts were mashed isothermally at 45, 55, 65, and 75oC for 2 hr. An increasing mashing temperature resulted in increased viscosity for the wort and the resulting beer. The viscosity of the wort from the well-modified malt was significantly low, due to its low initial malt ${\beta}$-glucan with increased solubility as well as a presumably sufficient ${\beta}$-glucanase activity during mashing.

Physicochemical Properties of Barley β-Glucan with Different Heating Temperatures (열처리 온도에 따른 보리 β-Glucan의 이화학적 특성)

  • This study was performed to investigate the changes of total and soluble ${\beta}$-glucan contents, purity, and physical characteristics of three heated barley varieties: Saessalbori (SSB), Saechalssalbori (SCSB), and Hinchalssalbori (HCSB). The barleys were heated at different temperatures of 110, 120, 130, 140 and $150^{\circ}C$ for 2 hours. The total ${\beta}$-glucan contents of raw SSB, SCSB, and HCSB were 8.40, 7.77 and 8.28%, and the soluble ${\beta}$-glucan contents were 4.79, 4.14, and 4.61%, respectively. After heating at $130^{\circ}C$, the total ${\beta}$-glucan contents increased to 11.59, 14.6, and 13.36%, as did the soluble ${\beta}$-glucan contents to 4.21, 7.96, and 7.23%, respectively. The purities of soluble ${\beta}$-glucan of the raw barleys were 35.11, 32.74 and 25.62%, but after heating at $150^{\circ}C$, it increased to 83.43, 91.02, and 88.01%, respectively. The molecular weight and viscosity of the ${\beta}$-glucan solution decreased with increasing heating temperature. The re-solubility of raw barley ${\beta}$-glucan was about 50%, but it was increased to 97% with increasing heating temperature. These results suggest that heating of ${\beta}$-glucan can improve the utilization of barley ${\beta}$-glucan.

${\beta}-Glucan$ Enrichment from Pearled Barley and Milled Barley Fractions (보리의 도정 및 제분분획을 이용한 ${\beta}-Glucan$의 강화)

  • Two hulled and two hull-less barley varieties were investigated for ${\beta}-glucan$ enrichment. Hull-less barleys contained higher levels of total ${\beta}-glucan$ than hulled barleys, and were thus suitable as starting materials for preparing ${\beta}-glucan-rich$ fractions. Particularly, a waxy type (Suweon-291) of hull-less barley was found to have high soluble dietary fiber content containing primarily ${\beta}-glucan$, compared to the other non-waxy barley varieties. ${\beta}-Glucan$ content of barley during pearling process was measured, and the highest value was observed at the pearling yield of approximately $70{\sim}75%$. The pearled barley grains were ground and sieved to yield ${\beta}-Glucan$ enriched fractions containing up to 22% ${\beta}-glucan$. In the meanwhile, whole barley samples were directly milled by $B{\ddot{u}}hler$ mill to produce bran, shorts, break flour and reduction flour. ${\beta}-Glucan$ contents in the bran and shorts from the milled stream were relatively high, and further concentration of ${\beta}-glucan$ could be accomplished by successive sieving of the bran and shorts fractions. Pearled barley and milled stream could be used to prepare barley fractions with ${\beta}-glucan$ concentrations $2.4{\sim}3.1$ times those of the original barley grain. Water solubility of barley ${\beta}-glucan$ from pearled barley and the milled stream was in the range of $40{\sim}81%$.

  • Phellinus baumii has been used in traditional oriental medicine for the treatment of various cancer types, such as lung cancer, ovarian cancer and malignant melanoma. It has strong anti-cancer, anti-inflammatory and antioxidant activities due to its polysaccharides including glucan, schizophyllan, heteroglycan and lentinan, as well as its polyphenolics such as protocatechuic acid, caffeic acid, coumaric acid. ${\beta}$-Glucan and polyphenolics may be the most important activ ecompounds in P. baumii. Therefore, researchers have focused on these two compounds to improve their contents in extracts. In this study, P. baumii was extracted with hot-water and ethanol at different pH conditions, and their ${\beta}$-glucan contents, antioxidant activity and antioxidant contents were determined. Extraction yield was highest for the 60% ethanol extract at pH 4. The ${\beta}$-glucan contents of the hot-water extract at pH 7 was higher than those of the ethanol extracts. The antioxidant contents and antioxidant activities of the ethanol extracts were higher than those of the hot-water extracts. Extraction with 60% ethanol at pH 7 was appropriate with respect to the antioxidant capacities.

Effect of Cultural Environments on ${\beta}-Glucan$ Contents and ${\beta}-Glucanase$ Activities on Malting Barley Varieties (재배환경(栽培環境)이 맥주맥(麥酒麥)의 ${\beta}-Glucan$ 함량(含量)과 ${\beta}-Glucanase$ 활성(活性)에 미치는 영향(影響))

  • The contents of water soluble ${\beta}-glucan$ were $1.4{\sim}4.0%$ in grains of 4 recommended varieties, and $0.3{\sim}0.6%$ in their malt. The ${\beta}-glucan$ content was subject to environmental factors such as cultivating region and nitrogen fertilizer level. The ${\beta}-glucan$ content of grains was positively correlated with the viscosity, but negatively with the protein and total gum contents. The ${\beta}-glucanase$ activity was $11.0{\sim}20.0$ sec as the reduced flow time in malt of 4 recommended varieties and was also subject to environmental factors. ${\beta}-Glucanase$ activity showed the highest level after the 6th day during malting. The ${\beta}-glucanase$ activity in malt was positively correlated with malt extract, but negatively with the residual ${\beta}-glucan$ content. The protein content in malt was negatively correlated with malt extract, but positively with the diastatic power.

Determination of Glucan Contents in the Fruiting Bodies and Mycelia of Lentinula edodes Cultivars

  • Shiitake mushrooms (Lentinula edodes) containing ${\beta}$-glucans may be beneficial for human health; they have been used in the treatment of cancer, hypertension, and high cholesterol levels. The objective of this study was to determine the ${\beta}$-glucan content in different sections of the fruiting bodies and mycelia of ten shiitake mushroom cultivars. The measured ${\beta}$-glucan content ranged from $20.06{\pm}1.76%$ to $44.21{\pm}0.13%$ in the pileus sections, and from $29.74{\pm}1.40%$ to $56.47{\pm}4.72%$ in the stipe sections. The results of this study indicate that the variance in ${\beta}$-glucan content dependent on the shiitake cultivar, and that the ${\beta}$-glucan content is higher in the stipe than in the pileus.

Characteristics of ${\beta}-Glucan$ Gums from Normal and Waxy Hull-less Barleys (찰성 및 메성 쌀보리 ${\beta}-Glucan$ Gum 특성 비교)

  • A normal and a waxy hull-less barley with similar ${\beta}-glucan$ contents were selected, and the effects of ${\beta}-glucans$ on rheological and pasting properties were investigated by using their flour extracts and isolated ${\beta}-glucan$ gum materials. ${\beta}-Glucans$ in the barley cultivars were extracted in a crude form with alkaline extraction, and the waxy hull-less barley produced more ${\beta}-glucan$ gum yield. The waxy barley also showed higher viscosities of water and alkaline flour extracts, compared to the normal barley. Both normal and waxy barley ${\beta}-glucan$ gums exhibited pseudoplastic flow behavior, and increasing ${\beta}-glucan$ concentration increased viscosity in a similar manner. The normal barley flour had a higher amylograph peak viscosity than did waxy barley flour. On the other hand, waxy barley flour with treatment of $HgCl_2$ demonstrated considerably higher increase in peak viscosity. Pasting characteristics of normal and waxy barley starches in the presence of ${\beta}-glucan$ gum solutions were tested using a rapid visco-analyzer (RVA). ${\beta}-Glucan$ gums increased the pasting viscosities of the barley starches, and the synergistic increase in viscosity appeared to be higher in the normal barley starch.

Beta glucan: Heath benefits of optimal ingredients via the mixture ratio of rice bran and Sarcodon aspratus

  • Beta-glucan is a polymerized polysaccharide on beta-1,3 chemical bonds. It has the main pharmacological action of various anticancer mushrooms and is colorless and odorless. In addition, it enhances phagocytosis of macrophages against mycobacterium tuberculosism and increases resistance of host against food poisoning bacteria. Owing to these advantages, beta-glucan was used in various health supplement foods such as immune boosters, hypotensive agent and hypoglycemic agent. Our study was aimed to investigate the effective components of beta-glucan, which was optimized via the contents of Sarcodon aspratus and rice bran. First, the mixture ratio condition of Sarcodon aspratus and rice bran were respectively 10:0, 7:3, 5:5, 3:7, and 0:10. The raw materials were fabricated using hot water extraction method. Distilled water of 100 mL was added to the raw material and extracted. After the extraction processed, the content of effective components was analyzed. The absorbance of beta-glucan was measured using a beta-glucan kit (Megazyme, (1-3) (1-4) BETA-D-GLUCAN ASSAY KIT). The absorbance analysis was repeated three times for accurate analysis. After the extracts were lyophilized, the yields of extracted raw materials according to the mixture ratio conditions of Sarcodon aspratus and rice bran (10:0, 7:3, 5:5, 3:7, and 0:10) were respectively 33.1%, 34.5%, 35.3%, 30.7%, and 24.3%. The absorbance was 1.52, 1.47, 1.50, 1.79, and 1.56, respectively. As a result, the optimum ratio of beta-glucan is 3:7 at Sarcodon aspratus and rice bran. This study suggested that the optimal amount of beta-glucan could be used as a health supplement foods and food additive such as immune boosters, hypotensive agent and hypoglycemic agent. However, the new condition (temperature, time) of hot water extraction to maximize the content of beta-glucan could be considered, could be necessary to compare with the existing extracts.

Physicochemical Properties of β-Glucan from Acid Hydrolyzed Barley

  • This study was performed to investigate changes in the content and purity, as well as physical characteristics of ${\beta}$-glucan extracted from acid hydrolyzed whole grain barleys. Waxy and non-waxy barleys (Hordeum vulgare) were hydrolyzed with different concentrations of HCl (0.1~0.5 N) for 1 h. As the HCl concentration increased, the contents of total and soluble ${\beta}$-glucan from acid hydrolyzed barley decreased. However the ratio of soluble/total ${\beta}$-glucan content and purities of ${\beta}$-glucan significantly increased. The ratio of ${\beta}-(1{\rightarrow}4)/{\beta}-(1{\rightarrow}3)$ linkages, molecular weight, and viscosity of soluble ${\beta}$-glucan of raw barleys were 2.28~2.52, $6.0{\sim}7.0{\times}10^5g/mol$, and 12.8~32.8 centipoise (cP). Those of isolated soluble ${\beta}$-glucan were significantly decreased to 2.05~2.15, $6.6{\sim}7.8{\times}10^3g/mol$, and 3.6~4.2 cP, respectively, with increasing acid concentration. The re-solubility of raw barley ${\beta}$-glucan was about 50%, but increased to 97% with increasing acid concentration. Acid hydrolysis was shown to be an effective method to produce ${\beta}$-glucan with high ratio of soluble ${\beta}$-glucan content, purity, water solubility, and low viscosity.
