Determination of 8-iso-PGF as Oxidative Stress Marker in Human Urine by High Performance Liquid Chromatography with Tandem Mass Spectrometry (LC/MS/MS를 이용한 산화성 스트레스 지표로써 소변 중 8-iso-PGF 분석)

  • This study aimed to develop analytical method for 8-isoprostanes as biomarkers for oxidative stress with LC/MS/MS technique and to apply the method for human urine samples. Analyzed compounds for urinary oxidative stress markers were 7 stereo-isomers of prostaglandins and the internal standard (iso-$PGF_{2{\alpha}}-d_4$) was used to adjust the recovery rate. The method for determining urinary iso-$PGF_{2{\alpha}}$ consisted of solid phase extraction and LC/MS/MS detection. Separation of isomers of prostaglandins completed by porous graphitic carbon column and buffer solution. Detection limits for urinary markers of oxidative stress, iso-$PGF_{2{\alpha}}$ with LC/MS/MS were 0.01 ng/ml by S/N ratio 3 and 0.028 ng/ml by calculated as to FDA method. The recovery (92.8~101.9%) and precision (8.8~20.7%) of analysis were feasible for detecting iso-$PGF_{2{\alpha}}$ in real human urine samples. We detected 4 isomers of prostaglandins in human urine samples. Mean (standard deviation) of urinary iso-$PGF_{2{\alpha}}$ concentration were 0.231 (0.117) ng/mg creatinine for smoking group and 0.154 (0.082) ng/mg creatinine for non-smoking group.

  • Dual scintillator for simultaneous alpha- and beta-ray counting used by detection materials of a surface contamination monitor was developed. In this study, preparation method was not a heat melting method but a solvent method, by which the counting material was manufactured by dissolving the polymer materials with solvent. It was simplified the preparation process. Plastic scintillator for beta-ray counting was prepared by solidifying the casting solution mixed with organic scintillator, polymer, and solvent. ZnS(Ag) scintillator layer was prepared by screen printing the paste solution mixed with ZnS(Ag), paste, and solvent onto the plastic layer. The good counting ability for alpha- and beta-ray using the ZnS(Ag)/plastic dual scintillator prepared and possibility for the counting material of surface contamination monitor was confirmed.

Optical Properties of α-In2S3:Co2+ Single Crystal (α-In2S3:Co2+ 단결정의 광학적 특성에 관한 연구)

  • The ${\alpha}-In_2S_3:Co^{2+}$ single crystal with a good quality and stabilized property were gained successfully by the CTR(Chemical Transport Reaction)method. XRD analysis showed that the grown single crystals were cubic structure. The optical absorption spectra of ${\alpha}-In_2S_3:Co^{2+}$ single crystal showed impurity absorption peaks due to cobalt impurity. These impurity absorption pesks were assigned to the ligand transition between the split energy levels of $Co^{2+}$ ions sited in $T_d$ symmetry of these semiconductor host lattice.

  • This study concerns the existence of positive solution for the following nonlinear system $$\{-div(|x|^{-ap}|{\nabla}u|^{p-2}{\nabla}u)=|x|^{-(a+1)p+c_1}({\alpha}_1f(v)+{\beta}_1h(u)),x{\in}{\Omega},\\-div(|x|^{-bq}|{\nabla}v|q^{-2}{\nabla}v)=|x|^{-(b+1)q+c_2}({\alpha}_2g(u)+{\beta}_2k(v)),x{\in}{\Omega},\\u=v=0,x{\in}{\partial}{\Omega}$$, where ${\Omega}$ is a bounded smooth domain of $\mathbb{R}^N$ with $0{\in}{\Omega}$, 1 < $p,q$ < N, $0{{\leq}}a<\frac{N-p}{p}$, $0{{\leq}}b<\frac{N-q}{q}$ and $c_1$, $c_2$, ${\alpha}_1$, ${\alpha}_2$, ${\beta}_1$, ${\beta}_2$ are positive parameters. Here $f,g,h,k$ : $[0,{\infty}){\rightarrow}[0,{\infty})$ are nondecresing continuous functions and $$\lim_{s{\rightarrow}{\infty}}\frac{f(Ag(s)^{\frac{1}{q-1}})}{s^{p-1}}=0$$ for every A > 0. We discuss the existence of positive solution when $f,g,h$ and $k$ satisfy certain additional conditions. We use the method of sub-super solutions to establish our results.

Adsorption and Biological Properties of Ni-treated PAN Based Activated Carbon Fiber (Ni가 처리된 PAN계 활성탄소섬유의 흡착특성과 생물학적 특성)

  • The study on the adsorption, the surface properties and the antibacterial effects of the Ni-treated PAN based activated carbon fibers was carried out. In the adsorption study on the Ni-treated PAN based ACFs, Type I isotherms for N1-N3 and Type II-Type III isotherms for N4-N6 were obtained, respectively. Futhermore, their adsorbed volumes slowly were decreased with the increase in the mole concentration of Ni on the treated PAN based ACFs. From the BET equation, the specific surface areas of the Ni-treated PAN based ACFs were in the range of $692.58-895.24m^2/g$. The micropore volumes obtained from ${\alpha}_s$-method using common-t value were $0.19-0.56cm^3/g$. The surfaces of PAN based ACFs partially blocked by metal after the treatment were observed from the SEM micrographs. Finally, from the antibacterial effects using Shake flask method against E. coli, the percentage of the effects was 92.5-100% and the antibacterial effect was increased with the increase in mole concentration of Ni treated.

Antifungal Activities of Essential Oil from the Roots of Angelica dahurica Bentham et Hooker f. (구릿대 뿌리 정유의 항진균 효과)

  • The essential oil was extracted from the roots of Angelica dahurica Bentham et Hooker f. by steam distillation and its composition was analyzed by GC-MS. The antifungal activities were evaluated by micro-dilution method against five Aspergillus and three Trichophyton species. The most abundant component was ${\alpha}$-pinene (17.21%) among 40 compounds identified in this oil. The essential oil fraction of A. dahurica and ${\alpha}$-pinene exhibited marked inhibiting activities against the tested Aspergillus and Trichophyton species with MICs (minimum inhibitory concentrations) between 0.12 mg/ml and 8 mg/ml.

  • The use of both an Alpha Hydroxy Acid, Citric Acid, and a Skin Moisturizer, Glycereth-26, formulated into a hand dishwashing detergent have been shown to be synergistic in their effects on certain skin health parameters. Each ingredient was evaluated alone and together in a hand dishwashing detergent via a 9-week use test. Panelists washed dishes using the sponge method commonly used in Asian markets and a 1:7 dilution of the concentrated dish detergent. Panelist's hands were evaluated initially and at 3-week intervals for nine weeks. After nine weeks panelist's hands showed significant improvements in Moisture Absorption and Transepidermal Water Loss (TEWL). A synergistic effect on TEWL was found between the AHA and the Moisturizer. These effects, showing an improvement in the condition of the panelist's skin are impressive, particularly since they were observed from a dishwashing product that is highly diluted and at near neutral pH during the washing process.

  • Like Alzheimer's disease, Parkinson's Disease(PD) is one of the most common neurodegenerative brain disorders. PD results from the deterioration of dopaminergic neurons in the brain region called the substantia nigra. Currently there is no cure for PD, but diagnosing in its early stage is important to provide treatments for relieving the symptoms and maintaining quality of life. Unlike many diagnosis methods of PD which use a single biomarker, we developed a diagnosis method that uses both biochemical biomarkers and imaging biomarkers. Our method uses ${\alpha}$-synuclein protein levels in the cerebrospinal fluid and diffusion tensor images(DTI). It achieved an accuracy over 91.3% in the 10-fold cross validation, and the best accuracy of 72% in an independent testing, which suggests a possibility for early detection of PD. We also analyzed the characteristics of the brain fiber pathways of Parkinson's disease patients and normal elderly people.

Detection of Bifidobacteria by ${\alpha}-Galactosidase$ activity (${\alpha}-Galactosidase$의 활력차이에 의한 Bifidobacteria의 선별)

  • This method using the synthesis substrate of $5-bromo-4-chloro-3-indolyl-{\alpha}-galactoside\;(X-{\alpha}-Gal)$ was examined for the differential enumeration of Bifidobacteria and lactic acid-producing bacteria. Bifidobacteria possess a high level of ${\alpha}-galactosidase$ activity. Bifidobacterium longum KCTC 3215 exhibited the highest ${\alpha}-galactosidase$ specific activity (8.57 units/mg protein). Determination of ${\alpha}-galactosidase$ activity using the PNPG procedure showed that Lactobacillus, Streptococcus, Pediococcus, and Leuconostoc strain had lower ${\alpha}-galactosidase$ activity as compared to Bifidobacteria. The $X-{\alpha}-Gal$ based medium is useful to identify Bifidobacteria among lactic acid-producing bacteria since the enzyme action of ${\alpha}-galactosidase$ spills $X-{\alpha}-Gal$ substrate and releases indol which impacts a blue color to Bifidobacterial colonies on agar plates. All strains of Bifidobacteria appeared as blue colonies on MRS agar medium supplemented with $100\;{\mu}M\;X-{\alpha}-Gal$ while colonies of other lactic acid-producing bacteria appeared white or light blue.
