  • 본 연구에서는 상아질의 항복인장강도 (UTS)나 탄성계수 (E)와 같은 물리적 특성이 적용된 용매틀의 각각에 대한 Hoy의 수소결합 용해도 매개변수에 반비례한다는 가설을 설정하고 실험한 결과 가설이 입증되었으며 이를 토대로 다음과 같은 결론을 유도하여 보았다. 첫째는 탈회된 상아질의 인장 특성 및 물성이 가해진 극성 용매의 수소결합능에 밀접히 연관되어 있다는 것이며, 둘째는 낮은 수소결합능을 가진 용매는 교원섬유 층 내에서 새로운 펩타이드간 수소결합을 유도함으로써 탈회된 상아질의 인장력 및 탄성계수를 증가시킨다는 결과이다. 셋째로는 이러한 결과들을 토대로 높은 수소결합능을 가진 용매들은 새로운 펩타이드간 수소결합의 형성을 차단하여 탈회된 상아질의 구조적 특성을 유지시킬 수 있다는 결론을 도출하였다.

막대형 Ni-Zn 페라이트 입자의 합성 및 특성 평가 (Synthesis and Characterization of Rod-Shaped Ni-Zn Ferrite Particles)

  • The rod-shaped $Ni_{0.5}Zn_{0.5}Fe_2O_4$ particles were synthesized via a topotactic reaction, in which goethite (${\alpha}-FeOOH$) particles are the main constituents. The phases, microstructures and magnetic properties of these particles were studied using XRD, FE-SEM and VSM. The precursor solution consisted of $NiSO_4{\cdot}xH_2O$, $ZnSO_4{\cdot}xH_2O$, goethite and D.I. water werereacted at four different temperatures (50, 70, 90, $100^{\circ}C$) to generate four differently precipitated particles respectively. During the co-precipitation reaction, the pH of the solution was maintained at 8.0 using NaOH. The particles co-precipitated and calcined at a temperature of $700^{\circ}C$, exhibited a rod-shape similar to its original goethite, which means that the shape of Ni-Zn ferrite particles can be topotactically controlled by the goethite. The particles synthesized at 70 and $90^{\circ}C$ have a saturation magnetization of 29 and 35 emu/g respectively; representing better values than the ones synthesized at the 50 and $100^{\circ}C$, in which some second phases such as $Fe_2O_3$ were observed.

  • Objectives: The present study is focused on the inhibitory effect of the rhizome hexane fraction extract of Zingiberis Rhizoma Crudus (ginger hexan extract; GHE) on the production of inflammatory mediators such as NO, $PGE_2$, and proinflammatory cytokines in lipopolysaccharide (LPS)-stimulated BV2 cells, a mouse microglial cell line. Methods: We separated the hexane fraction from Zingiberis Rhizoma Crudus's methanol extract. The inhibitory and anti-inflammatory effect of GHE was examined on microglial activation. Results: GHE significantly inhibited the excessive production of NO, $PGE_2$, TNF-${\alpha}$, and IL-1${\beta}$ in LPS-stimulated BV2 cells. In addition, GHE attenuated the mRNA expressions and protein levels of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and proinflammatory cytokines. Conclusion: The anti-inflammatory properties of GHE may make it useful as a therapeutic candidate for the treatment of human neurodegenerative diseases.

Blueberry, blackberry, and blackcurrant differentially affect plasma lipids and pro-inflammatory markers in diet-induced obesity mice

  • BACKGROUND/OBJECTIVES: Evidence indicates that berry anthocyanins are anti-atherogenic, antioxidant, and anti-inflammatory. However, berries differ vastly in their anthocyanin composition and thus potentially in their biological and metabolic effects. The present study compared hypolipidemic, antioxidant, and anti-inflammatory properties of blueberry (BB), blackberry (BK), and blackcurrant (BC) in a diet-induced obesity (DIO) mouse model. MATERIALS/METHODS: Male C57BL/6J mice were fed a high fat (HF; 35% fat, w/w) control diet or a HF diet supplemented with freeze-dried 5% BB, 6.3% BK or 5.7% BC for 12 weeks (10 mice/group) to achieve the same total anthocyanin content in each diet. Plasma lipids, antioxidant status and pro-inflammatory cytokines were measured. The expression of genes involved in antioxidant defense, inflammation, and lipid metabolism was determined in the liver, epididymal adipose tissue, proximal intestine, and skeletal muscle. Histological analysis was performed to identify crown-like structure (CLS) in epididymal fat pads to determine macrophage infiltration. RESULTS: No differences were noted between the control and any berry-fed groups in plasma levels of liver enzymes, insulin, glucose, ferric reducing antioxidant power, superoxide dismutase, and tumor necrosis factor ${\alpha}$. However, BK significantly lowered plasma triglyceride compared with the HF control and other berries, whereas BC significantly reduced F4/80 mRNA and the number of CLS in the epididymal fat pad, indicative of less macrophage infiltration. CONCLUSIONS: The present study provides evidence that BB, BK and BC with varying anthocyanin composition differentially affect plasma lipids and adipose macrophage infiltration in DIO mice, but with no differences in their antioxidant capacity and anti-inflammatory potential.

Elimination of Saturated Fatty Acids, Toxic Cyclic nonapeptide and Cyanogen Glycoside Components from Flax Seed Oil

  • Flax seed(Linseed, Linum usitatissimum L.) and its oil, a richest source of alpha-linolenic acid(ALA)(${\omega}-3$), contain saturated fatty acids, neurotoxic cyanogen glycosides and immuno-suppressive cyclic-nonapeptides. Present paper describes the development of two chemical processes, Process-A and -B, to remove saturated fatty acids and to destroy cyclic nonapeptides and cyanogen glycosides from flax seed oil. Process-A consists of three major steps, i.e., extraction of fatty acid mixture by alkaline saponification, removal of saturated fatty acid by urea-complexation, and triglyceride reconstruction of unsaturated fatty acid via fatty acyl-chloride activation using oxalyl chloride. Process-B consists of preparation of fatty acid ethyl ester by transesterification, elimination of saturated fatty acid ester by urea-complexation, and reconstruction of triglyceride by interesterification with glycerol-triacetate (triacetin). The destruction of lipophilic cyclic nonapeptide during saponification or transesterification processes could be demonstrated indirectly by the disappearance of antibacterial activity of bacitracin, an analogous cyclic-decapeptide. The cyanogen glycosides were found only in the dregs after hexane extraction, but not in the flax seed oil. The reconstructed triglyceride of flax seed oil, obtained by these two different pathways after elimination of saturated fatty acid and toxic components, showed agreeable properties as edible oil in terms of taste, acid value, iodine and peroxide value, glycerine content, and antioxidant activity.

Voltage Enhancement of ZnO Oxide Varistors for Various Y2O3 Doping Compositions

  • The microstructure and the electrical properties of a ZnO varistor, which was composed of a ZnO-$Bi_2O_3$-$Sb_2O_3$-CoO- $MnO_2$ -NiO-$Nd_2O_3$ system, were investigated at various $Y_2O_3$ addition concentrations. $Y_2O_3$ played a role in the inhibition of the grain growth. As the $Y_2O_3$ content increased, the average grain size decreased from $6.8{\mu}m$ to $4{\mu}m$, and the varistor voltage($V_{1mA}$) greatly increased from 275 to 400 V/mm. The nonlinearity coefficient ($\alpha$) decreased from 72 to 65 with increasing $Y_2O_3$ amount. On the other hand, the leakage current ($I_L$) increased from 0.2 to 0.9 ${\mu}A$. These results confirmed that doping the varistors with $Y_2O_3$ is a promising production route for production of a higher fine-grained varistor voltage ($V_{1mA}$) which can dramatically reduce the size of the varistors.

Effect of polishing and glazing on the color and spectral distribution of monolithic zirconia

  • PURPOSE. The aim of this study was to evaluate the effect of polishing and glazing on the color and spectral distribution of monolithic zirconia. MATERIALS AND METHODS. Forty-five monolithic zirconia specimens ($16.3mm{\times}16.4mm{\times}2.0mm$) were fabricated and divided into 5 groups according to the number of A2-coloring liquid applications (Group I to V). Each group was divided into 3 subgroups according to the method of surface treatments (n=3): N: no treatment; P: polishing; G: glazing. Color and spectral distribution of five different areas of each specimen were measured according to CIELAB color space in the reflectance mode relative to the standard illuminant D65 on a reflection spectrophotometer. Data were analyzed using one-way ANOVA followed by Tukey's HSD test, Pearson correlation and regression analysis (${\alpha}$=.05). RESULTS. There was a significant difference in CIE $L^*$ between Subgroup N and P, and in CIE $b^*$ between Subgroup P and G in each group. Spectral reflectance generally decreased in Subgroup P and G in comparison with Subgroup N. Color differences between Subgroup P and G were within the perceptibility threshold (${\Delta}E{^*}_{ab}$ < 3.7) in most groups. Highly significant correlation was found between CIE $b^*$ and each subgroups as the number of coloring liquid applications increased ($R^2$ >0.88, P<.001). CONCLUSION. A perceptible color difference can be detected after polishing of monolithic zirconia. Polishing decreases the lightness, and glazing also decreases the lightness, but increases the yellowness of monolithic zirconia.

SEOUL NATIONAL UNIVERSITY 4K×4K CAMERA (SNUCAM) FOR MAIDANAK OBSERVATORY

  • We present the characteristics of the Seoul National University 4k Camera (SNUCAM) and report its performance on the 1.5m telescope at the Maidanak observatory in Uzbekistan. SNUCAM is a CCD camera with a pixel scale of 0.266" in $4096{\times}4096$ format, covering $18.1'{\times}18.1'$ field of view on the 1.5m. The camera is currently equipped with Bessell UBVRI, $H{\alpha}$, SDSS ugriz, and Y-band filters, allowing us to carry out a variety of scientific programs ranging from exoplanet studies to survey of quasars at high redshift. We examine properties of SNUCAM such as the bias level and its temporal variation, the dark current, the readout noise, the gain, the linearity, the fringe patterns, the amplifier bias, and the bad pixels. From our observations, we also constructed the master fringe frames in I-, z-, and Y-band. We outline some of the current scientific programs being carried out with SNUCAM, and demonstrate that SNUCAM on the 1.5m can deliver excellent images that reach to the $5-{\sigma}$ detection limits of R~25.5 mag and z~22.7 mag in 1 hour total integration.

Purification and Physicochemical Characterization of a Recombinant Phospholipid Hydroperoxide Glutathione Peroxidase from Oryza sativa

  • Phospholipid hydroperoxide glutathione peroxidase (PHGPx) is an unique antioxidant enzyme that directly reduces lipid hydroperoxides in biomembranes. In the present work, the entire encoding region for Oryza sativa PHGPx was expressed in Escherichia coli M15, and the purified fusion protein showed a single band with 21.0 kD and pI = 8.5 on SDS- and IFE-PAGE, respectively. Judging from CD and fluorescence spectroscopy, this protein is considered to have a well-ordered structure with 12.2% $\alpha$-helix, 30.7%$\beta$-sheet, 18.5% $\delta$-turn, and 38.5% random coil. The optimum pH and temperature of the enzyme activity were pH 9.3 and 27$^{\circ}C$. The enzyme exhibited the highest affinity and catalytical efficiency to phospholipid hydroperoxide employing GSH or Trx as electron donor. Moreover, the protein displayed higher GSH-dependent activity towards t-Butyl-OOH and $H_2O_2$. These results show that OsPHGPx is an enzyme with broad specificity for hydroperoxide substrates and yielded significant insight into the physicochemical properties and the dynamics of OsPHGPx.

대맥종자의 Amylase 생성에 미치는 Gibberellic Acid의 촉진효과와 Indole-3-acetic Acid의 억제효과의 해석 (Studies on Gibberellic Acid-promoted and Indole-3-acetic Acid-repressed Amylase Synthesis of Barley Seeds)

  • Using barley seeds (Hordeum sativum Jess, var.), the influences of gibberellic acid (GA) and indole-3-acetic acid(IAA) on the amylase synthesis and that of the nucleic acid metabolism were investigated. 1. With the deembrynized barley seeds, the increase of amylase treated with a $10^{-5}M$ of GA and the decrease of amylase treated with $10^{-5}M$IAA were matched by a proportionate increase and decrease in the amount of RNA. The influence of the hormones on the RNA synthesis has appeared immediately after the treatment but on the amylase synthesis it has appeared 8 hours later. But no influence on the DNA synthesis was observed on both hormones. 2. The amylase from deembryonized barley seeds treated with GA and IAA have been fractionated by gel filteration on Sephadex G-100. The amylase components showed four fractions on both enzymes treated with GA and IAA. Fraction I(FI) was differed from fraction Ⅵ(FIV) in Km value and the effects of temperature, pH and metal ions. On the basis of their emzymatic properties, it was considered that the FI was $\beta$-amylase and FIV was $\alpha$-amylase. The influences of GA and IAA on each fractions appeared to be similar but on the amylase units per souble protein, IAA inhibited the production of amylase FIV while it promoted that of amylase FI. 3. An experiment was conducted to determine whether IAA inhibits GA-promoted amylase synthesis competitively or non-competitively. Using a Lineweaver-Burk plot, it was clear that IAA was acting in a non-competitive fashion. From this, IAA was probably not competing with GA at the same site, but it was acting at some other site which resutled in partial blocking of the action of GA on the amylase synthesis.
