혈당강하 기능 오미자 소스의 이화학적 품질 및 생리활성 (Physicochemical Quality and Hypoglycemic Effect of Omija Sauce)

  • 본 연구에서는 오미자 농축액 첨가량(0~5%)에 따른 소스의 품질특성과 혈당강하능 및 관능적 특성에 미치는 영향에 대해 조사하였다. 오미자 농축액 첨가량이 증가할수록 수분함량, 총 페놀함량 및 환원당은 증가하였고 pH는 감소하였다. 소스의 이화학 시험 결과 당도, 환원당, 총 페놀함량, ${\alpha}$-amylase 저해율 및 ${\alpha}$-glucosidase 저해율 등은 오미자 농축액의 첨가량에 따라 증가하였다. 소스의 색도는 오미자 농축액 첨가량이 증가함에 따라 명도는 감소하였고, 적색도, 황색도 및 ${\Delta}E$ 값은 증가하였다. 관능적 특성은 소스만 섭취 하였을 때 4% 첨가한 오미자 소스에서 가장 높은 기호도를 나타내었고, 닭 가슴살과 함께 섭취하였을 때 5% 첨가한 오미자 소스의 기호도가 가장 높았다.

  • Herb extracts commercially used in Korea were screened for PPAR-${\gamma}$ agonist test and ${\alpha}$-glucosidase inhibition assay. Total 16 herb plants had a PPAR-${\gamma}$ agonist activity. Specially, Alisma orientale Juz (108.41%), Ephedra sinica (98.22%), Sasa japonica Makino var. purpurascens Nakai (140.68%), Astragalus membranaceus Bunge (106.79%) and Cnidium officinale Makino (113.00%) showed high PPAR-${\gamma}$ agonist activity rate compared with rosiglitazone's (167.46%). And Cornus officinalis S. et Z. (90.3%), Cinnamomum cassia Blume (89.2%), Psoralea corylifolia L. (89.8%), Paeonia japonica (Makino) Miyabe (92.4%) and Paeonia suffruticosa Andr (93.2%), showed high ${\alpha}$-glucosidase inhibition rates. These results support previous reports of the efficacy of Oriental medicinal plants used for diabetes mellitus.

  • The major goal in the treatment of diabetes mellitus is to achieve near-normal glycemic control. To optimize both fasting blood glucose and postprandial glucose levels is important in keeping blood glucose levels as close to normal as possible. ${\alpha}-Glucosidase$ is the enzyme that digests dietary carbohydrate, and inhibition of this enzyme could suppress postprandial hyperglycemia. The purpose of this study was to test the inhibitory activity of methanol extract of Euonymus alatus on ${\alpha}-glucosidase$ in vitro and in vivo to evaluate its possible use as an anti-diabetic agent. Yeast ${\alpha}-glucosidase$ inhibitory activities of methanol extract of E. alatus were measured at concentrations of 0.50, 0.25, 0.10, and 0.05 mg/ml. The ability of E. alatus to lower postprandial glucose was studied in streptozotocin-induced diabetic rats. A starch solution (1 g/kg) with and without E. alatus extract (500 mg/kg) was administered to diabetic rats by gastric intubation after an overnight fast. Plasma glucose levels were measured at 30, 60, 90, 120, 180, and 240 min. Plasma glucose levels were expressed in increments from baseline, and incremental areas under the response curve were calculated. Extract of E. alatus, which had an $IC_{50}$ value of 0.272 mg/ml, inhibited yeast ${\alpha}-glucosidase$ activity in a concentration-dependent manner. A single oral dose of E. alatus extract significantly inhibited increases in blood glucose levels at 60 and 90 min (p<0.05) and significantly decreased incremental response areas under the glycemic response curve (p<0.05). These results suggest that E. alatus has an antihyperglycemic effect by inhibiting ${\alpha}-glucosidase$ activity in this animal model of diabetes mellitus.

자연발효 함초액의 이화학적 특성 및 생리활성 (Physicochemical Characteristics and Physiological Activities of Naturally Fermented Glasswort (Salicornia herbacea L.) Juice)

  • 본 연구는 함초를 이용한 응용제품의 하나로 자연발효된 함초발효액에 대한 기초자료를 제공하고자, 함초와 당을 혼합하여 2년 혹은 6년간 발효시킨 다음 여과하여 얻어진 여액을 상온에서 자연 발효시키며 함초발효액의 이화학적 특성 및 생리활성을 조사하였다. 함초발효액의 당 함량은 45%내외로 이는 당침출에 의한 제조 방법 때문이며, Na을 제외한 주요 무기질은 K, Mg 그리고 Ca으로, 그중 K 함량이 가장 높게 나타났다. 주요 유리아미노산은 alanine, proline, aspartic acid 그리고 lysine으로, 이들 일반성분들의 발효기간에 따른 차이는 거의 없었다. 총페놀성 화합물 함량은 약 50 CAE mg/100 mL이고, DPPH와 ABTS$^+$ radical-scavenging 활성은 ascorbic acid와 거의 유사할 정도의 활성을 보였다. 또한 함초발효액은 ACE 및 ${\alpha}$-glucosidase에 대해서도 저해 활성을 보였으며, 특히 ${\alpha}$-glucosidase 저해 활성은 장기 발효된 함초발효액(LFGJ)에서 좀 더 높은 활성을 나타냈다. 이에 장기 발효된 함초발효액(LFGJ)을 4%의 고염식이군 실험쥐(SD rat)에 급여한 결과, 일반적인 특성은 대조군(4% 고염식이군)과 차이가 없었으나, 함초발효액군(FGJ; 4% 고염식이+2% LFGJ)의 체중증가량이 통계적 유의성은 없었으나 더 낮은 경향을 보였고, 포도당 내성 실험 결과, 함초발효액군(FGJ)에서 포도당 내성이 더 개선되는 것으로 조사되었다.

  • 최근 들어 여주의 혈당개선의 약리학적 효능이 보고됨에 따라 기능성 채소로 각광받고 있다. 주로 여주 과실만이 식용으로 쓰이지만 여주씨 또한 다양한 가공과정을 거칠 경우(건여주 혹은 건조분말, 환, 엑기스 등) 필연적으로 함께 섭취되기도 한다. 본 연구에서는 과실이외의 특히, 연구결과가 미비한 여주씨에 대해 생리활성물질의 구명 및 혈당개선의 효과에 대한 연구를 수행하였다. 다양한 추출물 및 단일 활성성분의 혈당개선 in vitro검증법인 ${\alpha}$-glucosidase에 대한 저해효과를 측정하였고, 활성을 나타내는 추출물 또는 활성성분과의 상관관계를 구명하였다. 구체적으로는 95% 에탄올 추출물 및 비극성 용매 등에 다량 추출되는 생리활성물질인 ${\alpha}$-eleostearic acid는 우수한 저해효과를 나타내었다($IC_{50}=43.4{\mu}g/mL$). 또한 효소저해제의 함량을 HPLC를 통한 정량분석을 수행함으로써 여주씨에 함유된 기능성성분의 혈당개선 식 의약 소재로의 응용가능성을 연구하였다.

Cr3+ 또는 Selenium 첨가 배양액으로 재배한 치커리의 항산화활성 및 α-glucosidase저해효과 (Antioxidant activities and α-Glucosidase Inhibition Effects of Chicories Grown in Hydroponics Added with Cr3+ or Selenium)

  • $Cr^{3+}$ 및 Se을 첨가하여 수경 재배한 치커리 3종의 에탄올 추출물을 각각 제조하여 추출물별 항산화 활성을 비교 검토하였다. 그 결과 양액만으로 재배한 치커리보다 양액에 Se이나 $Cr^{3+}$을 첨가한 치커리 추출물에서 총 폴리페놀 함량, 총 플라보노이드 함량 및 FRAP 환원능이 증가하였으며, DPPH 라디칼 소거능 및 ABTS 라디칼 소거능도 증가하였다. 특히 Se을 첨가한 치커리보다 $Cr^{3+}$을 첨가한 치커리에서 더 우수한 항산화 활성을 나타내었다. 또한 $Cr^{3+}$을 첨가한 CLER 및 CLE의 경우 ${\alpha}$-glucosidase 저해활성이 있음을 알 수 있었다. 따라서 치커리 추출물의 수경재배 과정에서 양액에 $Cr^{3+}$, Se의 첨가는 치커리의 항산화 활성 및 ${\alpha}$-glucosidase 저해활성 증진에 영향을 주는 것으로 확인되었다.

The Inhibitory Activity of Polygonum Multiflorum Thunberg and its Effect on Postprandial Hyperglycemia in Streptozotocin-induced Diabetic Rats

The methanol extract of Polygonum multiflorum Thunberg was tested for inhibitoty activity against $\alpha$-glucosidase in vitro and in vivo. Polygonum multiflorum Thunberg extract inhibited yeast $\alpha$-glucosidase activity in a concentration-dependent manner. Polygonum multiflorum Thunberg showed an $IC_{50}$ value of 0.48 mg/mL. The ability of Polygonum multiflorum Thunberg extract to lower postprandial glucose was studied in streptozotocin-induced diabetic rats.   • Objectives: This study aimed to evaluate the hypoglycemic effects of an ethanol extract of Cassia abbreviata (ECA) bark and the possible mechanisms of its action in diabetic albino rats. Methods: ECA was prepared by soaking the powdered plant material in 70% ethanol. It was filtered and made solvent-free by evaporation on a rotary evaporator. Type 2 diabetes was induced in albino rats by injecting 35 mg/kg body weight (bw) of streptozotocin after having fed the rats a high-fat diet for 2 weeks. Diabetic rats were divided into ECA-150, ECA-300 and Metformin (MET)-180 groups, where the numbers are the doses in mg.kg.bw administered to the groups. Normal (NC) and diabetic (DC) controls were given distilled water. The animals had their fasting blood glucose levels and body weights determined every 7 days for 21 days. Oral glucose tolerance tests (OGTTs) were carried out in all animals at the beginning and the end of the experiment. Liver and kidney samples were harvested for glucose 6 phosphatase (G6Pase) and hexokinase activity analyses. Small intestines and diaphragms from normal rats were used for ${\alpha}-glucosidase$ and glucose uptake studies against the extract. Results: Two doses, 150 and 300 mg/kg bw, significantly reduced the fasting blood glucose levels in diabetic rats and helped them maintain normal body weights. The glucose level in DC rats significantly increased while their body weights decreased. The 150 mg/kg bw dose significantly increased hexokinase and decreased G6Pase activities in the liver and the kidneys. ECA inhibited ${\alpha}-glucosidase$ activity and promoted glucose uptake in the rats' hemi-diaphragms. Conclusion: This study revealed that ECA normalized blood glucose levels and body weights in type 2 diabetic rats. The normalization of the glucose levels may possibly be due to inhibition of ${\alpha}-glucosidase$, decreased G6Pase activity, increased hexokinase activity and improved glucose uptake by muscle tissues.

Water Chestnut (Trapa japonica Flerov.) Exerts Inhibitory Effect on Postprandial Glycemic Response in Rats and Free Radical Scavenging Activity in vitro

  • The ${\alpha}-glucosidase$ inhibitory and antioxidant effects of water chestnut (Trapa japonica Flerov.) were assessed to explore its possible use as an anti-diabetic agent. Methanol extracts of the fruit shell and meat of water chestnut were assayed for inhibitory activity against yeast ${\alpha}-glucosidase$ and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity. Effect of fruit shell extract on postprandial glucose response was assessed. Compared with fruit meat, shell extract showed stronger inhibition against ${\alpha}-glucosidase$ with an $IC_{50}$ of 273 ${\mu}g/mL$. Oral administration of fruit shell extract (500 mg/kg) significantly lowered the postprandial area under the glucose response curve to starch (1 g/kg) in streptozotocin (STZ)-induced diabetic rats (p<0.01). Compared with fruit meat, shell extract showed stronger scavenging activity against DPPH, with an $IC_{50}$ of 27.1 ${\mu}g/mL$. The results indicate that the fruit shell of water chestnut was effective in controlling postprandial hyperglycemia and exerted an antioxidant effect. Therefore, water chestnut may be useful in treating diabetes.

  • An inhibitory activity against ${\alpha}$-glucosidase was identified in extracts of medicinal lichens, Parmelia austrosinensis and P. praesorediosa. The extracts retained almost all of their original activities when treated with heat, acid and alkaline conditions, and some hydrolytic enzymes. Partially purified inhibitor showed strong inhibition against disaccharide hydrolytic enzymes of mammalian and mold origin, but weak or no inhibition against polysaccharide hydrolytic enzymes except glucoamylase. The inhibitors from the two Parmelia sp. showed almost same retention time in HPLC. The inhibitor suppressed elevation of blood glucose level in rats after oral administration of soluble starch or sucrose.
