The Preliminary Study on the Structure of Cop Protein by CD and NMR

  • Cop protein is the transcription repressor protein in rolling circle replication plasmid. With antisense RNA, Cop protein controls the copy number of plasmid. Cop family proteins have been found in various plasmids. Among Cop family proteins, Cop studied in this paper consists of 55 amino acids (Mw. 6,400), and was known to have trimer structure. Since no structural facts are elucidated, we have carried out preliminary experiments aimed at the elucidation of its three dimensional structure. The secondary structure of Cop is studied by CD and NMR. To solve the aggregation of Cop at high concentration, we tested various detergents and salts. The addition of detergents and salts could not solve the aggregation problem. However, we found that concentration is important in solving the aggregation problem. We knew that 0.18mM in 50mM potassium phosphate without any other ingredients is maximum concentration not to aggregate. Wa also investigated the pH dependence of Cop protein, and knew that Cop protein is more stable in acid state. At various temperatures, 15N-1H HSQC spectra were measured in order to find the optimal experimental condition. To enhance the peak resolution, 3D NOESY-HSQC spectrum is acquired. Since there are NOE peaks in the NH-NH region, we knew that Cop protein has $\alpha$-helical content, which was also confirmed by CD.

Synthetic Coprisin Analog Peptide, D-CopA3 has Antimicrobial Activity and Pro-Apoptotic Effects in Human Leukemia Cells

  • Recently, we reported that the synthetic Coprisin analog peptide 9-mer dimer CopA3 (consisted of all-L amino acid sequence) was designed based on a defensin-like peptide, Coprisin isolated from Copris tripartitus. The 9-mer dimer CopA3 (L-CopA3) had antibacterial activity and induced apoptosis in human leukemia cells via a caspase-independent pathway. In this study, all of amino acid sequences of L-CopA3 were modified to all D-form amino acids (DCopA3) to develop a more effective antimicrobial peptide. We investigated whether D-CopA3 had antimicrobial activities against pathogenic microorganisms and pro-apoptotic effects in human leukemia cells (U937, Jurkat, and AML-2). The synthetic peptide D-CopA3 had antimicrobial activities against various pathogenic bacteria and yeast fungus with MIC values in the 4~64 ${\mu}M$ range. Moreover, D-CopA3 caused cell growth inhibition, and increased the chromosomal DNA fragmentation and the expression of inflammatory cytokines, TNF-${\alpha}$ and IL1-${\beta}$, transcripts in human leukemia cells. The all-D amino acid peptide DCopA3 proved as effective as the L-CopA3 reported previously. These results provide the basis for developing D-CopA3 as a new antibiotic peptide.

Anticancer activity of CopA3 dimer peptide in human gastric cancer cells

  • CopA3 is a homodimeric ${\alpha}$-helical peptide derived from coprisin which is a defensin-like antimicrobial peptide that was identified from the dung beetle, Copris tripartitus. CopA3 has been reported to have anticancer activity against leukemia cancer cells. In the present study, we investigated the anticancer activity of CopA3 in human gastric cancer cells. CopA3 reduced cell viability and it was cytotoxic to gastric cancer cells in the MTS and LDH release assay, respectively. CopA3 was shown to induce necrotic cell death of the gastric cancer cells by flow cytometric analysis and acridine orange/ethidium bromide staining. CopA3-induced cell death was mediated by specific interactions with phosphatidylserine, a membrane component of cancer cells. Taken together, these data indicated that CopA3 mainly caused necrosis of gastric cancer cells, probably through interactions with phosphatidylserine, which suggests the potential utility of CopA3 as a cancer therapeutic. Effects of Fatigue Induction on Ground Reaction Force Components, Postural Stability, and Vertical Jump Performance in Taekwondo Athletes

  • Objective: The purpose of this study was to investigate the effect of fatigue induction on ground reaction force (GRF) components, postural stability, and vertical jump performance in Taekwondo athletes. Method: Ten Taekwondo athletes (5 men, 5 women; mean age, $22.30{\pm}2.62years$; mean height, $174.21{\pm}9.20cm$; mean body weight, $67.28{\pm}12.56kg$) participated in this study. Fatigue was induced by a short period of strenuous exercise performed on a motorized treadmill. The analyzed variables included vertical jump performance, static stability (mediolateral [ML], center of pressure [COP], anteroposterior [AP] COP, ${\Delta}COPx$, ${\Delta}COPy$, and COP area), postural stability index values (ML stability index [MLSI], AP stability index [APSI], vertical stability index [VSI], dynamic postural stability index [DPSI]), and GRF components (ML force, AP force, peak vertical force [PVF], and loading rate). To analyze the variables measured in this study, PASW version 22.0 was used to calculate the mean and standard deviation, while a paired t-test was used to evaluate the pre- versus post-fatigue results. Pearson's correlation coefficients among variables were also analyzed. The statistical significance level was set at ${\alpha}$ = .05. Results: Vertical jump performance decreased significantly after the induction of fatigue, while AP COP, ${\Delta}COPx$, COP area, APSI, VSI, and DPSI increased significantly. PVF and loading rate increased significantly after the induction of fatigue, while the postural stability variables (AP COP, ${\Delta}COPy$, COP area, APSI, VSI, DPSI) were similarly correlated with GRF components (PVF, loading rate) after fatigue was achieved (r = .600, $R^2$ = 37%). Conclusion: These results suggest that the induction of fatigue can decrease postural stability and exercise performance of Taekwondo athletes during training and competition sessions.

Effect of uterine histotroph on embryo development in pigs

  • The aim of this study was to investigate the effect of uterine histotroph on embryo development and the expression of cysteine-rich protein 2 (CRP2), coatomer subunit gamma-2 (G2COP), myoglobin (MYG), vascular endothelial growth factor D (VEGFD), collagen alpha 4 chain (COL4) and galactoside 3-L-fucosyltransferase 4 (FUT4) proteins in porcine embryo during pre-implantation. Uterine histotroph (UH) was collected from uterine horn on corpus albican phase, and embryos were cultured in porcine zygote medium with UH for 168 hours. Cleavage and blastocyst formation of embryo were detected at 168 hours after in vitro fertilization. And CRP2, G2COP, MYG, VEGFD, COL4 and FUT4 proteins were observed using confocal laser microscope. In results, embryo cleavage rate was not significantly changed by UH, but blastocyst rate was significantly (P<0.05) decreased in UH-treated embryos. Moreover, CRP2, G2COP, MYG, VEGFD, COL4 and FUT4 proteins were expressed in blastomere. CRP2 in embryo was significantly overexpressed (P<0.05), but not G2COP, MYG, VEGFD, COL4 and FUT4 proteins. In summary, UH on corpus albican phase was increased CRP2 protein in embryo, and inhibited blastocyst formation in preimplantation porcine embryos, suggesting that CRP2 may play an interrupter on embryo development in pigs.

  • COPI vesicles are essential to the retrograde transport of proteins in the early secretory pathway. The COPI coatomer complex consists of seven subunits, termed ${\alpha}-$, ${\beta}-$, ${\beta}^{\prime}-$, ${\gamma}-$, ${\delta}-$, ${\varepsilon}-$, and ${\zeta}$-COP, in yeast and mammals. Plant genomes have homologs of these subunits, but the essentiality of their cellular functions has hampered the functional characterization of the subunit genes in plants. Here we have employed virus-induced gene silencing (VIGS) and dexamethasone (DEX)-inducible RNAi of the COPI subunit genes to study the in vivo functions of the COPI coatomer complex in plants. The ${\beta}^{\prime}-$, ${\gamma}-$, and ${\delta}$-COP subunits localized to the Golgi as GFP-fusion proteins and interacted with each other in the Golgi. Silencing of ${\beta}^{\prime}-$, ${\gamma}-$, and ${\delta}$-COP by VIGS resulted in growth arrest and acute plant death in Nicotiana benthamiana, with the affected leaf cells exhibiting morphological markers of programmed cell death. Depletion of the COPI subunits resulted in disruption of the Golgi structure and accumulation of autolysosome-like structures in earlier stages of gene silencing. In tobacco BY-2 cells, DEX-inducible RNAi of ${\beta}^{\prime}$-COP caused aberrant cell plate formation during cytokinesis. Collectively, these results suggest that COPI vesicles are essential to plant growth and survival by maintaining the Golgi apparatus and modulating cell plate formation.

  • Allele rescue and sequence analysis of soo1-1 allele in Saccharomyces cerevisiae mutant LP0353 revealed that soo1-1 is identical to the previously reported ret1-1 allele, which has a base substitution of A for $G^{681}$ leading to an amino acid substitution of aspartic acid for $glycine^{227}$ in Soolp. However, it was revealed that the addition of osmotic stabilizer, such as 1.2M sorbitol can rescue the temperature sensitive phenotype of the ret1-1 mutant and that the soo1-1/ret1-1 mutation may confer defects in post-translational modification of proteins involved in the yeast cell wall biogenesis. Evidence for a putative role of 5th WD40 domain of the Soo1p/$\alpha$-COP in the construction and maintenance of cell walls was also presented by complementation test with deletion constructs of the SOOl.

Design Effect of Different Components and Economic Evaluation of an Adsorption Chiller on the System Performance

  • A conventional silica gel/water adsorption chiller has been analyzed numerically. A novel non-dimensional mathematical model has been presented to analyze the design effect of different components of an adsorption chiller. The design parameters of this system are characterized by the number of transfer unit, NTU, of different components and the inert material alpha number, ${\alpha}$of different components of the systems. Results show that condenser NTU$\sub$a/ has the most influential effect on the system performance, which is fellowed by absorber NTU$\sub$e/. It is also seen that coefficient of performance (COP) and non-dimensional specific cooling capacity increases with the increase of NTU$\sub$a/ and NTU$\sub$e/, but decreases with the increase of inert material alpha number. A thermo-economic data of the adsorption chiller and some other heat pump systems those are in practical operation are also presented.

  • The performance and applicability to food concentration of heat pump were investigated. During heating the water of spa from $43^{\circ}C$ to $51^{\circ}C$, COP's of heat pump (R-12, 150 HP) were 4.03 at heating part and 3.5 at cooling part. And, the efficiency of compressor (${\alpha}$) was 0.477 While the city water was heated to $39^{\circ}C$ by heat pump (R-22, 10 HP), its COP's were 3.0 at heating part and 1.87 at cooling part. During concentrations sucrose solution by centrifugal evaporator (ALFA-LAVAL CO, CTIB) with heat pump, heat capacity for condensating water vapor was required greater 15% than the latent heat for concentrating and then the overall heat transfer coefficient was $1196\;Kcal/m^{2}.\;h.\;^{\circ}C$. When low temperature concentration ($30-35^{\circ}C$, 28-40 Torr) of garlic extract was carried out by the water of $60^{\circ}C$ and $15^{\circ}C$ adjusted by heat pump, the ratio of heat capacity for concentrating vs. that for condensating of water vapor was 0.961.

  • This study is intended to identify how quick disconnect coupling which connects with refrigerant piping of air-conditioner using R-22 refrigerant has effect on characteristics of flux. in the case where the air-conditioner installs utilizes quick disconnect coupling, COP has an effect on the quantity of cooling load because of changing flow rate and physical properties of refrigerant which flow into an entrance of expansion valve from coupling. Variation of flow rate can be regulated by changing expansion-contraction angle; $\alpha$ of an entrance and an exit of coupling.
