In this study, we investigated the inhibitory effect of four solvent fractions of Alnus firma on ${\alpha}-amylase$, ${\alpha}-glucosidase$ and aldose reductase activities. The inhibitory test showed that methanol (MeOH) extract and hexane (HX) fraction strongly inhibited pork pancreatin and salivary ${\alpha}-amylase$ activity. The MeOH extract and HX fraction of Alnus firma at the concentration of 4 mg/ml inhibited more than 70% of pancreatin and salivary ${\alpha}-amylase$ activity. The inhibitory effect of fractions has different specificities against ${\alpha}-amylase$ from pancreatin and salivary. In addition, the MeOH extract and butanol (BuOH) fraction showed the highest inhibitory activity on yeast ${\alpha}-glucosidase$ at values of $IC_{50}$$137.36\;{\mu}g/ml$ and $115.14\;{\mu}g/ml$ respectively. The MeOH extract and BuOH fraction showed the highest inhibitory activity on yeast ${\alpha}-glucosidase$ than commercial agent such as 1-deoxynorjirimycin and acarbose. Inhibition kinetics of solvent fractions showed that ${\alpha}-glucosidase$ has been inhibited noncompetitively by the MeOH, EA and BuOH fraction. The aldose reductase from human muscle cell had been inhibited strongly by the MeOH extract and EA fraction at 57.996% and 83.293% at the concentration of $50\;{\mu}g/ml$, respectively. These findings may contribute to biological significance in that ${\alpha}-amylase$, ${\alpha}-glucosidase$ and aldose reductase inhibitory compounds could be used as a functional food and a drug for the symptomatic treatment of antidiabetic disease in the future.
We investigated the biological activities of ethanol extracts obtained from five endive cultivars. Free radical-scavenging activities of extracts from Redoria, Halifax, Vintor, Kibora, and Metafora cultivars were 94.2%, 60.8%, 66.6%, 70.0% and 67.4%, respectively. The extract from Redoria showed the highest activity among all the tested samples including BHT (Butylated hydroxyanisole) and vitamine C, known as antioxidant. Endive extracts slightly (17.5 - 38.6%) inhibited $\alpha$-glucosidase activity. However, there is no inhibitory activity against $\alpha$-amylase. In terms of proteolytic activity, all endive extracts showed strong activity than pancreatin (used as a positive control), and the extract of Redoria also exhibited stronger activity than other extracts as well as free radical scavenging activity. These results indicate that endive, especially the Redoria cultivar, can be used as a natural resource for development of antioxidants or materials aiding digestion.
The biological activities of water extracts from different fruit parts, such as peel, flesh, and placenta, of oriental melon were investigated. The concentrations of total phenolic in fruit extracts were $816.37\;{\mu}g/ml$, $385.23\;{\mu}g/ml$, and $925.56\;{\mu}g/ml$, respectively. Whereas the total flavonoid content in the peel extracts was $231.21\;{\mu}g/ml$, those in the extract of flesh and placenta were $8.16\;{\mu}g/ml$ and $36.07\;{\mu}g/ml$, respectively. The DPPH free radical scavenging activity of each fruit extract at 10,000 ppm was 34.84% for peel, 10.70% for placenta and 9.26% flesh. The ABTS radical cation decolorizing activity of each fruit extract at 10,000 ppm was in fruit extracts were 72.92% for peel, 48.0% for flesh and 74.31% for placenta. In addition, xanthine oxidase inhibitory activity, ${\alpha}-Glucosidase$ inhibition activity, and tyrosinase inhibition activity of the peel extracts appeared to be higher than those of placenta and flesh. Taken together, these results indicated that the peel part of oriental melon contained higher level of total flavonoid content, and several physiological activities including antioxidation, ${\alpha}-Glucosidase$ inhibition activity, and tyrosinase inhibition activity than did the flesh and placenta parts, and suggested that the peel might have a potential to be applicable as a source for functional foods.
To evaluate the anti-oxidant and anti-tumor potential of the green pepper (Capsicum annuum L. cv. DangZo), total polyphenol content, radical scavenging activities and anti-tumor properties were measured. The total polyphenol content of the 70% ethanol extracts from green pepper (Capsicum annuum L. cv. DangZo) was 30.29 mg gallic acid equivalent/g extract. The DPPH radical and hydroxyl radical scavenging activities of 70% ethanol extracts of green pepper (Capsicum annuum L. cv. DangZo) were documented at 2.87 and 10.55, respectively. For ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibitory activity, 70% ethanol extracts of green pepper (Capsicum annuum L. cv. DangZo) were documented at 35.67% and 58.41% respectively. The green pepper (Capsicum annuum L. cv. DangZo) demonstrated greater capability in terms of anti-neoplastic activity vis-a-vis colon cancer cell lines when compared to other cancer cell lines.s. er (Capsicum annuum L. cv. This study was conducted to investigate changes in the proximate composition, antioxidant activities, and ${\alpha}$-glucosidase inhibitory activity of Opuntia ficus-indica (OFI) cladodes cultivated in Jeju (JJ1, JJ2, JJ3) and Jeonnam (JN1, JN2). The difference in the proximate composition (crude protein, lipid and ash content) of OFI between the two regions was not significant. Ca, Mg and Na were the major mineral components of OFI. The ascorbic acid content of OFI ranged from 57.87 to 143.72 mg/100 g. A 70% ethanol extract was used to investigate the antioxidant content and activity as well as the ${\alpha}$-glucosidase inhibitory activity. The total polyphenol and flavonoid contents of OFI were 38.69~55.29 and 3.33~4.03 mg/g, respectively. The antioxidant activities based on the DPPH and ABTS free radical scavenging assays were 45.19~61.52% and 39.15~51.96%, respectively, at a concentration of 1 mg/mL. The inhibitory activity of OFI extracts against rat intestinal ${\alpha}$-glucosidase was 29.72~45.73% at 1 mg/mL concentration, and JN1 showed the highest ${\alpha}$-glucosidase inhibitory activity. This information could be very useful for authentication of Opuntia species with the highest potential as sources of nutritional and therapeutic elements.
Currently many studies aimed at enhancing efficacy of medicinal food on biological activity using bioconversion technology including fermentation process. In this study, the quality characteristics and antioxidative activity of fermented Crataegi fructus was investigated. The antioxidant activity of fermented Crataegi Fructus was assessed by various radical scavenging assays using DPPH (2,2-Diphenyl-1-picrylhydrazyl), FRAP (Ferric ion reducing antioxidant power), Reducing power and ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)). Moisture content of fermented Crataegi Fructus was $39.3{\pm}0.06%$. Contents of crude ash, crude protein, and crude fat were $0.20{\pm}0.01$, $1.77{\pm}0.04$, and $1.40{\pm}0.59%$, respectively. Moreover, the hunter's color values of fermented Crataegi Fructus were 79.24 (lightnees), 1.58 (redness), and 31.25 (yellowness), respectively. Total phenolic contents of fermented Crataegi Fructus were $3,015{\pm}250$ GAE ${\mu}g/g$. The antioxidative activities of fermented Crataegi Fructus significantly increased in a dose dependent manner. In addition, fermented Crataegi Fructus slightly (10.4%) inhibited ${\alpha}$-glucosidase activity; however, there was no inhibitory activity against ${\alpha}$-amylase. In terms of proteolytic activity, fermented Crataegi Fructus showed a strong activity than pancreatin (used as a positive control). These results indicate that fermented Crataegi Fructus can be used as a natural resource for material aiding digestion.
The $\beta$-glucosidase enzyme was purified from E. coli carrying Cellulomonas fimi $\beta$-glucosidase gene. SDS-PAGE and analytical gel filtration revealed that molecular weight of this enzyme was 56,000 dalton and consisted of a single polypeptide.Inhibition caused by heavy metals and activation by dithiothreitol suggest the existence of essential thiol group in the enzyme. The enzyme was not active on maltose (glucose $\alpha$-1,4-glucose) which has a $\alpha$-linkage, whereas it was active on lactose (glucose $\beta$-1,4-glucose), PNPG (p-nitrophenyl $\beta$-D-glucopyranoside) and PNPC (p-nitrophenyl $\beta$-D-cellobioside), although its reaction rates were different.
For development of new high-value Korean traditional fermented food by using bioactive fungi and bacteria, Nuruk, Makgeolli and Cheonggukjang were prepared by mold, yeasts and bacteria from Korean traditional fermented foods and their physiological functionalities were investigated. Aspergillus oryzae N152-1 Nuruk showed the highest antihypertensive angiotensin I-converting enzyme inhibitory activity(57.2%), and Makgeolli made by Saccharomyces cerevisiae Y111-5 and commercial JS Ipguk (solid cultures of saccharifying enzyme-producing mold) was showed 42.0% of anti-obesity ${\alpha}$-glucosidase inhibitory activity. Among various Cheonggukjang, No 463 Cheonggukjang made by Brevibacterium iodinum NCDO 613(T) was showed the highest fibrinolytic activity (size of clear zone: 28 mm) and good anti-obesity ${\alpha}$-glucosidase inhibitory activity.

This study investigated the enzyme inhibitory effects of ethanol extracts from the different parts of rough rice (Oryza sativar L.) from its germination period. Rough rice was germinated at $37^{\circ}C$ for 6 days, then separated into hull+sprout and brown rice. ${\alpha}$-Glucosidase inhibitory activity had the highest value (39.38%) in hull+sprout extracts after 5 days of germination. ${\alpha}$-Amylase and DPP-4 inhibitory activity had the highest values (75.32% and 47.77%, respectively) in hull+sprout extracts after germination for 5 days. ACE inhibitory activity of hull+ sprout extracts increased from 43.16% at the beginning to 58.60% at 5 days, while brown rice extracts increased this activity from 0.88% at the beginning to 14.50% at 4 days. The xanthine oxidase inhibitory activity of hull+ sprout extracts increased from 62.02% at the beginning to 64.49% at one day, and then decreased. Lipase inhibitory activity had its highest value (55.17%) in hull+sprout extracts after germination for 5 days. These results indicate that the optimal germination period for increasing enzyme inhibitory activities may be 5 days, and that hull+sprout extracts have a higher enzyme inhibition activity than brown rice.

Hemp seed (Cannabis sativa L.; HS), an annual herbaceous plant in the Cannabis genus, has been reported to play various biological functions in immunity increase, atherosclerosis, constipation, hyperlipidemia prevention, anti-inflammatory, and anti-cancer. In recently years, as superfood, the growing interest in the health care benefits of hemp seed has led to increased consumption. In this study, we investigated the effect of an ethanol extract of HS fermented with lactic acid bacteria (Lactobacillus plantarum KCTC 3107, L. plantarum KCTC 3108, L. brevis BHN-LAB128, L. paracasei BHN-LAB129). An antibacterial activity against Staphylococcus aureus and Bacillus cereus were 13.99 mm and 15.17 mm, respectively. The ethanol extracts of fermented hemp seed by lactic acid bacteria that the contents of total polyphenol, total flavonoid content, DPPH radical scavenging activity, SOD-like activity, and ${\alpha}$-glucosidase inhibitory activity were increased compared to non-fermented hemp seed. Also, tyrosinase inhibitory activity of the fermented hemp seed (FHS), known to melanin increasing substance was increased. In these results, we suggested that FHS have effects of anti-oxidant, ${\alpha}$-glucosidase inhibitory activity, and tyrosinase inhibitory activity. Hence, we proposed that FHS has possible to development as functional foods and cosmetics.
