Inhibitory Effects of Four Solvent Fractions of Alnus firma on α-Amylase and α-Glucosidase. (사방오리나무 추출물의 α-amylase 및 α-glucosidase 저해활성)

  • In this study, we investigated the inhibitory effect of four solvent fractions of Alnus firma on ${\alpha}-amylase$, ${\alpha}-glucosidase$ and aldose reductase activities. The inhibitory test showed that methanol (MeOH) extract and hexane (HX) fraction strongly inhibited pork pancreatin and salivary ${\alpha}-amylase$ activity. The MeOH extract and HX fraction of Alnus firma at the concentration of 4 mg/ml inhibited more than 70% of pancreatin and salivary ${\alpha}-amylase$ activity. The inhibitory effect of fractions has different specificities against ${\alpha}-amylase$ from pancreatin and salivary. In addition, the MeOH extract and butanol (BuOH) fraction showed the highest inhibitory activity on yeast ${\alpha}-glucosidase$ at values of $IC_{50}$ $137.36\;{\mu}g/ml$ and $115.14\;{\mu}g/ml$ respectively. The MeOH extract and BuOH fraction showed the highest inhibitory activity on yeast ${\alpha}-glucosidase$ than commercial agent such as 1-deoxynorjirimycin and acarbose. Inhibition kinetics of solvent fractions showed that ${\alpha}-glucosidase$ has been inhibited noncompetitively by the MeOH, EA and BuOH fraction. The aldose reductase from human muscle cell had been inhibited strongly by the MeOH extract and EA fraction at 57.996% and 83.293% at the concentration of $50\;{\mu}g/ml$, respectively. These findings may contribute to biological significance in that ${\alpha}-amylase$, ${\alpha}-glucosidase$ and aldose reductase inhibitory compounds could be used as a functional food and a drug for the symptomatic treatment of antidiabetic disease in the future.

Isolation and Characterization of $\alpha$-Amylase Producing Bacillus sp. AIV 1940 and Properties of Starch Synthetic Wastewater Degradation ($\alpha$-Amylase 생성균주 Bacillus sp. AIV 1940의 분리, 특성 및 합성폐수분해능)

  • $\alpha$-Amylase producing bacteria were isolated from activated sludge of corn processing wastewater plant and paddy field soil samples and selected by the direct iodine reaction. The isolate was identified as Bacillus sp. after morphology, API system and fatty acid analyses. To enchance $\alpha$-amylase productivity, a successive mutation of Bacillus sp. AIV 19 was performed using the treatment of nitrosoguanidine(NTG).The mutant, Bacillus sp. AIV 1940, showed about 1.8-fold level of amylase activity compared with parental strain. The isolate was Gram-positive and rod (2.8-3.0 $\mu$m long, 0.5-0.6 $\mu$m wide) type. The strain increased the bacterial mass at 3000 mg/l starch concentration. Organic substance removal rate was 40.2, 72.3% respectively after 1 and 3 day reaction using starch synthetic wastewater (intial CODcr was 4,455 mg/l).

Physiological Characteristics and Anti-Obesity Effect of Lactobacillus plantarum K6 isolated from Kimchi (김치에서 분리한 Lactobacillus plantarum K6의 생리적 특성 및 비만억제효과)

  • This study aimed to investigate the physiological characteristics and anti-obesity effects of a newly isolated bacterium, Lactobacillus plantarum K6. L. plantarum K6 showed good ${\alpha}-amylase$ inhibitory activity ($96.78{\pm}3.29%$), ${\alpha}-glucosidase$ inhibitory activity ($92.55{\pm}9.62%$), and lipase inhibitory activity ($85.17{\pm}0.79%$), and the strain inhibited the adipocyte differentiation of 3T3-L1 cells ($27.4{\pm}1.4%$) when present at a concentration of $100{\mu}g/mL$. L. plantarum K6 was isolated from kimchi and its physiological characteristics were investigated. A comparison of the sensitivity of the isolate to 15 different antibiotics showed that L. plantarum K6 is highly sensitive to erythromycin and highly resistant to vancomycin, ampicillin, and polymyxin B. This strain also showed high arylamidase and ${\beta}-galactosidase$ activities. Moreover, it was relatively tolerant to bile acid and low pH, and displayed resistance to Escherichia coli, Salmonella Typhimurium, Listeria monocytogenes, and Staphylococcus aureus, with rates of 51.8%, 42.4%, 61.6%, and 54.9%, respectively. No bio genic amines were produced. L. plantarum K6 also showed high adhesion activity to HT-29 cells compared to L. rhamnosus GG. These results demonstrate that Lactobacillus plantarum K6 has potential as a probiotic with anti-obesity effects.

Antioxidant and Antidiabetic Activities of Eucommia ulmoides Bark

  • Eucommia ulmoides bark extracts by cold water, boiling water, 100% EtOH, 70% EtOH, 100% MeOH, 70% MeOH and $CHCl_3$ were assayed for their medicinal effects. The antioxidant activity of the extracts ranged from $IC_{50}$ 125.2 to $IC_{50}\;872.7{\mu}g/ml$ in the 1,1-diphenyl-2-picrylhydrazyl (DDPH) free radical-scavenging assay, and cold water extracts had the highest antioxidant activity. $CHCl_3$ extracts had the highest inhibitory effect on angiotensin I-converting enzyme (ACE) giving inhibition of up to 56.4% at a concentration of 1 mg/ml. Extracts in 100% EtOH had the greatest inhibitory effect on $\acute{a}-amylase$ activity ($IC_{50}=174.6{\mu}g/ml$), and 70% MeOH extracts had the greatest inhibitory effect on ${\alpha}-glucosidase$ activity ($IC_{50}=14.0{\mu}g/ml$). Taken together, these results provided the in vitro evidence on the ACE, amylase and glucosidase inhibitory actions of E. ulmoides bark that form the pharmacological basis for its antihypertensive and antidiabetic action.

Inhibitory effects of extract from the Schizandra chinensis on rat small intestinal ${\alpha}-amylase$ activity and postprandial blood glucose

  • Postprandial hyperglycemia plays an important role in the development of type 2 diabetes and complications associated with the disease such micro-and macro-vascular disease. The present study investigated the effect and action mechanism of a ethanolic extract from the Schizandra chinensis(SC-E) on hypeglycemia in vivo and in vitro. In vitro, SE-E demonstrated a potent inhibitory effects on ${\alpha}-amylase$ activity ($IC_{50}$ : 4 ${\mu}g/ml$). Its inhibition on ${\alpha}-amylase$ was determined to be competitive type. Oral administration of SE-E markedly lowered plasma glucose levels in non-fasted streptozotocin induced diabetic rats (45 mg/kg BW). In addition when it was orally administrated to rats with starch (2g/kg BW), SC-E (50 and 100 mg/kg BW) significantly suppressed the increase of blood glucose levels after starch loading . These results suggest that some edible plants merit further evaluation for clinical usefulness as anti-diabetic drugs.

Hydrolysis Characteristics of Amylase from Alkaline-Tolerant Bacillus sp. on the Raw Starch (알칼리 내성 Bacillus sp.가 생산하는 Amylase의 생전분 분해 특성)

  • The raw starch hydrolysis by amylase prepared from alkaline-tolerant Bacillus sp. were investigated. Degree of hydrolysis(%) of 5%(w/v) raw rice, corn and potato starch by this enzyme were about 40, 25 and 20%, respectively. The hydrolysis action on raw starch by change of blue value was similar to the action pattern of exo ${\beta}$-amylase. The hydrolysis products of rice starch were mainly glucose and maltose. Oligosaccarides were also detected. From the above results, this enzyme was considered as exo type ${\alpha}$-amylase. This enzyme activity on the raw starch and the gelatinized starch were 28.40 and 86.60 IU/mg protein, respectively, and the ratio of raw starch-digesting activity to gelatinized starch-digesting activity (raw starch digestivity) was about 32%. The Km values for the raw and the gelatinized starch were 4.22 and 3.0mg/mL, respectively, and the VmaX values were 0.20 and 0.31mg/mL/min, respectively.

Physiobiochemical Characteristics of Hybrid Rice (1대 잡종벼의 생리생화학적 특성)

  • This study was conducted to determine physiobiochemical basis of heterosis using rice hybrids such as Shanyou 63 (Zhenshan 97As Minhui 63) and Teyou 63 (Longtepu A Minhui 63) as compared with inbred rice like Milyang 23. Seed protein patterns of rice hybrid showed complementary genetic characteristics inherited from their parents. Hybrid rice had larger embryo and higher $\alpha$-amylase activity than those of inbred rice. The larger embryo of hybrid was significantly correlated with tillering ability and high number of low node tillers jplant increased by 60~70% in Shanyou 63, leading to higher productive tillers/plant which directly influenced on grain yield of hybrid rice. These characters were further supported by high chlorophyll content in hybrids. Exogenous application of GA$_3$ (0.02 ppm) on inbred rice like Milyang 23, increased significantly $\alpha$-amylase activity, but no effect of GA$_3$ on hybrid rice was observed, indicating that sufficient amount of GA$_3$ is endogenously present in hybrid rice, showing 1 to 3.5 fold higher activity of $\alpha$-amylase in hybrid rice, which trigger heterosis from the germinating stage. Further, activity of cytochrome c oxidase was 2.66 to 5.52 fold higher in hybrid rice than that of inbred rice, indicating that rice hybrids have very active metabolism reflecting vigorous growth starting from the germinating stage, in turn leading to higher tillering ability.

  • The objective of this research was to evaluate the inhibitory activities of phenolic compounds isolated from Crataegi Fructus on ${\alpha}-amylase$ and ${\alpha}-glucosidase$. The content of total phenolic compounds of water extract from Crataegi Fructus was 22.5 mg/g. The inhibitory activity of the water extract (200 ${\mu}g/ml$) from Crataegi Fructus on ${\alpha}-amylase$ and ${\alpha}-glucosidase$ was determined to be 100% and 82.6%, respectively. Isolation of inhibitory compounds was carried out on Sephadex LH-20 and MCI-gel CHP-20 column chromatography using a gradient elution procedure of increasing MeOH in $H_2O$. The chemical structure of the inhibitory compound against ${\alpha}-amylase$ and ${\alpha}-glucosidase$ was confirmed as chlorogenic acid by spectroscopic analysis of FAB-MS, NMR and IR spectrum.

Anti-Oxidative and Anti-Proliferative Effect of 70% Ethanol Extracts from Green Pepper (Capsicum annuum L. cv. DangZo) (당조고추 70% 에탄올추출물의 항산화 및 항암활성측정)

  • To evaluate the anti-oxidant and anti-tumor potential of the green pepper (Capsicum annuum L. cv. DangZo), total polyphenol content, radical scavenging activities and anti-tumor properties were measured. The total polyphenol content of the 70% ethanol extracts from green pepper (Capsicum annuum L. cv. DangZo) was 30.29 mg gallic acid equivalent/g extract. The DPPH radical and hydroxyl radical scavenging activities of 70% ethanol extracts of green pepper (Capsicum annuum L. cv. DangZo) were documented at 2.87 and 10.55, respectively. For ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibitory activity, 70% ethanol extracts of green pepper (Capsicum annuum L. cv. DangZo) were documented at 35.67% and 58.41% respectively. The green pepper (Capsicum annuum L. cv. DangZo) demonstrated greater capability in terms of anti-neoplastic activity vis-a-vis colon cancer cell lines when compared to other cancer cell lines.s. er (Capsicum annuum L. cv. DangZo) higher activities of anticancer activities on colon cancer cell lines compared to other cancer cell lines.

Hypoglycemic Activity of the Hexane Extract of Oriental Melon (Cucumis melo L. var. makuwa Makino) Seeds and Its Active Compounds

  • The aim of this work is to evaluate the potential of oriental melon (Cucumis melo L. var. makuwa Makino) seeds for the management of type 2 diabetes by controlling glucose absorption. The ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibitory effects of the hexane extracts from oriental melon seeds were investigated. A bioassay-guided fractionation technique was used to elucidate the principal active components. The results show that the hexane extract from oriental melon seeds exhibited high inhibitory activities against ${\alpha}$-glucosidase and ${\alpha}$-amylase. The hexane extract was further fractionated into four sub-fractions. Among them, the sub-fraction F-1 exhibited the most potent anti-diabetic effect. The active components were isolated and identified by gas chromatography/mass spectroscopy (GC-MS). Free fatty acids showed significant hypoglycemic activity (p < 0.001) and fatty acid composition influenced enzyme inhibitory activities. These results suggest that oriental melon seeds could be used to prevent type 2 diabetes.
