  • Deuterium NMR studies have been carried out for two kinds of main- chain dimer liquid crystals $\alpha$.$\omega$-bis[(4,4`-cyanobipheny0oxy] alkane (CBA-n, n=9,100.The H-NMR spectra were recorded on a JEOP JNM-GSX-500 spectrometer by using deuterium labelled CBA-n at various temperatures. The RIS analysis of the NMR spectra was performed so as to elucidate the conformational characteristics of the spacer in the nematic phase. Following the previous treatment, the single-ordering-matrix model was adopted, in which the molecular axis was defined parallel to the line connecting the centers of the terminal mesogenic cores. Conformer fractions of the spacer were estimated by simulation so as to reproduce the observed NMR profile. The conformational entropy changes at both CN and NI interphases were estimated on the basis of the nematic conformations taken from the conformation map as well as those derived from the simulation. In these calculations the spacer was assumed th by in the all-trans conformation and in the random coil stats in the crystal and isotropic phases respectively. The esimated conformational entropy change values were then compared with the corresponding constant-volume entropies obtained from PVT measurements. The correspondence between both entropy values was found to be quite good in consideration of the uncertainties involved in both experiment and calculations.

Metabolic Discrimination of Rice Cultivars and Relative Quantification of Major Sugar Compounds Using 1H NMR Spectroscopy Combined by Multivariate Statistical Analysis (1H NMR 스펙트럼 데이터의 다변량 통계분석에 의한 벼 품종의 구분 및 주요 당 화합물의 정량분석)

  • Discrimination of 5 rice cultivars (Sangjubyeo , Dongjinbyeo Simbaekbyeo , Hwamanbyeo , and Simbaek-hetero ) using metabolic profiling was carried out. Whole cell extracts from each cultivar were subjected to $^1H$ NMR spectroscopy. When spectral data were analyzed by principal component analysis, 5 cultivars were clustered into 3 groups: SJ, DJ + SB, and HM + SH. Thecultivars showed great difference in carbohydrate region of $^1H$ NMR spectra, suggesting that qualitative and quantitative differences in carbohydrate compounds play a major role in discrimination of the cultivars. In addition, it was readily possible to determine relative quantification of major carbohydrates including sucrose, glucose, maltose from spectral data of the cultivars. SJ showed 2 to 4 times higher content of maltose than the other rice cultivars. Overall results indicate that metabolic discrimination of rice cultivars using $^1H$ NMR spectroscopy combined by multivariate statistical analysis can be used for rapid discrimination of numerous rice cultivars and simple quantitative analysis system of major carbohydrate compounds in rice grains.

Identification of Xanthium Sibiricum Components using LC-SPE-NMR-MS Hyphenated System

  • Xanthium sibiricum is used as a traditional folk medicine for the treatment of cancer, fever, headache, nasal sinusitis, and skin pruritus. This study aimed to identify components from Xanthium sibiricum extracts using an SPE-800MHz NMR-MS hyphenated system. The simultaneous acquisition of MS and NMR spectra from the same chromatographic peaks significantly increases the depth of information acquired for the compound and allows the elucidation of structures that would not be possible using MS or NMR data alone. LC -NMR analysis was conducted using a HPLC separation system coupled to 800 MHz spectrometer equipped with a cryoprobe, and a SPE unit was used to automatically trap chromatographic peaks using a HPLC pump. LC-MS analysis was conducted with a Q-TOF MS instrument using ESI ionization in the negative ion mode. Using the hyphenated analysis, several secondary metabolites were identified, such as 3',5'-O-dicaffeoylquinic acid, 1',5'-O-dicaffeoyl- quinic acid, and ethyl caffeate. These results demonstrate that the SPE-800MHz NMR-MS hyphenated system can be used to identify metabolites within natural products that have complex mixtures.

  • High-fat diets (HFD) and high-carbohydrate diets (HCD)-induced obesity through different pathways, but the metabolic differences between these diets are not fully understood. Therefore, we applied proton nuclear magnetic resonance ($^1H$ NMR)-based metabolomics to compare the metabolic patterns between C57BL/6 mice fed HCD and those fed HFD. Principal component analysis derived from $^1H$ NMR spectra of urine showed a clear separation between the HCD and HFD groups. Based on the changes in urinary metabolites, the slow rate of weight gain in mice fed the HCD related to activation of the tricarboxylic acid cycle (resulting in increased levels of citrate and succinate in HCD mice), while the HFD affected nicotinamide metabolism (increased levels of 1-methylnicotineamide, nicotinamide-N-oxide in HFD mice), which leads to systemic oxidative stress. In addition, perturbation of gut microflora metabolism was also related to different metabolic patterns of those two diets. These findings demonstrate that $^1H$ NMR-based metabolomics can identify diet-dependent perturbations in biological pathways.

A Study on the Synthesis ann Pyrolytic Properties of SiC/Ti Hybrid Ceramic Precursor by Hydrolysis (가수분해에 의한 탄화규소/티타늄 혼성 세라믹 전구체 합성과 열분해 특성에 관한 연구)

  • In order to increase the thermal stability at high temperatures, new hybrid ceramic percursors were synthesized by chemical modification of polycarbosilane (PCS). The structure of hybrid ceramic percursors were investigated by using FT-IR and $^1$H-NMR spectrometers. The syntheses of hybrid ceramic precursors were confirmed by monitoring the change of the adsorption peaks appearing at 0893, 1092, 609 $cm^{-1}$ / on the FT-IR spectra, and also by the presence of peaks at 3.8, 2.0, 0.6 ppm on the $^1$H-NMR spectra. The conversion of hybrid ceramic percursor was around 74 and 10 wt% higher than that of the pure PCS. After the heat-treatment at 150$0^{\circ}C$, the crystalline peaks for $\beta$-SiC were observed at 2$\theta$=35.7, 42.2, 61.0$^{\circ}$ on the X-ray powder diffractogram. It showed the conversion of hybrid ceramic percursor to crystalline $\beta$-SiC.

Identification of 1H-NMR characteristics for black ginger specimens from different places of origin

  • Black ginger (Kaempferia parviflora) is a short-lived ginger plant with dark purple colored root and is known to be effective in treating diabetes and obesity. To find out the difference in the characteristics of the black ginger according to the variety of production, 1D proton NMR experiments were performed on 4 types of black gingers from different regions. The NMR spectra of all black ginger showed the characteristic peaks of the polymethoxy flavone compounds, and the chemical shifts and intensity of peaks showed slight differences depending of the type of black ginger implying the difference in molecular environment. These initial NMR experiments can be applied to the identification of the diversity of black ginger in physiological function according to the climate of regions using SNIF-NMR (Site-specific Natural Isotope Fractionation studied by NMR).

Quantitative Analysis of t-Cinnamaldehyde of Cinnamomum cassia by $^1H-NMR$ Spectrometry ($^1H-NMR$을 이용한 계피의 t-cinnamaldehyde 정량분석)

  • trans-Cinnamaldehyde, a major component of Cinnamomum cassia, was quantitatively analyzed using the $^1H-NMR$ spectrometry. Applicability of this method was confirmed through observing the variation of chemical shift in the $^1H-NMR$ spectrum of t-cinnamaldehyde and the integration value according to various sample concentrations or running temperatures. When the $^1H-NMR$ spectrometry was run for t-cinnamaldehyde (7.1429 mg/ml) at 19, 25, 30, 40 and $50^{\circ}C$, the chemical shifts of the doublet methine signal due to an aldehyde group were observed at 9.7202, 9.7184, 9.7169, 9.7142 and 9.7124 ppm, respectively, to imply that the running temperature had no significant variation in the chemical shift of the signal. The integration values of the signal were $1.37\;(19^{\circ}C),\;1.37\;(25^{\circ}C),\;1.37\;(30^{\circ}C),\;1.37(40^{\circ}C)$ and $1.37(50^{\circ}C)$, respectively, to also indicate running temperature gave no effect on the integration value. When the sample solutions with various concentrations such as 0.4464, 0.8929, 1.7857, 3.5714, 7.1429 and 14.286 mg/ml were respectively measured for the $^1H-NMR$ at $25^{\circ}C$, the chemical shifts of the aldehyde group were observed at 9.7206, 9.7201, 9.7196, 9.7192, 9.7185 and 9.7174 ppm. Even though the signal was slightly shifted to the high field in proportion to the increase of sample concentration, the alteration was not significant enough to applicate this method. The calibration curve for integration values of the doublet methine signal due to the aldehyde group vs the sample concentration was linear and showed very high regression rate ($r^2=1.0000$). Meantime, the $^1H-NMR$ spectra (7.1429 mg/ml $CDCl_3,\;25^{\circ}C$) of t-cinnamaldehyde and t-2-methoxycinnamaldehyde, another constituent of Cinnamomum cassia, showed the chemical shifts of the aldehyde group as ${\delta}_H$ 9.7174 (9.7078, 9.7270) for the former compound and ${\delta}_H$ 9.6936 (9.6839, 9.7032) for the latter one. The difference of the chemical shift between two compounds was big enough to be distinguished using the NMR spectrometer with 0.45 Hz of resolution. The contents of cinnamaldehyde in Cinnamomum cassia, which were respectively extracted with n-hexane, $CHCl_3$, and EtOAc, were determiend as 94.2 \;mg/g (0.94%), 137.6 mg/g (1.38%) and 140.1 mg/g(1.40%) t-cinnamaldehyde in each extract, respectively, by using the above method.

  • From the catalytic reductions of the respective 2-amino-, and 2-nitro-3,4-dihydro-5,6-dimethoxy-1(2H)-naphthalenone, a major reduction product of 2-amino-5,6-dimethoxy-1,2,3,4-tetrahydro-naphthalene was obtained, along with a minute amount of rearranged product, 5,6-dimethoxy-1,2,3,4-tetrahydro-2(1H)-naphthalenone. The unusual formation of 5,6-dimethoxy-1,2,3,4-tetrahydro-2(1H)-naphthalenone was verified on the basis of ir, and nmr spectra, and elemental analysis. A plausible mechanism for the rearrangement is proposed.

  • 6- and 7-Benzylpteridine derivatives have been converted to the corresponding 6- and 7-benzoylpteridines by the oxidation reaction with KMnO4. The newly synthesized compounds have been characterized by pKa determinations, UV, and 1H-NMR spectra.

Synthesis and fluorescent property investigation of novel fluoroionophores

  • Novel fluoroionophore of dibenzothiazolyl-dibenzo-crown ethers were synthesized from diformal-dibenzo 18-crown-6 (24-crown-8) with 2-aminothiophenol, and they were characterizated by $^1H$-NMR, $^{13}C$-NMR, IR spectrum, Mass spectrum, elemental analyses, respectively. The fluorescent properties of the newly synthesized crown ether were examined with $Li^+$, $Na^+$, $K^+$, $Rb^+$, $Cs^+$, $NH_4^+$ and $CF_3COOH$, respectively. With protonation using $CF_3COOH$, the absorption bands of the new crown ethers are further blue shifted, the maximum emission wavelengths further red shifted.
