Hepatoprotective and free radical scavenging activities of Lagerstroemia speciosa Linn. leaf extract

  • The present study deals with the amelioration by Lagerstroemia speciosa Linn. leaf extract against hepatotoxicity induced by carbon tetrachloride ($CCl_4$), which was evaluated in terms of serum marker enzymes like serum glutamate pyruvate transaminase, serum glutamate oxaloacetate transaminase, alkaline phosphatase, serum total bilirubin, total protein levels along with concomitant hepatic and antioxidants like superoxide dismutase, catalase, glutathione, glutathione peroxidase and lipid peroxidation enzymes were monitored. These biochemical parameters altered by the single dose level of $CCl_4$ (0.75 ml/kg body weight, i.p). Pre treatment with L. speciosa prior to the administration of $CCl_4$, at the doses of 50 and 250 mg/kg. body weight/day, p.o. for 7 days, significantly restored all the serum and liver tissue parameters near to the normal levels, respectively. Silymarin was used as a reference standard, prior to the administration of $CCl_4$ to rats. These findings indicate the protective potential of L. speciosa against hepato toxicity which possibly involve mechanism related to its ability of selective inhibitors of (reactive oxygen species like antioxidants brought about significant inhibition of TBARS suggesting possible involvement of $O_2{\cdot}-$, $HO_2{\cdot}$, and ${\cdot}OH$. In conclusion, the amelioration may be attributed to the synergistic effects of its constituents rather than to any single factor as the leaves are rich in tannins, sterols, flavonoids, saponins etc.

  • The present study was investigated on the DNA damage by the peroxidation of polar and non-polar lipid fractionated from mackerel lipid to elucidate the DNA damage mechanism by fish oil peroxidation. The degree of DNA damage by polar lipid peroxidation became greater with the increase of its concentration, and such DNA damage was induced below 100 millieq./kg in POV for 4 days incubation. Among the polar lipid peroxidation products, singlet oxygen $^1O_2$ and superoxide anion ${\cdot}O_2^-$ greatly affected to the DNA damage than hydrogen peroxide $H_2O_2$ and hydroxyl radical ${\cdot}OH$. Non-polar lipid peroxidation also induced the DNA damage with the increase of its concentration, but such effect was lower than the case of total lipid and polar lipid. And, the effects of active crygens on the DNA damage by non-polar lipid peroxidation was the same as in the case of total and polar lipid peroxidation.

  • The DNA damage mechanism by fish oil peroxidation was investigated through the model system of a DNA-mackerel lipid at $37^{\circ}C$. Mackerel lipid peroxidation products induced a great DNA damage with the increment of its concentration, and such DNA damage in all systems examined occurred below $100millieq{\cdot}/kg$ in POV (peroxide value) Singlet oxygen $(^1O_2)$ and superoxide anion${\cdot}O_2^-$ greatly participated in the DNA damage during peroxidation of mackerel lipid, while hydrogen peroxide$(H_2O_2)$ and hydroxyl radical $({\cdot}OH)$ did little show the DNA damage. From the results of the addition of several active oxygen scavengers to the DNA-lipid systems, singlet oxygen ana superoxide anion greatly affected to the increase of POV ana to the DNA damage by mackerel lipid peroxidation, respectively. It indicates that there was a close relationship between the effects of active oxygens in the mackerel lipid peroxidation and its DNA damage mechanism.

Application of Electrochemical Technology for Reusing Biologically Treated Water (생물학적 처리수 재이용을 위한 전기화학 기술의 적용)

  • Laboratory experiments were conducted to investigate disinfection as well as removal of color and residual organics from reclaimed municipal wastewater by electrochemical processes with Nb/Pt anode installed. RNO was rapidly bleached by OH$\cdot$ and the second order rate constants of RNO removal were $\frac{0.223l}{mg{\cdot}min}$, $\frac{1.679l}{mg{\cdot}min}$ and $\frac{2.322l}{mg{\cdot}min}$ with for 5 A, 10 A, and 15 A, respectively, with r$^2$ of > 96%. In batch electrochemical processes, after 15 min at currency of 15 A and initial pH of 5, 7,5 and 9, COD$_{Mn}$ was below 4 mg/L, color unit below 5 degree and general bacteria was not detected, the concentration of which are suitable for drinking water regulation. In the continuous electrochemical process, with HRT of 3.7$\sim$49.2 min, free chlorine were 0.2$\sim$0.7 mg/L, general bacteria was not detected, color unit below 5 degree and THMs was 0.017 mg/L. Therefore, electrochemical process with Nb/Pt anode was employed satisfactory to meet for reusing biologically treated water as well as disinfection.

Antioxidative Properties of Artemisia princeps Pamp (약쑥(Artemisia princeps Pamp)의 항산화작용)

  • Antioxidant properties of Artemisia princeps Pamp were determined using in vitro assay systems against 1,1-diphenyl-picrylhydrazyl (DPPH) radical, superoxide ($O_2{^-}$), hydrogen peroxide ($H_2O_2$), hydroxyl radical ($HO{\cdot}$) and nitric oxide (NO) radical, as well as rat liver microsomal lipid peroxidation. Among the five solvent fractions, ethyl acetate fractions showed the highest total polyphenol and flavonoid contents at $311.35{\mu}g/mg\;and\;92.73{\mu}g/mg$, respectively. Ethyl acetate fractions, except for $H_2O_2$ scavenging activity, also showed the highest scavenging activity; the 50% inhibitory concentration ($IC_{50},\;{\mu}g/mg$) values for DPPH, superoxide, $HO{\cdot}$ and NO radical scavenging were 52.71, 26.47, 58.92 and 65.94, respectively. Additionally, the highest inhibition of rat liver microsomal lipid peroxidation was observed by ethyl a cetate fraction.

Effects of Edible Lentinus tuber-regium on Oxidative Stress and Defense System in Serum of SD Rats (유용버섯 Lentinus tuber-regium이 산화적 스트레스 및 방어체계에 미치는 영향)

  • Oxidative stress and defense system of SD-rats were studied with an edible Nigerian mushroom, namely, Lentinus tuber-regium (Fries) Singer. Experimental diets prepared with Lentinus tuber-regium (LTR) instead of carbohydrates were fed to SD rats for 6 weeks. Hydrolxyl radical $({\cdot}OH)$ formations were significantly inhibited (21.7% and 16.4%, respectively). In LTR-50 and LTR-100 groups used instead of carbohydrates, and hydrogen peroxide and nitric oxide (NO) were also significantly inhibited by 10%, and $6{\sim}10%$, respectively compared with control group, but there was no significant changes in superoxide radical $({O_2}^-)$ formations in these groups. Lipid peroxide (LPO) and oxidized protein (OP) levels as an oxidative stress were desirably inhibited ($6{\sim}12%\;and\;5{\sim}13%$, respectively) in these LTR groups compared with control group. Superoxide dismutase (SOD), glutathione peroxidase (GSHPx) and catalase (CAT) activities were significantly increased ($15{\sim}50%,\;10{\sim}25%\;and\;60{\sim}90%$, respectively) in these LTR groups. These results suggest that an edible mushroom, Lentinus tuber-regium may inhibit an oxygen radicals and oxidative stresses, but may also effectively modulate an aging processes.

  • In this study, we investigated the biological activity of antioxidant and antibacterial activity of citrus by-product ferment. Hot water extracts and 80% methanol extracts from citrus byproduct of ferment were screened for antibacterial activity pathogenic bacteria by paper disc method. Among the various hot water extracts and 80% methanol extracts the Bacillus subtilis showed relatively strong antibacterial activities in the order. The reducing activity on the 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical and ${\cdot}OH$ radical scavenging potential were sequentially screened, in search for antioxidant activities of citrus by-product ferment.

  • Acetylsalicylic acid (ASA) has been issued recently in contaminated water environments because of potential impacts on ecosystem and public health. This study was aimed at investigating the possibility of ASA degradation using gamma ray irradiation. In addition, the use of sodium persulfate, hydrogen peroxide, ferrous sulfate were tested in order to examine a synergistic effect with gamma ray. The absorbed dose was ranged from 0.2 to 10 kGy and the concentration of oxidants were from 0.1 to 10 mM in this study. The concentration of ASA was gradually decreased corresponding to the increase of the absorbed dose. When soudium persulfate was simultaneously applied, most of the parent compound was completely degraded even at a low dose of 0.8 kGy. The removal efficiency of total organic carbon was 90% even at the highest dose of 10 kGy without sodium persulfate. However, the efficiency was dramatically enhanced up to 98% at the same dose by adding 10 mM of oxidants. It was suggested that hydroxyl radical ($OH{\cdot}$) and sulfate radical ($SO{_4}^-{\cdot}$) were formed in the system and made roles in degrading ASA at the same time.

Treatment of TNT Red Water by the Ozone-based Advanced Oxidation Processes (오존을 산화제로 사용한 다양한 고급산화 공정에 의한 TNT Red Water의 처리)

  • Several combinations of ozone based advanced oxidation processes were tested for the treatment of red water (RW) containing recalcitrant chemical pollutants produced from 2,4,6-trinitrotoluene (TNT) manufacturing process. $O_3$, $UV/O_3$, $UV/O_3/H_2O_2$, $UV/O_3/H_2O_2/Fe^{2+}$ processes were tested for the treatment of RW. The order of organic and color removal efficiency was found to be : $O_3{\leq}UV/O_3$ < $UV/O_3/H_2O_2$ < $UV/O_3/H_2O_2/Fe^{2+}$. The optimum conditions for the removal of organic and color in the $UV/O_3/H_2O_2/Fe^{2+}$ process were 0.053 g/min of ozone flow rate, 10 mM of $H_2O_2$ concentration and 0.1 mM of $FeSO_4$ concentration. Organic and color removal efficiencies were 96 and 100 % respectively in the $UV/O_3/H_2O_2/Fe^{2+}$ process. tert-butyl alcohol (t-buOH) was used as the hydroxyl radical scavenger. Enhancement of hydroxyl radical production was achieved by the combination of ozone with several oxidants such as UV, $H_2O_2$, $Fe^{2+}$.

Electron Beam Mediated Simple Synthetic Route to Preparing Layered Zinc Hydroxide

  • We have developed a novel and eco-friendly synthetic route for the preparation of a two-dimensional layered zinc hydroxide with intercalated nitrate anions. The layered zinc hydroxide nitrate, called 'zinc basic salt', was, in general, successfully synthesized, using an electron beam irradiation technique. The 2-propanol solutions containing hydrated zinc nitrate were directly irradiated with an electron-beam at room temperature, under atmospheric conditions, without stabilizers or base molecules. Under electron beam irradiation, the reactive OH radicals were generated by radiolysis of water molecules in precursor metal salts. After further radiolytic processes, the hydroxyl anions might be formed by the reaction of solvated electrons and the OH radical. Finally, the $Zn_5(OH)_8(NO_3)_2{\cdot}2H_2O$ was precipitated by the reaction of zinc cation and hydroxyl anions. Structure and morphology of obtained compounds were characterized by powder X-ray diffraction (XRD), field emission scanning electron microscopy (FE-SEM), and high resolution transmission electron microscopy (HR-TEM). The chemical components of the products were determined by Fourier transform infrared spectroscopy (FT-IR) and elemental analysis (EA). The thermal behavior of products was studied by thermogravimetric (TG) and differential thermal analysis (DTA).
