Rhizobium meliloti 102F51 Mutants Defective in Heme Synthesis (Heme 합성특성이 다른 Rhizobium meliloti 102F51 Mutant의 선별)

  • Rhizobium meliloti 102 F 51, the symbiotic partner of alfalfa, was mutagenized with N-methyl-N'-nitro-N-nitrosoguanidine (NTG) and UV-irradiation. Three group of mutants which form white, white-pink and red nodules were selected. The adetylene reduction activity, nodulation activity, amount of heme synthesis during the nodulation, and ${\delta}-aminolevulinic$ acid synthetase (ALAS) and ${\delta}-aminolevulinic$ acid dehydratase (ALAD) activities in free living rhizobia and bacteroid states of the each group of mutants were compared. The mutants forming white nodules showed lower acetylene reduction activity compared to those of red nodule forming mutants. The two key enzymes for the heme synthetic pathway, ALAS and ALAD activities of the mutants forming red nodules was much higher than those of the mutants forming white nodules in bacteroid state, however no significant difference was observed in free living state. In the nodules the ALAS was detected only in bacteroid fraction, while ALAD was detected both in bacteroid and plant fraction. ALAS was dramatically increased with the heme synthesis during the nodulation, while ALAD was decreased in plant fraction but slight increase was observed in bacteroid fraction.

  • The effects of S-23142{N-(4-chloro-2-fluoro-5-propargyloxyphenyl)-3, 4, 5, 6-tetrahydrophtalimide}, on protoporphyrin IX(PPIX) biosynthesis in Spinacia oleracea L, leaf in vivo and in vitro condition were investigated by reversed-phase HPLC with fluorescence detector. The stroma and the membrane fraction of spinach chloroplast were isolated by osmotic regulation. The conversion of ${\delta}$-aminolevulinic acid(ALA) to PPIX occured more in the stroma than in the membrane fraction. It suggested that the enzymes that catalyse PPIX biosynthesis from ALA were localized in the stroma. Also, the synthesized PPIX content from ALA was completely inhibited by $10^{-8}M$ of S-23412 or $10^{-7}M$ of acifluorfen in the stroma but not in the membrane fractions. Therefore, these results suggested that the target site of S-23142 and acifluorfen may exist in the stroma fraction of spinach chloroplast.

  • Present experiments were undertaken in order to clarify the clinico-pathological characteristics of lead poisoning in goats. Twenty goats were divided into three experimental groups(A, B and C) and a control(D). The three experimental groups received diets contaminated artificially with 10(A group), 200(B group) and 1,000(C group) ${\mu}g/$ of lead, for 70 days respectively. The control group received normal diets. Blood samples were collected 1 or 2 weeks interval and were examined for anemia(erythrocyte counts, hemoglobin concentrations and hematocrit values) and lead contents of erythrocyte and serum. Urine samples collected similarly with blood were examined for delta-aminolevulinic acid and lead content. Collected samples were analyzed for lead content by atomic absorption spectrophotometry. From these experiments following results were obtained : In group B and C, marked decreases in body weight and feed intake, and diarrhea were observed from the $30^{th}$ day of experimental periods. The B and C groups showed pronounced anemia(decrease in erythrocyte count, hemoglobin concentration and hematocrit value) from the $21^{st}$ or $42^{nd}$ day. In group B and C, the lead content of erythrocytes was increased significantly from the $28^{th}$ or $14^{th}$ day. The lead content of serum was increased significantly from the $42^{nd}$ or $14^{th}$ day in B and C groups. The lead content of urine was increased significantly from the first day in both B and C groups. The urinary delta-aminolevulinic acid content was increased significantly from $14^{th}$ day in both B and C groups.

Clinicopathological and Histopathological Findings of Experimental Lead Poisoning in Dogs (실험적 납 중독견의 임상병리학적 및 조직병리학적 소견)

  • The study of present study was to determine the valuable laboratory tests for the differential diagnosis of lead poisoning in dogs. Sixteen mongrel dogs were divided into 2 experimental groups (A and B) and a control group (C). The A and B groups were administered orally 2 mg and 20 mg of lead per kilogram of body weight for 49 days, respectively. In addition to clinical observation, blood, urine and hair samples were collected on appointed day and examined for hematological changes, lead content of serum, whole blood and hair, and urinary $\delta$-aminolevulinic acid concentrations. All dogs were necropside on 49th day and examined for the lead content and histological changes of organs. The results obtained were summarized as follows: The group B showed digestive and nervous signs, and weight loss. The group A showed no significant hematological changes except polychromatophilla on the 7th day. But group B showed polychromatophilia as well as mild anemia and nucleated erythrocyte on the 7th and 35th day. Basophlic stippling erythrocytes were observed in some of the group B on the 14th day. The lead content of whole blood was increased significantly in both A and B groups on the 21the day. The urinary $\delta$-aminolevulinic acid content was increased in both A and B groups on the 7th day. The hair lead content of A and B groups was increased significantly on the 49th and 21th day, respectively. The lead contents of organs including liver, kidney, spleen, muscle and bone were increased significantly in group B. Histopathologic changes were characterized by hemorrhages, necrosis and intranuclear inclusion body in the epithelial cells of convoluted tubles of kidney, cloudy swelling and degeneration and/or necrosis of liver, enlargement of Virchow-Robin space, and swelling of endothelial cells and hyperplasia of the pericytes of brain. From these results it may be concluded that examination of nucleated erythrocyte/polychromatophilia, urinary $\delta$ -aminolevulinic acid, and whole blood and hair lead contents is a reliable clinico-pathological diagnostic methods, and that examination of the Virchow-Robin space, endothelial cells and pericytes of brain as well as intranuclear inclusion body in the epithelial cells of convoluted tubles of kidney is valualble postmortem diagnostic methods for lead poisoning in dogs.

Effects of Dietary Irons Supplementation on ${\delta}-Aminolevulinic$ Acid Dehydratase Activity and Detoxification of the Pb-administered Rats (철보충이 납중독된 흰쥐의 ${\delta}-Aminolevulinic$ Acid Dehydratase 활성과 해독에 미치는 영향)

  • This study was designed to investigate the effects of dietary iron supplementationon ${\delta}-aminolevulinic$ acid dehydratase (DALAD) activity and liver damage of the lead(Pb)-administered rats. The iron (Fe) supplement levels were 100 ppm(Fe100) and 500ppm(Fe500) and the Pb-exposed rats were given 2, 000ppm-Pb in drinking water, while control rats were given neigher iron nor lead. Hematocrit was lower in the Pb, Fe100 -Pb, Fe500-Pb group than in the control, but was not affected by the Pb adminstration when the rats fed the Fe supplementation diet. DALAD activity were reduced by Pb added but, were higher in the Fe500-Pb group than in the Fe100 -Pb group. No significant difference was found in serum Pb content due to Pb administration and Fe supplementation. The liver Pb contentwas higher in the Fe supplementation group than in the Pb-group. Level of serum FE was lower in the Pb added groups than in the control group. Liver Fe contents were increased with Pb administration and higher in the Fe supplement groups than in the Pb-group. Levels of serum and liver copper was decreased with the Fe supplementation. Aminotransferase activity of serum and liver were increased in the Pb group.

Supplementation of δ-aminolevulinic acid to sows' diet from day 100 of gestation to lactation improves the feed intake and red blood cells of sows and improves the birth weight of offspring

  • This experiment was conducted to evaluate the effects of δ-aminolevulinic acid (ALA) when added to sows' diet on their reproductive performance and growth performance and on the hematology parameters of the sows and their piglets. Sixteen multiparous sows (Yorkshire × Landrace) were allotted into two treatment groups and fed basal diets (CON, piglets were injected with iron dextran) or the basal diet containing 0.1% ALA (ALA, piglets were not injected with iron dextran) from day 100 of gestation to day 28 of lactation. Supplementation of ALA had no effect on the body weight (BW), backfat thickness (BFT), or litter sizes of sows in the present experiment. However, the average daily feed intake (ADFI) of the sows was significantly improved (p < 0.05) in the ALA group. Supplementation of ALA had no effect on the growth performance or survival of suckling piglets but had a significant effect on the birth weight (p < 0.05). With regard to the blood profiles, serum concentrations of iron were unaffected in sows and piglets as compared to the control group. Red blood cell (RBC) counts were significantly improved (p < 0.05) in sows during late gestation to the time before farrowing period and in piglets at weaning. In summary, these results suggest that dietary supplementation of ALA can have positive effects by improving growth performance and blood RBC in sows and suckling piglets.

Evaluation of δ-Aminolevulinic Acid on Serum Iron Status, Blood Characteristics, Egg Performance and Quality in Laying Hens

  • Effects of dietary ${\delta}$-aminolevulinic acid (ALA) supplementation on serum iron status, blood characteristics, egg production and quality were examined in laying hens in an 8-week feeding trail. Two hundred and forty (Hy-line brown, 40-week-old) layers were randomly assigned to four dietary treatments with ten replications (six layers in adjacent three cages). Dietary treatments included: 1) CON (basal diet), 2) ALA1 (CON+ALA 5 ppm), 3) ALA2 (CON+ALA 10 ppm) and 4) ALA3 (CON+ALA 15 ppm). All nutrient levels of diets were formulated to meet or exceed NRC (1994) recommendations for laying hens. During the entire experimental period, differences of serum iron concentration and total iron binding capacity (TIBC) were significantly increased in ALA1 supplemented treatment (quadratic effect, p<0.05). The difference of total protein between 8 and 0 weeks was significantly higher in ALA2 treatment than CON treatment (quadratic effect, p<0.05). No significant effects were observed on hemoglobin, WBC, RBC, lymphocyte and albumin concentrations. Egg production and egg weight were not influenced by the ALA supplementation. Egg yolk index was also significantly higher in ALA3 treatment than CON treatment at the end of 4 and 8 weeks (linear effect, p<0.05). Haugh unit was increased in ALA3 treatment compared to CON and ALA1 treatments at the end of 8 weeks (linear effect, p<0.05). However, egg shell thickness, breaking strength and yolk color unit were not affected by the ALA supplementation. In conclusion, dietary ALA supplementation at a level of 5 ppm can affect iron concentration in serum while higher levels (10 or 15 ppm) have some beneficial influences on blood profiles and egg quality.

Measurement of δ-Aminolevulinic Acid in Urine by Fluorometric HPLC and Colorimetric Methods (비색법과 HPLC 법에 의한 요중 δ-Aminolevulinic acid의 측정치 비교)

  • The urinary excretion of ${\delta}$-aminolevulinic acid has been widely used as a measure of the biological effect of lead in lead exposed workers. It is usually measured by colorimetric method based on the color reaction of ALA-pyrrole with Ehrlich's reagent. But the results of ${\delta}$-ALA in urine by this method are somewhat artificially higher than expected due to the urinary ALA-like compound such as aminoacetone. On the other hand, the recently developed fluorometric HPLC method is very sensitive and specific for the measuring urinary ALA. In order to compare the data obtained by two methods and to investigate the interrelation between two methods, 117 lead workers with different lead exposure were checked urinary ${\delta}$-ALA, blood lead and other lead exposure related indices. The results obtained are as follows; 1. Urinary excretion of ${\delta}$-ALA by colorimetric method is 2.15 times higher than HPLC method in overall, revealing 2.47 times in workers of blood lead less than $20{\mu}g/dl$, 2.53 times in workers of blood lead $21-40{\mu}g/dl$ and 1.86 times in workers of blood lead over $41{\mu}g/dl$, respectively. 2. While the correlation coefficients of ${\delta}$-ALA measured by colorimetric method with blood lead and blood ZPP was 0.571 and 0.629, those of ${\delta}$-ALA measured by HPLC with blood lead and blood ZPP were 0.6l0 and 0.637. All the correlation coefficients were statistically significant, but there was no statistical difference of correlation coefficients between two methods. 3. The correlation coefficient of urinary excretion of ${\delta}$-ALA between two method was 0.838 without any correction, but it was 0.852 with the correction of specific gravity 1.024. 4. Simple linear regression of ${\delta}$-ALA measured by HPLC method on ${\delta}$-ALA measured by colorimetric method was (ALA-UPH)=-0.245+0.536 (ALA-UCO) without any correction and it was (SP ALA)=-0.525+0.598 (SP ALA-UCO) with the correction of specific gravity 1.024. With above results, it is recommended that the diagnostic criteria of ${\delta}$-ALA for lead poisoning needed to be revised if ${\delta}$-ALA is measured by HPLC rather than colorimetric method.

Evaluation of Dietary Supplementation of Delta-aminolevulinic Acid and Chito-oligosaccharide on Production Performance, Egg Quality and Hematological Characteristics in Laying Hens

  • The effects of delta-aminolevulinic acid (DALA) or chito-oligosaccharide (COS) in layer diets on production performance, egg quality and hematological characteristics were investigated in this 8-week trial. Two hundred and seventy 27-week-old (Hy-Line brown) layers were randomly assigned to 5 treatments with 9 replications per treatment. Dietary treatments were i) Control (basal diet); ii) DALA1 (basal diet+DALA 2 mg/kg); iii) DALA2 (basal diet+DALA 4 mg/kg); iv) COS1 (basal diet+COS 100 mg/kg) and v) COS2 (basal diet+COS 200 mg/kg). In this study, no significant difference was observed in feed intake and egg shell quality (eggshell breaking strength and egg shell thickness) among treatments. The inclusion of DALA increased egg production during the first 4 weeks. Egg weight was increased in both DALA (quadratic, p<0.05) and COS (linear, p<0.05) treatments compared with the control treatment. Laying hens fed the DALA treatment had an increased yolk color unit (quadratic, p<0.05) and haugh unit (linear, p<0.05) compared with the control group. Similarly, these characteristics were also affected by COS treatments, with both values being linearly increased (p<0.05) in COS treatments compared with the control treatment. Additionally, birds fed DALA treatments significantly increased (quadratic, p<0.05) the number of RBC, WBC and lymphocytes compared with the control treatment. Dietary DALA supplementation linearly increased (p<0.05) the serum iron concentration at the end of the 8th week. The inclusion of COS increased (linear, p<0.05) the concentration of RBC, WBC and lymphocytes compared with the control treatment. In conclusion, dietary DALA at the lower dosage (2 mg/kg) could exert better effects in laying hens than higher dosage (4 mg/kg). Birds fed DALA supplemented diet had an increased iron availability, egg weight, eggshell quality and immunity. Moreover, the inclusion of COS (200 mg/kg) can increase egg weight, eggshell quality and immunity in laying hens. Therefore, both the utilization of COS and DALA could be considered as a new strategy for optimizing egg quality and health condition of laying hens.
