  • We use some known contiguous function relations for $_2F_1$ to show how simply the following three recurrence relations for Jacobi polynomials $P_n^{({\alpha},{\beta)}(x)$: (i) $({\alpha}+{\beta}+n)P_n^{({\alpha},{\beta})}(x)=({\beta}+n)P_n^{({\alpha},{\beta}-1)}(x)+({\alpha}+n)P_n^{({\alpha}-1,{\beta})}(x);$ (ii) $2P_n^{({\alpha},{\beta})}(x)=(1+x)P_n^{({\alpha},{\beta}+1)}(x)+(1-x)P_n^{({\alpha}+1,{\beta})}(x);$ (iii) $P_{n-1}^{({\alpha},{\beta})}(x)=P_n^{({\alpha},{\beta}-1)}(x)+P_n^{({\alpha}-1,{\beta})}(x)$ can be established.

A Study on the Effect of Coffee Shop Service Quality on Perceived Value and Behavioral Intention - Focusing on Busan-Area College Students - (커피전문점 서비스품질이 지각된 가치와 행동의도에 미치는 영향에 관한 연구 - 부산지역 대학생을 중심으로 -)

  • This study used SPSSS 18.0 on 237 university students in Busan for frequency, factorial and reliability, correlation, and regression analyses to determine the effect relationship of coffee shop service quality on perceived value and behavioral intention. Multiple regression analysis for hypothesis testing showed that among the four service quality factors, confidence in employees and primary coffee quality had a non-significant effect on esthetic value, while menu characteristics besides coffee (${\beta}$=.293, p<.001) and physical environment (${\beta}$=.293, p<.001) were analyzed to be significant, partially supporting the study hypothesis. Regarding the effect of service quality on practical value, confidence in employees (${\beta}$=.264 p<.001), primary coffee quality (${\beta}$=.463, p<.001), menu characteristics besides coffee (${\beta}$=.139, p<.05) and physical environment (${\beta}$=.110, p<.05) were all significantly analyzed, supporting the study hypothesis. Regarding the effect of service quality on behavioral intention, confidence in employees (${\beta}$=.262 p<.001), primary coffee quality (${\beta}$=.411, p<.001), menu characteristics besides coffee (${\beta}$=.157, p<.01) and physical environment (${\beta}$=.137, p<.05) were all significantly analyzed, supporting the study hypothesis. In addition, regarding the effect of perceived value on behavioral intention, esthetic value (${\beta}$=.265, p<.001) and practical value (${\beta}$=.536, p<.001) were both significantly analyzed, showing a causal relationship with behavioral intention.

  • In order to study the physiological properties of the intestinal bacteria, we isolated the intestinal bacteria of Koreans and tested the enzymatic patterns. Isolated Bifidobacterium sp. Int-57 had the higher activity of $\alpha$-glucosidase, $\beta$-glucosidase, $\alpha$-galactosidase, $\beta$-galactosidase. $\beta$-xylosidase and $\alpha$-arabinofuranosidase than other intestinal microorganisms. The effect of the carbon sources on the production of each enzymes of Bijidobacterium sp. Int-57 was investigated. The most suitable carbon source for the production of $\beta$-glucosidase was maltose, for a-glucosidase cellobiose, for $\alpha$-galactosidase raffinose, for $\beta$-galactosidase lactose, and for $\beta$-xylosidase and $\alpha$-arabinofuranosidase xylose, respectively. In addition, we investigated the optimal conditions and pH stability of each crude enzymes. The optimal condition of a-glucosidase was pH 6.0 and $40^{\circ}C$. that of Jj-glucosidase pH 7.0 and 50oe, that of $\beta$-galactosidase pH 7.0 and $50^{\circ}C$, that of $\beta$-xylosidase pH 6.0 and $40^{\circ}C$ , and that of $\alpha$-arabinofuranosidase pH 5.0 and $50^{\circ}C$. respectively. a-Glucosidase was stable at pH 4.0-9.0. Jj-glucosidase at pH 4.0-7.0. $\beta$-galactosidase at pH 4.0-9.0, $\beta$-xylosidase at pH 4.0-6.0, and /3-arabinofuranosidase at pH 7.0-9.0, respectively.

Novel substrate specificity of a thermostable β-glucosidase from the hyperthermophilic archaeon, Thermococcus pacificus P-4 (초고온 고세균 Thermococcus pacificus P-4로부터 내열성 β-glucosidase의 새로운 기질 특이성)

  • Based on the genomic analysis of Thermococcus pacificus P-4, we identified a putative GH1 ${\beta}$-glucosidase-encoding gene (Tpa-glu). The gene revealed a 1,464 bp encoding 487 amino acid residues, and the deduced amino acid residues exhibited 77% identity with Pyrococcus furiosus ${\beta}$-glucosidase (accession no. NP_577802). The gene was cloned and expressed in Escherichia coli system. The recombinant protein was purified by metal affinity chromatography and characterized. Tpa-Glu showed optimum activity at pH 7.5 and $75^{\circ}C$, and thermostability with a half life of 6 h at $90^{\circ}C$. Tpa-Glu exhibited hydrolyzing activity against various pNP-glycopyranosides, with kcat/Km values in the order of pNP-${\beta}$-glucopyranoside, pNP-${\beta}$-galactopyranoside, pNP-${\beta}$-mannopyranoside, and pNP-${\beta}$-xylopyranoside. In addition, the enzyme exhibited exo-hydrolyzing activity toward ${\beta}$-1,3-linked polysaccharide (laminarin) and ${\beta}$-1,3- and ${\beta}$-1,4-linked oligosaccharides. This is the first description of an enzyme from hyperthermophilic archaea that displays exo-hydrolyzing activity toward ${\beta}$-1,3-linked polysaccharides and could be applied in combination with ${\beta}$-1,3-endoglucanase for saccharification of laminarin.

Cloning of $\beta$-Glucosidase Gene from Streptomyces coelicolor A3(2) and Characterization of the Recombinant $\beta$-Glucosidase Expressed in Escherichia coli (Streptomyces coelicolor A3(2)로 부터 $\beta$-Glucosidase 유전자 클로닝 및 재조합 효소의 특성)

  • The $\beta$-glucosidase gene from Streptomyces coelicolor A3(2) was cloned and expressed in Escherichia coli. The ORF consisted of 1377 nucleotides encoding 51 kDa in a predicted molecular weight. Effects of pH indicated that the $\beta$-glucosidase showed similar activity using $\alpha$-pNPG($\rho$-nitrophenyl-$\alpha$-D-glucopyranoside), $\beta$-pNPG($\rho$-nitrophenyl-$\beta$-D-glucopyranoside), and $\beta$-pNPF($\rho$-nitrophenyl-$\beta$-D-fucopyranoside) at range of pH 3 to 10, and high activity using $\beta$-pNPGA ($\rho$-nitrophenyl-$\beta$-D-galactopyranoside) from pH 5 to 10, especially, 3.3 times higher activity at pH 9. Effects of temperature indicated that the $\beta$-glucosidase showed low activity using $\alpha$-pNPG, $\beta$-pNPG, and $\beta$-pNPF from $20^{\circ}C$ to $70^{\circ}C$, and increased activity using $\beta$-pNPGA from $30^{\circ}C$ to $50^{\circ}C$, 1.8 times higher activity at $50^{\circ}C$ than at $30^{\circ}C$. According to activity determination of other substrates, the enzyme was active on daidzin, genistin, and glycitin, inactive on esculin and apigenin-7-glucose. The EDTA and DTT as reducing agents inhibited $\beta$-glucosidase activity, but SDS and mercaptoethanol did not inhibit. Monovalent or divalent metal ions such as $MnSO_4$, $CaCl_2$, KCl, and $MgSO_4$ did not inhibited $\beta$-glucosidase activity. $CuSO_4$ and NaCl showed low inhibition, and $ZnSO_4$ inhibited 3.3 times higher than control.

  • The rate constants of the hydrolysis of ${\alpha}$-Cyano-${\beta}$-piperonylacrylic acid were determined by Ultraviolet spectrophotometry at various pH and a rate equation which can be applied over wide pH range was obtained. The reaction mechanism of hydrolysis of ${\alpha}$-Cyano-${\beta}$-piperonylic acid and especially the catalytic contribution of hydroxide ion which not studied carefully before in acidic media, can be fully explained by the rate equation obtained. The rate equation reveals that; below pH 4.0, the reaction is initiated by the addition of water molecule to ${\alpha}$-Cyano-${\beta}$-piperonyl acrylic acid. At pH $5.0{\sim}7.5$, ${\alpha}$-Cyano-${\beta}$-piperonylacrylic acid compete with ${\alpha}$-Cyano-${\beta}$-piperonyl acrylate ion in adding of water. At pH 8.0, water is the only nucleophile for ${\alpha}$-Cyano-${\beta}$-piperonylacrylate ion, however, above pH 12.0, hydroxide ion is an addendum and the accepter is ${\alpha}$-Cyano-${\beta}$-piperonylacrylate ion.

  • In this paper, two weight conditions are introduced and the multiple weighted strong and weak characterizations of the multilinear fractional new maximal operator 𝓜ϕ,β are established. Meanwhile, we introduce the ${\mathcal{S}}_{({\vec{p}},q),{\beta}}({\varphi})$ and $B_{({\vec{p}},q),{\beta}}({\varphi})$ conditions and obtain the characterization of two-weighted inequalities for 𝓜ϕ,β. Finally, the relationships of the conditions ${\mathcal{S}}_{({\vec{p}},q),{\beta}}({\varphi}),\,{\mathcal{A}}_{({\vec{p}},q),{\beta}}({\varphi})$ and $B_{({\vec{p}},q),{\beta}}({\varphi})$ and the characterization of the one-weight $A_{({\vec{p}},q),{\beta}}({\varphi})$ are given.

Studies on the Variation of Monoterpene Composition of the Subgenus Diploxylon of the Genus Pinus in Korea (우리나라 소나무속(屬)의 Monoterpene 조성(組成)의 변이(變異)에 관(關)한 연구(硏究))

  • Monotupenes of the oleoresin of P. densiflora S. et Z., P. densi-thunber gii Uyeki, P. thunbergii Parl., and P. densiflora for. erecta Uyeki of the subgenus Diploxylon of the genus Pinus in Korea were analysed by means of gas liquid chromatography, and following results were obtained. Monoterpene of P. densiflora, P. densi-thunbergii, P. thunbergii, and P. densiflora for. erecta consisted of ${\alpha}$-pinene, camphene, ${\beta}$-pinene, myrcene, limonene, ${\beta}$-phellandrene, terpinolene and two unknown components. Major monoterpene components of P. densiflora and it's for. erecta were ${\alpha}$-pinene, ${\beta}$-phellandrene, myrcene, and terpinolene. Major monoterpene components of P. densi-thunbergii were ${\beta}$-pinene, ${\beta}$-phellandrene, ${\alpha}$-pinene, and terpinolene. Major monoterpene components of P. thunbergii were ${\beta}$-pinene, ${\alpha}$-pinene, ${\beta}$-phellandre, and limonene. Monoterpene components in P. densiflora, P. densi-thunbergii, P. thundergii, and P.densiflora for, erecta showed a range of variation by the individual trees. Monoterpene contents of P. densi-thunbergii were intermediate between those of P. densiflora and P. thunbergii. ${\alpha}$-pinene, ${\beta}$-pinene, and myrcene appeared to be the best taxonomio characteristics for separating P. densiflora from P. thunbergii, and the former had higher content of ${\alpha}$-pinene than ${\beta}$-pinene while those of the latter were vise versa. There was a significant difference of ${\alpha}$-pinene components of P. densiflora between that of Anmyun Island and Mt. Chiak, but it seemed to be caused by the difference of the individual trees rather than provenancial difference.

Purification and Characterization of a Thermostable ${\beta}-Glycosidase$ from Thermus caldophilus GK24

  • A ${\beta}-glycosidase$ enzyme with $\beta$-D-fucosidase, ${\beta}-D-galactosidase$, and $\beta$-D-glucosidase activities has been purified from Thermus caldophilus GK24. The enzyme was monomeric with a molecular mass of 49 kDa, as evidenced by SDS-PAGE. The $K_m$ values for p-nitrophenyl ${\beta}-D-fucopyranoside$ (p-NPFuc), p-nitrophenyl ${\beta}-D-galactopyranoside$ (p-NPGal), and p-nitrophenyl ${\beta}-D-glucopyranoside$ (p-NPGlu) were 0.23 mM, 6.25 mM, and 0.28 mM, respectively. The enzyme showed optimal pH ranging between 5.5-6.5 and maximum temperature in the range of $85-90^{\circ}C$ for all the above mentioned activities. The half-life of the enzyme in sodium phosphate buffer (pH 6.0) at $80^{\circ}C$ was approximately 7 h. The p-NPGal hydrolyzing activity of Tca ${\beta}-glycosidase$ was strongly activated by L-histidine, while the p-NPFuc and p-NPGlu hydrolyzing activities of Tca ${\beta}-glycosidase$ were not affected at all by the amino acid. These results suggest differences in the conformation or in the reactive residues at the active site of Tca ${\beta}-glycosidase$.
