원유를 두유에 첨가한 혼합액으로 Mozzarella Cheese Analogue 제조에 관한 연구 (Production of a Mozzarella Cheese Analogue by Using Raw Milk and Soy Milk Blends)

  • The study was conducted to evaluate the feasibility of Mozzarella cheese analogue generation by using a mixture of soy milk and raw milk and to compare the quality of the resultant cheese with that of Mozzarella cheese manufactured using the traditional method. The mixtures showed increase in protein and decrease in lactose and SNF in a dose-dependent manner with the addition of soy milk. The Mozzarella cheese analogue had lower total solids content than the control cheese product, while the fat content was similar between both. The analogue cheese had lower ash content than the traditionally prepared cheese; the content was proportional to the amount of soy milk in the mixture. Higher soy milk quantities within mixtures also resulted in proportionally higher levels of fat content within analogue cheese. Water-soluble nitrogen content was lower in the analogue cheese than in the control cheese. While the WSN level increased in the control cheese, it was almost constant in the analogue cheese. The control cheese had much higher actual and predicted yield than the analogue cheese, while the analogue cheese had a higher stability level. The control cheese had a higher transfer rate than the analogue cheese, with the exception of lactose. Electrophoresis analysis showed bands for Mozzarella cheese analogues that were present in addition to the normal ${\alpha}$-casein and ${\beta}$-casein bands. Physical characteristic analysis showed that hardness was affected by the addition of soy milk to cheese, while cohesiveness and brittleness were affected by the addition of raw milk, and elasticity was barely affected by milk composition. The meltability of the control cheese was higher than that of the analogue cheese and increased during 30 days of storage at $4^{\circ}C$. Browning, oiling-off, and stretching characteristics were almost identical between the 2 types of cheeses. Sensory evaluation findings showed that the control cheese had much better body texture, appearance, and flavor than the analogue cheese.

  • 다양한 건강 증진 및 의료적인 능력을 가진 영양 식품이 많은 관심을 받아 오고 있다. 이러한 이유 때문에 kefir가 다른 발효유제품보다 소비가 증가되고 있는 실정이다. 왜냐하면 기능성 식품 및 치료적인 활력이 다른 발효유제품보다 매우 높은 kefir 및 kefir 관련 제품이기 때문이다. Kefir의 유익한 건강 특성은 단백질, 비타민, 산화 방지제, 미네랄, 특정 생물기원 물질에 기인한다. Kefir와 관련된 건강 관련 특성으로는 위장 촉진, 항균 능력, 항암 효과, hypocholesterolemic 효과, 항당뇨병 효과, 항돌연변이 활성, ${\beta}$-galactosidase 활력, scavenging 활력, 유산 함량, 지질 및 혈압 제어 효과, 세포 사멸에 대한 보호, 항알레르기 특성, 항염증 작용, 세균 정착능, 면역 시스템 활성 등이 알려져 있다. 따라서 kefir는 장내 미생물을 정상화 할 수 있으며, 유아부터 성인까지 매우 적합한 발효유제품이다. Kefir는 제품 제조 시 스타터 균주로서 사용되는 독특하고 다종의 자연적인 kefir grain 때문에 특유의 발효유제품이다. Kefir의 미생물학적 및 화학적 조성은 고유의 유산균과 효모에 의한 복합적인 probiotics 효과를 제공한다. 발효 중에 생성되는 대사 물질들은 약효 식품(neutraceutical) 활력이 입증되었다. 이러한 긍정적인 건강 속성은 kefir는 매우 가치 있는 식품에 근거해서 추가적인 실험과 임상 연구를 통해서만 가능할 것으로 사료된다.

산양유의 조성과 그 식품영양학적 의의 (Major Components of Caprine Milk and Its Significance for Human Nutrition)

  • 산양유는 단백질 구성과 개별 지방산의 구성이 모유에 가깝기 때문에 우유에 비하여 소화율이 높다. 특히 casein 단백질은 모유의 단백질과 분자구조가 흡사하여 높은 소화 흡수력을 보이므로 설사 또는 복통 등과 같은 소화장애를 일으키지 않는다. 뿐만 아니라 지방산 함량에 있어서도 우유보다 중간사슬지방산이 우유보다 2배정도 많이 들어있어 소화 흡수 시 분해되는 속도가 빠르다. 산양유에는 질 좋은 수용성 유청단백질, 비타민 및 미네랄이 다량 함유되어 있고, 특히 비타민 A, 콜린, 이노시톨의 함량이 높고 흡수율도 양호한 편이다. 산양유에 포함된 유당과 올리고당은 우유보다 모유에 더 가까운 효과를 기대할 수 있다. 유당은 장 내 산성을 증가시켜 유익한 박테리아의 증식을 돕는 한편 장내에서의 칼슘의 흡수를 도와주거나 인(P)과 마그네슘(Mg)의 흡수를 촉진시키는 역할을 담당한다. 또한 올리고당류는 장내유산균의 생육을 돕는 prebiotic 효과를 발휘하여 장내 유익균이 정착하는데 도움을 준다는 실험결과로 판단할 때 실제로 유아에게 정장작용을 기대할 수 있다. 그리고 산양유에 다량 함유된 미네랄 성분 또한 소화 및 흡수를 촉진시키는 역할을 하기 때문에 산양유는 우유 섭취 시 유당불 내증을 호소하던 소비자들도 어렵지 않게 섭취할 수 있다. 이처럼 산양유가 여러 가지 측면에서 우유보다 우수한 식품학적 생리적 특성을 가지고 있음에도 불구하고 국내 산양유가공 산업은 거의 불모지라 해도 과언이 아니다. 게다가 산양유의 정확한 영양학적 가치가 일반 소비자들에게 전달되지 못하고 있는 실정이다. 따라서 우유보다 그 성분과 조성이 모유와 흡사한 산양유의 식품학적 특성, 안전성 및 기능성에 대한 체계적이며 과학적 연구가 시급히 활성화되어야 할 것이다.

Escherichia coli O157 : H7, Staphyloccus aureus 및 Candida albicans에 대한 키토산의 항균 효과 (Antimicrobial Effects of Chitosans on Escherichia coli 0157 : H7, Staphyloccus aureus and Candida of albicans)

  • 분자량이 상이한 세 종류의 키토산의 항균활성을 E. coli O157 : H7, S. aureus 및 C. albicans 균주를 이용하여 측정, 분석하였다. E. coli O157 : H7와 S. aureus에 대해서는 분자량 10,000인 키토산이 가장 강한 항균활성을 보였으며 C. albicans에 대해서는 6량체의 키토산 올리고당이 가장 강한 활성을 나타내었다. 키토산 첨가농도는 E. coil O157 : H7와 S. aureus의 경우 0.1 mg/mL의 농도에서, C. albicans의 경우는 chitohexaose 1 mg/mL의 농도에서 항균활성이 가장 높았다. 모든 키토산 처리구에서 미생물 사멸 속도는 키토산 처리 1시간 이내에서 가장 높게 나타났으며 그 이후로는 점차 낮은 속도를 보였다. 사멸되었거나 파손된 미생물 세포로부터 유래되는 단백질, 핵산물질 및 $Ca^{2+}$ 량은 키토산 처리 시간 이내에 가장 많았으며 ${\beta}-galactosidase$ 활성도 같은 시간대에서 가장 빠른 속도로 중대되는 것으로 나타났다. S. aureus에 대한 세포막 손상 정도를 측정하여 본 결과 전체 미생물균체중 약 10%에 상당하는 균수가 막손상을 가져 왔다. 따라서, 키토산은 고유의 양이온성 성질을 이용하여 미생물의 세포벽과 세포막에 결합하여 그 결과로 세포 내 물질의 세포 외로의 유출내지는 세포막 대사의 저해 등의 효과를 나타냄으로써 항균활성을 나타내는 것으로 추측되었다.

Dasania marina gen. nov., sp. nov., of the Order Pseudomonadales, Isolated from Arctic Marine Sediment

  • An obligately aerobic bacterium, strain KOPRI $20902^T$, was isolated from a marine sediment in Ny-${\AA}$lesund, Spitsbergen Islands, Norway. Cells were irregular rods and motile with polar monotrichous flagellum. The optimum growth temperature was $17-22^{\circ}C$. Cells grew best in pH 7.0-10.0 and 3-4% sea salts (corresponding to 2.3-3.1% NaCl). The novel strain required $Ca^{2+}$ or $Mg^{2+}$ in addition to NaCl for growth. Sequence analysis of 16S rRNA gene revealed that the Arctic isolate is distantly related with established species (<92.4% sequence similarity) and formed a monophyletic group with Cellvibrio, which formed a distinct phylogenetic lineage in the order Pseudomonadales. Predominant cellular fatty acids [$C_{16:1}\;{\omega}7c/15:0$ iso 2OH (45.3%), $C_{16:0}$ (18.4%), ECL 11.799 (11.2%), $C_{10:0}$ 3OH (10.4%)]; DNA G+C content (37.0 mol%); nitrate reduction to nitrogen; absence of aesculin hydrolysis, N-acetyl-${\beta}$-glucosaminidase and esterase; no assimilation of arabinose, galactose, glucose, lactose, maltose, and trehalose differentiated the strain from the genus Cellvibrio. Based on the phylogenetic and phenotypic characteristics, Dasania marina gen. nov., sp. nov. is proposed in the order Pseudomonadales. Strain KOPRI $20902^T$ (=KCTC $12566^T$=JCM $13441^T$) is the type strain of Dasania marina.

Effects of Using Monensin and Different Levels of Crude Protein on Milk Production, Blood Metabolites and Digestion of Dairy Cows

  • Twenty-four Holstein dairy cows were used to evaluate the single and combined effects of different levels of crude protein (CP) and monensin treatment during early lactation on blood metabolites, milk yield and digestion of dairy cows. The experiment was designed as a completely randomized block with a $3{\times}2$ factorial arrangement of treatments. The factors were three concentrations of CP supplement (19.5, 21.4, and 23.4% of dry matter) and two levels of monensin (0 and 350 mg per cow per day). The experiment consisted of three phases and each phase was 3 wk in length. Monensin did not affect milk yield, lactose, solids-non-fat (SNF), blood glucose, triglyceride and DMI, but increased blood cholesterol, blood urea nitrogen (BUN), insulin and reduced blood ${\beta}$-hydroxybutyrate (BHBA), milk fat and protein percentage. Monensin premix significantly decreased rumen ammonia, but rumen pH and microbial protein synthesis were not affected by monensin treatment. Increasing dietary CP improved milk and protein production, but did not alter the other components of milk. Digestibility of NDF, ADF, CP were improved by increasing dietary CP. Increasing dietary CP from 19.5 to 21.4% had no significant effect on ruminal ammonia, but increasing CP to 23.4% significantly increased ruminal ammonia. There was a linear relationship between level of crude protein in the diet and volume of urine excretion. Microbial protein synthesis was affected by increasing CP level; in this way maximum protein synthesis was achieved at 23.4% CP.

오메프라졸-에칠렌디아민 복합체를 이용한 제제설계 (Formulation of Omeprazole Preparations using Omeprazole-Ethylendiamine Complex)

  • The study was carried out to develop useful formulation for omeprazole(OMP) through OMP-ethylendiamine complex(OMPED), and the pharmaceutical properties of formula were tested to find out the difference in vivo behaviors of formulations between the free and complexed OMP. Oral and suppository dosage forms were also formulated and the dissolution profiles and pharmacokinetic parameters were measured to observe the difference in bioavailability between the free and complex form, and the correlation between dissolution rate and bioavailability was evaluated. The results are summarized as follows; In the case of formulation for oral administration, the release of OMP from enteric OMPED pellets was found satisfactory to the requirement standard and no decomposition of OMP in the pellets was found in acidic solution. Therefore the enteric OMPED pellets are anticipated to be a stable formulation. The release of OMP from OMPED tablet with chitosan as excipient and coated with cellulose acetate phthalate was found to be significantly retarded. The results of bioavailability test for OMP and OMPED tablets with lactose-excipient showed that the AUC value of OMP tablet was $116.89\;{\mu}g\;{\cdot}\;min/ml$, that of OMPED tablet was $161.10\;{\mu}g\;{\cdot}\;min/ml$, respectively. The reason why was thought that OMP decomposes more readily in body than OMPED, and the AUC of the tablet with chitosan-excipient and coated with cellulose acetate phthalate was most enhanced. In the case of bioavailability for suppositories with OMP, $OMP-{\beta}\;-cyclodextrin$ complex and OMPED, the AUC of OMPED suppository was most increased. From the above results, it is thought that the more stable and bioavailable oral or rectal dosage forms could be developed by using the OMPED as a potential OMP complex.

Effects of Exogenous Bovine Somatotropin on Mammary Function of Late Lactating Crossbred Holstein Cows

  • The objective of the present study was to determine the effect of exogenous bovine somatotropin on the mammary function in late lactating crossbred Holstein cows. Twelve 87.5% late lactating Holstein cows, approximately 30 weeks postpartum, were divided into two groups of 6 animals each. Animals in the control group were given sodium bicarbonate buffer by subcutaneous injection, while animals in the treated group were given recombinant bovine somatotropin (bST) by subcutaneous injection with 500 mg of bST (14 day prolonged-release bST). After bST injection, milk yield significantly increased from the control level on day 8 to day 20 (p<0.05) with a concomitant increase in mammary blood flow (p<0.01). An increase in mammary blood flow in response to bST treatment was greater than an increase in milk production. An increased plasma concentration of IGF-I coincided with an increase in mammary blood flow in animals treated with bST. There were no significant changes in the concentration of arterial plasma glucose concentration, the arteriovenous concentration difference (A-V difference) and mammary extraction ratio while the mammary glucose uptake increased when compared to the control group. The concentration of arterial plasma triglyceride decreased throughout the experimental period in animals give bST. The plasma concentration of acetate, and the mammary uptake for acetate significantly increased (p<0.05) after bST treatment. The action of bST did not affect the plasma concentration, A-V difference and extraction ratio across the mammary gland for $\beta$-hydroxybutyrate. The concentrations of milk fat and lactose tended to increase during bST treatment. Milk protein concentration initially increased in the first few days and decreased after bST injection when compared to the pretreated period. The present results indicated that bST could affect the mammary function in late lactating cows by increase in milk yield involving changes in both extra-mammary and intra-mammary mechanisms. The exogenous bST exerted its galactopoietic action through an increase in circulating IGF-I of the late lactating Crossbred Holstein cattle.

API STAPH system을 이용한 돼지, 닭 및 소유래 Staphylococcus hyicus subsp. hyicus의 동정 (Identification of Staphylococcus hyicus subsp. hyicus of swine, poultry and bovine origin with the API STAPH system)

  • The API STAPH system was compared with conventional methods for identification of 214 strains of Staphylococcus hyicus subsp. hyicus isolated from cases of exudative epidermitis in piglets, skin of healthy pigs, skin of healthy chickens and bovine intramammary infections, and biochemical characteristics among the swine, avian and bovine strains were also compared. All of the swine and bovine strains produced acid within 24 hours from fructose, lactose and trehalose by conventional methods, but some of the avian strains showed a delayed positive reaction in these carbohydrates. These delayed positive strains in conventional methods gave usually negative results for them in the API STAPH system. With the API STAPH system, eighteen different profile numbers were encountered in 214 strains of swine, avian and bovine origin. The swine and bovine strains, respectively, were distributed among 4 profiles, while the avian strains were distributed among 17 profiles. The profile number observed most frequently in the strains of each animal species was uniformly 6 516 153. By conventional methods, approximately 96% of the swine strains were positive for ${\beta}$-glucuronidase, but not in any strains from chickens and cattle. For hyaluronidase production determined by degradation of sodium hyaluronidate in a solid culture medium, all the swine and bovine strains were positive, but only 37.5% of the avian strains were positive for it. From these findings, there were differences in the production of extracellular active substances between swine strains of Staphylococcus hyicus subsp. hyicus and those isolated from chickens and cattle.

Systematic Review on Application of Whey Towards Production of Galacto-oligosaccharide Using β-Galactosidase Enzyme from Pichia pastoris

  • Galacto-oligosaccharides(GOS)는 프로바이오틱스 미생물의 성장을 증진시켜 인류 건강에 유익한 작용을 갖게 하는 프리바이오틱스이며 식품 산업에서 다양한 활용성을 갖는다. GOS는 보통 β-galactosidase에 의해 촉매 반응이 일어난 lactose로부터 생성된다. 한편, 세포 표면 발현은 살아있는 세포 표면의 펩타이드와 단백질을 세포의 기능성 성분에 융합시켜 발현시키는 것이다. 표층 발현 세포는 다양한 잠재적 이용가치를 갖는다. N 말단 부근에 위치하는 것으로 생각되는 Flo1p 응집 functional domain은 세포의 flocs로의 가역적인 응집을 유발하면서 α-mannan carbohydrates와 같은 세포벽 성분과 비공유결합을 한다. 한외여과한 유청을 농축, 분무건조한 유청막투과액(Whey Permeate, WP)을 이용하여 β-galactosidase 재조합 Pichia pastoris (P. pastoris) 로 표층 발현 처리 (surface engineering)하는 GOS의 합성법은 폐기물을 활용하는 새로운 효율적인 방법이라 할 수 있다.
