Effects of Paprika (Capsicum annuum L.) on Serum Lipid Profile and $\alpha$-Tocopherol Concentration in Rats Fed a High-Cholesterol Diet (파프리카 급여가 고콜레스테롤 식이 흰쥐의 혈청 지질과 $\alpha$-Tocopherol 농도 변화에 미치는 영향)

  • In conclusion, rats were fed diets containing either NC, HC, HC-FDP, HC-WEP, or HC-EEP for 6 weeks. At the 6th week, increases in body weight and visceral fat were lower in the paprika fed groups as compared to the HC group. Serum levels of triglycerides, total cholesterol, low density lipoprotein cholesterol, and atherogenic index values were significantly lower in the paprika diet fed groups than the HC group (p<0.05). In particular, the lipid lowering effects in the HC-EEP group were superior among the paprika fed groups. Also, serum $\alpha$-tocopherol levels were lower in the control group compared to the paprika fed groups. The supplementation of paprika may exert lipid lowering effects and saving effect of $\alpha$-tocopherol in the serum of high-cholesterol diet fed rats. However, it should be noted that the results are based on very small sample numbers and a short experimental period.

  • Starch solid model system was employed to investigate the effect of lipoxygenase, ${\beta}-carotene$, ${\alpha}-tocopherol$ and water activity on the oxidation of linoleic acid. The rate of oxidation of linoleic acid by lipoxygenase was increased with the increase in water activity, Addition of ${\beta}-carotene$ and ${\alpha}-tocopherol$ to this system has been shown to inhibit the oxidation of linoleic acid and ${\alpha}-tocopherol$ was more effective antioxidant than ${\beta}-carotene$. However, an increase in the concentration of ${\beta}-carotene$ was found to have a strong antioxidant effect in the solid model system. And also the antioxidative action of ${\beta}-carotene$ was increased with increasing water activity in this system.

Effects of Korean Ginseng and Tocopherol on Testosterone Content, Protein Metabolism and Stamina Promotion in Rats (흰쥐의 Testosterone 함량(含量), Protein 대사(代謝) 및 스테미너 증진(增進)에 미치는 인삼(人蔘)과 Tocopherol의 영향(影響))

  • This experiment was conducted to study the effects of Korean ginseng and tocopherol on serum testosterone level, protein metabolism and swimming endurance in rats. Powdered ginseng (2.5 g/kg) and ${\alpha}-tocopherol$ (250 mg/kg) were added directly into the basal diet of rats feeding for 6 weeks. The serum testosterone level of ginseng-treated rats had no difference from that of control rats, but the level was slightly increased in tocopherol-treated rats. In the case of ginseng-treated rats, there were no significant difference in the contents of albumin, ${\alpha}_1-globulin$, ${\alpha}_2-globulin$, ${\gamma}-globulin$ and A/G ratio compared with those of the control rats, however, the levels of total protein and ${\beta}-globulin$ were increased significantly(p<0.005). Total protein value of tocopherol-treated rats was higher than that of the control rats(p<0.005). In contrast, albumin content was slightly decreased. The contents of ${\alpha}_1-globulin$, ${\alpha}_2-globulin$, ${\beta}-globuiln$ and ${\gamma}-globulin$ were higher in the rats fed tocopherol diet that those obtained from the control rats. Accordingly A/G value was markedly decreased (p<0.001). In swimming endurance test, both Korean ginseng and ${\alpha}-tocopherol$ did not appear to have any effect on rats' stamina.

Effect of Dietary Vitamin E Supplementation on Serum α-Tocopherol and Immune Status of Crossbred Calves

  • An experiment was conducted with twenty crossbred male calves (7-15 days old) divided into 4 different experimental groups on the basis of body weights to study the effect of vitamin E supplementation on the serum ${\alpha}$-tocopherol concentration and immune response of the calves. All the calves were fed on milk and calf starter up to 13 weeks and afterwards, they were fed on concentrate mixture and oat hay up to 32 weeks of age. In addition, the calves in groups I, II, III and IV were supplemented with 0, 125, 250 and 500 IU feed grade DL-${\alpha}$-tocopheryl acetate, respectively. Blood samples were collected at 0 day and subsequently at 1, 2, 3, 4, 6 and 8 months of age to monitor the serum ${\alpha}$-tocopherol concentration in crossbred calves. After 24 weeks of experimental feeding, 4 animals from each group were intramuscularly inoculated with single dose (3 ml) of Haemorrhagic septiceaemia (Pasteurella multocida P52 strain) oil adjuvant vaccine. The cumulative group mean serum ${\alpha}$-tocopherol concentration (${\mu}g/100ml$) was 88.12, 210.11, 235.21 and 294.02 in-groups I, II, III and IV, respectively and differed significantly (p<0.001) among the four groups. Lymphocyte stimulation indices (LSI) did not differ among the groups significantly. The pooled mean ELISA antibody titer against Pasteurella multocida (P52 strain) was 788.02, 926.85, 1,214.00 and 1,109.51 for group I, II, III and IV, respectively, which indicated higher antibody titer in groups supplemented with vitamin E as compared to the control group. It may be concluded that vitamin E supplementation increased the ${\alpha}$-tocopherol concentration in serum and dietary supplementation of vitamin E at higher level has a humoral immune enhancing effect against killed bacterial antigen.

Preparation of Valuable Compounds Encapsulated Polymer Nanoparticles with High Payload Using Core-crosslinked Amphiphilic Polymer Nanoparticles (코아 가교 양친성 고분자 나노입자를 이용한 고함량 유용 약물 담지 고분자 나노입자 제조)

  • In this study, core-crosslinked amphiphilic polymer (CCAP) nanoparticles prepared using a reactive amphiphilic polymer precursor (RARP) were used for preparing some valuable compounds encapsulated polymer nanoparticles with high payload through nanoprecipitation process. Various solvents (acetone, ethanol, and THF) having different polarity and CCAP nanoparticles prepared using different amphiphilicity were used for the preparation of ${\alpha}$-tocopherol encapsulated polymer nanoparticles to investigate their effects on the encapsulation efficiency, payload, nanoparticle size, and stability. CCAP dissolved in hydrophobic solvent, THF, could form ${\alpha}$-tocopherol encapsulated polymer nanoparticles dispersed in water with the high payload of ${\alpha}$-tocopherol and encapsulation efficiency. Because of their physically and chemically robust nano-structure originated from crosslinking of the hydrophobic core, CCAP nanoparticles could encapsulate ${\alpha}$-tocopherol with the high payload (33 wt%) and encapsulation efficiency (97%), and form 70 nm-sized stable nanoparticles in water.

Deodorization of Fish Oil Using Adsorption Method (흡착법을 이용한 어유의 탈취)

  • Instead of deodorization appararus of fish oil, an adsorbent such as activated charcoal, activated alumina, silicagel, bamboo charcoal was packed in column alone or mixed with preparative ratio, and then test the effective deodorization with bleaching. In the progress of degumming, the effective method was 18 ml of 2.5% oxalic acid per 100m1 of crude large anchovy oil. The optical condition to deacidified was treating for 30 min at $40^{\circ}C$ with 2.5% sodium hydroride solution. The effective deodorization was added with 3% silicagel under the alone treating adsorbent, and mixed treating was 30% activated alumina and 10% silicagel but added to green tea powder was not effective. The major fatty acid of total lipid were 16:0, 20:5n-3, 18:1n-9, 16:1n-7 and 22:6n-3 after treatment of degumming, deacidfication and deodorizing in the large anchovy oil. The oxidative stability of refined anchovy oil added to $\alpha$-tocopherol was validated 20 days under the control, and 30 days in the case of $\alpha$-tocopherol. The 0.01% $\alpha$-tocopherol was more effective than 0.02% $\alpha$-tocopherol.

  • The aim of this study was to evaluate the effect of ${\alpha}$-tocopherol on blastocysts development and subsequent cryosurvival of the vitrification. The ${\alpha}$-tocopherol(0, 100, 200, 400 ${\mu}M$) was added in to culture medium for the bovine embryos. The blasocysts from the ${\alpha}$-tocopherol and untreated control groups were then frozen-thawed, and their cryosurvival was assessed by in vitro culture for 48 h. There were no differences in the overall cleavage rate($56.14{\pm}4.66$, $58.18{\pm}4.70$, $62.97{\pm}6.86$ and $51.17{\pm}7.28$) among four treatment groups. However, in blastocyst development and total cell number were significantly higher in ${\alpha}$-tocopherol 200 ${\mu}M$ ($38.60{\pm}7.12$; $106.33{\pm}3.50$) to culture medium than other treatment groups($29.30{\pm}5.24$, $31.60{\pm}7.12$ and $26.37{\pm}4.18$; $101.36{\pm}5.12$, $97.27{\pm}2.87$, and $91.23{\pm}7.52$ respectively). Before and after vitrification, the total cell number and blastocyst development of embryo were significantly higher in July to August than September to October. In conclusion, addition of ${\alpha}$-tocopherol 200 ${\mu}M$ to in vitro bovine embryo culture medium was beneficial for improving embryo quality by decreasing the embryo damage blsstocysts cell number and improving the tolerance of the embryos to cryopreservation.

Analysis of Serum Antioxidant Materials Concentration and Their Relation with Blood Lipids and Anthropometric Indices in Middle-Aged Adults in Korea (중년기 성인의 혈중 항산화 물질 분석 및 혈중 지질과 신체계측치와의 상관성)

  • The serum levels of antioxidant materials (${\alpha}$-carotene, ${\beta}$-carotene, ${\beta}$-cryptoxanthin, lutein, lycopene, ${\alpha}$-tocopherol, ${\gamma}$-tocopherol, retinol) of the healthy Korean middle-aged adults (n = 373) were measured and their relationships with the serum lipids and anthropometric indices were analyzed. The serum levels of ${\beta}$-cryptoxanthin, ${\beta}$-carotene, lutein were higher than those of lycopene and ${\alpha}$-carotene. The levels of all measured carotenoids except lutein were significantly higher in females than in males, but retinol level was vice versa. There was a tendency of increase in serum levels of antioxidant vitamins with increasing age. The serum carotenoid levels had a positive correlation with serum cholesterol and a negative correlation with serum triglyceride. The serum levels of tocopherols or retinol showed a significantly positive relationship with blood cholesterol or triglyceride. In overall, anthropometric indices showed negative relations with serum carotenoids levels, but vice versa with serum tocopherol or retinol levels. Particularly, ${\beta}$-carotene and lutein levels showed a significantly negative relation with blood pressure in male subjects. The serum levels of ${\alpha}$-carotene, ${\beta}$-carotene and ${\beta}$-cryptoxanthin had significantly negative relations with body fat-related indices in female subjects. The results demonstrated that blood antioxidants levels differed by sex and age, and had significant relations with blood lipid levels and anthropometric indices. Therefore, the rationale and significance of the relationships need to be elucidated in the future study related to dietary intakes and life style.

Status of Antioxidant Enzymes in Normal Cycling and α-Tocopherol Supplemented Anestrus Buffalo Heifers (Bubalus bubalis)

  • The present investigation was undertaken to study status of erythrocytic antioxidant enzymes in normal cycling and $\alpha$- tocopherol supplemented anestrus buffalo heifers. The pre-supplementation erythrocytic activities of superoxide dismutase (U/mg Hb), glutathione peroxidase (U/mg Hb) and glucose-6-phosphate dehydrogenase (U/g Hb) upregulated significantly (p<0.05) in anestrus heifers ($10.08{\pm}0.09$, $14.09{\pm}0.54$, $9.25{\pm}0.29$) when compared to normal cycling ones ($6.93{\pm}0.04$, $11.61{\pm}0.19$, $5.58{\pm}0.26$). The oral supplementation of $\alpha$-tocopherol (a) 3,000 mg per week per animal in anestrus heifers declined erythrocytic superoxide dismutase and glucose-6-phosphate dehydrogenase activities significantly (p<0.01) but led to non-significant increase in erythrocytic glutathione peroxidase activity. Results indicated that supplementation of $\alpha$-tocopherol to anestrus buffalo heifers mitigated the effects of oxidative stress to improve their antioxidant status.

Influences of $\alpha$-Tocopherol on the Toxicity of Vitamin A Acetate in Rats (랏트에 있어서 Vitamin A Acetate의 독성에 미치는 $\alpha$-Tocopherol의 영향)

  • Influences of u-tocopherol on the toxicity of vitamin A acetate in male rats were studied. The obtained results are as follows; 1) The administration of vitamin A acetate 500,000 IU/Kg i.p. twice at 3 days interval decreased the liver weight/body weight and increased the spleen weight/body weight, and increased activities of SGOT and alkaline phosphatase, and also increased BUN and creatinine. 2) ${\alpha}$-Tocopherol administered together with vitamin A acetate as given as the above 1) poteniated the increase of SGOT activity caused by vitamin A acetate and reduced the increase of alkaline phosphatase activity and creatinine which were caused by vitamin A acetate. 3) The administration of vitamin A acetate 500,000 IU/Kg i.p. twice a week for 4 weeks showed remarkable decrease of body weight gain and the effect of it was larger in later stage than in early. It increased significantly liver weight/body weight and further increased the activities of SGOT, SGPT and alkaline phosphatase, and showed no influnence on BUN and creatinine. 4) ${\alpha}$-Tocopherol administered together with vitamin A acetate as given as the above 3) reduced the decrease of body weight gain caused by vitamin A acetate, and potentiated remarkably the increased activities of SGOT and alkaline phosphatase which were caused by vitamin A acetate.
