$\alpha$-tocopherol의 항산화작용에 미치는 인삼사포닌분획의 영향 (The Effect of Ginseng Saponin Fraction on Antioxidant Activity of $\alpha$-Tocopherol)

  • 인삼 사포닌 분획이 $\alpha$-tocopherol의 항산화작용에 미치는 영향을 시헌관내 실험과 동물실험으로 관찰하였다. 흰쥐 (Wister, 수컷, 180~200g)의 간 미크로좀 분획을 $Fe^{3+}$, NADPH, ADP, $\alpha$-tocopherol($50{\mu}M}$), 인삼 사포닌 분획(최종농도 $10^{-4}$%, 대조군은 사포닌 분획 대신 같은 부피의 물)을 함유한 혼합물에서 반응을 진행시키고 생성된 malondialdehyde를 분석한 결과, 사포닌은 $\alpha$-tocopherol의 항산화작용을 상승적으로 증가시켰다. 인삼 saponin 분획이 함유된 $\alpha$-tocopherol의 alcohol용액을 쥐에게 경구투여한 후 적당 시간 후에 간에서의 과산화수소량을 분석하여 대조군과 비교한 결과, 인삼 saponin 은 $\alpha$-tocopherol의 흡수를 촉진하므로서 $\alpha$-tocopherol의 항산화활성이 더욱 효율적으로 일어나게 하는 것으로 해석된다.

Alpha-Tocopherol Contents of Peanut Seeds Depend on Varieties and Processing Methods

  • The purpose of this study is to establish an extraction and analysis method for $\alpha$-tocopherol, and to then distinguish among varieties. The $\alpha$-tocopherol contents of 22 varieties of peanut seeds were analyzed by HPLC. Peanut seeds of cv. Palkwang were processed in two ways, by boiling and roasting. The $\alpha$-tocopherol contents of the two types of peanuts were compared with fresh seeds without any processing. $\alpha$-Tocopherol was detected at a retention time of 2.95 minutes. Five standards of $\alpha$-tocopherol covering a range of $20{\sim}100{\mu}g/ml$ were made up in 2% isopropyl alcohol/n-hexane and analyzed in duplicate. The $\alpha$-tocopherol contents differed according to extraction temperature. The contents were $85{\mu}g/g$ or less at $10^{\circ}C\;and\;20^{\circ}C$ and $94{\mu}g/g$ at $30^{\circ}C$, but they were decreased at $40^{\circ}C$ or higher. The $\alpha$-tocopherol contents in 22 peanut varieties were $61.36{\sim}96.80{\mu}g/g$ according to variety. Fresh peanuts contained $106.7{\mu}g/g$ of $\alpha$-tocopherol, while boiled peanuts contained $108.8{\mu}g/g$ of $\alpha$-tocopherol, and roasted peanuts contained $109.2{\mu}g/g$ of $\alpha$-tocopherol.

  • 한국산 인삼(Panax ginseng C. A. Meyer) 뿌리에서 추출한 사포닌 성분이 ${\alpha}$-tocopherol의 항산화 활성에 미치는 영향을 실험관 내 실험과 동물실험으로 관찰하였다. 흰쥐(Sprague-Dawley, male, $18{\~}200g$)의 마이크로좀 분획을 NADPH, $Fe^{+3}$, ATP $\alpha^{-+}$-ocopherol 과 인삼 사포닌 분획 (대조군은 사포닌 분획대신 같은 부피의 물)을 함유한 혼합물에서 30분간 반응을 진행시키고 생성되는 molondialdehyde를 분석한 결과 사포닌 분획은 ${\alpha}$-tocopherol의 항산화작용을 상승적으로 증가시켰다. 그리고 ${\alpha}$-tocopherol의 항 산화작용에 상승적 효과는 반응액에서의 인삼 사포닌 분획의 농도가 $10^{-5}\%$ 전후에서 특히 현저하였다. 술로 과산화물을 유발케 한 쥐에게 ${\alpha}$-tocopherol만을 또는 ${\alpha}$-tocopherol과 인삼 사포닌 분획의 혼합물을 경구투여하고 일정 시간 후에 간의 과산화 지질을 thiobarbituric acid 방법으로 분석한 결과 사포닌 분획이 동물체내에서도 ${\alpha}$-tocopherol의 항 산화작용을 크게 촉진시키는 것으로 관찰되었다. 또한 사포닌 분획은 ${\alpha}$-tocopherol의 흡수를 촉진시키는 것으로 보고되고 있으며 본 실험에서도 이것을 확인하였고 이와 같은 사포닌 분획의 ${\alpha}$-tocopherol흡수 촉진은 ${\alpha}$-tocopherol의 항 산화작용을 더욱 효과적으로 촉진 시킬 것으로 생각된다.

Characterization of Thermal Products of Alpha-Tocopherol

  • Alpha-tocopherol was thermally oxidized and degraded at high temperatures and the resulting products were chromatographically separated and identified by LC-MS. Alpha-tocopherol dissolved in glycerol was heated at 200^{\circ}C for 30 min. The thermal products were separated by hexane extraction and analyzed by HPLC using a reversed phase $\mu$-Bondapak $C_{18}-column$ with two kinds of elution solvents: a mixture of acetonitrile and methanol (3:2), and of acetonitrile, methanol, 2-propanol, chloroform and methylene chloride (3:2:5:0.5:0.5) in a gradient mode. The isolated thermal. products of alpha-tocopherol were more viscous than alpha-tocopherol, and dark brown in color. Major thermal degradation products of alpha-tocopherol were identified by LC-MS, and the structures of thermal products were proposed. Alpha-tocopherol and its thermal degradation products were degraded into fragments, mainly at the non-aromatic parts. The degradation products of alpha-tocopherol. were combined with oxidized product (tocopherylquinone) to make thermal. products through dimerization.

  • The concentrations of $\alpha$-tocopherol in the brain, liver, and serum were studied with and without saponification process between control and vitamin E supplemented rats. Young rats, 80-120g body weight, were fed control and vitamin E supplemented diets, ad libitum, for four weeks. $\alpha$-Tocopherol concentrations were determined by high pressure liquid chromatography. The $\alpha$-tocopherol concentration per wet weight base in the brain tissue was significantly lower than that in the liver. Vitamin E supplementation had no effect on brain $\alpha$-tocopherol levels in contrast to the significant increase in lover $\alpha$-tocopherol concentration with and without saponification is significantly greater in the brain than in the liver or serum. Further study is needed to clarify the nature of interaction or /and binding between $\alpha$-tocopherol and the complex membrane system in brain tissue. It can be speculated from this and other studies that the metabolism and the nature of interaction of $\alpha$-tocopherol with the complex membrane system in brain tissue rich in polyunsaturated fatty acids seems different from that in liver tissue or serum.

  • liposome에 결합되어 있는 ${\alpha}-Tocopherol$의 형광을 이용하여 liposome의 상전이온도를 측정할 수 있었다. 이는 ${\alpha}-Tocopherol$의 vitamin E로서의 중요성 이외에 형광성 프로브로서의 새로운 가치의 발견이다. 형광성 프로브로서 그 응용의 확대를 위한 필수적인 기초자료의 수집을 위해, 분자분광학적성질을 조사하였다. 용액중의 ${\alpha}-Tocopherol$은 monomer와 dimer의 두가지 형태로 존재하며, 단파장흡수대$(291{\sim}294nm)$의 monomer는 형광성인데 반하여 dimer는 298nm 부근을 흡수하는 비형광성이고 분자간수소결합에 의하여 형성되었음이 밝혀졌다. liposome에 결합된 ${\alpha}-Tocopherol$은 각종 유기용매 중에서의 ${\alpha}-Tocopherol$과는 판이한 형광성질을 갖고 있었고 chromanolate이온형태의 화학종으로부터 생긴 것으로 해석되었다.

Effect of Allopurinol on the Ethanol-induced Oxidative Stress : Mechanism of Allopurinol Action

  • An acute ethanol load(50mmol/kg , i.p) resulted in an increase in peroxidation and a decrease in the levels of $\alpha$-tocopherol and ascorbate in rat cerebellum. Pretreatement with allopurinol(146$\mu$mol/kg, i.p) prevented the ethnol-induced increment in lipid peroxidation and decrease in $\alpha$-tocopherol content. However, the decrease of ascorbate was of greater magnitude when allopurinol was associated with ethanol. These results suggested that allopurinol. besides its action as a radical scavenger and xanthine oxidase inhibitor, might favor the regeneration of $\alpha$-tocopherol antioxidant acitviity was studied using ${\gamma}$-radiolysis in aerated ethanolic solutions. Even though allopurinol did not react by itself with $\alpha$-hydroxyethyl-peroxyl radicals [H3C-CH(OH)OO] , it enhance the $\alpha$-hydroxyethyl-peroxyl radical scavenging properties of $\alpha$tocopherol. The regeneration of $\alpha$-tocopherol from the $\alpha$-hydroxyethyl-peroxyl radical scavenging properties of $\alpha$-tocophero. The regeneration of $\alpha$-tocopherol from the $\alpha$-tocopherol radical by ascorbate remained as efficient in the presence of allopurinol as in its absence. The effects of allopurinol on the Vitamin E oxidation-reduction mechanism could be involoved in the beneficial effectof allopurinol on the biological cellular damages linked to free radical reactions.

한국 성인 남자의 혈장 Tocopherol 수준과 관련 요인 분석 (Association between Plasma Tocopherol Levels and Related Factors in Middle-Aged Korean Men)

  • Vitamin E in the body system plays an important role in preventing chronic diseases by decreasing the oxidative stress by free-radicals. However, there are not enough researches on analyzing the primary factors affecting vitamin E levels in the blood in Korean adults. Therefore, the purpose of this research was to examine blood tocopherol levels and the primary factors affecting the status. A complete lifestyle survey was performed on 314 Korean adult men and surveyed their smoking, drinking and exercising habits. The average plasma level of ${\alpha}-\;and\;{\gamma}-tocopherol$ showed similar mutual relations with plasma total cholesterol (TC), triglyceride (TG), or low density lipoprotein cholesterol (LDL-C) levels (p<0.001). Plasma ${\alpha}-tocopherol$ level of the subjects did not show any difference as smoking, drinking and exercising habits changed. However, ${\gamma}-tocopherol$ per TG showed much lower figure in smokers than non smokers (p < 0.05). Amongst diet factors, plasma ${\alpha}-tocopherol$ level showed negative correlations with Vitamin E intake, while ${\gamma}-tocopherol$ level showed positive correlations with Vitamin E intake. Erythrocyte superoxide dismutase (SOD) activity and plasma tocopherol showed negative correlations, and catalase activity and plasma ${\alpha}-tocopherol$ showed positive correlationship. The level of cell DNA damage of Iymphocyte and plasma ${\alpha}-\;or\;{\gamma}-tocopherol$ showed negative correlations. As a result of this research, the factors that affect Korean adult men's plasma ${\alpha}-tocopherol$ level are plasma TG, LDL-C and cell DNA damage in Iymphocyte, while the factors that affect ${\gamma}-tocopherol$ level are plasma TG, LDL-C and vitamin E intake based on multiple regression analysis. These findings implies that the level of different types of tocopherol depends on slightly different factors. A further research is needed on the factors involved in the differentiation of the types of tocopherol.

α-Tocopherol과 Selenium의 첨가 급여가 육계의 생산성 및 닭 다리육의 이화학적 특성에 미치는 영향 (Effects of Dietary Supplementation of α-Tocopherol and Selenium on Performance and Physicochemical Properties of Chicken Thigh Meat)

  • 본 실험은 ${\alpha}$-tocopherol(0, 50, 100, 200 IU/kg)과 selenium(0.3 ppm) 및 ${\alpha}$-tocopherol 100 IU/kg과 selenium 0.3 ppm을 혼합 급여한 육계의 생산성, 계육의 일반 성분, pH, TBARS, WHC, 전단력, 육색 및 지방산 조성에 미치는 영향을 알아보고자 수행하였다. 육계 360수를 공시하여 대조구와 ${\alpha}$-tocopherol 50 IU/kg 급여구를 T1, ${\alpha}$-tocopherol 100 IU/kg 급여구를 T2, ${\alpha}$-tocopherol 200 IU/kg 급여구를 T3, selenium 0.3 ppm/kg 급여구를 T4, ${\alpha}$-tocopherol 100 IU/kg과 selenium 0.3 ppm/kg 급여구를 T5 등 6개 처리구로 나누어 5주간 사양한 후 다리살 근육을 분석하였다. 육계의 증체량, 사료 섭취량 및 사료 요구율은 처리구간에 유의성이 없었다. 계육의 일반 성분중에 수분과 조단백질, 조회분은 처리구간에 유의성이 없었고, 조지방 함량은 대조구와 T1보다 처리구에서 높았다. pH, 보수성과 전단력은 모든 처리구에서 유의성은 없었다. TBARS는 대조구와 T1은 유의한 차이가 없으나, ${\alpha}$-tocopherol과 selenium 급여구에서 낮았으며, 특히 혼합급여구인 T5에서 TBARS 값이 유의하게 낮았다. 육색중 CIE $L^*$값과 $b^*$값은 처리구간에 유의성이 인정되지 않았으나, CIE $a^*$값은 T2, T3, T5에서 높았다(P<0.05). 지방산의 분석 결과, ${\alpha}$-tocopherol의 급여로 palmitic acid는 감소하고, oleic acid, linileic acid는 증가하여 포화지방산 함량은 줄고, 불포화지방산 함량이 증가하는 결과로 나타났다(P<0.05). 이상의 결과를 종합적으로 고찰해 볼때 ${\alpha}$-tocopherol과 selenium를 육계에 급여하면 육색과 지방의 산화를 억제하는 효과가 있음을 확인할 수 있어 계육의 저장성을 개선할 가능성이 있을 것으로 기대된다.

Role of ${\alpha}$-tocopherol in cellular signaling: ${\alpha}$-tocopherol inhibits stress-induced mitogen-activated protein kinase activation

  • Tocopherols belong to the plant-derived poly phenolic compounds known for antioxidant functions in plants and animals. Activation of mitogen-activated protein kinases (MAPK) is a common reaction of plant cells in defense-related signal transduction pathways. We report a novel non-antioxidant function of ${\alpha}$-tocopherol in higher plants linking the physiological role of tocopherol with stress signalling pathways. Pre-incubation of a low concentration of $50{\mu}M$ ${\alpha}$-tocopherol negatively interferes with MAPK activation in elicitor-treated tobacco BY2 suspension culture cells and wounded tobacco leaves, whereas pre-incubated BY2 cells with ${\alpha}$-tocopherol phosphate did not show the inhibitory effect on stimuli-induced MAPK activation. The decreased MAPK activity was neither due to a direct inhibitory effect of ${\alpha}$-tocopherol nor due to the induction of an inhibitory or inactivating activity directly affecting MAPK activity. The data support that the target of ${\alpha}$-tocopherol negatively regulates an upstream component of the signaling pathways that leads to stress dependent MAPK activation.
