식혜의 이소말토올리고당에 관한 연구(II) -효소적 분석- (A Study on Sugars in Korean Sweet Rice Drink "Sikhye"(II) -Enzymatic Analysis of Isomaltooligosaccharides and Rice Residue-)

  • 식혜에 함유된 이소말토올리고당과 밥알에 30unit/ml의 $\alpha$-아밀라아제, 글루코아밀라아제, $\alpha$-글루코시다아제, $\beta$-아밀라아제를 가하여 작용시킨 결과 글루코아밀라아제 외에는 일부밖에 가수분해하지 못하였다. 그리고, 인체의 효소인 $\alpha$-아밀라아제와 $\alpha$-글루코시다아제를 함께 작용시켜도 25% 이상 가수분해하지 못하므로 비피두스균의 활성인자로 작용할 것으로 보인다. 밥알은 물에 녹지 않기 때문에 식이섬유로도 작용할 것으로 보인다.

효모 Cloning Vector와 전분발효 효모의 개발 (Yeast Cloning Vectors and their Application to the Development of Starch-fermenting Yeast)

  • Transformed, hybrid strains of the yeast Saccharomyces capable of simultaneous secretion of both glucoamylase and ${\alpha}-amylase$ have been produced. These strains can carry out direct, one-step assimilation of starch with conversion efficiency greater than 93% during a 5 day growth period. One of the transformants converts 92.8% of available starch into reducing sugars in only 2 days. Glucoamylase secretion by these strains results from expression of one or more chromosomal STA genes derived from Saccharomyces diastaticus. The strains were transformed by a plasmid(pMS12) containing mouse salivary ${\alpha}-amylase$ cDNA in an expression vector containing yeast alcohol dehydrogenase promoter and a segment of yeast $2{\mu}$ plasmid. The major starch hydrolysis product produced by crude amylases found in culture broths is glucose, indicating that ${\alpha}-amylase$ and glucoamylase act cooperatively.

한국산 곡류와 두류 중 $\alpha$-Amylase 저해물질의 검색 및 특성 (Screening and Characterization of $\alpha$-Amylase Inhibitors from Cereals and Legumes in Korea)

  • To investigate characterization of the ${\alpha}$-amylase inhibitors from cereals and legumes produced in Korea, inhibitory activities against ${\alpha}$-amylase with the inhibitor from barley(Hordeum vulgare), wheat(Triticum aestivun), black bean(Glycine max), bean(Cajanus cajon) and pea(Pisum sativum) were measured. Among the samples tested, inhibitors from naked barley and black bean(sabong) which showed the highest inhibitor activities of cereals and legumes, respectively, were characterized according to treatment condition. The results obtained were summarized as follows. During the germination of naked barley and black bean, ${\alpha}$-amylase activities were gradually increased but inhibitory activities against ${\alpha}$-amylases were decreased. Both activities were gradually decreased when naked barley and black bean were stored. More than 50% of activities of the inhibitors from naked barley and black bean were remained at 100$^{\circ}C$ for 15 min and 20 min, respectively, indicating that the inhibitor from black bean was more stable to heat than that of barley.

고온성 방선균에 의한 효소생산에 관한 연구 제 1보 $\alpha$-Amylase생산균의 분이 및 효소생산 조건 (Studies on the Production of Enzymes by Thermophilic Actinomycetes Part I. Isolation and Culture Conditions of Thermostable $\alpha$-Amylase Producing Actinomycete)

  • In the course of studies on the production of thermostable amylases by thermophilic actinomycetes isolated from soils the investigation was carried out on the production of $\alpha$-amylase by G-1011 strain which had presented the most remarkable $\alpha$-amylase formation ability among 128 amylolytic isolates. The results were as follows : 1. Characteristics of G-1011 strain were compared with those descriptions of thermophilic actinomycetes given in Bergey's Manual. The strain was identical to these species of actinomycetes. The details of physiological properties of the strain luould he published in near future. 2. The optimum temperature for incubation of the cell growth of G-1011 strain and $\alpha$-amylase production by the strain was revealed to 5$0^{\circ}C$. 3. The effective medium for $\alpha$-amylase formation by the strain was consisted of 3.0%, soluble starch, 1.0%, peptone, 0.5%, yeast extract, 0.5%, NaCl, 0.1%, MgSO$_4$ㆍ7$H_2O$ 0.02%, $K_2$MPO$_4$and 0.002% FeSO$_4$ㆍ7$H_2O$. The pH of the medium was ajusted to 7.0 with phosphate buffer solution. 4. The maximum production of $\alpha$-amylase (3420 D. U/ml) by G-1011 strain resulted when it was grown for 16 hours with the culture of reciprocal shaking.

한국산 검정콩 및 쌀보리 $\alpha$-Amylase 저해물질의 이화학적 특성 (The Physicochemical Properties of $\alpha$-Amylase Inhibitors from Black Bean and Naked Barey in Korea)

  • The physicochemical properties of the $\alpha$-amylase inhibitors from black bean and naked barley is Korea were investigated. Preincubation time for maximum inhibition was 30min and no activity change was seen after that time. Optimum pH of the $\alpha$-amylase inhibitors from the black bean and naked barley was pH 7.0 and the inhibitory activities were stable in the range of pH 6.0~8.0 in both phosphate and Tris-HCI buffer solutions. Both inhibitors maintained more than 50% of activity after incubation for 17 min at 7$0^{\circ}C$. The inhibitors from the black bean and naked barley maintained more than 50% of activities after treatment for 40 min and 30 min with pepsin, and 30 min and 50 min with trypsin, respectively. Both inhibitors functioned via a noncompetitive mechanism and were active against porcine pancreatic and human salivary $\alpha$-amylases. The activities of both inhibitors were linear for the ionic stength ranging from 0 to 0.9. The addition of 70 mM maltose to the reaction mixture caused a maximum increase in the relative activities of both inhibitors, but it did not affect the dissociation of the EI complex. The activities of both inhibitors were significantly enhanced by adding 1mM of K+ or Mg2+.

A New α-Amylase from Reticulitermes speratus KMT1

  • Termites are wood pests that cause vast economic damage every year. They digest both cellulose and starch, but the enzymes for starch digestion have not been well characterized. We obtained complete amino acid sequence information on the KME1 ${\alpha}$-amylase from Reticulitermes speratus KMT1 through analysis of total mRNA sequences. The KME1 enzyme has two ${\alpha}$-amylase domains and is 68% identical to the ${\alpha}$-amylase from Blattellager manica, its closest relative in the GenBank database. Some unique features of its conserved region and its distant evolutionary relationship to other insect ${\alpha}$-amylases suggest that KME1 is a new type of ${\alpha}$-amylase.

  • 보리 맥아로부터 발견된 서로 다른 알파아밀라제 동질효소(AMY1, AMY2)는 80%에 달하는 높은 아미노산 서열의 상동성을 보이지만, 두 효소의 특성은 서로 달라 AMY1 효소는 낮은 농도의 칼슘 조건에서 최대 활성을 보이는 반면, AMY2 효소는 높은 칼슘이온 농도에서 높은 활성을 나타낸다. 또한 BASI (Barley ${\alpha}$-Amylase/Subtilisin Inhibitor) 단백질은 AMY2 효소만을 특이적으로 저해한다. 따라서 본 연구에서는 AMY1과 AMY2 효소의 유전자를 I, II, III의 세 부위로 나눈 후, 제한효소 처리에 의해 일부 부위를 상호 치환한 4종의 chimera 효소를 추가로 제조하고, Pichia pastoris 균주에서 대량 발현하였다. 이들 효소의 특성을 비교한 결과, 제 I 부위만이 상호치환된 AMY211 및 AMY122 효소의 경우, AMY1과 AMY2의 중간적 칼슘 의존성을 나타내었으며, BASI에 의한 저해효과는 AMY2의 제 I, II 부위를 포함하는 AMY221 효소에서만 관찰되었다. 따라서 보리 아밀라제의 제 I 부위 및 제 II 부위에 존재하는 아미노산 잔기들이 칼슘 의존성 및 BASI와의 결합에 중요한 역할을 담당하는 반면 제 III 부위는 이들 효소의 활성 차이에 영향을 미치지 않음을 확인하였다.

Biochemical Properties of Starch Granule Non-Digestive Enzyme(SGNA) of Bacillus polymyxa No.26

  • A $\alpha$-l, 4-D-glucan maltohydrolase $(\beta$-amylase), secreted by the mesophilic aerobic bacterium Bacillus polymyxa No.26, was purified and characterized. The enzyme production was increased after a logarithmic phase of bacterial growth and paralleled with the onset of bacterial sporulation. By applying anion exchange chromatography and gel filtration the enzyme was purified 16.7-fold and had a specific activity of 285.7 units/mg. Two enzyme activities were eluted on a column of DEAE-Sephadex chromatography, and they were designated as E-I for a major enzyme peak and E-II for a minor peak. Of them, E-I enzyme peak was further purified by using gel chromatography. The molecular mass of this enzyme was determined to be 64, 000 daltons and consisted of a single subunit, showing an isoelectric point of 8.9. The enzyme was able to attack specifically the $\alpha$-l, 4-glycosidic linkages in soluble starch and caused its complete hydrolysis to maltose and $\beta$-limited dextrin. This amylolytic enzyme displayed a temperature optimum at $45^\circ{C}$ and a pH optimum at 7.0. The amino acid composition of the purified enzyme was quite similar to the other bacterial $\beta$-amylases reported. Surprisingly, the purified enzyme from this aerobe only exhibited hydrolytic activity on soluble starch, not on starch granules. The degradation of from starch by $\beta$-amylase was greatly stimulated by pullulanase addition. These results differentiated from other $\beta$-amylases reported. Based on a previous result that showed the enzyme system involves in effective degradation of raw starch granules, this result strongly suggested that the purified enzyme (E-I) can be a synergistic part of starch granule-digestion and E-II plays a crucial role in digestion of starch granules.

A New Protein of ${\alpha}$-Amylase Activity from Lactococcus lactis

  • An extracellular ${\alpha}$-amylase from Lactococcus lactis IBB500 was purified and characterized. The optimum conditions for the enzyme activity were a pH of 4.5, temperature of $35^{\circ}C$, and enzyme molecular mass of 121 kDa. The genome analysis and a plasmid curing experiment indicated that $amy^+$ genes were located in a plasmid of 30 kb. An analysis of the phylogenetic relationships strongly supported a hypothesis of horizontal gene transfer. A strong homology was found for the peptides with the sequence of ${\alpha}$-amylases from Ralstonia pikettii and Ralstonia solanacearum. The protein with ${\alpha}$-amylase activity purified in this study is the first one described for the Lactococcus lactis species, and this paper is the first report on a Lactococcus lactis strain belonging to the amylolytic lactic acid bacteria (ALAB).

Herpetosiphon geysericola 균주의 Amylase 부분정제 및 특성 (Partial Purification and Characteristics of Amylases from Herpetosiphon geysericola)

  • 고온성 cellulose 분해이용세균인 Herpetosiphon geysericola CUM 317 균주가 생성하는 ${\alpha}-amylase$, ${\beta}-amylase$ 및 glucoamylase를 황산암모늄 염석, DEAE-cellulose chromatography, CM-cellulose chromatography 방법으로 각각 부분정제하였다. 이들 ${\alpha}-amylase$, ${\beta}-amylase$ 및 glucoamylase의 감자 전분에 대한 Km치는 $2.31mg/m{\ell}$, $7.69mg/m{\ell}$$8.33mg/m{\ell}$였으며, 각 분자량은 84,000 dalton, 76,000 dalton 및 80,000 dalton의 크기로 나타났다.
