Changes in the Expressional Levels of Sarcoplasmic Reticulum $Ca^{2+}-regulatory$ Proteins in the Postnatal Developing Rat Heart

  • In the present study, the postnatal developmental changes in the expressional levels of cardiac sarcoplasmic reticulum (SR) $Ca^{2+}$ regulatory proteins, i.e. $Ca^{2+}-ATPase,$ phospholamban, and $Ca^{2+}$ release channel, were investigated. Both SR $Ca^{2+}-ATPase$ and phospholamban mRNA levels were about 35% of adult levels at birth and gradually increased to adult levels. Protein levels of both SR $Ca^{2+}-ATPase$ and phospholamban, which were measured by quantitative immunoblotting, were closely correlated with the mRNA levels. The initial rates of $Ca^{2+}$ uptake at birth were about 40% of adult rates and also increased gradually during the myocardial development. Consequently, the relative phospholamban/$Ca^{2+}-ATPase$ ratio was 1 in developmental hearts. $Ca^{2+}$ release channel (ryanodine receptor) mRNA was about $50{\sim}60%$ at birth and increased gradually to adult level throughout the postnatal rat heart development. $^3[H]ryanodine$ binding increased gradually during postnatal myocardial development, which was closely correlated with ryanodine mRNA expression levels during the development except the ryanodine mRNA level at birth. These findings indicate that cardiac SR $Ca^{2+}-ATPase,$ phospholamban, and $Ca^{2+}$ release channel are expressed coordinately, which may be necessary for intracellular $Ca^{2+}$ regulation during the rat heart development.

Inhibitory Effect of Thapsigargin on the Activities of $H^+-ATPases$ in Tomato Roots (토마토 뿌리조직 $H^+-ATPase$ 활성에 미치는 Thapsigargin의 저해효과)

  • Thapsigargin is a specific antagonist of SR/ER-type $Ca^{2+}-ATPase$ in animal tissue, and it was used to characterize the microsomal ATPases prepared from the roots of tomato. When $10\;{\mu}M$ thapsigargin was added, it inhibited the microsomal ATPase activity by 30%. The thapsigargin-induced inhibition was dose-dependent. Since the activity of $Ca^{2+}-ATPase$ is very low in the roots of tomato tissue, it is possible that thapsigargin inhibits the activities of major $H^+-ATPases$ located in plasma and vacuolar membranes. The inhibitory effect of thapsigargin was reduced when the vacuolar $H^+-ATPase$ activity was inhibited by ${NO_3}^-$. However, the effect of thapsigargin was not observed on the $H^+-ATPase$ activity located in the plasma membrane. These results suggest that thapsigargin inhibits the vacuolar $H^+-ATPase$ activity in the roots of tomato.

  • The effects of various sedative cyclopeptides and peptide alkaloids from the Zizyphus species on calmodulin-dependent $Ca^{2+}$ -ATPase and phosphodiesterase were Investigated. Calmodulin-induced activation of $Ca^{2+}$-ATPase was strongly inhibited by sanjoinine-A dialdehyde (IC_{50}$, 2.3$\mu\textrm{m}$), -Ah1 (IC_{50}$, 4.0$\mu\textrm{m}$), -A (IC, 4.6$\mu\textrm{m}$), and -G2 (IC_{50}$, 7.2$\mu\textrm{m}$), while calmodulin-induced activation of phosphodiesterase was strongly inhibited by both deachuine- S10 (IC_{50}$, 4.9$\mu\textrm{m}$) and sanjoinine-D (IC_{50}$, 9.0$\mu\textrm{m}$). The inhibitory activity of the various cyclopeptides and peptide alkaloids on $Ca^{2+}$-ATPase was found to correlate well with their Sedative activity.

  • The $(Ca^{2+}+Mg^{2+})$-ATPase has been purified homogeneously from sarcoplasmic reticulum of rat skeletal muscle by sucrose density gradient centrifugation. The purified enzyme has a molecular weight of 115,000 as judged by polyacrylamide gel electrophoresis in the presence of sodium dedecyl sulfate, and therefore has the same size of the enzyme in rabbit and chick skeletal muscle. $Ca^{2+}, Mg^{2+}, Fe^{2+}, Co^{2+}, and Mn^{2+}$ at 50 $\\muM$ show stimulatory effect on the ATP-ase, while $Zn^{2+}, Cu^{2+}, and Hg^{2+}$ inhibit it at the same concentration. The ATPase activity is insensitive to antimalarial drugs such as quinine and quinacrine, but is sensitive to inhibition by p-hydroxymecurie benzoate and phenylmethylsulfonylfluoride. The enzyme has optimum pH of 6 to 7 and Km value for ATP is estimated to be 98 $\\muM$. Thus, a number of biochemical properties of this enzyme appear to be different from those of the enzyme that have been isolated from rabbit skeletal muscle. The $(Ca^{2+}+Mg^{2+})$-ATPase appears to be selectively degraded in microsomal fraction. The activity of metalloendoprotease is evident in the microsomal preparation when assayed by radioactively labeled protein substrate, such as $^{3}H-casein and $^{125}I$-insulin. However, it is presently unclear whether the metalloendoprotease is responsible for the degradation of the $(Ca^{2+}+Mg^{2+})$-ATPase.

  • The Ca uptake by the fragmented sarcoplasmic reticulum of the rabbit skeletal muscle was measured under various concentrations of K, Mg, Caffeine, procaine and quinine. The ATPase activity of this reticular membrane was measured under the same conditions simultaneously. The saturation of Ca uptake was almost completed within 1 minute. The Ca uptake was inhibited by high concentrations of K (above 50 mM) and Mg (above 1 mM)in the absence of ATP. When ATP is present, however, the Ca uptake did not reflect the concentration of K, while it increased greatly as the concentration of Mg was increased. Caffeine and procaine caused the inhibition of Ca uptake in the presence of ATP, but quinine did not. The ATPase activity of the membrane was little affected by the concentration of K, while it was enhanced in the presence of Mg. Caffeine, procaine and quinine did not influence the ATPase activity.

  • Myotibrillar ATPase activity, thermostability and composition of muscle protein were investigated to elucidate biochemical properties regard with rearing period and sex of olive flounder. Myofibrillar ATPase activity of male olive flounder reared for 6, 12 and 20 months was stronger than that of female one. $Mg^{2+}\;(+Ca^{2+})-ATPase$ activity of both female and male fish decreased with the elapse of rearing period, and the activity of male was higher than that of female far beyond the rearing periods. The high correlationship between the weight gain and myofibrillar ATPase activity was observed. The thor mostability of male myofibrillar protein was higher than that of female. Subunit composition of the myofibrillar and sarcoplasmic protein did not show difference between the both sex of the fish.

  • Investigation on the extractability, Mg2+-, Ca2+ , EDTA-ATPase activity and solubility of actomyosin prepared from leg and breast muscle of silky fowol were as follows. The extractability of actomyosin in leg and breast muscle was 779mg/100g and 1, 318mg/100g respectively, breast muscle was higher than leg muscle . Mg2+-ATPase activity of actomyosin was high inionic strength 0.02-0.10 and Mg2+ATPase activity of low ionic strength was higher than high ionic strength not related to the part. Ca2+ ATPase activity was high in ionic strength 0.05-0.13, the activity of leg muscle was higher that breast muscle. And EDTA-ATPase activity showed low in low ionic strength and showed high in high ionic strength, and increased greatly depend ionic strength up to 0.4. The solubility of actomyosin was not different in leg and breast muscle , the solution started in KCI concentration of 0.3M and ended in DCI concentration of 0.4M.

  • The effect of adenosine 3', 5'-cyclic monophosphate on the ATPase activity and on the active transport of Ca of the sarcoplasmic reticulum fragments of the rabbit skeletal muscle was studied. Cyclic AMP (cAMP) had no effect on the ATPase activity of the fragments (8,000 ~ 20,000 $\times$ G and 20,000 ~ 36,000 $\times$ G fractions). $N^6$, O^{2'} -Dibutyryl cAMP (DBcAMP) had either no effect on the activity. On the other hand, theophylline (1 mM) increased the activity by about 20%. The active uptake of Ca by the sarcoplasmic reticulum fragments was inhibited by the presence of 1$\times$$10^{-6}$ ~ 1 $\times$ $10^{-3}$M of cAMP. The presence of DBcAMP or theophylline also inhibited the uptake. It is, therefore, concluded that the Ca uptake of the sarcoplasmic reticulum seems to be controlled by cAMP.

The Effect of Cryoprotectants on the Properties of Pacific Sand Lance Ammodytes personatus Girard Surimi During Frozen Storage

  • We investigate the effects of cryoprotectant mixtures on the quality of sand lance surimi (SLS) during storage at $-30^{\circ}C$. We monitored freeze-induced denaturation of myofibrillar protein in SLS and examined the texture profile of SLS gel. Freeze-induced denaturation was assessed by evaluating SLS $Ca^{+2}$-ATPase activity. SLS gels prepared with sorbitol or sucrose and a mixture of both as cryoprotectant. Higher concentrations of cryoprotectants resulted in significantly higher residual SLS $Ca^{+2}$-ATPase activity at the same storage time (P < 0.05). Residual $Ca^{2+}$-ATPase activity of SLS prepared with sorbitol was higher than that of sucrose when cryoprotectant concentration and storage period were same. A blend of sorbitol and sucrose resulted in a stronger cryoprptective effect of SLS myofibrillar protein than did sorbitol or sucrose alone. The presence of a phosphate compound in SOP (3% sorbitol + 0.2% phosphate compound) resulted in higher SLS $Ca^{2+}$-ATPase activity than that of did 5% sorbitol. The hardness, brittleness, and elasticity values and a folding test of the SLS gels were significantly affected by cryoprotectant concentrations and the storage time. Preference scores and acceptance for texture in a sensory evaluation of the SLS gels increased with increasing sorbitol or sucrose concentration.
