Rice (Oryza sativa L.) is the world's most important cereal crop. In crop plant, chlorophyll content and leaf senescence could affect grain filling and yield. We analyzed a QTL associated with chlorophyll content and delayed leaf senescence using high chlorophyll near isogenic line (HC-NIL). HC-NIL derived from a cross between Oryza sativa cv. Hwaseong as a recurrent parent and wild species O. grandiglumis as a donor parent showed higher chlorophyll content than Hwaseong. To identify QTL associated with chlorophyll content, 58 $F_3$ and 38 $F_4$ lines were developed from a cross between HC-NIL and Hwaseong. For QTL analysis, simple sequence repeat (SSR) markers were used for genotyping and one-way ANOVA was conducted. A QTL for chlorophyll content (qCC2) was detected in chromosome 2 and explained 24.63% of phenotypic variation. The senescence effect of the qCC2 was examined in dark-induced incubation (DII). Detached leaves from Hwaseong and HC-NIL were incubated on 3mM MES buffer (pH 5.8) at $27^{\circ}C$ under complete dark condition. After 3 days of incubation, the Hwaseong leaves turned yellow, but the HC-NIL leaves were green. HC-NIL has higher chlorophyll content with delayed senescence than Hwaseong. These results indicated that qCC2 is associated with stay-green phenotype. To know whether the qCC2 is responsible for leaf functionality, ion leakage test and Fv/Fm measurement were performed. Both experiment results showed that differences were observed between Hwaseong and HC-NIL but it was not statistically significant. These results might suggest that the qCC2 is possibly related to chlorophyll content and non-functional stay-green phenotype.

A comparative study on rumen bacterial and protozoal population and fungal zoospores in cattle (Brahman$\times$Native) and swamp buffalo (Bubalus bubalis) was conducted. Forty animals, twenty of each, with same sex and similar age which were raised under similar condition in the Northeast of Thailand, were used. Rumen digesta were sampled immediately post slaughtering for total microscopic counts of bacteria, protozoa and fungal zoospores. It was found that total bacterial population were higher in swamp buffalo that those in cattle (1.6 vs $1.36{\times}10^{8}cells/ml$) having more population of cocci, rods and ovals. Lower rumen protozoal pupulation in swamp buffalo with lower numbers of Holotrichs and Entodiniomorphs were found as compared to those in cattle. Significant higher fungal zoospore counts were in swamp buffalo than those in cattle being 7.30 and $3.78{\times}10^6$, respectively. Study under electron microscope, revealed Anaeromyces sp. with acuminate apex were more predominant in the rumen of swamp buffalo. With these findings, cattle and swamp buffaloes showing differences in rumen bacterial, protozoal population and fungal zoospore counts, offer new additional information as why swamp buffaloes exhibit conditionally better than cattle especially during long dry season without green grass.
Hairy roots of carrot were induced by Agrobacterium rhizogenes $A_4$ strain within about 2-4 weeks after inoculated from root disc. Early axonic culture is established in RCM agar medium and following is in MS rigid medium. After 15 days culture, the hairy roots were vigrous growth in about 10 times of initial inoculum. Anthocyanin contents of hairy roots were more than of ordinary roots. 2, 4-D ($10^{-4}mg/ l$), sucrose (5%), nitrogen source (0.03M) contained medium was optimized to growth of hairy root and contents of anthocyanin. Phenotypic alterations of leaves are observed in transformed plants and determined the transformation of hairy roots and the transformed plants by opine assay.

A Superconductor Flywheel Energy Storage System(SFES) mainly consists of a pair of non-contacting High Temperature Superconductor(HTS) bearings that provide very low frictional losses, a composite flywheel with high energy storage density. The HTS bearings, which offer dynamic stability without active control, are the key technology that distinguishes the SFES from other flywheel energy storage devices, and great effort is being put into developing this technology. The Superconductor Journal Bearing(SJB) mainly consists of HTS bulks and a stator, which holds the HTS bulks and also acts as a cold head. Static properties of HTS bearings provide data to solve problems which may occur easily in a running system. Since stiffness to counter vibration is the main parameter in designing an HTS bearing system, we investigate SJB magnetic force through static properties between the Permanent Magnet(PM) and HTS. We measure stiffness in static condition and the results are used to determine the optimal number of HTS bulks for a 100kWh SFES.
Spherical reticulated shells are widely applied in structural engineering due to their good bearing capability and attractive appearance. Parametric modeling of spherical reticulated shells is the basis of internal analysis and optimization design. In the present study, generation methods of nodes and the corresponding connection methods of rod elements are proposed. Modeling programs are compiled by adopting the ANSYS Parametric Design Language (APDL). A shape optimization method based on the two-stage algorithm is presented, and the corresponding optimization program is compiled in FORTRAN environment. Shape optimization is carried out based on the objective function of the minimum total steel consumption and the restriction condition of strength, stiffness, slenderness ratio, stability. The shape optimization of four typical Schwedler spherical reticulated shells is calculated with the span of 30 m~80 m and rise to span ratio of 1/7~1/2. Compared with the shape optimization results, the variation rules of total steel consumption along with the span and rise to span ratio are discussed. The results show that: (1) The left and right rod-Schwedler spherical reticulated shell is the most optimized and should be preferentially adopted in structural engineering. (2) The left diagonal rod-Schwedler spherical reticulated shell is second only to left and right rod regarding the mechanical behavior and optimized results. It can be applied to medium and small-span structures. (3) Double slash rod-Schwedler spherical reticulated shell is advantageous in mechanical behavior but with the largest total weight. Thus, this type can be used in large-span structures as far as possible. (4) The mechanical performance of no latitudinal rod-Schwedler spherical reticulated shell is the worst and with the second largest weight. Thus, this spherical reticulated shell should not be adopted generally in engineering.
This study was performed to identify the level of stress recognized by the parents as a support system for their infant patients who were hospitalized in an emergency department, thus to provide the resulting data as the basic material of care intervention for the families of infant patients. This study subjected the parents with infant patients who were hospitalized in emergency department of C University Hospital in Daejeon, and the data was collected from questionnaires for them, dating from Mar. 20, to Jun. 24, 2001 This study used the tool(30 questions) which was originally developed by Ji, Dong-ok (1992) for measuring the stress of families of infant patients in emergency room, and then was modified and complemented by this researcher suitably for infant patients. Collected data was statistically analyzed with frequency, percentage, mean, standard deviation, t-test, ANOVA, by using SPSS WIN10.0 program, and the results were as follows: 1. The stress of parents with infant patients hospitalized in emergency department averaged 3.31 on the basis of 5-point measure, which means that they felt stress beyond means. 2. The factor with high average out of stress factors the parents of infant patients recognized included the followings: 'about the pain of infant patients due to examination(3.91±1.00)'; 'about rare opportunity to be in contact with physician(3.78±1.09)'; 'delay in emergent treatment required for infant patients(3.75±1.31)'; 'delay in the general treatment of infant patients(3.72±1.32)'. Factors of the stress level includes the followings: 'care and medical treatment'(3.46±.72); 'body and diseases'(3.41±.97)'; 'lack of information and supportable resources (3.25±.77)'; and so on. 3. For the stress level according to general characteristics, there were statistically significant differences in the result of the condition of infant patients, medical treatment(P<.01), religion, the procedure of treatment(P<.05). With little preceding studies for infant patients hospitalized in emergency department, this study looked through the level of stress recognized by the parents of infant patients. Based on the comprehension on the parents with infant patients and the knowledge on stress factors recognized by the parents, it is expected that we can seek the methods of care intervention such as explanation of care and treatment procedures, unit policies, continuous interest and emotional supports as well as the provision of information to understand the responses of parents and reduce accompanied stresses.

Varietal difference in root distribution and other root characteristics were investigated under fold and water culture condition. The results were as follows: 1. IR667 showed funnel type of root distribution in soil profit while Jinheung had barrel type, and each type appearance was more distinguishable with fertilizer application. 2. Root weight per tiller was smaller in IR667 than in Jinheung and IR667 had more root in 0 to 5cm of soil depth but Jinheung had more in 5 to l0cm depth. 3. Horizontal distribution of root was dencer near to stem base without fertilizer than with fertilizer in both IR667 and Jinheung indicating structural construction for intensive nutrient uptake. Between varieties this 'dence to stem base' trend accompaning 'dence to wide spacing side' was greater in IR667 without fertilizer and these were quite true with fertilizer in Jinheung. 4. The decreasing rates of root and ear weight by fertilizer application were greater in IR667 than in Jinheung. This and other characteristics indicated that the root of IR667 is likely to be panicle-number type comparing with Jinheung. 5. The root of IR667 had lower oxidizing power of ${\alpha}-naphthylamine$ than that of Jinheung indicating weaker resistance to reductive soil but cation exchange capacity of water-cultured root was higher in IR667 suggesting stronger nutrient uptake. 6. The content of phosphorus and especially potassium in root were higher with fertilizer but lower without fertilizer in IR667 than in Jinheung indicating that IR667 is more sensitive to root environment. 7. The contents of N, K and CEC were increasing toward root tip while P content was decreasing. The root from surface soil had higher N and K content than that from subsoil. The contents of N,P,K, and CEC of root at harvesting stage were about 1.0%, 0.1%, 0.5% and 15me/100g at dry weight base, respectively.
The Dissolved Organic Carbon (DOC) existing in a water includes both hydrophobic and hydrophilic substances however, most of the discussion focuses on hydrophobic substances. The hydrophobic fraction was easily removed by absorption or coagulation more than hydrophilic fraction. Therefore, control of the hydrophilic fraction is very important in water treatment process. This study is to determine the variation of DOC, the removal efficiency of DOC, and Trihalomethane formation potential (THMFP) after each stage of water treatment process by fractionating Natural Organic Matters (NOM) into hydrophobic and hydrophilic substance. DOC from raw water was fractionated at acidic pH (pH<2) using XAD 8 resin column, into two fraction : hydrophobic substance (i.e. humic substance) adsorbed on XAD 8 and hydrophilic substance which represent the organics contained in the final effluent. THMFP was carried out according to the following set condition: Cl2/DOC=4 mg/mg, incubation at $25^{\circ}C$ in darkness, pH 7 adjust with HCl or NaOH as necessary, and 72hour-contact time. THMs analyzed in this study were chloroform, bromodichloromethane, dibromochloromethan, and bromoform. Sewage was almost evenly split between the hydrophobic (56%) and hydrophilic fraction (44%). But, Aldrich humic substance (AHS) was found to contain less hydrophilics (14%) than hydrophobics (86%). The formation of THMs may depend on the source which is characterized by the composition of organic matters such as AHS and sewage. The THMFP yield of sewage and AHS were assessed as follows. The value of the THMFP reaction yield, AHS $172.65{\mu}g/mg$, is much higher than that of sewage $41.68{\mu}g/mg$. This illustrates possible significant difference in THMFP according to the component type and the proportion of organic matter existing in water source. Apparently AHS react with chlorine to produce more THMFP than do the smaller molecules found in sewage. Water treatment process may reduce THMFP, nevertheless residual DOC (the more hydrophilic substance) has significant THMFP. Further reduction in organic halide precursors requires application of alternative treatment techniques.
A vector harboring double cassettes; a heterologous gene expression cassette of pHCE-InaN-antigen and a ghost formation cassette of pAPR-cI-E lysis 37 SDM was constructed and introduced to E. coli DH5a. For the production of a bacterial ghost vaccine, bacterial ghosts from E. coli / Streptococcus iniae with four different types of antigens - enolase, GAPDH, sagA and piaA - were produced by the optimization of fermentation parameters such as a glucose concentration of 1 g/l, agitation of 300 rpm and aeration of 1 vvm. Efficiency of ghost bacteria formation was evaluated with cultures of OD$_{600}$=1.0, 2.0 and 3.0. The efficiency of the ghost bacteria formation was 99.54, 99.67, 99.99 and 99.99% with inductions at OD$_{600}$=3.0, 1.0, 2.0 and 1.0 for E. coli/S. iniae antigens enolase, piaA, GAPDH and sagA, respectively. Ghost bacteria as a vaccine was harvested by centrifugation. The antigen protein expressions were analyzed by SDS-PAGE and western blot analysis, and the molecular weights of the enolase, piaA, GAPDH and sagA were 78, 26, 67 and 26 kDa, respectively. The molecular weights of the expressed antigens were consistent with theoretical sizes obtained from the amino acid sequences.
Gonadal development, gametogenesis, reproductive cycle, egg-diameter and composition, condition factor, and the first sexual maturity of the clam, Potamocorbula amurensis were investigated by histological observation. Samples were collected monthly from the tidal flat of Moonpo, Puan-gun, Chollabuk-do, west coast of Korea from November 1996 to October 1997. P. amurensis is dioecious and oviparous. The gonads were composed of a number of gametogenic follicles. The oogonia and fully ripe oocytes were $9\~12\mu$m and $50\~60\mu$m in diameter, respectively. Each of the spermatogenic follicle formed stratified layers composed of spermatogonia, spermatocytes spermatids, and spermatozoa in groups on the follicular wall. The reproductive cycle of P. amurensis could be classified into five successive stages: early active, late active, ripe, partially spawned, and recovery. Spawning occurred twice a year from May to July and from September to October, the main spawning seasons also appeared twice a year between May and June, and in October when the water temperatures reached above $18^{\circ}C$. The monthly changes in the condition factor were closely related with the reproductive cycle. Minimum size for the sexual maturation of female and male were 8.1 mm in shell length. There were two patterns for the gametogenesis: 1. After spawning, the undischarged ripe oocytes and spermatozoa in the follicles were degenerated and absorbed, but in part, the existing follicles were not contracted significantly and then they took part in new gametogenesis within one or two months (especially, in summer). 2. After spawning, each follicle was contracted, thereafter gametogenesis again occurred in newly formed follicles.
