치자색소의 염료화 및 염색성 (The Acquisition and dyeability of Gardenia jasminoides Colorant)

  • The acquistion and dyeability of the Gardenia jasminoides were examined to establish the optimum condition for extraction and storage in the process of obtaining the natural dye, Gardenia jasminoides colorant. Also the dyeability and colorfastness of Gardenia jasminoides were investigated. The results of this study are as follows. The optimum part-removed seed. In all experiments, the part of fat-removed seed and pericarp was used. The optimum condition for extraction of Gardenia jasminoides colorant was at 40℃ and for 90 min. in methanol. As storage temperature was higher, the absorbance of colorant extract decreased rapidly. The Gardenia jasminoides colorant exhibited dyeability to cotton, silk, wool, and nylon. The dyeability was the greatest in wool, and then nylon, silk, and cotton. Both wool and nylon had the greatest K/S value at pH3, however, nylon and cotton at pH 6 and pH 8 respectively. In addition, the increase in K/S value corresponded to temperature in wool and nylon, but the silk and cotton had the greatest K/S value at 60℃. Also, the K/S value increased in concomitant with the increased number of dyeing repetition. In the most cases, colorfastness of light was weak but colorfastness of laundry, sweat was relatively excellent.

Rhizopus nigricans로부터 원형질체의 분리 (Isolation of Protoplasts from Rhizopus nigricans)

  • Rhizopus nigricans의 포자와 균사체로부터 원형질체 생성을 위한 최적 조건을 조사하였다. 5% 2-deoxy-D-glucose가 첨가된 액체 배지에서 14시간 배양시킨 균사체보다 동일배지에서 4시간 배양하여 팽창시킨 포자로부터 더 많은 원형질체가 생성되었으며, Novozym 234(2%)를 단독으로 처리하였을 때보다는 5000 unit/ml 의 ${\beta}-glucuronidase$와 혼합하였을 때 원형질체의 생성율이 더 높았다. 삼투 안정제로 0.6 M $MgSO_4$를 사용한 복합 효소 용액(pH 6.0)에 2시간 처리하였을 때 팽창된 포자로부터 단위ml당 $8.0{\times}10^6$의 원형질체를 분리할 수 있었다.

신증후군 환아 어머니의 부담감, 사회적 지지 및 삶의 질 (A Study Burden, Social Support and Quality of Life in Mothers of a Child with Nephrotic Syndrome)

  • The purpose of this study was to identity the level of burden, social support and quality of life of the subjects. The subjects of this study were 68 mothers of nephrotic syndrome patients whose children were hospitalized in one pediatric ward of the University Hospital in Seoul. The data was collected using questionnaires, and the period of the data collection was from Nov. 15 to Dec. 31, 1999. The instruments used for this study were the Burden Measurement Instrument developed by Montgomery et. al(1985), social support measurement instrument designed Brandt an Weinert(1978) and Quality of life scale designed by Ro,Yoo JA(1988). The data analysis was done by SPSS, t-test, ANOVA and the Pearson correlation coefficient. The results of were as follows. 1. The level of burden showed a mean score of 54.47, the level of social support, a mean score of 86.00 and the level quality of life, a mean score of 140.20. 2. The level of burden differed according to mother's religion, patient's purpose for admission and perceived patient's condition by mothers. 3. The level of social support and the level of quality of life differed according to perceived patient's condition by mothers. 4. There was a negative correlation between burden and social support(r=-.348, p<.001). Also, burden was negatively related with quality of life(r=-3.97, p<.001). Social support was positively related with quality of life(r=.064, p<.001).

Effects of Electric Current and Potential on the Electrokinetic Removal of Heavy Metals from an Abandoned Mine Tailings

  • In the removal of heavy metals from the mine deposit using electrokinetic processes, the effects of operation under both constant current and constant potential conditions were estimated. The results of soil pH distributions for DDW-20 V and DDW-100 mA cases after the electrokinetic remediation tests were observed. In the former case, soil pH was not much changed and kept to almost constant value just little higher than initial soil pH of 3.52, except near the cathode, which was about pH 5. While in the latter case, soil pHs of anode and the cathode regions were less than pH 3 and about 6, respectively. The electroosmotic flow to the cathode increased rapidly till 10 hrs and decreased steadily and then maintained to constant rate until the end of operation at constant current condition. Electric potential gradient was continuously increased to as much as 34.375 V/cm. At the steady state, values of the apparent electric conductivity for DDW-20 V and DDW-100 mA were around 40 ${\mu}\textrm{s}$/cm and 30 ${\mu}\textrm{s}$/cm, respectively. In the DDW-100mA test, Cu, Cd, and Zn except Pb showed the tendency of moving toward the cathode. While in the DDW-20 V case, it was observed that Cu, Zn, and Pb except Cd were not moved to any directions. The results of the tests demonstrated that the electrokinetic soil remediation process could be operated better under constant current condition than constant electric potential condition.

구강환경과 구취와의 관련성 (Relationship between oral environment and halitosis)

  • Objective : This study was performed in order to provide basic data for halitosis prevention and establish a device to efficiently eliminate halitosis and to analyze the factors that affect the halitosis. Methods : Oral examination on the Gingival index, CPITN, Tongue Plaque index, and OHI-S as well as halitosis measurement among 293 rural residents. Results : Gingival index was high at mild on female and at moderate on male(p=0.025). Sorting the result by age, mild was 54.1% in the 40s, and moderate was 49.5%, 42.0% and 70.0% each in the 50s, 60s, and the 70s(p=0.005). The need for dental plaque management was 100%. The need for scaling was high with 78.3%, 93.0%, 89.9%, and 90.0% each for the 40s, 50s, 60s, and 70s. The need for complex periodontal treatment was also high with 32.4%, 47.5%, 48.7% and 60.0%, each for the 40s, 50s, 60s, and 70s(p=0.050). The highest bad condition for OHI-S was 69.5%, and good being 18.9%, and very bad being 11.6%. For moderate tongue plaque index was 74.4% as the highest. As the level of education increased, the slight tongue plague was increased, but in contrast, the moderate and higher tongue plague index was decreased(p=0.010). OG under 50ppm was 61.1% on male and OG over 50ppm was 50.9% on female(p=0.041). In OG over 50ppm, CPITN was 52.1% and 41.9% in scaling and complex periodontal treatment group(p=0.018). OHI-S, in bad and very bad condition with OG, over 50ppm, was 48.7% and 46.9%(p=0.019). The higher tongue plague index showed significant amount of increase at OG and EG above 50ppm(p=0.006). $NH_3$, as the tongue plaque index increased, the wider range of distribution was shown(p=0.000). As for the multiple regression analysis result, there have been selected females and tongue plaque index as factors affecting OG. There have been selected age and tongue plaque index as factors affecting EG and there have been selected females as factors affecting on $NH_3$. Conclusions : With the aforementioned results in mind, the status of halitosis among rural residents is considered to bare a close relation with oral environments. we have to focus on correct tooth brushing methods and tongue brushing, with using tongue cleaner to remove fur of tongue plaque. Also, in order to analyze exactly the factors of individual halitosis, we need continuous and systematic study.

대기 $CO_2$ 상승시 인산공급이 식물체의 광합성 및 질소고정에 미치는 영향 (Effect of Phosphorus Stress on Photosynthesis and Nitrogen Fixation of Soybean Plant under $CO_2$ Enrichment)

  • 대기 $CO_2$ 상승시 인산공급이 대두식물체의 광합성 및 질소고정에 미치는 영향을 살펴보고저 Bradyrhizobium japonicum MN110을 접종한 대두 식물체를 인산결핍구(0.05 m-P) 및 대조구(1.0 mM-P)를 처리하여 정상 $CO_2(400\;{\mu}l/L)$와 상승 $CO_2(800\;{\mu}l/L)$의 조건인 phytotron에서 35일 동안 재배하였다. 인산결핍은 건물량을 정상 $CO_2$ 처리시 51%, 상승 $CO_2$ 처리시 64%를 감소시켰고 총 엽면적도 유의성있게 감소시켰으나 specific leaf weight는 증가시켰다. 광합성 속도 및 잎내부의 $CO_2$농도는 인산결집구에서 감소하였고 그 정도는 $CO_2$ 상승처리시 더욱 컸으며 질소고정능과 근류생체량도 인산결핍시 감소하였으나 잎의 질소농도는 인산대조구가 30% 감소하였다. 이러한 결과는 $CO_2$ 상승조건에서도 인산이 결핍될 경우 식물생육을 촉진시키지 못하며 인산은 정상 $CO_2$ 농도시 보다 $CO_2$ 상승조건에서 근류의 성장과 작용에 더욱 중요한 역할을 하는것으로 유추된다.

감귤원 토양에서 분리한 인산염 가용화 미생물 Bacillus sphaericus PSB-13의 특성 (Characteristics of Bacillus sphaericus PSB-13 as Phosphate Solublizing Bacterium Isolated from Citrus Orchard Soil)

  • 본 연구는 감귤원 토양유형별로 고정되어 있는 난용성 인산염함량을 파악하고 이를 생물학적으로 이용하고자 인산가용화 우수미생물을 선발하여 특성을 조사하였다. 토양중 난용성 인산염의 형태별 분포는 Al-P>Ca-P>Fe-P 순으로 화산회토 토양이 비화산회토 보다 많았다. PDA-P배지를 이용하여 인산염 가용화능이 우수한 Bacillus sphaericus를 분리하였으며 pH 4~5와 $30^{\circ}C$에서 생육이 제일 좋았다. Bacillus sphaericus은 $AlPO_4$에서 324.5 ppm, $Ca_{10}(PO_4)_6(OH)_2$에서 334.8 ppm, $Ca_3(PO_4)_2$에서 207.0 ppm의 유리인산을 생성하였으며 인산효소활성은 $35^{\circ}C$에서 배양할 때 높았다. Bacillus sphaericus를 Bentonite 등 담체에 고정 후 온도를 달리하여 7개월 보존기간 동안 밀도변화를 조사한 결과 $10^5c.f.u.\;g^{-1}$ 수준과 인산가용화능을 유지하여 추후 생물비료로 이용이 가능할 것으로 기대된다.

Bioactive Sphingolipids as Major Regulators of Coronary Artery Disease

  • Atherosclerosis is the deposition of plaque in the main arteries. It is an inflammatory condition involving the accumulation of macrophages and various lipids (low-density lipoprotein [LDL] cholesterol, ceramide, S1P). Moreover, endothelial cells, macrophages, leukocytes, and smooth muscle cells are the major players in the atherogenic process. Sphingolipids are now emerging as important regulators in various pathophysiological processes, including the atherogenic process. Various sphingolipids exist, such as the ceramides, ceramide-1-phosphate, sphingosine, sphinganine, sphingosine-1-phosphate (S1P), sphingomyelin, and hundreds of glycosphingolipids. Among these, ceramides, glycosphingolipids, and S1P play important roles in the atherogenic processes. The atherosclerotic plaque consists of higher amounts of ceramide, glycosphingolipids, and sphingomyelin. The inhibition of the de novo ceramide biosynthesis reduces the development of atherosclerosis. S1P regulates atherogenesis via binding to the S1P receptor (S1PR). Among the five S1PRs (S1PR1-5), S1PR1 and S1PR3 mainly exert anti-atherosclerotic properties. This review mainly focuses on the effects of ceramide and S1P via the S1PR in the development of atherosclerosis. Moreover, it discusses the recent findings and potential therapeutic implications in atherosclerosis.

시각 흐름 속도에 따른 가상현실 프로그램이 뇌졸중 환자의 보행에 미치는 영향 (The Effects of Visual Flow Speed's Modulation-Based Virtual Reality Program on Gait Function in Stroke Patients)

  • 본 연구는 시각흐름 속도에 따른 가상현실 프로그램이 뇌졸중 환자의 보행에 미치는 영향을 알아보고자 시행하였다. 연구 대상자는 서울시 소재 M재활전문병원에 입원 중인 뇌졸중 환자 중 무작위로 31명을 선정하였고 대상자는 가상현실 프로그램에 몰입하여 보행분석기 위에서 보행을 실시하였으며 가상현실 프로그램의 4가지 조건은 정상 보행과 0.25배, 1배, 2배의 시각흐름 속도변환 가상현실 프로그램으로 구성되었다. 중재동안 보행분석은 GaitRite보행분석시스템을 이용하여 보행 속도, 분속수, 활보장, 보장, 단하지지지 시간, 양하지지지 시간을 측정하였다. 그 결과 0.25배의 느린 시각흐름 가상현실 프로그램에서 연구 대상자는 보행속도, 분속수, 환측 활보장, 건측 활보장, 환측 보장, 건측 활보장이 통계적으로 유의하게 감소하였고(p<.05), 환측 단하지지지시간, 건측 단하지지지시간, 환측 양하 지지지시간, 건측 양하지지지시간이 통계적으로 유의하게 증가하였다(p<.05). 또한 2배의 빠른 시각흐름 가상현실 프로그램에서 연구 대상자는 보행속도, 분속수, 환측 활보장, 건측 활보장, 환측 보장, 건측 활보장, 환측 단하지지지시간이 유의하게 증가하였고(p<.05), 건측 단하지지지시간, 환측 양하지지지시간, 건측 양하지지지시간이 유의하게 감소하였다(p<.05). 따라서 가상현실 보행 프로그램의 적용 속도에 따라서 뇌졸중 환자의 보행 능력이 유의하게 차이를 나타낸다는 것을 알았다. 그러므로 뇌졸중 환자의 운동 프로그램을 계획할 때 보행 훈련 프로그램을 증가시킬 수 있는 다양한 훈련 프로그램이 요구된다.

반응표면분석법을 이용한 광학활성 phenyl oxirane의 회분식생산 최적화 (Optimization of Batch Production of Chiral Phenyl Oxirane by Response Surface Analysis)

  • 라세믹 phenyl oxirane 기질에 대한 asymmetric biohydrolysis 활성이 우수한 Rhodosporidium toruloides SJ-4를 생촉매로 이용하여 입체선택적 가수분해 반응을 통해 라세믹 phenyl oxirane 기질로부터 광학활성 (S)-phenyl oxirane를 회분식으로 생산하는 실험을 수행하였다. (R)-phenyl oxirane 이성질체에 대한 초기 가수분해 속도에 영향을 주는 실험인자들인 pH, 반응온도, DMSO cosolvent 첨가량 등에 대해 중심합성계획법을 이용한 반응표면 분석을 통해 가수분해반응 속도를 향상시킬 수 있는 최적 반응조건을 결정하였다. pH 7.4, 반응온도 34℃ 및 DMSO 첨가량 2.3%(v/v)의 조건에서 라세믹 기질 초기 농도 100mM로부터 약 10시간 정도의 반응을 통해 ee 값이 100%인 광학적으로 순수한 (S)-phenyl oxirane를 24% 정도 (이론수율 = 50%)의 높은 수율로 얻을 수 있었다. ^u Batch production of (S)-phenyl oxirane was investigated using epoxide hydrolase activity of Rhodosporidium toruloides SJ-4. Effect of reaction condition of asymmetric biohydrolysis of racemic phenyl oxirane was analyzed and optimized by response surface methodology. The optimal conditions of pH, temperature and DMSO cosolvent ratio were 7.4, 34 ℃, and 2.3%(v/v), respectively. The final yield was enhanced up to 67%, and reaction times required to reach 99% ee (enatiomeric excess) decreased down to 50% by response surface methodology Enantiopure (S)-phenyl oxirane with 100% enantiopurity and 24% yield (theoretical yield = 50%) was obtained from racemic substrate.
