  • Saprolegnia diclina, which was the pathogen causing death in snake fishes(Channa argus) at culture farm, was investigated using scanning electron microscope. It was found that Saprolegnia diclina infection caused snake fishes to fail gas change in the gills. Cell lysis as well as edematous disease and hyperplasia as a result of Saprolegnea diclina attachment on the surface of gills were observed. The granules, the mean diameters of which ranged from 6 to $7\;{\mu}m$, attaching on the surface of gills were found to be secondary zoospores of Saprolegnia diclina. The failures of gas exchange in the gill cells and circulation as a result of the osmotic dilution of the blood were supposed to be the main cause of death.

  • The active antifouling compounds coumarin and its derivatives were isolated from the cinnamon tree Cinnamomum loureiroi. The antifouling activities were determined using representative soft fouling organisms: the seaweed Ulvapertusa and diatom Navicula annexa. The chemical constituents with antifouling activities were identified as coumarin, hydroxylcoumarin, coumaric acid, and cinnamaldehyde by interpreting nuclear magnetic resonance, and high-resolution mass spectroscopy data. These compounds were isolated from 1.09 g of crude Cinnamomum sp. methanol extract, yielding approximately 18.4, 6.3, 9.8, and 14.7 mg of coumarin, hydroxylcoumarin, coumaric acid, and cinnamaldehyde, respectively. The compounds inhibited U. pertusa zoospores with $EC_{50}$ values of $0.13-0.25{\mu}g/mL$, and the diatom N. annexa with $EC_{50}$ of $0.21-0.81{\mu}g/mL$.

  • The population dynamics of Ecklonia stolonifera was investigated at Tongyeong coastal area in the South Sea of Korea. The blade length and width, stipe length and diameter, mean total length and mean weight were measured from randomly collected fifty individuals in every month. The plants in Tongyeong population were relatively bigger than those of Busan Korea and Aomori Japan in terms of their blade length and width. The population biomass was low in winter and high in spring and summer. The sporangial sori were found from August to December but prominent in September and October. About 90 % of the Tongyeong population was consisted of one- and two-year old plants and the zoosporangial sori were observed mostly in two-year old plants. It was different from the results of Busan and Aomori population in which the zoosporangial sori were observed from the plants older than three years. The new populations were developed from the zoospores of two-year old plants and their generation time was relatively shorter than those of Busan and Aomori.

  • Zoospore progenies of Phytophthora cactorum were relatively uniform and similar to their respective parent in the rate of linear extension, whereas oospore progenies were greatly various. Also, the character of colony pattern was quantitatively various in oospore progenies but not zoospore progenies. Therefore, these results suggested that multiple genes were involved in determining growth rate and colony morphology of P. cactorum, and support the hypothesis that species of Phytophthora are diploid during the vegetative phase.

  • Ultrastructural organization of encysting zoospores of Allomyces macrogynus was examined using the methods of cryofixation and freeze substitution. During enxcystment, obvious changes were observed at the surface of the plasma membrane and in the structure of gamma particles. Many multivesicular bodies associated with the plasma membrane were observed at early stages of encystment. After induction of encystment, vesicles were found within the gamma particles. These vesicles appeared to leave gamma particles after forming multivesicular bodies. This study suggests that the cell wall formation during encystment is mediated by the fusion of multivesicular bodies with the plasma membrane.

  • The objective of this study was to determine the effects of protein sources and roughage (R) to concentrate (C) ratio on in vitro fermentation parameters using a gas production technique. The experimental design was a $2{\times}5$ factorial arrangement in a completely randomized design (CRD). Factor A was 2 levels of protein sources yeast fermented cassava chip protein (YEFECAP) and soybean meal (SBM) and factor B was 5 levels of roughage to concentrate (R:C) ratio at 80:20, 60:40, 40:60, 20:80, and 0:100, respectively. Rice straw was used as a roughage source. It was found that gas production from the insoluble fraction (b) of YEFECAP supplemented group was significantly higher (p<0.05) than those in SBM supplemented group. Moreover, the intercept value (a), gas production from the insoluble fraction (b), gas production rate constants for the insoluble fraction (c), potential extent of gas production (a+b) and cumulative gas production at 96 h were influenced (p<0.01) by R:C ratio. In addition, protein source had no effect (p>0.05) on ether in vitro digestibility of dry matter (IVDMD) and organic (IVOMD) while R:C ratio affected the IVDMD and IVOMD (p<0.01). Moreover, YEFECAP supplanted group showed a significantly increased (p<0.05) total VFA and $C_3$ while $C_2$, $C_2:C_3$ and $CH_4$ production were decreased when compared with SBM supplemented group. In addition, a decreasing R:C ratio had a significant effect (p<0.05) on increasing total VFA, $C_3$ and $NH_3$-N, but decreasing the $C_2$, $C_2:C_3$ and CH4 production (p<0.01). Furthermore, total bacteria, Fibrobacter succinogenes, Ruminococcus flavefaciens and Ruminococcus albus populations in YEFECAP supplemented group were significantly higher (p<0.05) than those in the SBM supplemented group while fungal zoospores, methanogens and protozoal population remained unchanged (p>0.05) as compared between the two sources of protein. Moreover, fungal zoospores and total bacteria population were significantly increased (p<0.01) while, F. succinogenes, R. flavefaciens, R. albus, methanogens and protozoal population were decreased (p<0.01) with decreasing R:C ratio. In conclusion, YEFECAP has a potential for use as a protein source for improving rumen fermentation efficiency in ruminants.

  • In 2004 and 2005, Phytophthora rot on Luffa cylindrica which had not been reported in Korea occurred in the experimental field at Gyeongsangnam-do Agricultural Research and Extension Services. The disease initiated on leaves and fruits of the plant with small watersoaked dark brown spots and progressed rapidly. The causal pathogen isolated from diseased tissues was identified as a Phytophthora sp. because of aseptate mycelia and zoospores released directly from sporangia. The fungus grew well on PDA and 10% V-8 juice agar showing an arachnoid or rosaceous colony pattern. Sporangia formed abundantly in water and were conspicuously papillate, noncaducous, ovoid to globose, and sized $26\sim62\times19\sim38{\mu}m$. The fungus was heterothallic as producing sexual reproduction structures only when mated with only A2 standard mating type strain. Oogonia and oospores were spherical, smooth walled, and measured as $20\sim28{\mu}m\;and\;16\sim24{\mu}m$, respectively. Oospores were aplerotic and antheridia were amphigynous, unicellula and spherical. Chlamydospores were globose and $20\sim38{\mu}m$ in diameter. Optimum temperature for growth was around $28\sim30^{\circ}C$. The fungus caused similar symptoms on artificially inoculated plant and could be re-isolated thereby proving Koch's postulation. Based on the mycological criteria investigated in this study, the causal fungus of Luffa sylindrica rot was identified as Phytophthora nicotianae. This is the first report of Phytophthora rot of Luffa cylindrica caused by P. nicotianae in Korea.

  • An in vitro study was carried out to investigate the differences in rumen microbes and fiber degradation capacity between sheep and goats. Three local male sheep and three Inner Mongolia male cashmere goats (aged 1.5 to 2 years; weight 25.0 to 32.0 kg) were each fitted with a permanent rumen cannula used to provide rumen fluid. Cycloheximide was used to eliminate rumen anaerobic fungi. The results showed that the quantities of fungal zoospores in the culture fluid of the control group were significantly greater in the sheep than in the goats; however, bacteria and protozoa counts were significantly higher in goats than in sheep. The digestibility of straw dry matter did not differ significantly between the two species before elimination of fungi, but tended to be higher for sheep (55.4%) than for goats (53.3%). The results also indicated that bacteria counts increased significantly after elimination of anaerobic fungi; however, the digestibility of straw dry matter significantly decreased by 12.1% and 8.6% for sheep and goats respectively. This indicated that the anaerobic fungi of the rumen played an important role in degradation of fiber.

  • Ginseng (Panax ginseng) is one of the most widely cultivated medicinal herbs in Korea. During 3 or 5 years cultivation of ginseng, yield losses can reach as high as 30-60% due to numerous diseases in Korea. Among 106 Bacillus strains isolated from various plant internal roots, we selected three promising biocontrol agents by screening against root rot caused by Cylindrocarpon destructan in a greenhouse. Preinoculation of selected isolates to seed or one-year-old root resulted in stimulation of shoot and/or root growth of seedlings, and control of root rot in infested soils with Cylindronrpon destructans (P=0.05). Furthermore, drenching of selected isolates on seedling-growing pots reduced the incidence of Phytophthora blight when the seedlings were challenged with zoospores of Phytophthora cactorum (P=0.05). However, isolates B1141 and B1142 did not show any antifungal activity against various soilborne pathogens while B1146 did in vitro. Our results provide an insight that rhizobacteria can induce resistance against various plant diseases on ginseng even if any resistant breeds have been unknown on ginseng yet.

  • Yuzu (Citrus junos) gummosis disease, caused by Phytophthora nicotianae, was first reported in 1997. As known in citrus, Phytophthora is the most fastidious soil-borne pathogen to control. In order to minimize its damage to Citrus spp., integrated pest management (IPM) approach, including fungicide chemicals and resistant cultivars, is necessary. Therefore, in this study we tried to evaluate tolerance of yuzu cultivars and its related species against yuzu Phytophthora. Trifoliate orange was evaluated as a susceptible host to yuzu Phytophthora by both mycelial growth onto extract media and immature fruit inoculation. However, in zoospores spray-inoculation on 2-year-old cuttings tree, trifoliate orange appeared to have a resistant property as showing less than 6% diseased leaf rate. Among yuzu cultivars only 'Namhae No. 1' appeared resistant property against both P. nicotianae and P. citrophthora. The 'Namhae No. 1' showed 5.7% and 10.6% diseased leaf ratio by P. nicotianae and P. citrophthora, respectively. Clearly, in order to reduce damages caused by two yuzu Phytophthora, we suggest that growers may utilize a trifoliate orange as a rootstock and 'Namhae No. 1' as a scion for fruit production.
