  • This study was carried out to get a industrial information about a possibility of IgY antibody production, antimicrobial activity and properties of IgY antibody in egg yolk. The residual antibody activities of IgY were 91.5% after heating for 30min at 6$0^{\circ}C$. At the same time. these activities, were 73.2% after heating 15min at $65^{\circ}C$ and decreased vapidly at 7$0^{\circ}C$ and little antibody activity was left after heating for 15min at 8$0^{\circ}C$. When the prepared IgY was incubated at various pH ranges from 7 to 2 for 5hr at 37$^{\circ}C$., the antibody activity was stable from pH 7 to 4 and remained to 69.8% at pH 3.

  • It has been recognized that the hen, like its mammalian counterparts, provides young chicks with antibodies as protection against hostile invaders. This system facilitates the transfer of specific antibodies from serum to egg yolk, and provides a supply of antibodies called immunoglobulin Y(IgY) to the developing embryo and the hatched chick. The protection against pathogens that the relatively immune-incompetent newly hatched chick has, is through transmission of antibodies from the mother via the egg. Egg yolk, therefore, can be loaded with a large amount of IgY against pathogens which can immobilize the existing or invading pathogens during the embryo development or in day-old chicks. Thus, the immunization of laying hens to various pathogens results in production of different antigen-specific IgY in eggs. Egg yolk contains 8∼20 mg of jmmunoglobulins (IgY) per ml or 136∼340 mg per yolk suggesting that more than 30 g of IgY can be obtained from one immunized hen in a year. By immunizing laying hens with antigens and collecting IgY from egg yolk, low cost antibodies at less than $10 per g compared to more than $20,000 per g of mammalian IgG can be obtained. This IgY technology opens new potential market applications in medicine, public health, veterinary medicine and food safety. A broader use of IgY technology could be applied as biological or diagnostic tool, nutraceutical or functional food development, oral-supplementation for prophylaxis, and as pathogen-specific antimicrobial agents for infectious disease control. This paper has emphasized that when IgY-loaded chicken eggs are produced and consumed, the specific antibody binds, immobilizes and consequently reduces or inhibits the growth or colony forming abilities of microbial pathogens. This concept could serve as an alternative agent to replace the use of antibiotics, since today, more and more antibiotics are less effective in the treatment of infections, due to the emergence of drug-resistant bacteria.

  • This study was carried out to get a industrial information about a possibility of IgY antibody production, antimicrobial activity and Properties of IgY antibody in egg yolk. After the initial immunization the anti-Salmonella typhimurium IgY antibody level gradually were decreased from firth week to tenth week. On the other hand, the antibody level in the serum were increased from the first week, reaching its peak in the sixth week. Molecular weights of IgY were estimated approximately 72-75KD in a heavy chain and 30-40KD in a light chain by electrophoresis.

  • The three yolk polypeptides have been isolated and partially characterized. Their molecular weights of YPI, YP2, and YP3 were 48, 000, 47, 000, and 46, 000, respectivelv, as judged by SDS-polyacrvlamide gel electrophoresis. They have different digestion products upon in situ peptide mapping by limited proteolysis. Two-dimensional gel electrophoresis showed that their isoelectric points were heterogeneous from 5.92 to 6.54. And thew showed three different antigen-antibody reactions when each polvpeptides is reacted with antisera made to a mixture of all of three. These data reported here indicate that the yolk proteins are consisted of distinctive polypeptides in Drosophlla sp. (robusta species group).

  • Preventive and therapeutic effects of egg yolk antibody, immunoglobulin Y (IgY), against canine parvovirus (CPV) was evaluated in 25 pups orally challenged with CPV-2a. Oral administration of IgY using powder, paste and coated paste delivery systems was compared. Each type of IgY was administered orally for 17 days from 3 days before challenge. The group of pups administered coated IgY showed mild symptoms such as a moderate decrease in total white blood cell count, no depression, vomiting and diarrhea when compared with other groups. The overall clinical score of the group of pups administered coated IgY was significantly lower than that of the challenge control group. However, mortality did not differ among groups because not all pups received symptomatic treatment. These results implied that oral treatment of coated IgY could improve therapeutic effects against CPV challenge if pups received symptomatic treatment.

Nutritional Value Evaluation of Spray-Dried Egg Protein Containing Egg Yolk Antibody (난황항체를 함유한 분무건조전난의 영양적 가치 평가)

  • For the Exp. 1, twelve Duroc $\times$ Yorkshire $\times$ Landrace pigs (6.12 $\pm$ 0.24 kg average initial body weight and 21 d average age) were used in an 8-d metabolic assay to determine the effects of replacing spray-dried plasma protein (SDPP) with spray-dried egg Protein containing egg yolk antibody (SDCEP) on nutrient and amino acids digestibility in weaning Pigs. Experimental animals were fed diets containing SDPP, SDCEP (spray-dried commercial egg protein) and SDAEP. Protein content in the SDPP, SDCEP and SDAEP were 58.20, 45.83 and 41.85%, respectively. Pigs fed the SDPP diet tended to increase the apparent digestibility of dry matter and nitrogen compared to pigs fed the SDAEP diets without significant differences. The ileal digestibility of lysine and methionine for the SDAEP were greater than those for the SDPP, however, there are no significant differences between groups. For Exp.2, 36 Duroc $\times$ Yorkshire $\times$ Landrace pigs (4.11 $\pm$ 0.05 kg average initial body weight and 14 d average age) were used in a 10-day growth assay to determine the effects on growth performance and nutrient digestibility of replacing SDPP with SDAEP in early-weaning pigs. Experimental animals were fed diets containing CON (corn-dried whey-SBM based diet), SDAEP3 (corn-dried whey-SBM based diet + 3% SDAEP), SDAEP6 (corn-dried whey-SBM based diet + 6% SDAEP). The average daily weight gain of the pigs fed the SDAEP3 diet was higher than that for the pigs fed the CON and SDAEP6 diets (p < 0.05). SDAEP3 significantly increased the digestibility of dry matter and nitrogen compared to the CON and SDAEP6 diets (p < 0.05).

Effects of Egg Yolk Antibody Powder (IgY) Supplementation on Growth Performance, Blood Component Profile, Intestinal Microflora, and Immunoglobulin G in Meat Ducks (난황항체제제(IgY) 첨가가 육용오리의 생산성, 혈액성상, 장내 미생물 및 면역글로불린 함량에 미치는 영향)

  • This study was conducted to investigate the effect of dietary egg yolk antibody (IgY) powder supplementation on the growth performance, blood component profile, intestinal microflora, and immunoglobulin G in ducks. A total of 300 1-day-old ducks (Cherry Valley) were randomly divided into 5 groups, with 3 replicates of 20 birds. The treatment groups were the negative (NC), positive (PC), egg yolk antibody powder 0.1% (T1), egg yolk antibody powder 0.5% (T2), and egg yolk antibody powder 1.0% (T3) groups. In the growth performance of ducks during the entire experimental period, the IgY groups and positive control group (PC) showed significantly higher (P<0.05) body weights and body weight gains compared to the negative control group (NC). However, no significant differences were observed in the feed intake and feed conversion ratio. The blood component profiles showed that the IgY 0.1 and 1.0% groups decreased in total cholesterol content compared to the NC group. The aspartate aminotransferase (AST) and alanine aminotranferease (ALT) contents were lower in the IgY 1.0% group, but there were no significant differences. Regrading the leukocyte content after feeding IgY, the heterophil: lymphocyte ratio decreased in the IgY groups, especially in the IgY 0.1% group, which had a lower content than the other groups. However, these results showed no significant differences. The Lactobacillus count in the intestines significantly increased (P<0.05) in the IgY 0.1 and 0.5% groups, the level of IgY increased, and the Escherichia coli count decreased. However, no significant difference was observed in the total plate count. The immunoglobulin G content was lower in the IgY groups than in the NC group, and compared with the IgY groups, the IgY 0.5% had a lower content, which was not a significant difference.

Production of Specific Egg Yolk Antibodies in Chicken against Recombinant Fowl Adenovirus Fiber 2 Protein (재조합 가금 아데노바이러스 Fiber 2 단백질을 이용한 특이 난황 항체 생산)

  • Fowl adenovirus (FAV) is an important cause of several diseases, which result in considerable economic losses to the poultry farm. An outer capsid protein of FAV, fiber 2 is essential for virus growth, assembly or spread. This study was performed to produce about 22 kDa of recombinant fiber 2 protein and to immunize in laying hens to acquire the specific IgY antibody against the recombinant fiber 2. Laying hens were immunized with the recombinant fiber 2 intramuscularly in the breast muscle by injection 4 times at intervals of three weeks. At 12 weeks, serum- and egg yolk-antibody titers of hens against fiber 2 were increased up to 430,000 and 414,000, respectively. The recombinant fiber 2 could be recognized be the anti-His monoclonal antibody. Anti-fiber 2-IgY antibody could recognize the fiber 2 specifically in western blot analysis. These results suggested that the recombinant fiber 2 antigen could be used as an immunogen to elicit IgY antibody against fiber 2 and the anti-fiber 2-IgY could neutralize fowl adenovirus fiber 2 effectively.

  • Although several methods have been developed and applied to control swine respiratory diseases, the disease induces severe economic impact to swine industry worldwide. As one of the new trials, application of egg yolk antibody(IgY) was attempted for the purposes and immune response in sera and egg yolk was analysed with ELISA in previous study. In this study, immunological specificity of the IgY was analysed by Western blot analysis. In the analysis of causative agents of atrophic rhinits, B bronchiseptica and P multocida 4D, proteins of 33, 40, 43, 67 and 141 kDa were specifically reacted with IgY Also, 40 and 110 kDa proteins were identified as the major immunogens in P multocida 3A. In A pleuropneumoniae serotypes 2 and 5, 40 kDa and 47 kDa proteins were found to be the major reactive ones. These results suggested that egg yolk antibodies from immunized hens was specific with antigens injected into hens and partially purified antigens, outer membrane proteins and dermonecrotic toxin, were more effective than bacterin for the production of specific antibody.

Simulation of IgY(Immunoglobulin Yolk) Purification by SMB(Simulated Moving Bed) (SMB(Simulated Moving Bed)를 이용한 IgY(Immunoglobulin Yolk) 분리의 전산모사)

  • IgY(Immunoglobulin Yolk) is a specific antibody in egg yolk, and it protects human body from virus and antigen. There are a lot of egg yolk components such as lipoprotein and protein. To separate IgY, HPLC(High Performance Liquid Chromatography) and precipitation were used in a batch mode and SMB(Simulated Moving Bed) was adopted for continuous purification of yolk proteins. IgY and other proteins in yolk were separated by using three-zone SMB chromatography. Before performing SMB experiments, batch chromatography and PIM(pulse input method) were performed to find operation parameters and adsorption isotherms. The results of batch chromatography were compared with simulated results using Aspen chromatography. To find the most suitable separation condition in SMB chromatography, simulations in $m_2$-$m_3$ plane on the triangle theory were carried out. $m_2$ = 0.18, $m_3$ = 1.0 and ${\Delta}$t = 419 s are the best conditions for the highest purity of IgY. With this operating parameters(flow rate in three zone and switching time), the purity of raffinate results in 98.39% from Aspen chromatography simulation. Most of the simulation reached steadystate within second recycle.
