Ras homolog enriched in brain (Rheb) and FK506 binding protein 38 (FKBP38) are two important regulatory proteins in the mammalian target of rapamycin (mTOR) pathway. There are contradictory data on the interaction between Rheb and FKBP38 in human cells, but this association has not been examined in cashmere goat cells. To investigate the interaction between Rheb and FKBP38, we overexpressed goat Rheb and FKBP38 in goat fetal fibroblasts, extracted whole proteins, and performed coimmunoprecipitation to detect them by western blot. We found Rheb binds directly to FKBP38. Then, we constructed bait vectors (pGBKT7-Rheb/FKBP38) and prey vectors (pGADT7-Rheb/FKBP38), and examined their interaction by yeast two-hybrid assay. Their direct interaction was observed, regardless of which plasmid served as the prey or bait vector. These results indicate that the 2 proteins interact directly in vivo. Novel evidence is presented on the mTOR signal pathway in Cashmere goat cells.
본 연구에서는 Candida antarctica로부터 genomic DNA을 추출하여 lipase A(CalA) 유전자를 PCR 증폭하였고, 재조합 pColdIII/CalA, $pPICZ{\alpha}A$/CalA, $pPICZ{\alpha}A$/CalA$his{\times}6$을 구축하였다. 재조합 CalA 유전자의 기능적 발현을 위해 최적화된 시스템을 구축하고자 Escherichia coli와 Pichia pastoris 시스템에서 각각 수행하여 비교, 분석하였다. SDS PAGE gel을 통해 CalA의 발현의 여부 및 발현양을 확인하였고, pNPP를 기질로 한 가수분해 반응을 통해 활성을 측정하였다. E. coli 발현 시스템은 형질전환 방법이 간단하고, 미생물의 성장 속도가 빠르다는 장점을 갖지만 CalA의 활성이 0.02 Unit/ml으로 비교적 낮았으며 세포질 (cytoplasm)에서 발현되므로 비목적 단백질과의 분리 및 정제과정이 필요하다. 재조합 $pPICZ{\alpha}A$/CalA을 P. pastoris 시스템에서 발현한 경우 높은 발현양 뿐만 아니라 분비작용으로 인해 고순도 발현이 용이하였고, 활성 또한 약 0.7 Unit/ml으로 가장 높았다. 결론적으로 CalA의 기능적 발현을 위해 P. pastoris 시스템을 구축하는 것이 가장 적합함을 확인하였다.
To improve conventional yeast one-hybrid screening, we have developed an efficient mammalian one-hybrid system that allows rapid isolation of com-plementary DNAs which are able to induce human p14$^{ARF}$. tumor suppressor gene. A 1.5 kb promoter region of p14$^{ARF}$ was fused to EGFP to generate ARF promoter-EGFP reporter vector. This reporter plasmid was stably trans-fected into NIH3T3 cells for generation of reporter cell line. When the reporter cell line was infected with E2F-1 together with excess amounts of empty vector, the cells that received the positive modulator were readily identifiable by green fluorescence using FACS. The GFP-positive cells were cloned directly from the cultured cells and expanded in bulk culture. The genomic DNAs from GFP-positive cells were prepared and the CDNA insert in integrated retroviral genome was recovered by PCR using primers annealing to the retroviral vector sequences flanking the insert-cloning site. This system should be useful for efficient screening of expression CDNA libraries in mammalian cells to identify novel upstream regulators for spe-cific genes by one-hybrid interaction.ion.
More and more available fungal genome sequence data reveal a large amount of secondary metabolite (SM) biosynthetic 'dark matter' to be discovered. Heterogeneous expression is one of the most effective approaches to exploit these novel natural products, but it is limited by having to clone entire biosynthetic gene clusters (BGCs) without errors. So far, few effective technologies have been developed to manipulate the specific large DNA fragments in filamentous fungi. Here, we developed a fungal BGC-capturing system based on CRISPR/Cas9 cleavage in vitro. In our system, Cas9 protein was purified and CRISPR guide sequences in combination with in vivo yeast assembly were rationally designed. Using targeted cleavages of plasmid DNAs with linear (8.5 kb) or circular (8.5 kb and 28 kb) states, we were able to cleave the plasmids precisely, demonstrating the high efficiency of this system. Furthermore, we successfully captured the entire Nrc gene cluster from the genomic DNA of Neosartorya fischeri. Our results provide an easy and efficient approach to manipulate fungal genomic DNA based on the in vitro application of Cas9 endonuclease. Our methodology will lay a foundation for capturing entire groups of BGCs in filamentous fungi and accelerate fungal SMs mining.
효모의 RNAI유전자는 RNA processing에 관여 하는지 혹은 RNA transport에 관여 하는지 아직까지 유전자의 기능이 정확히 알려져 있지 않은 실정이다. 효모의 RNAI 유전자의 기능을 파악하기 위한 방법으로 본 연구에서는 rna1-1 mutant gene을 cloning하여 이에 대한 DNA sequence를 조사함으로써 RNAI 유전자와 rna1-1 유전자의 차이점을 이해하고자 하였다. rna1-1 marker를 갖는 yeast strain(R49)로 부터 genomic DNA를 추출하여 이를 BglII로 절단하여 genomic southern blotting을 행한 결과 wild type의 경우와 동일하게 3.4 kb에서 hybridization되는 signal을 얻었으며, RNAl 및 rna1-1이 yeast genome내에 single site로 존재함을 보여 주는 결과를 얻었다. mutant strain으로 부터 얻은 3.4 kb의 BglI fragment를 pUC19의 BamHI site에 subcloning하여 transformant들을 얻었고, wild type RNAl 유전자를 probe로 하여 rna1-1 mutant 유전자를 cloning할 수 있었다. pUC19에 cloning된 RNA1유전자 및 rna1-1유전자로부터 다양한 Ba131유도체를 얻어 이들에 대한 염기 서열을 비교한 결과 transcription initation site에서부터 down stream쪽으로 17 아미노산위치에 TCC가 TTC로 대치되어 있었으며 그 결과 serine이 phenylalanine으로 변환되는 결과를 얻었다. Wild type RNAI gene의 5'-region에는 3군데의 TATA-like sequence가 true TATA box인지 확인하기 위하여 Bal3I deletion에 의해 -103nt까지 deletion된 유도체를 얻었으며 ${\Delta}RNAI$, rna1-1, 81-2-6 clone이 rna1-1 allele와 complementation한지 확인하였으나 ${\Delta}RNAI$은 TS-complementation을 하지 못하였다. 따라서 현재까지 TATA-box라고 알려진 부분은 promoter로 작용하지 못함을 확인하였다.
분열효모 S. pombe의 포자형성은 배지상의 질소원의 고갈에 의하여 유도되어진다. 감수분열로부터 포자형성에 도달하는 과정에는 다수의 특이적인 유전자가 기능을 하고 있다. 본 연구에서는, 전포자막 구축에 필수적인 유전자 spo 5의 발현조절과 유전자의 메커니즘에 관하여 조사하였다. spo 5 유전자를 보유하는 약 5kb의 Hind III DNA 단편을 cloning 하였다. 이 단편으로부터 제한효소지도를 작성하여 얻어진 DNA 단편을 probe로 하여, RNA blot-hybridization를 이행하였다. 이 결과, 최소배지의 hetro matting-type 균주 (CD16-1)로 부터 조제한 mRNA가 검출되었다. 그리고 이 전사산물을 전사레밸에서 해석하기 위하여, homo matting-type (CD16-3) 균주를 질소원이 함유되지 않은 포자형성배지에서 배양한 후, 동일한 방법으로 mRNA를 조제하여 Northern hybridization으로 조사하였다. 그 결과, 이들 세포에서는 3.2kb에서만 전사산물이 검출되었으며, 2.5kb의 mRNA는 검출되지 않았다. 이상의 결과로 부터 spo 5 유전자를 coding하는 전사산물인 2.5kb의 mRNA는 질소원의 고갈된 상태하에서, 접합형 유전자좌의 hetro 접합성을 요구하는 것으로 입증하였다. spo 5 유전자의 전사발현은 질소원이 결핍과 접합형 유전자좌의 구성에 따른 환경요인과 유전적 요인에 의해서 제어되어지고 있다는 것을 입증하였다.
유전자의 재조합 균주의 생산성을 향상시키기 위한 발효시스템 개발을 목적으로 regulated promoter 인 SUC2 gene 갖는 유전자 재조합 S. cerebisiae를 모델로하여 유전자 산물인 Invertase 발현에 미치는 글루코오스 농도의 영향, 발효 중 플라스미드의 불안정성, 생육특성 및 continuous fed batch system을 연구하였다. 유전자 재조합 균주는 biphasic growth 현상을 보였으며 글루코오스 농도가 0.9g/$\ell$에서 2.2g/L로 증가함에 따라 비증식 속도는 0.224 h$^{-1}$에서 0.226 h$^{-1}$로 증가했으며 숙주효모보다 낮은 값을 나타내었다. 유전자 재조합 균주의 invertase 생산은 발효조내의 글루코오스 농도에 크게 영향을 받아 글루코오스 농도가 0.25~0.4g/L로 감소될 때 invertase 생산이 시작되었으며 회분발효중 플라스미드의 분리는 글루코오스 자화기간 동안에 빈번히 일어났으나 에탄을 자화기간에는 완만해지는 경향을 보였다. 또한 통기를 해주지 않고 배지의 용존산소만으로 배양시킨 결과 통기를 한 경우에 비하여 invertase의 비활성과 총활성이 각각 1.5 및 1.3 배 증가되었다. 균체의 증식단계와 유전자의 발현단계로 구분하여 발효시키기 위하여 영양분을 함유한 글루코오스 용액을 48m1/h(0.096g 글루코오스/48L의 유량으로 12시간 연속적으로 공급하여 fed batch 배양한 결과 invertase의 비활성과 총활성이 비통기적 회분배양 보다 각각 1.74, 2.74배 증가되었다.
치오레독신 환원효소(TrxR)를 encoding하는 Schizosaccharomyces pombe의 유일한 $TrxR^+$ 유전자는 스트레스 반응 전사인자인 Pap1의 매개에 의하여 스트레스 유발 인자들에 의하여 양성적으로 조절됨이 발견되었다. 본 연구에서는, TrxR 과잉 발현 재조합 플라스미드 pHSM10을 사용하여 S. pombe TrxR의 방어적 역할들이 평가되었다. 과산화수소($H_2O_2$)와 superoxide anion을 생성하는 menadione (MD)의 존재 하에서, S. pombe TrxR은 세포성장과 총 글루타치온 (GSH) 수준을 증강시키나, 세포 내 활성산소종(ROS) 수준은 감소시켰다. $H_2O_2$와 MD에 의하여 크게 영향 받지 않는 일산화질소(NO) 수준에는 유의성 없는 효과를 보였다. S. pombe TrxR은 sodium nitroprusside(SNP)에 의하여 생성되는 NO를 소거할 수 있었으나, SNP에 노출된 세포들의 성장, ROS 수준이나 총 글루타치온 수준에는 영향을 보이지 않았다. S. pombe TrxR은 질소 결핍(nitrogen starvation)에 의하여 감소되는 세포 성장 및 총 글루타치온 수준을 증가시키며, 질소 결핍에 의하여 생성되는 ROS와 NO를 소거하였다. 요약하건대, S. pombe TrxR은 산화적, 일산화질소 및 영양 스트레스로부터 효모 세포를 보호하지만, 공통적인 기전에 의하지는 않는다.
세포주기조절에서 유전자 발현의 조절은 매우 중요한 부분이다. 본 연구에서는 인간의 유전자인 CIP29/Hcc1과 상동성을 가지는 분열형 효모의 새로운 유전자 mas1+을 분리하였다. 중합효소연쇄반응을 수행하여 cDNA를 얻고 이 cDNA의 염기서열을 분석한 결과 mas1+의 전체 염기서열은 735 bp로서, 245개의 아미노산을 암호화하고 있다. mas1+의 프로모터에서는 M-G1에 특이적인 전사를 보이는 유전자들에 보존되어 있는 PCB 서열이 발견되었다. 세포주기별 mas1+의 전사 수준을 분석한 결과 격막이 형성된 세포수의 빈도를 나타내는 격막 세포지표 의 양상과 유사하게 발현하는 것을 확인하였다. mas1+ 결손 돌연변이를 $25^{\circ}C$와 $36^{\circ}C$에서 배양한 결과, 세포질 분열과정이 늦어진 다중격막 세포의 빈도가 증가하였다. 이를 FACS로 분석하여 DNA 함량이 2C, 4C와 6C등이 형성됨을 확인하였다. mas1+결손 돌연변이 세포를 질소 결핍 배양액에서 배양한 결과 다중격막 세포의 형성이 확연히 증가하였는데 이는 질소 결핍에 따른 세포분열의 가속화 단계에서 mas1+의 결손이 특히 부정적 영향을 초래함을 시사한다. mas1+ 유전자 결손 돌연변이 세포에 mas1+을 포함한 plasmid를 형질전환한 후 mas1+의 발현을 유도한 결과 정상의 세포 형태로 전환됨을 확인하였다. Mas1 단백질에 EGFP를 융합시켜 발현을 유도한 결과 핵내에서 위치함을 분열형 효모와 인간 배양세포인 HeLa에서 확인하였다. 또한, mas1+ 결손 돌연변이에서 상동성을 가지는 인간 유전자 CIP29/Hcc1을 발현시킨 결과 multi-septate 세포가 줄어들었다. 한편, 생쥐의 배발달 단계에 따른 CIP29 유전자의 전사체 수준은 세포 분열이 활발한 시기에 증가하였다. 이상의 결과들은 Mas1은 인간의 핵단백질인 유전자 CIP29/Hcc1과 구조 기능적으로 상동성을 가지며, 세포주기 중 M-G1에 속하는 세포질 분열에 연관되어 있음을 시사한다.
