비 생물학적 스트레스 시 벼에서 OsABF3 유전자 분리와 ABA 신호전달 대한 연구 (Studies on OsABF3 Gene Isolation and ABA Signal Transduction in Rice Plants Against Abiotic Stress)

  • ABA는 식물에서 비 생물학적 스트레스 내성에 관여하는 중요한 식물 호르몬이다. 애기장대의 group A bZIP 전사인자는 ABA 신호전달 과정에 중요한 역할을 한다고 알려져 있다. 그러나 벼에서는 group A bZIP 전사인자의 기능이 잘 알려져 있지 않다. 따라서 우리는 벼에서 group A bZIP 전사인자인 OsABF3(Oryza sativa ABA responsive element binding factor 3)를 연구하였다. 이를 위해 벼의 다양한 조직과 다양한 스트레스(가뭄, 염분, 저온, ABA, 산화 스트레스)에 따른OsABF3발현 패턴을 분석하였다. 또한 maize의 원형질체에서 GFP fusion 벡터를 사용한 세포 내 위치 분석을 통해 OsABF3가 핵단백질이라는 것을 확인하였다. Yeast one-hybrid 실험을 통해 OsABF3의 C-terminal 부분이 ABREs에 결합한다는 것과 N-terminal 부분이 하위 유전자의 transactivation 하는데 필요하다는 것을 알 수 있었다. 그리고 T-DNA가 삽입된 OsABF3의 homozygous 돌연변이체가 야생형과 과발현체에 비해 발아와 발아 후 단계에서 고농도의 ABA에 대한 민감도가 더 감소한 것을 알 수 있었다. 결과적으로 종합해 볼 때 OsABF3는 ABA의 의존적인 경로를 통해 비 생물학적 스트레스에 반응하는 유전자의 발현을 조절하는 기능을 하는 전사 조절자이다. 또한 OsABF3의 transactivation을 측정하는 실험에 있어서 억제 domain이 존재한다는 결과를 얻었다.

  • 식물에서 재조합 단백질을 생산하는 것은 여러 가지 장점이 있다. 식물은 인간 병원체에 감염되지 않으며, 박테리아와 달리 내독소를 생산하지 않는다. 엽록체 형질전환은 핵 형질전환에 비해 안정적으로 많은 유전자를 발현시킬 수 있는 등 다양한 이점이 있다. 항균펩타이드(AMP)는 많은 동물들이 가지고 있는 선천면역의 일종으로, 소량이라도 항균력을 가지며, 기존 항생제와 다르게 쉽게 내성균이 생기지 않는다. 항균펩타이드인 moricin은 누에나방의 한 종류인 Bombyx mori에서 분리되었으며, C-말단은 염기성 아미노산이 모여 있고, N-말단은 α-helix 구조를 가지고 있다. Moricin을 생산할 때 SUMO와 6xHis tag를 융합하여 사용하였다. 발현된 moricin의 용해성과 안정성을 높이기 위해 SUMO를, 발현된 moricin을 정제하기 위하여 6xHis tag를 이용하였다. 본 연구에서 담배 엽록체와 대장균에서 항균펩타이드를 발현하기 위한 형질전환벡터를 제작하였다. 또한, 엽록체와 박테리아의 전사 및 번역의 유사성을 이용하여 대장균에서 단백질의 발현을 확인하였다. 발현된 moricin을 Ni 컬럼 및 SUMOase를 처리하여 정제하고 agar diffusion assay를 이용하여 항균 활성을 확인하였다.

Enhanced and Targeted Expression of Fungal Phytase in Saccharomyces cerevisiae

  • Phytase improves the bioavailability of phytate phosphorus in plant foods to humans and animals, and reduces the phosphorus pollution of animal waste. In order to express a high level of fungal phytase in Saccharomyces cerevisiae, various expression vectors were constructed with different combinations of promoters, translation enhancers, signal peptides, and terminator. Three different promoters fused to the phytase gene (phyA) from Aspergillus niger were tested: a galactokinase (GAL1) promoter, glyceraldehyde-3-phosphate dehydrogenase (GPD) promoter, and yeast hybrid ADH2-GPD promoter consisting of alcohol dehydrogenase II (ADH2) and a GPD promoter. The signal peptides of phytase, glucose oxidase (GO), and rice amylase 1A(RAmy1A) were included. Plus, the translation enhancers of the ${\Omega}$ sequence and UTR70 from the tobacco mosaic virus (TMV) and spinach, respectively, were also tested. Among the recombinant vectors, pGphyA06 containing the GPD promoter, the ${\Omega}$ sequence, RAmy1A, and GAL7 terminator expressed the highest phytase activity in a culture filtrate, which was estimated at 20 IU/ml. An intracellular localization of the expressed phytase activity in a culture filtrate, which was estimated at 20 IU/ml. An intracellular localization of the expressed phytase was also performed by inserting an endoplasmic reticulum (ER) retention signal, KDEL sequence, into the C-terminus of the phytase within the vector pHphyA-6. It appeared that the KDEL sequence directed most of the early expression of phytase into the intracellular compartment yet more than $60\%$ of the total phytase activity was still retained within the cell even after the prolonged (>3 days) incubation of the transformant. However, the intracellular enzyme activity of the transformant without a KDEL sequence was as high as that of the extracellular one, thereby strongly suggesting that the secretion of phytase in S. cerevisiae appeared to be the rate-limiting step for the expression of a large amount of extracellular recombinant phytase, when compared with other yeasts.

Isolation, Characterization, and Molecular Cloning of the cDNA Encoding a Novel Phytase from Aspergillus niger 113 and High Expression in Pichia pastoris

  • Phytases catalyze the release of phosphate from phytic acid. Phytase-producing microorganisms were selected by culturing the soil extracts on agar plates containing phytic acid. Two hundred colonies that exhibited potential phytase activity were selected for further study. The colony showing the highest phytase activity was identified as Aspergillus niger and designated strain 113. The phytase gene from A. niger 113 (phyI1) was isolated, cloned, and characterized. The nucleotide and deduced amino acid sequence identity between phyI1 and phyA from NRRL3135 were 90% and 98%, respectively. The identity between phyI1 and phyA from SK-57 was 89% and 96%. A synthetic phytase gene, phyI1s, was synthesized by successive PCR and transformed into the yeast expression vector carrying a signal peptide that was designed and synthesized using P. pastoris biased codon. For the phytase expression and secretion, the construct was integrated into the genome of P. pastoris by homologous recombination. Over-expressing strains were selected and fermented. It was discovered that ~4.2 g phytase could be purified from one liter of culture fluid. The activity of the resulting phytase was 9.5 U/mg. Due to the heavy glycosylation, the expressed phytase varied in size (120, 95, 85, and 64 kDa), but could be deglycosylated to a homogeneous 64 kDa species. An enzymatic kinetics analysis showed that the phytase had two pH optima (pH 2.0 and pH 5.0) and an optimum temperature of $60^{\circ}C$.

  • Kluyveromyces marxianus 유래의 exoinulinase (INU1) 분비 서열과 GAL10 promoter를 이용하여 Clostridium thermocellum endoglucanase A gene (celA)의 과발현과 분비 성능 검증연구를 수행하였다. 자체 분비 서열을 가지는 pYEG- CT1 과 INU1 분비 서열을 가지는 pYInu-CT1, 두개의 plasmid는 S. cer-evisiae SEY2102와 S. cerevisiae 2805에 각각 형질전환시켜 ur-acil이 결핍된 배지에서 선별하였다. celA gene 자체 분비 서열보다 INU1 분비 서열을 사용했을 때 발현량과 분비효율은 각각 약 $18{\sim}22%$ 와 11% 향상되었다. 이 중 361 unit/l의 발현율과 89%의 plasmid 안정성, 그리고 70%의 분비효율을 보인 S. cerevisiae 2805/pYinu-CT1 효모 형질전환체를 cellolose를 효과적으로 분해하는 재조합 효모 생균체로 선별하였다. Galactose 배지내에서 S. cerevisiae 2805/ pYInu-CT1의 발효조 회분배양 결과, 총발현량과 분비효율은 각각 418 unit/l 와 73% 로 나타났다. 또한 분비된 endoglucanase A는 분자량 100kDa 이상에서 활성 밴드를 보여, N-linked 당쇄부가에 의해 상당한 비율의 당쇄가 부가됨을 추측할 수 있었다.

Construction of High Sensitive Detection System for Endocrine Disruptors with Yeast n-Alkane-assimilating Yarrowia lipolytica

  • To construct a highly sensitive detection system for endocrine disruptors (EDs), we have compared the activity of promoters with the n-alkane-inducible cytochrome P450 gene (ALK1), isocitrate lyase gene (ICL1), ribosomal protein S7 gene (RPS7), and the translation elongation factor-1${\alpha}$ gene (TEF1) for the heterologous gene in Yarrowia lipolytica. The promoters were introduced into the upstream of the lacZ or hERa reporter genes, respectively, and the activity was evaluated by ${\beta}$-galactosidase assay for lacZ and Western blot analysis for hER${\alpha}$. The expression analysis revealed that the ALK1 and ICL1 promoters were induced by n-decane and by EtOH, respectively. The constitutive promoter of RPS7 and TEF1 showed mostly a high level of expression in the presence of glucose and glycerol, respectively. In particular, the TEF1 promoter showed the highest ${\beta}$-galactosidase activity and a significant signal by Western blotting with the anti-estrogen receptor, compared with the other promoters. Moreover, the detection system was constructed with promoters linked to the upstream of the expression vector for the hER${\alpha}$ gene transformed into the Y. lipolytica with a chromosome-integrated lacZ reporter gene under the control of estrogen response elements (EREs). It was indicated that a combination of pTEF1p-hER${\alpha}$ and CXAU1-2XERE was the most effective system for the $E_2$-dependent induction of the ${\beta}$-galactosidase activity. This system showed the highest ${\beta}$-galactosidase activity at $10^{-6}\;M\;E_2$, and the activity could be detected at even the concentration of $10^{-10}\;M\;E_2$. As a result, we have constructed a strongly sensitive detection system with Y. lipolitica to evaluate recognized/suspected ED chemicals, such as natural/synthetic hormones, pesticides, and commercial chemicals. The results demonstrate the utility, sensitivity, and reproducibility of the system for identifying and characterizing environmental estrogens.

Functional Expression of the Internal Rotenone-Insensitive NADH-Quinone Oxidoreductase (NDI1) Gene of Saccharomyces cerevisiae in Human HeLa Cells

  • Many studies propose that dysfunction of mitochondrial proton-translocating NADH-ubiquinone oxidoreductase (complex I) is associated with neurodegenerative disorders, such as Parkinson's disease and Huntington's disease. Mammalian mitochondrial proton-translocating NADH-quinone oxidoreductase (complex I) consists of at least 46 different subunits. In contrast, the NDI1 gene of Saccharomyces cerevisiae is a single subunit rotenone-insensitive NADH-quinone oxidoreductase that is located on the matrix side of the inner mitochondrial membrane. With a recombinant adeno-associated virus vector carrying the NDI1 gene (rAAV-NDI1) as the gene delivery method, we were able to attain high transduction efficiencies even in the human epithelial cervical cancer cells that are difficult to transfect by lipofection or calcium phosphate precipitation methods. Using a rAAV-NDI1, we demonstrated that the Ndi1 enzyme is successfully expressed in HeLa cells. The expressed Ndi1 enzyme was recognized to be localized in mitochondria by confocal immunofluorescence microscopic analyses and immunoblotting. Using digitonin-permeabilized cells, it was shown that the NADH oxidase activity of the NDI1-transduced HeLa cells were not affected by rotenone which is inhibitor of complex I, but was inhibited by flavone and antimycin A. The NDI1-transduced cells were able to grow in media containing rotenone. In contrast, control cells that did not receive the NDI1 gene failed to survive. In particular, in the NDI1-transduced cells, the yeast enzyme becomes integrated into the human respiratory chain. It is concluded that the NDI1 gene provides a potentially useful tool for gene therapy of mitochondrial diseases caused by complex I deficiency.

Expression of Hepatitis B Virus S Gene in Pichia pastoris and Application of the Product for Detection of Anti-HBs Antibody

  • Antibody to hepatitis B surface antigen (HBsAb) is the important serological marker of the hepatitis B virus (HBV) infection. Conventionally, the hepatitis B surface antigen (HBsAg) obtained from the plasma of HBV carriers is used as the diagnostic antigen for detection of HBsAb. This blood-origin antigen has some disadvantages involved in high cost, over-elaborate preparation, risk of infection, et al. In an attempt to explore the suitable recombinant HBsAg for the diagnostic purpose, the HBV S gene was expressed in Pichia pastoris and the product was applied for detection of HBsAb. Hepatitis B virus S gene was inserted into the yeast vector and the expressed product was analyzed by sodium dodecyl sulphate polyacrolamide gel electrophoresis (SDS-PAGE), immunoblot, electronic microscope and enzyme linked immunosorbent assay (ELISA). The preparations of synthesized S protein were applied to detect HBsAb by sandwich ELISA. The S gene encoding the 226 amino acid of HBsAg carrying ahexa-histidine tag at C terminus was successfully expressed in Pichia pastoris. The His-Tagged S protein in this strain was expressed at a level of about 14.5% of total cell protein. Immunoblot showed the recombinant HBsAg recognized by monoclonal HBsAb and there was no cross reaction between all proteins from the host and normal sera. HBsAb detection indicated that the sensitivity reached 10 mIu (micro international unit)/ml and the specificity was 100% with HBsAb standard of National Center for Clinical Laboratories. A total of 293 random sera were assayed using recombinant S protein and a commercial HBsAb ELISA kit (produced by blood-origin HBsAg), 35 HBsAb positive sera and 258 HBsAb negative sera were examined. The same results were obtained with two different reagents and there was no significant difference in the value of S/CO between the two reagents. The recombinant HBV S protein with good immunoreactivity and specificity was successfully expressed in Pichia pastoris. The reagent for HBsAb detection prepared by Pichia pastoris-derived S protein showed high sensitivity and specificity for detection of HBsAb standard. And a good correlation was obtained between the reagent produced by recombinant S protein and commercial kit produced by blood-origin HBsAg in random samples.

Candida fermentati SI의 exo-β-(1,3)-glucanase유전자의 클로닝 및 그 특성 (Molecular Cloning and Functional Expression of Extracellular Exo-β-(1,3)-Glucanase from Candida fermentati SI)

  • 본 연구에서는 C. fermentati SI가 생산하는 isoflavone 배당체 가수분해 효소를 클로닝하여 염기 서열을 밝힌 뒤 P. pastoris X-33에 형질전환하여 재조합 효소의 과발현을 시켰고, 또한 재조합 isoflavone 가수분해 효소의 효소학적 특성을 조사하였다. 재조합 isoflavone 가수분해 효소의 분자량은 약 50.4 kDa이었으며, Meyerozyma guilliermondii ATCC 6260의 exo-1,3-β-glucanase와 96%로 가장 높은 homology를 나타내었다. exo-1,3-β-glucanase의 ORF는 pPICZA 벡터로 클로닝 후 P. pastoris X-33으로 형질전환을 하였으며, His6-tag을 이용하여 효소를 정제하였다. 정제된 효소는 citrate phosphate buffer pH 4.5에서 최적 활성을 나타내었으며, 효소의 최적 활성 온도는 40℃로 나타났다. 40℃이상에서는 효소의 활성이 급격하게 감소함을 확인 하였으며, pH 안정성을 조사한 결과 비교적 넓은 범위인 4−8 사이에서 80%이상의 활성을 유지하였다. 따라서, 재조합 효소의 과발현을 통해 isoflavone aglycone의 효율적인 생산에 이용할 수 있을 것으로 사료된다.

Schizosaccharomyces pombe 포자형성유전자 (spo 5)의 발현조절기구의 해석 (Expression and Regulatory Analysis of Sporulation Gene (spo 5) in Schizosaccharomyces pombe)

  • 분열효모 S. pombe의 포자형성은 배지상의 질소원의 고갈에 의하여 유도되어진다. 감수분열로부터 포자형성에 도달하는 과정에는 다수의 특이적인 유전자가 기능을 하고 있다. 본 연구에서는, 전포자막 구축에 필수적인 유전자 spo 5의 발현조절과 유전자의 메커니즘에 관하여 조사하였다. spo 5 유전자를 보유하는 약 5kb의 Hind III DNA 단편을 cloning 하였다. 이 단편으로부터 제한효소지도를 작성하여 얻어진 DNA 단편을 probe로 하여, RNA blot-hybridization를 이행하였다. 이 결과, 최소배지의 hetro matting-type 균주 (CD16-1)로 부터 조제한 mRNA가 검출되었다. 그리고 이 전사산물을 전사레밸에서 해석하기 위하여, homo matting-type (CD16-3) 균주를 질소원이 함유되지 않은 포자형성배지에서 배양한 후, 동일한 방법으로 mRNA를 조제하여 Northern hybridization으로 조사하였다. 그 결과, 이들 세포에서는 3.2kb에서만 전사산물이 검출되었으며, 2.5kb의 mRNA는 검출되지 않았다. 이상의 결과로 부터 spo 5 유전자를 coding하는 전사산물인 2.5kb의 mRNA는 질소원의 고갈된 상태하에서, 접합형 유전자좌의 hetro 접합성을 요구하는 것으로 입증하였다. spo 5 유전자의 전사발현은 질소원이 결핍과 접합형 유전자좌의 구성에 따른 환경요인과 유전적 요인에 의해서 제어되어지고 있다는 것을 입증하였다.
