경구개 창상 치유과정에 있어서의 비만세포의 분포

  • The author observed the distribution of mast cells in the hard palate wound healing of healthy male albino rats weighing about 100gm. The mild wounds were made antero-posterior linealy by sugical knife. The results were as follow : 1. At the from three days to five days after wound, the number of mast cells were markedly increased. 2. In the wound healing process, mast cells were more distributed in the lamina propria than in the submucosa.

백서에서 스테로이드 투여가 창상 파열장력에 미치는 영향 (The Effect of Administration of Steroid on the Wound Breaking Tensile Strength in Rats)

  • Purpose: The successful wound healing means the scarless wound with adequate strength. It has been shown in vivo and in vitro that steroid retard the collagen synthesis. We studied the effect of steroid on the wound breaking strength in rats. Methods: 40 Sprague - Dawley rats were evenly assigned to two groups. One group was served as control, the other group was experimental. We made dorsal midline incision and closed the wound. In the experimental group, we medicated methylprednisolone (0.15 mg/g/day) for 1 week. Then, we compared the differences of the breaking strength and microscopic histology between control and experimental group at 2, 4, 6 and 8 week. Results: Up to 4 week, the breaking strength of the experimental group was markedly decreased than that of control group, while at 6 week the strength of experimental group attained to that of control group nearly. In histologic findings, control group demonstrated dense organization of collagen to experimental group at 4th week. Conclusion: In this experiment, steroid significantly inhibited the strength gain in wound at early period of the wound healing process. When using steroid after the surgery or the wound formation, it is desirable to administrate carefully and need thorough wound management to prevent delayed wound healing.

술전 및 술후 방사선조사가 백서 배부 창상치유에 미치는 영향 (EFFECTS OF PREOPERATIVE AND POSTOPERATIVE RADIATION ON THE HEALING OF SURGICAL WOUND)

  • This study was performed to evaluate the effects of preoperative and postoperative radiation on the healing of surgical wound and the relationship between surgery-radiation interaval histopathologically. Experimental animals were 64 rats of Sprague-Dawely strain weighing about 180grams. In postoperative radiation group, a single dose of 1000 rads irradiation was delivered on 1,2,3, weeks after incision and 24 animals were sacrificed on the 1st, 3rd, 7th, and 14th day after radiation. In preoperative radiation group incision was performed on 1,2,3,4 weeks after a single dose of 1000 rads and 32 animals were sacrificed on the 1st, 3rd, 7th, and 14th after incision. Tissue specimens were prepared as usual methods and stained with hematoxyline-eosin for ordinary light microscopy. Histopathologic study revealed the following favorable results : 1. In 2 and 3 weeks radiation group after incision, the healing process was unaffected by radiation. 2. In 1 week radiation group after incision, the healing process was slightly retarded, as compared with 2,3 weeks radiation group after incision. 3. In 1,2 and 3 weeks incision group after radiation, the healing process appeared about 7 days later than that of control group. 4. In 4 weeks incision after radiation, the healing process was unaffected by radiation.

백서에서 Pulsed Nd : YAG Laser에 의한 치은절제술후의 치유양상 (Wound Healing After Gingivectomy Using Pulsed Nd : YAG Laser In The Rat)

  • Recently, dental laser have been applied for removal of soft tissues, hemostasis and blood coagulation, removal of benign and malignant tumor, treatment of leukoplakia, aphthous ulcer and herpetic lesion, implant second surgery, removal of granulation tissue, frenectomy, clinical crown lengthening, gingivectomy, gingivoplasty, and treatment of dentin hypersensitivity. Even though the frequency of laser treatment is increasing, the research on the healing process after gingivectomy using pulsed Nd : YAG laser is very rare. The purpose of this study was to observe and compare the wound healing after gingivectomy using scalpel and pulsed Nd : YAG laser in the rat. Gingivectomy was performed using pulsed Nd : YAG laser(SUNRISE Technologies, U.S.A., 1.5 Watts, 10 pps) on the buccal gingiva of right maxillary first molar and using scalpel(No.12) on the contralateral side. Those sites treated by surgical scalpel were designated as the control, and by pulsed Nd : YAG laser as the experimental group. Animals were sacrificed at 1, 2, 3, 5, 7, 11 and 14 days postoperatively, and specimens were histologically observed under light microscope. The results were as follows : 1. Clinical observation Normal color and shape were observed at the 5th day ill the control group and the 7th day in the experimental group. 2. Histologic findings 1) In the control group, denser inflammatory infiltration was observed. 2) Epithelialization started at the 2nd day in the control group, similar to the experimental group, and completed at the 11th to the 14th day postoperatively. 3) In the experimental group, connective tissue showed the vacuole formation and degenerative change during early healing period. Healing of connective tissue was slower in the experimental group than in the control group by 2 days. 4) In the both groups, wound healing was completed at the 2nd week. From this study, gingivectomy using pulsed Nd : YAG laser seems to result in a little delayed wound healing process, compared to the gingivectomy using scalpel. Considering the clinical advantages of laser surgery, pulsed Nd : YAG laser might be useful device for gingivectomy.

세포 이동능력 분석을 위한 96-Well Plate 전용 Lab-Made Wound Maker (A Lab-Made Wound Maker for Analysis of Cell Migration in a 96-Well Plate)

  • Cell migration은 embryogenesis 혹은 cancer metastasis 이외에, 물리적 손상에 의한 상처의 수복을 위해서 손상된 부위로 세포가 이동하는 매우 흔하게 관찰되는 현상 중 하나 이다. Wound healing assay는 in vitro의 이차원 평면상에서 세포의 이동을 관찰할 수 있는 기본적인 연구 기법이다. In vitro상에서 물리적 손상을 재현하는 가장 손쉬운 접근법으로서, 세포의 confluent monolayer 표면에 날카로운 도구를 이용하여 기계적인 스크레치를 내는 방법이 사용되고 있다. 완충 스프링이 탑재된 금속 핀을 96-well plate를 기반으로 하는 wound maker에 장착하여 multi-well plate 바닥 표면의 고르지 못한 굴곡과 스크레칭 팁 사이에 직각을 이루는 접촉면에서의 미세한 조절이 가능하도록 하였다. 실험용 팁으로 confluent monolayer위에 스크래치를 내었을 때에는 다양한 지그재그 패턴이 그려진 반면에, 직접 제작한 wound maker에서는 동일한 형태의 선형 wounds가 fibroblast가 seeding된 96-well plate의 각 well의 중심부에 그려짐을 확인하였다. 상용화 되어있는 몇몇 multi-well plate가 본 실험에서 제작된 wound maker와 호환되는 것을 고려하여 보았을 때에, 실시간 wound healing을 관찰하는 high content screening (HCS)실험에 있어서의 활용적인 측면에서 기존의 전형적인 polypropylene 파이펫 팁을 이용한 스크래칭 방법보다 더욱 용이한 방법임을 알 수 있다.

황기약침이 전층피부손상 마우스의 치유에 미치는 영향 (Effect of Astragali Radix Pharmacocupuncture for Wound Healing of Full-Thickness Skin Wound in Mouse)

  • Objectives : In this study, we investigated whether Astragali Radix Pharmacocupuncture (ARP) has an effective on the full thickness defect wound healing process of mouse. Methods : A total of 50 mice (ICR mouse, 7 week-old male) were divided into control group and ARP group. A single full thickness skin defect was made on the dorsal side of the each mouse using an 8mm diameter biopsy punch. Control group were treated with 0.2㎖ saline and ARP group were treat with 0.2ml ARP at 8 points around the wound every three days total 4 times during the experimental period. The change in wound size, contraction rate, healing rate, and epithelization rate was measured by digital images taken on days 3, 6, 9, and 13, and evaluated using a digital image analysis program. Tissues were collected for histological analysis, RT-PCR, and Western blot on days 3, 6, 9, and 15. Results : The results are as follows. ARP group accelerated the rate of wound contraction, wound healing and epithelization compared to the control group. ARP group showed the decrease of inflammatory cells in early inflammatory phase compared to the control group. ARP group upregulated PECAM-1 mRNA and protein expression compared to the control group. ARP group inhibited the scar width and area compared to the control group. Conclusions: ARP showed positive effects on wound healing through the inhibition of inflammatory reaction and increase of PECAM-1 expression related to the wound healing process.

커큐민과 지방 기질세포가 흰 쥐의 창상치유에 미치는 영향 (The Effect of Curcumin and Human Adipose-derived Stromal Cells on Wound Healing of Lewis Rats)

  • Purpose: On this study, we investigated the effects of curcumin and adipose-derived stromal cells (ADSCs) in wound healing process, especially in the aspect of synergic effects when they were administrated simultaneously. Methods: Curcumin (40 mg/kg) and/or $1.0{\times}10^6$ ADSCs were applied to an $1.5{\times}1.5\;cm$-sized full thickness wound on the backs of male Lewis rats (n=5 in each group). In control group (n=5), saline was administrated instead of curcumin and ASCs. The wound size was followed by computer planimetry in 5, 7, and 14 days, and wounds were harvested for histological analysis in 7 and 14 days. Results: The dimensions of wounds of curcumin, ADSCs, and curcumin-ADSCs group significantly decreased in 5, 7, 14 days compared with those of control group (p<0.05), but there were no significant differences among three groups. The wound sizes were lowest in curcumin-ADSCs group compared with the other groups, but the differences were insignificant (p>0.05). There were infiltration of more epithelization and more precisely organization of extracellular matrix in curcumin, ADSCs, and curcumin-ADSCs group compared with those of control group. Conclustion: The results suggest that curcumin and ADSCs have beneficial effects in the acceleration of wound healing. Although the simultaneous application of curcmin and ADSCs also has beneficial effects on wound healing, there are no significant synergic effects.

Synthesis and optimization of immunomodulating hydrogel for biomedical application

  • Treatment towards wound healing, a complex and dynamic process, has been given a great deal of efforts in the last few decades. Focus has been imposed on developing wound dressings that meet the requirements for proper wound healing. In this study, hydrogel made from blends of poly (vinyl alcohol) and ${\beta}$-1,6-branched-${\beta}$-1,3-glucan (beta-glucan) were synthesized by modified solvent casting method for wound dressing application. Optimization of hydrogel composition and analysis of wound dressing parameters such as stability and fluid uptake capacity (in the presence of water, saline and different pH solutions) has been studied. The result indicated that the PVA/beta-glucan hydrogel hold its structural integrity even at alkaline pH (pH~9) and upholds fluids four times of its original weight. Thus, the developed hydrogel is expected to be a promising candidate as wound dressing.

수술칼, $CO_2$ 및 Nd:YAG 레이저에 의한 절개 창상의 치유에 관한 비교 연구 (A COMPARATIVE STUDY OF WOUND HEALING AFTER SKIN INCISION WITH SCALPEL, $CO_2$, AND Nd:YAG LASER IN RATS)

  • The purpose of this study was to compare the wound healing process after skin incision using scalpel, $CO_2$ laser and pulsed Nd:YAG laser in rats. After skin on the back was incised 3 cm long, rats were sacrificed at 1, 3, 7, 14, 21 and 28 days. Macroscopic, histologic and immunohistochemical examinations using the collagen type IV and the CD34 antibodies which are necessary to the forming process of new capillary were performed. Results obtained were as follows ; Macroscopically the initial wound healing of the laser group was about $1{\sim}2$ weeks slower than that of the scalpel group. There weren't however any remarkable differences in all groups in 4 weeks after incision. By histologic finding, acute inflammatory cells were more prominent during the initial wound healing in the scalpel group than in the other groups. Epithelialization started in the order of scalpel, $CO_2$ and Nd:YAG laser group after skin incision. By the Masson's trichrome stain, collagen synthesis in the Nd:YAG laser group was more slowly initiated than in the other groups. But it was completed at the $3{\sim}4$ weeks in all groups. Immunohistochemically, collagen type IV and CD34 expression were markedly increased at 2 weeks in the scalpel and $CO_2$ laser group. Meanwhile, in the Nd:YAG laser group, these reactions were observed later tan the other groups. Collagen type IV and CD34 expression were decreased in all groups after 4 weeks. These results suggest that $CO_2$ and Nd:YAG laser showed similar healing process compared with scalpel and a potential substitute for scalpel in skin incision.

스트렙토조토신 유도 당뇨 흰쥐에서 전기자극이 상처치유와 피부 비만세포에 미치는 영향 (Effects of Electrical Stimulation on Wound Healing and Skin Mast Cells in Streptozotocin-Induced Diabetic Rats)

  • The aim of this study was to investigate the effect of electrical stimulation on healing of impaired wound and alteration of mast cells in experimental diabetic rats. Thirty male Sprague-Dawley rats were divided into three groups : incision (control), diabetes+incision (diabetes) and diabetes + incision + electrical stimulation (D/ES). Diabetes was induced in rats by streptozotocin (STZ) injection (60 mg/kg, one time) and 20 mm length incision wounds were created on the back after shaving hair. The electrical stimulation rats were treated with a current intensity of 30~50 V at 120 pps and $140{\mu}s$ for 10 days from 3 days after STZ injection. The lesion and adjacent skin tissues were fixed with 10% buffered formalin, embedded with paraffin. For wound healing analysis, hematoxylin-eosin (HE) and picrosirius red staining were performed. Mast cells (MC) were stained with toluidine blue (pH 0.5) and quantified at ${\times}200$ using a light microscope. The density of keratinocyte proliferation and microvessels in skin tissues were analyzed using a computerized image analysis system on sections immunostained with proliferative cell nuclear antigen (PCNA) and ${\alpha}$-smooth muscle actin (${\alpha}$-SMA), respectively. The results showed that the wound healing rate, collagen density and neoepidermis thickness, density of PCNA-positive cells and density of ${\alpha}$-SMA-positive vessels were significantly higher in D/ES rats than in diabetic rats. The density of MCs and degranulated MCs in D/ES rats were also significantly higher than those in diabetic rats. These findings suggest that the electrical stimulation may promote the tissue repair process by accelerating collagen production, keratinocyte proliferation and angiogenesis in the diabetic rats, and MCs are required for wound healing of skin in rats.
