Cellulase Production in the Digestive Organs of Reticulitermes speratus, a Native Termite from Milyang, Korea

  • This study investigated on enzyme production in the digestive organs of the native termite (Reticulitermes speratus) in Milyang, Korea. Four types of major cellulases [EG (endo-1,4-${\beta}$-glucanase), BGL (${\beta}$-glucosidase), CBH (cellobiohydrolase) and BXL (${\beta}$-1,4-xylosidase)] were present in the digestive organs of the termite. The strong enzyme activity for BGL was found from the native termite, and also shown that the enzyme was distributed in the salivary gland, foregut, and hindgut. BXL, which breaks down hemicellulose near the amorphous region, was detected mainly from salivary gland, foregut, and midgut. However, CBH was distributed mainly in the hindgut. Meanwhile, EG which degrades cellulose, was found mainly in the hindgut and salivary glands. These facts indicate that celluases production patterns are differ from different sites compare to the same species found in Japan, suggesting that enzyme production in the digestive organs of termites is changed according to their habitats.

Pattern Classification of Acoustic Emission Signals During Wood Drying by Artificial Neural Network (인공신경망을 이용한 목재건조 중 발생하는 음향방출 신호 패턴분류)

  • This study was Performed to classify the acoustic emission(AE) signal due to surface cracking and moisture movement in the flat-sawn boards of oak(Quercus Variablilis) during drying using the principal component analysis(PCA) and artificial neural network(ANN). To reduce the multicollinearity among AE parameters such as peak amplitude, ring-down count event duration, ring-down count divided by event duration, energy, rise time, and peak amplitude divided by rise time and to extract the significant AE parameters, correlation analysis was performed. Over 96 of the variance of AE parameters could be accounted for by the first and second principal components. An ANN analysis was successfully used to classify the Af signals into two patterns. The ANN classifier based on PCA appeared to be a promising tool to classify the AE signals from wood drying.

Ultrastructural Observation of Bacterial Attacks on the Waterlogged Archaeological Woods (세균에 의한 수침고목재 피해양태의 초미시구조적 관찰)

  • Micromorphological changes in waterlogged archaeological woods excavated from Sweden and Germany were investigated. Especially bacterial attacks on those wood samples under near anaerobic conditions were examined by transmission electron microscopy(TEM). The major feature of micromorphological alterations in those wood samples was the preferential destruction of secondary wood cell wall. In contrast, the middle lamella was not extensively degraded. Three distinct degradation patterns by bacteria were observed : erosion, cavitation and tunnelling bacteria. Erosion and cavitation bacteria attacked primarily $S_2$ layer, whereas tunnelling bacteria made the tunnel-like degradation along the $S_1$ layer. Tunnelling bacteria, in some samples, were able to degrade tunnel in the lignin-rich areas, such as middle lamella, suggesting that these bacteria had the capacity to degrade the lignin. IR spectra indicate that hemicellulose and cellulose in the waterlogged woods were preferentially decomposed. Breakdown of the lignin, on the other hand, was much slower.

  • This research was performed to investigate the expression patterns of odorant-binding proteins (OBPs) migrating hydrophobic semiochemicals such as pheromone to the olfactory receptors in a termite (Reticulitermes speratus). Antennas and legs were cut from soldier and worker termites, respectively, and RT-PCR were conducted to investigate the existence of the OBPs reported up to now. Blast search suggested that the OBPs obtained were highly homologues of the OBPs reported. In worker termites, OBP-1 was expressed in both antennas and legs, OBP-2 and OBP-3 were observed only in antennas. And in soldier termites, OBP-1 was shown in both antennas and legs, OBP-2 were not observed, and OBP-3 was found in both antennas and legs. The differences of expression patterns of OBPs between worker and soldier termites may be explained by their specialized peculiarity.

A Literature Review on the Stagnation Pattern - Focusing on Liver-Qi Stagnation - (울증(鬱證)에 대한 문헌적 고찰 - 간기울결(肝氣鬱結)을 중심으로 -)

  • Objectives : The objective of this paper is to study the literature on stagnation pattern, to further determine reasoning for the emphasis on Liver-qi stagnation out of all the Five Zang stagnation patterns. Methods : Stagnation patterns in literature from the Xian-Qin/Liang-Han period, Sui-Tang-Song period, Jin-Yuan period and Ming-Qing period were studied and analyzed. Results & Conclusions : Based on chronological analysis on discussions of stagnation pattern, the Five stagnation in the Neijing were related to the excessiveness and deficiency of the Five circuits. After the discussions of the Four Masters of the Jin-Yuan period, WangAndao explained that stagnation could happen without relation to external contraction in his discussion of Benqiziyubing. Yutuan focused on the inter-relationship between the six stagnation patterns, emphasizing qi stagnation, while Sunyikui discussed stagnation pattern within the framework of the Five Zang stagnation. Zhaoxianke argued for the first time that one of the treatment methods of Wood stagnation could replace the five treatment methods, focusing on Wood stagnation which could be the clue to stagnation pattern. Of current Zangfu diagnostic terminology, the influential Liver qi stagnation remains to this day.

Development of the Automatic Machining Technology for Boat's Wooden Patterns (레저보트 목형가공 자동화에 관한 연구)

  • The cutting automation of boat's wooden pattern is strongly required to improve the productivity and quality of boats in leisure boat industry. This paper is concerned with the development of wooden pattern machining technology by the machining center. The leisure boat is designed with a 3 dimensional design s/w. The NC cutting data are generated in a CAM s/w and are verified using verification s/w. The cutting forces are monitored to analyse the cutting process. The surface characteristics of machined surface are investigated at various cutting conditions such as spindle speed, feed speed, the cutting direction of wood, and wood material.

Preparation and Screen Printing of Natural Dye Powders (천연염료 분말 제조 및 날염)

  • The purpose of this study is to obtain the dyed-fabrics of the elaborate patterns using the natural dye powders extracted from Galla Rhois, Sophorae Flos Immaturus, Clove, Sappan Wood, Madder, Log Wood and Japanese Green Alder. Specific informations on the separation and powder-making of each colorant were disclosed. Appropriate printing paste preparation and printing conditions including the viscosity and mordant concentration in the printing paste were investigated. The dyeability and colorfastness of the screen-printed silk fabrics with the colorant powders were discussed in terms of practical applicability. The deodorization and antimicrobial activities of the silk fabrics screen-printed with natural dyes were also assessed.

Tree-Ring Dating for a Medicine Cabinet in Seoul Museum of History (서울역사박물관 소장 가께수리 약장의 연륜연대 분석)

  • Tree-ring dating can be used to date scientifically prehistoric timbers, historical buildings or woodcrafts. It gives a calendar year to each tree ring and produces the felling dates of logs or wood panels. In this study, we applied tree-ring dating to a medicine cabinet in Seoul Museum of History. We obtained tree-ring data from radial or cross section by taking photographs with a digital camera. The cabinet were dated A.D. 1821 to the last ring. It was about 50 year older than the previous medicine cabinets. Tree-ring patterns indicated that the origin of woods for the cabinet would be near Sorak mountains.

Enzyme Activities and Cellulose Degradation of Domestic Softwoods in Shaking Culture of Fomitopsis palustris (국내산 침엽수 목분의 진탕배양에서 나타난 Fomitopsis palustris의 효소 활성 및 셀룰로오스 분해)

  • Activities of the extracellular enzyme from Fomitopsis palustris, a brown-rot fungi, and by which crystallinity changes of cellulose in the various softwoods, such as Larix leptolepsis, Finns rigida, Finns koraiensis and Finns densiflora by liquid culture, were investigated. Activity of Cellobiohydrolase (CBH) from F. palustris was detected in the every test softwoods culture, showing activities of the Endoglucanase (EG), $\beta$-glucosidase (BGL) and $\beta$-1,4-xylosidase (BXL). It was shown high enzyme activities in the sapwood culture than heartwood of the same wood species, However, the enzyme activities in most of test wood cultures increased with longer incubation time, indicating a possibility of intermix sapwood and heartwood for degradation process by enzyme. Also it was shown that protein patterns of the extracellular enzyme from F. palustris in wood particle substrate of the several domestic softwoods were similar with each other wood species, which suggested the possibility of mixing all softwoods in saccharification by enzyme from F. palustris. Crystallinity reduction value of cellulose by F. palustris was 4.2~20.4% in 4 weeks cultivation, 12.9~28.9% in 8 weeks.

A study on log diameter classes of Korean softwood log (국산 침엽수 원목의 경급구분 기준에 관한 연구)

  • Log grading rules are essential tools to ensure the quality of logs in distribution structure. The rules should reflect the long experience and accepted usage practice in the market. A gap between the rules and market should be improved based on analysis of log qualities that produced and market demand. In this study more than ten millions logs which were produced by 5 Regional Forest Services in 2010~2011 period, were analyzed in their qualities including diameters and lengths by species. A proposal was driven to improve the current log grading rules in terms of log diameter classes and length. The followings are the summary of this study. Most of domestic softwood logs are belong to small diameter class of 100~160 mm, which imply the diameter classes of current log grading rules are immoderate. Distributions of log diameter shows distinctive patterns by species, which indicate a necessity of differentiated diameter classes by species in an improved rules. Lengths of logs in productions do not corresponding to the demands and preferences in sawmills. Therefore it is highly recommended to include log length term in an improved log grading system. Based on these findings, 6 log grading systems for 3 species groups of softwood are newly proposed to improve current log grading rules. Limits of log diameter and log length are also proposed for each log grading system.
