The Expression of Egg Plant Flavonoid 3',5'-Hydroxylase Gene in Tobacco Plants (Nicotiana tabacum cv. Xanthi)

  • The anthocyanin gene encoding flavonoid 3',5'-hydroxylase(F3,5H) was normally expressed in Nicotiana tobacco (Xanthi) plants cocultivated with Agrobacterium tumefaciens LBA4404 carrying egg plant flavonoid 3',5'-hydroxylase cDNA. Northern blot analysis showed the normal expression of F3', 5'H gene from transgenic plants. Here we found the phenotypic differences between transgenic plants and wild-type plants. The petal shape of transgenic plants showed more round shape and around petal tube area was compared to that of wild-type tobacco plants. And the petal color of transgenic plants was much lighter than that of wild-type tobacco plants.

Responses of an herbaceous community to wild boar (Sus scrofa coreanus Heude) disturbance in a Quercus mongolica forest at Mt. Jeombong, Korea

  • This research identified a disturbance regime caused by wild boar in a mature Quercus mongolica forest and analyzed the impact of this disturbance on the structure and distribution of herbaceous plants in Mt. Jeombong, Korea. We demonstrate that disturbance by wild boar was most frequent from winter to early spring, but also occurred year round. Areas which were frequently disturbed by wild boar included the mountain ridge, the mild slope on the north face, and sites with high concentration of Erythronium japonicum. The disturbance cycle by the wild boar in this region was estimated at approximately 2.8 years. The wild boar's reduced the community's species diversity and herbaceous coverage, and increased its evenness. This disturbance reduced the coverage of spring ephemeral; Veratrum nigrum var. ussuriense, Symplocarpus niponnicus, Anemone koraiensis and Corydalis turtschaninovii were particularly sensitive. In addition, summer green herbaceous plants such as Astilbe chinensis, Ainsliaea acerifolia, Meehania urticifolia, and Pimpinella brachycarpa were sensitive to the wild boar's. It was found that wild boar ate E. japonicum most selectively of all plants in this investigation area. In conclusion, together with micro-topography, wind, formation of gaps of a forest and rearrangement of litter layer, wild boar's disturbance is an important factor influencing the dynamic changes of an herbaceous community in a mature temperate hardwood forest.

  • The wild Pyrus plants belong to the deciduous woody plants in Rosaceae family. Most are cultivated for fruit productions, but some for ornamental plants. There are 5 Pyrus species including Pyrus ussuriensis and 8 varieties cultivated in Korea. In Korea, these plants are populated near villages or on the hills, and they are easily propagated since their fruit are very juicy and quite big in size. Plant breeding to get improved cultivars is not difficult because the Pyrus plants are easy to cross breeding among its genus. Because not only this reason but there have been shown lots of variation in wild groups, the Pyrus plants inhabited in Korea are excellent deciduous woody plants and favorable fruit trees to develop ornamental plants. It is necessary to develop many kinds of fruit trees and ornamental trees containing genetically different character, so that it will be satisfied for many different people's taste and demands. Collecting lots of wild plant resource with diverse genes is necessary for plant breeding.

  • Lam Dong Province of Vietnam has an exceptional diversity of orchids and herbs. At least 920 herb species are widely mentioned in Vietnamese traditional medicinal literature and over 239 Lam Dong wild orchid species are used for attention under the Conservation Vietnamese wild orchids projects. In collaboration with Department of Plant Biotechnology, Dongguk University - South Korea working under supported ODA funds from Korea government (MIFAFF), we will consider how to collecting and preserving these plants in situ or conservating in vitro as a genetic resource.

Damping-off of Edible Aster Caused by Rhizoctonia solani AG-4

  • In August 2021, we surveyed diseases of wild vegetables grown in Taebaek, Gangwon Province, Korea. During the disease survey, we observed severe damping-off symptoms in young edible aster (Aster scaber) plants in a vinyl greenhouse investigated. The incidence of the disease in the plants ranged from 5% to 20%. Diseased plants of edible aster were collected from the vinyl greenhouse, and fungi were isolated from petiole lesions of the diseased plants. Rhizoctonia sp. was consistently isolated from the petiole lesions. We examined morphological characteristics and anastomosis groups of nine Rhizoctonia sp. isolates obtained from the petiole lesions. The examination results revealed that all the isolates corresponded to Rhizoctonia solani AG-4 based on the morphological characteristics and anastomosis test. Three isolates of R. solani AG-4 were tested for their pathogenicity on edible aster plants by artificial inoculation. Inoculation tests showed that the tested isolates caused damping-off symptoms on the inoculated plants. The induced symptoms were similar to those observed in the vinyl greenhouse investigated. Damping-off of edible aster caused by R. solani AG-4 is first reported in this study.

The Germination Characteristics of Seeds by Temperature Conditions in Artemisa annua L. (온도 조건에 따른 개똥쑥(Artemisa annua L.) 종자의 발아특성)

  • 본 실험은 다양한 생리활성이 보고되어 약용으로 널리 쓰이는 개똥쑥(Artemisia annua L.) 종자의 온도에 따른 발아 특성을 조사하기 위해 진행하였다. 미세온도구배 발아기를 이용하여 낮과 밤의 온도를 각각 5 ~ 35℃ 범위에서 낮과 밤의 시간을 12시간으로 고정하고, 낮 온도가 밤 온도보다 크거나 같은 조건을 설정하여 총 27개의 온도조건으로 개똥쑥 종자의 최종 발아율 및 발아율과의 관계를 분석하였다. 실험결과, 개똥쑥 종자는 실험에 사용한 모든 온도조건에서 발아가 가능한 것으로 나타났으며, 25/10℃(낮/밤) 조건에서 90%로 가장 높게 조사되었다. 또한, 발아율 조사결과를 통해 일평균온도뿐만 아니라 일교차온도도 발아율에 영향을 미치는 것으로 판단되어 일평균온도와 일교차온도로 나누어 발아율과의 관계를 분석하였다. 온도조건과 발아율과의 연속적인 발아특성을 분석하기 위해 다중회귀분석과 비선형 회귀분석을 이용하여 온도 조건과 상대적 발아율의 관계를 수식으로 표현하였다. 일평균온도를 기준으로 발아율과의 관계를 분석한 결과, 5 ~ 35℃의 모든 일평균 온도범위에서 유의성이 나타났으며 5, 7.5, 32.5, 35℃는 상대적인 음의 영향력을, 나머지 조건에서는 상대적인 양의 영향력을 가진 것으로 분석되었다. 일교차온도를 기준으로 발아율과의 관계를 분석한 결과, 0 ~ 25℃의 모든 일교차 온도범위에서 양의 영향력을 가진 것으로 분석되었다. 일평균온도는 19.3℃에 가까울수록, 일교차온도는 14.9℃에 가까울수록 발아에 대한 영향력이 큰 것으로 조사되었다. 각 수식을 통해 도출된 수치화된 온도에 따른 개똥쑥 종자의 일일누적 온도 영향력을 temperature score(TS)로 설정하였다. 본 연구에서 도출된 수식을 통해 누적 TS를 계산한 결과, 14.9 TS가 누적되었을 때 발아율이 85% 이상으로 나타날 것으로 예측되었다.

  • To evaluate nutritional values and to detect pharmacologically active and antibacterial components of wild edible plants in Korea, 20 species were examined. 1) The essential free amino acid compositions of these plants were determined by TLC. The plants contained threonine, leucine, valine and methionine. 2) In antibacterial tests of 17 species the plants Hemerocallis sp. and Plantago asiatica showed an antibacterial activity against Staphylococcus, Bacillus, Eschericia, and Salmonella species. 4) Rutin. quercitrin, $k\ddot{a}mpferol$, chlorogenic acid and caffeic acid were also identified.

Effect of B-complex vitamins on the antifatigue activity and bioavailability of ginsenoside Re after oral administration

  • Background: Both ginsenoside Re and B-complex vitamins are widely used as nutritional supplements. They are often taken together so as to fully utilize their antifatigue and refreshing effects, respectively. Whether actually a drug-nutrient interaction exists between ginsenoside Re and B-complex vitamins is still unknown. The objective of this study was to simultaneously investigate the effect of B-complex vitamins on the antifatigue activity and bioavailability of ginsenoside Re after their oral administration. The study results will provide valuable theoretical guidance for the combined utilization of ginseng and B-complex vitamins. Methods: Ginsenoside Re with or without B-complex vitamins was orally administered to mice to evaluate its antifatigue effects and to rats to evaluate its bioavailability. The antifatigue activity was evaluated by the weight-loaded swimming test and biochemical parameters, including hepatic glycogen, plasma urea nitrogen, and blood lactic acid. The concentration of ginsenoside Re in plasma was determined by liquid chromatography-tandem mass spectrometry. Results: No antifatigue effect of ginsenoside Re was noted when ginsenoside Re in combination with B-complex vitamins was orally administered to mice. B-complex vitamins caused to a reduction in the bioavailability of ginsenoside Re with the area under the concentration-time curve from zero to infinity markedly decreasing from $11,830.85{\pm}2,366.47h{\cdot}ng/mL$ to $890.55{\pm}372.94h{\cdot}ng/mL$. Conclusion: The results suggested that there were pharmacokinetic and pharmacodynamic drug-nutrient interactions between ginsenoside Re and B-complex vitamins. B-complex vitamins can significantly weaken the antifatigue effect and decrease the bioavailability of ginsenoside Re when simultaneously administered orally.

Damping-off of Indian Lettuce Caused by Rhizoctonia solani AG-4

  • Damping-off symptoms were frequently observed on young plants of Indian lettuce (Lactuca indica) grown in a farmer's vinyl greenhouse located in Goseong, Gangwon province of Korea during a disease survey in June 2019. The incidence of diseased plants in the vinyl greenhouse investigated was 30-50%. Four isolates of Rhizoctonia sp. were obtained from the diseased plants and identified as Rhizoctonia solani AG-4 based on the morphological characteristics and anastomosis test. Three isolates of R. solani AG-4 were tested for pathogenicity on Indian lettuce by artificial inoculation. All the tested isolates induced damping-off symptoms on the inoculated plants. The symptoms were similar to those observed in plants from the farmer's vinyl greenhouse investigated. This is the first report of R. solani AG-4 causing damping-off in Indian lettuce.
