Diversity and Inheritance of AFLP Markers in Wild and Cultivated Soybeans (AFLP marker를 이용한 콩의 유전적 다양성과 유전분리 분석)

  • Genetic variation is the basis of crop improvement. Limited genetic diversity in a crop species may restrict the amount of genetic improvement that can be achieved through plant breeding. Soybean is one of the world's most important crops. A potential source of genetic variability for the cultivated soybean is the wild species G. soja Sieb. & Zucc. Amplified fragment length polymorphism (AFLP) analysis is a PCR-based technique, which can detect a 10-fold greater nubmer of loci than other DNA marker analysis. Twenty cultivated soybeans and two-hundred wild soybeans were used to determine genetic vatiations by AFLPs and evaluate the usefulness of AFLPs as DNA markers. Six-hundred and ten fragments were detected with an average of 56 AFLP fragments produced per primer in a total of 11 AFLP primer pairs. The number of polymorphic loci detected per primer ranged from 7 to 20 and the polymorphism was greater in wild than in cultivated soybean. F$_2$ segregation analysis of four AFLP fragments in combination of Hwaeomputkong ${\times}$ PI 417479 indicated that they segregate as stable Mendelian loci with 3 : 1. This results strongly suggest that the AFLP analysis is a good technique for the detection of genetic polymorphism in a wide plant species.

AtHAP3b Plays a Crucial Role in the Regulation of Flowering Time in Arabidopsis during Osmotic Stress

  • The HAP complex has been found in many eukaryotic organisms. HAP recognizes the CCAAT box present in the promoters of 30% of all eukaryotic genes. The HAP complex consists of three subunits - HAP2, HAP3 and HAP5. In this paper, we report the biological function of the AtHAP3b gene that encodes one of the HAP3 subunits in Arabidopsis. Compared with wild-type plants, hap3b-1 and hap3b-2 mutants exhibited a delayed flowering time under long-day photoperiod conditions. Moreover, the transcription levels of FT were substantially lower in the mutants than in the wild-type plants. These results imply that AtHAP3b may function in the control of flowering time by regulating the expression of FT in Arabidopsis. In a subsequent study, AtHAP3b was found to be induced by osmotic stress. Under osmotic stress conditions, the hap3b- 1 and hap3b-2 mutants flowered considerably later than the wild-type plants. These results suggest that the AtHAP3b gene plays more important roles in the control of flowering under osmotic stress in Arabidopsis.

  • The experiments tvere carried out to different temperature which affected growth, yield, proximate and antitumor activity in the Codonopsis lanceolata. Growth of aerial part and subterranean partwere best at 2$0^{\circ}C$ and lower 3$0^{\circ}C$. Components of fat, protein and fiber were best at 3$0^{\circ}C$. Wild C.fonceoforo had higher contents of fat, protein, fiber and ash as compared with cultur'ed C.lanceolataArginin was predominant amino acid in both wild and cultivated C.laceolata No significant differ-ence in the mineral contents was found between the wild root and the cultivated at 30'c inbiotronroom. No minerals difference in the contents was found between the cultivated temperature.The content of elements of inorganic metal differs according to the part. C. lanceolata were subjectedto preliminary antitumer screening test with Sarcoma 180 ascites and screening on V-79 cell. Thisexperiments were conducted in accordance with the total packed cell volume method and cytotoxicity method.

Absence of AVP1 transcripts in wild type watermelon scions grafted onto transgenic bottle gourd rootstocks

  • In this study we confirmed the stable integration of Arabidopsis AVP1 in the genomes of bottle gourd $T_3$ homozygous lines and its transcription, and additionally evaluated possibility of translocation of the AVP1 mRNA from transgenic bottle gourd rootstocks to wild type watermelon scions. Each AVP1 gene in two bottle gourd T3 lines is abundantly expressed under a field condition. Given the grafting between wild type watermelon scions and AVP1-expressing bottle gourd rootstocks, no translocation of the AVP1 mRNA was detected in leaves, both sexual flowers, and fruits of the scions.

  • Proximate compositions of the roots of cultured and wild Codonopsis lanceolata were examined as tile basic reasearch for the study of their source of processed foods.No significant difference in the contents was found between the wild root and thecultivated at 3$0^{\circ}C$ in biotronroom. In view of the results to have measured mineralswhich is included in 14 sorts of Codonopsis lanceolata and surveyed their distribution.12 kinds of minerals including T1, Co, Ge, Sm, Mo, Sc, Be, Cd, As, Ga, Bi, ph are ne-vel or little included in almost source. Other twenty-one sorts of minerals (Ni, Se,Ba, Sb, Si, Ti, B, Li, Ifs, Ca, Sr, Mn, Fe, Cu, Zn, p, Al, Na, V, Cr, K) are more or less in-cluded in all source and Ca, Mg, p, K, and Fe are metals that are included in large qu-entities in comparison with others. No minerals difference in the contents was foundbetween the cultivated temperature. The content of elements of inorganic metal differsaccording to the part of C. lanceolata.

Damping-off of Coastal Hogfennel Caused by Rhizoctonia solani AG-4

  • Damping-off symptoms were frequently observed on young plants of coastal hogfennel (Peucedanum japonicum) grown in a farmer's vinyl greenhouse located in Goseong, Gangwon Province, Korea during a disease survey in June 2019. Incidence of the diseased plants was 50-70% in the vinyl greenhouse investigated during the disease survey. Eight isolates of Rhizoctonia sp. were obtained from the diseased plants. All the isolates were identified as Rhizoctonia solani AG-4 based on the morphological characteristics and anastomosis test. Three isolates of R. solani AG-4 were tested for pathogenicity on coastal hogfennel by artificial inoculation. All the tested isolates induced damping-off symptoms on the inoculated plants. The symptoms were similar to those observed in the farmer's vinyl greenhouse investigated. This is the first report of R. solani AG-4 causing damping-off in coastal hogfennel.

Selection of Nitrate-nonutilizing Mutants of Hypoxylon atropunctatum, A Fungal Pathogen on Oak Species

  • Latent infections of healthy-appearing oaks of Hypoxylon atropunctatum complicates field studies by interfering with inoculation experiments to follow pathogenesis, fungal development and reproduction of this canker rot fungus. Mutants with unique and easily scorable phenotypes would be useful for inoculation studies. There is a broad range in the capacity of wild-type isolates to utilize nitrate as a sole nitrogen sources. Several types of nitrate-nonutilization mutants (nit1, Nit3, NitM) were selected from nitrate-utilizing wild-type isolates. Also, a few mutants of Hypoxylon atropunctatum were selected that could only grow poorly on basal medium supplemented with various nitrogen sources and even on yeast extract agar. These unknown mutants need to be characterized further. Nit mutants of Hypoxylon atropunctatum were readily selected, grew well and were recovered after inoculation into oak stems. These results suggest that nit mutants could be useful for inoculation studies in trees that contain latent infections.
