Multiple shoot induction and callus regeneration in Sarcostemma brevistigma Wight & Arnott, a rare medicinal plant

  • An efficient micropropagation protocol based on multiple shoot induction and callus regeneration has been standardized in Sarcostemma brevistigma, a rare medicinal plant. The nodal cuttings were cultured on MS medium supplemented with BA ($0.5-8{\mu}M$) or Kn ($0.5-8{\mu}M$) alone or in combination with NAA ($0.5-1.5{\mu}M$). Maximum multiple shoot induction was observed on MS medium supplemented with $4{\mu}M$ BA. On this medium, 100% cultures responded with an average number of 11.3 shoots per explant. However, the average shoot length was limited to only 0.9 cm on this medium. The addition of $1{\mu}M$ NAA along with $4{\mu}M$ BA gave rise to an average number of 10.9 shoots with an average shoot length of 1.8 cm. Luxuriantly growing callus was obtained on MS medium supplemented with BA ($5{\mu}M$) and 2,4-D ($2{\mu}M$). The callus was subcultured on MS medium supplemented with BA ($2-15{\mu}M$) or Kn ($2-15{\mu}M$) alone or in combination with NAA ($0.5-2{\mu}M$) for shoot organogenesis. Optimum callus regeneration was obtained on MS medium supplemented with $10{\mu}M$ BA and $1{\mu}M$ NAA. On this medium, 100% cultures responded with an average number of 13.4 shoots per culture. The shoots obtained via multiple shoot induction and organogenesis were rooted on half-strength MS medium supplemented with NAA ($1-7{\mu}M$) or IBA ($1-7{\mu}M$). IBA was better than NAA in terms of both the percentage of cultures that responded and the average number of roots per explant. The rooted shoots were successfully transplanted to soil with 86% success. This standardized protocol will help to conserve this rare medicinal plant.

Effect of Feeding Basal Diet Supplemented with Mugwort Powder on the Serum Components in Rat (쑥 첨가급식이 흰쥐의 혈청 성분에 미치는 영향)

  • This study was designed to observe the body growth and components of edible mugwort(Artemisia sp.) and medicinal mugwort(Kanghwa medicinal mugwort) . Twenty-four young rats of Sprague Dawley strain, body weight of about 89g were used in this study. They were fed on the basal diet(control diet) supplemented with 5% edible mugwort powder ( EM diet) and 5% medicinal mugwort powder( MM diet) for 4 weeks respectively. In proximate composition of nutrients of mugwort in dry basis(100g). crude protein (16.4g) and crude ash(11.8g) contents of EM were higher to about 2% than that of MM, but crude lipid content(4.3g) of EM was lower to about 2% than that of MM. However, the contents in calcium(6.9g) of MM was higher to 5.3 times than that of EM. but in Mn(17mg), Zn(0.5mg), Fe(131mg), Mg(337mg) of EM were higher to 2.8∼2.3 times and vitamin A(39,776 IU) of EM was higher to 2.9 times than that of MM respectively. Body wight gain rate and diet efficiency ratio of EM and MM diet group were similar to that of the control group. The contents of total protein, albumin, urea nitrogen. creatinine, uric acid, total cholesterol, HDL-C, LDL-C, glucose, amylase, transaminase (GOT, GPT) in serum exhibited no remarkable difference among of the EM and MM diet group but the level of LDH activity of MM diet group were significantly lower than that of the control group and EM diet group.

Effect of Ethanol on Aerobic Biodegradation of Benzene, Toluene, and Ethylbenzene by Rhodococcus sp. EH831 (Rhodococcus sp. EH831에 의한 벤젠, 톨루엔 및 에틸벤젠의 호기성 생분해에 미치는 에탄올의 영향)

  • The usage of ethanol (EtOH)-blended gasoline (gasohol), has been increasing in recent years. EtOH has influence on the distribution and biodegradation of aromatic compounds such as BTEX (benzene (B), toluene (T), ethylbenzene (B), and xylene (X)) that are gasoline compositions. In this study, the effect of EtOH on the aerobic biodegradation of B, T and E was investigated using a BTEX and EtOH-degrading bacterium, Rhodococcus sp. EH831. The degradation rates of B in the conditions of 1:1, 1:4, and 1:0.25 mixtures with EtOH (B:EtOH, mol:mol) were ranged from $3.82{\pm}0.20$ to $5.00{\pm}0.37{\mu}mol{\cdot}g-dry$ cell wight $(DCW)^{-1}{\cdot}h^{-1}$. The degradation rate of T was the fastest in the 1:0.25 mixture ($6.63{\pm}0.06{\mu}mol{\cdot}g-DCW^{-1}{\cdot}h^{-1}$), and it was the lowest in the 1:4 mixture ($4.41{\pm}0.04{\mu}mol{\cdot}DCW^{-1}{\cdot}h^{-1}$). The degradation rates of E were increased with increasing the addition amount of EtOH: The degradation rate of E was the highest in the 1:4 mixture ($1.60{\pm}0.03{\mu}mol{\cdot}g-DCW^{-1}{\cdot}h^{-1}$), and the rates were $1.42{\pm}0.06$, $1.30{\pm}0.01$, and $1.01{\pm}0.30{\mu}mol{\cdot}g-DCW^{-1}{\cdot}h^{-1}$ in the 1:1, 1:0.25, 1.0 mixtures, respectively. In conclusion, the biodegradation of B, T, E by Rhodococcus sp. EH831 was not significantly inhibited by the co-existence of EtOH.

Uptake Capacity of Heavy Metals by Water Plants (수생식물의 중금속 흡수능에 관한 연구)

  • In this study, we investgated the uptake capacity of several water plants for heavy metals (lead and cadmium) in soil of rivers where are adjacent to a industrial complex in Chun-An city and in A-San city. We also examined the deposition pattern of heavy metal in plants. The results are as follows: 1. The soil of river in Chun-An city was polluted more serious than that of A-San city. In Chun-An city, mean values of lead and cadmium contents in soil were 26.224 $\pm$ 28.037 $\mu$g/g, and 0.854 $\pm$ 1. 127 $\mu$g/g, respectively. 2. Water plants examined in this study were Slum suave KITAGAWA, Persicaria thunbergii H. GROSS, Phragmiles japonica STEUD, Echinochloa crus-galli var. frumentacea WIGHT and Persicaria hydropiper SPACH. Both metal contents of several water plants distributed in Chun-An city were higher than those in A-San city. In these plants, Slum suave showed the highest uptake capacity for lead and cadmium. The mean values of lead and cadmium contents in Slum suave were 40.957 $\pm$ 29.577 $\mu$g/g and 1. 930 $\pm$ 1. 076 $\mu$g/g, respectively. Persicaria thunbergii also showed a relatively high uptake capacity for both metal. 3. Correlation between metal contents in soil and water plants was high. In both cases of Sium suave and Persicaria thunbergii correlation coefficients were 0.605 and 0.549, respectively. 4. We analyzed lead and cadmium contents in root, stem and leaf of several water plants. Both metals were mostly deposited in root. Much of both metals were also deposited in leaf. From the results, we suggest that Slum suave KITAGAWA and Persicaria thunbegii H. GROSS can be used to reduce heavy metals from industrial waste water.

  • Wild plants which have potential availability as pasture plants were collected from around Kyungpook province. Plants collected were cultivated in the university farm and their characteristics were investigated and recorded. Seeds of pasture plant resources collected were selected, weighed for 1,000 grains, packaged and stored at $4^{\circ}C$. Species of collected were as follows. Gramineous plants are Cymbopogon totilis var, goeringii Hand Mazz, Pennisetum akioecyriudes L., Dactylis glomerata L., Agropyron tsukushience var transiens Ohwi, Arundinella hirta Tanaka, Themeda triandra var., japonica Makino, Miscanthus sinensis Anderss, Setaria virides L., Digitaria sanguinalis L., Imperata cylindrica var. Koenigii Retz, Cynodon dactylon Pers, Echinochloa crus-galli var. frumentacea Wight, and Avena fatua L., and leguminous plants are Vicia angustrifolia var. segetilis Koch, Kummerowia stipulacea Makino, Kummerowia striata Schindl, Trifolium repens L., Melilotus officinalish L., and Astragalus sinicus L..

Comparative floristic diversity of Southwest Primorye and neighboring areas of the Russian Far East

  • Southwest Primorye is located in the southern most part of the Russian Far East. The flora of this area is one of the most thoroughly studied and contains 1,530 species of vascular plants, belonging to 622 genera and 154 families, representing 55.6% of the flora of the Primorsky Territory. The flora native to Southwest Primorye encompasses 1,356 species from 547 genera and 148 families. Adventitious plants are represented by 174 species from 127 genera and 37 families. Among other areas of the Russian Far East, only Southwest Primorye has flora in common with the Korean Peninsula, which contains plant species in the subtropical and tropical latitudes: Mitrasacme indica Wight, Halosciastrum melanotilingia [Boissieu] M. Pimen. et V. Tichomirov, Streptolirion volubile Edgew., Lipocarpha microcephala [R. Br.] Kunth, Pueraria lobata [Willd.] Ohwi, Belamcanda chinensis [L.] DC., Zoysia japonica Steud., Deinostema violacea [Maxim.] Yamazaki and Parthenocissus tricuspidata [Siebold et Zucc.] Planch. Most of the flora is represented by the species restricted to Southwest Primorye (seven endemic species) or the Korean Peninsula (seven hemiendemic species). A total of 50.2% of the 10 largest families are nemoral (preboreal) floras, located in the transitional zone of the boreal and nemoral floras. In terms of the structure of the ranked taxonomic spectrum of the largest families, the flora of Southwest Primorye is more similar to that of Northeast China with influence of the Korean flora.

Effects of Stocking Density and Feeding Frequency on the Growth of the Pacific Cod, Gadus macrocephalus (사육밀도와 먹이 공급 횟수가 대구(Gadus macrocephalus)의 성장에 미치는 영향)

  • This study investigated the effects of stocking density and feeding frequency (three and five times per week) on the growth of Pacific cod, Gadus macrocephalus. In the first experiment, experimental groups of fish (mean weight $167.8{\pm}41.4\;g$) were reared at three stocking densities (4, 8 and 12 kg/$m^3$) for 16 weeks. The total weight gain was 17.8, 17.1 and 35.4% at 4, 8 and 12 kg/$m^3$, respectively. The specific growth rates, survival rates and feeding efficiency increased with the density, but there were no significant differences (P>0.05). The daily feed intake decreased as the stocking density increased, whereas the condition factors increased with the stocking density (P<0.05). In the second experiment, groups of fish ($194.1{\pm}68.3\;g$) were fed either three or five times a week. The total wight gain was 14.4 and 18.5% for the fish fed three and five times per week, respectively. The specific growth rates of Pacific cod fed to satiation increased with the feeding frequency, although the difference was not significant (P>0.05). The survival rates, daily feed intake, feed efficiency, and condition factors of the Pacific cod were not significantly different (P>0.05). Based on these results, the optimum stocking density for juvenile cod is above 12 kg/$m^3$, and the best feeding frequency is three times a week, under the conditions used in this experiment.

Effects of estrogen on the ovariectomized rat (에스트로겐이 난소가 제거된 흰쥐에 미치는 영향)

  • The aim of this study was to determine the effects of body weight, organ weight, hematological values and biochemical parameters by estrogen on the ovariectomized (OVX) rats. The animals were divided into 4 groups. Intact group (n=10) received no treatment and operation. Sham group (n=10) received only sham operation and no treatment. OVX group (n=10) received operation and no treatment. OVX+E2(n=10) received operation and estrogen. The body weight of each group increased, but that of OVX+E2 group decreased. There were very significant differences (P<0.001) of body weights between OVX+E2 group and the other groups. Also, the wight of important organs such as heart, liver, spleen and kidney was measured. The heart weight was significantly lower(P<0.05) in the OVX+E2 group than the Intact group. The liver weight of the OVX+E2 group was significantly different in comparison with those of the other groups. On the other hand, there were no significantly differences in the organ weight of spleen and kidney between the OVX+E2 group and the other remaining groups. The hematological values of WBC, RBC, MeV, MCH and MCHC were not significantly different among the groups. The serum total protein and albumin decreased significantly in the OVX+E2 group as compared to those in the intact group. But, there were no significant differences in Ca, IP and Mg among the groups. We conclude that estrogen was significantly decreased the body weight in the ovariectomized rats. Our findings suggest that estrogen may influence the process of lipid packaging and absorption in the ovariectomized rats.

A Study on the Development of Traditional Korean Paper Changphanji by the Bast Fiber of Paper Mulberry (닥나무의 인피섬유(靭皮纖維)를 이용(利用)한 전통(傳統) 한지장판지(韓紙壯版紙) 개발(開發)에 관(關)한 연구(硏究))

  • This study was carried out to develop the traditional korean paper changphanji from the bast fiber of paper mulberry The results obtained were as follows: 1. The variations in the basis weight were remarkable, but there were no significant adverse effects on the tensile strength and thickness. 2. The stamping have contributed to the improvement of smoothness and the control of thickness. The automatic pressing system recommended to obtain good control of thickness. 3. The average density of the traditional korean paper changphanji was 1.06g/$cm^3$. 4. The changphanji developed in this study gave a clude, but excellent toughness and warm feeling. Some fiber lumps, dark spots and bamboo, screen marks were also obserded. 5. The tensile strength of four-fold sheet changphanji is highest and those of two-fold sheet changphanji is lowest. the lengthwise tensile strength was higher than crosswise one due to the fiber. 6. The elongation of three-fold sheet changphanji was highest 7. The water absorption of all products ranged from 11.3g/$m^2$ to 17.5g/$m^2$. 8. The wight loss by abrasion was 34.0%(four-fold sheet) 31.6%(three fold sheet), 31.0% (two-fold sheet)respectively. 9. Based on the results obtained. tensile strength and elongation, the four-fold sheet was excellent. the use of the bast fiber of paper mulberry as a raw material is attractive point which can not obtained from the commercial changphanji made from other raw materials.

Root vs. Shoot Genotype Effects on Growth Characters and Seed to Pod-Shell Ratio in Grafted Soybean Plants (콩 유전자형간 상호접목이 지상부 생육과 협실비율에 미치는 영향)

  • The partitioning of dry matter into seed and pod-shell is important because yield increase can be achieved by improving the distribution rate of assimilation products to seeds. The present study was undertaken to characterize whether the partitioning of dry matter into seed was shoot- or root-controlled through graftiong techniques. Self- and reciprocal-grafts were made among four soybean genotypes, which were 'Baekunkong', 'Suwon 168', and two local soybeans with black seed coat (hereafter referred to as the 'black soybean'), 'Kangleungjarae' and 'Keumleungjarae'. Self-grafted black soybeans showed lower ratio of seed to pod-shell dry wight than self-grafted Baekunkong and Suwon 168. Varying the shoot genotypes in grafts resulted in significant differences in growth characters as well as pod and seed dry weight per plant at physiological maturity. There were significant effects of shoot genotypes on the ratio of seed to pod-shell dry weight, indication that the partitioning of dry matter into seed and pod-shell should be shoot- rather than root-controlled. The grafts with large pod-shell dry weight tended to partition relatively smaller dry matter into seed than pod-shell.
