  • 홍삼(紅蔘) 및 백삼분말제품(白蔘粉末製品)에 대(對)한 방사선(放射線)($^{60}Co-{\gamma}$ 선(線)) 조사(照射) 및 조사선량(照射線量)과 장기저장(長期貯藏)이 이화학적(理化學的) 특성(特性)과 외관품질(外觀品質) 변이(變異)에 미치는 영향을 $25^{\circ}C$에서 4개월간(個月間) 저장하면서 조사하였다. 양시료(兩試料) 모두에서 조사선량(照射線量)과 장기저장(長期貯藏)이 따른 변이(變異)가 거의 없었다. 이화학적(理化學的) 특성(特性)에서 수분(水分), pH, 아미노태질소(態窒素), buthanol extract, 환원당(還元糖)등과 사포닌분획별 패턴에도 대질(大窒)없었으나 백삼분말(白蔘粉末)의 경우 특히 50% ethanol extract의 수율(收率)이 약간 증가(增加)되는 경향(傾向)을 보였다. 외관품질(外觀品質)의 경우 50% ethanol extract의 색농도(色濃度)에서는 대차(大差)없었으나 분말자체(粉末自體)의 색도(色度)에서는 시료간(試料間)에 약간 차이(差異)가 있었다. 홍삼분말(紅蔘粉末)의 경우 L치(値)와 a치(値)는 감소(減少)되었고 b치(値)는 증가(增加)되어 다갈색화(茶褐色化) 되는 경향(傾向)이었고 백삼분말(白蓼粉末)에서는 L치(値)가 감소(減少)되고 a치(値) b치(値)는 선량(線量)에 따라 증가(增加)되나 저장기간(貯藏期間)에서는 감소(減少)와 증가(增加)를 보여 농황색화(濃黃色化)되는 경향(傾向)이 있으나 식별가능정도(識別可能程度)의 색도변이(色度變異)범위는 아니었다. 전체적(全體的)으로 품질(品質)의 안정성(安定性)을 유지(維持)하고 있음을 보여주었다.

Wild Ginseng Prevents the Onset of High-Fat Diet Induced Hyperglycemia and Obesity in ICR Mice

  • Ginseng is a shade-loving perennial herb that is cultivated mainly in Korea, Japan, and China. The ginseng root has been used as a tonic remedy, and its antidiabetic activity has been demonstrated as early as 1920s. Although wild ginseng was anecdotally thought to be superior to cultivated ginseng as far as pharmacological properties were concerned, there have been no prior reports on the antidiabetic effect of wild ginseng. In this study, we investigated the preventative anti-diabetic and anti-obese effects of wild ginseng ethanol extract (WGEE). In the preventive experiment, WGEE co-administered with a high fat diet significantly inhibited body weight gain, fasting blood glucose, triglyceride, and free fatty acid levels in a dose dependent manner. WGEE-treated mice at doses of 250 and 500 mg/kg improved the insulin resistance index by 55% and 61% compared to the high fat diet (HFD) control, respectively. Diameters of white and brown adipocytes were also decreased by 62% and 46% in the WG500-treated group compared to those in HFD fed control mice. Taken together, WGEE has potential as a preventive agent for type 2 diabetes mellitus (and possibly obesity) and deserves clinical trial in the near future.

  • This study was conducted to elucidate the effects of light intensity using polyethylene (p.E) net shading on the specific gravity, red ginseng quality, sugar and saponin contents of ginseng root. The specific gravity significantly increased in the ginseng roots grown under the P.E net shading as compared with that of common straw shading. The red ginseng quality under the P.E net shading was improved in order of 10, 5, 15, 20, 30% of light intensity and the inside cavity and inside white part decreased remarkably as compared with those of common straw shading. The ginseng roots grown under the P.E net shading at 10% and 15% light intensity showed a significant increase in the total sugar content but a significant decrease in the reducing sugar content at 15% light intensity as compared with those of common straw shading. The amount of total saponin of ginseng roots was increased under the P.E net shading at high light intensity as compared with that of common straw shading and the ginseng roots grown under the P.E net shading at 10% light intensity showed an increase in the diol group saponin but the ratio of PT/PD was decreased. Extract contents of ginseng root under the P.E net shading was higher than those of common straw shading and the roots grown under the P.E net shading at 15% and 20% light intensity resulted in a remarkable increase in extract contents.

온도 및 당의 첨가가 인삼차의 향미에 미치는 영향 (Effects of Temperature and Sugar Addition on The Flavor of Ginseng Tea)

  • 인삼차의 관능적 향미의 성질을 밝히고져 백미삼(白尾蓼)과 홍미삼(紅尾蓼)에서 얻어진 농축 추출액에 당(糖)을 혼합시켜 제조한 백삼차와 홍삼차를 마실 때의 온도와 설탕의 첨가가 향미에 미치는 영향을 조사하였다. 인삼차의 향미를 표현하는 12가지의 묘사를 선택한 뒤 QDA방법에 의하여 백삼차와 홍삼차를 비교한 결과 백삼차는 흙냄새, 마른나무냄새, 아린맛. 쓴맛등이 강하여 전반적인 향미가 홍삼차보다 강함을 보여 주었다. 온도가 증가하면서 냄새는 전반적으로 현저한 증가를 보인 반면 맛은 쓴맛과 아린맛의 증가외에 뚜렷한 변화의 경향을 보여주지 않았다. 한편, 마실 때의 설탕의 첨가는 냄새에 큰 영향이 없었으나 쓴맛, 아린맛. 떫은맛등 전반적인 맛의 감소를 나타내었다.

Ameliorative effect of black ginseng extract against oxidative stress-induced cellular damages in mouse hepatocytes

  • Background: Oxidative stress induces the production of reactive oxygen species (ROS), which play important causative roles in various pathological conditions. Black ginseng (BG), a type of steam-processed ginseng, has drawn significant attention due to its biological activity, and is more potent than white ginseng (WG) or red ginseng (RG). Methods: We evaluated the protective effects of BG extract (BGE) against oxidative stress-induced cellular damage, in comparison with WG extract (WGE) and RG extract (RGE) in a cell culture model. Ethanolic extracts of WG, RG, and BG were used to evaluate ginsenoside profiles, total polyphenols, flavonoid contents, and antioxidant activity. Using AML-12 cells treated with $H_2O_2$, the protective effects of WGE, RGE, and BGE on cellular redox status, DNA, protein, lipid damage, and apoptosis levels were investigated. Results: BGE exhibited significantly enhanced antioxidant potential, as well as total flavonoid and polyphenol contents. ATP levels were significantly higher in BGE-treated cells than in control; ROS generation and glutathione disulfide levels were lower but glutathione (GSH) and NADPH levels were higher in BGE-treated cells than in other groups. Pretreatment with BGE inhibited apoptosis and therefore protected cells from oxidative stress-induced cellular damage, probably through ROS scavenging. Conclusion: Collectively, our results demonstrate that BGE protects AML-12 cells from oxidative stress-induced cellular damages more effectively than WGE or RGE, through ROS scavenging, maintenance of redox status, and activation of the antioxidant defense system.

동충하초 추출물과 백삼추출물 투여가 방사선을 조사한 생쥐 간에서의 glutathione함량에 미치는 영향 (The Effect of glutathione contents of White Ginseng(WG) Extracts and Entomopathogenic Fugi Extracts on the liver in Mice that was irradiated by radiation)

  • 본 연구는 선형가속기의 고에너지 엑스선을 조사한 생쥐 간에 대한 백삼과 동충하초의 방사선방호효과를 연구하였다. ICR계 수컷생쥐 군에 7일 동안에 경구적으로 백삼(150 mg/kg/day)과 동충하초(200 mg/kg/day)를 각각 방사선조사 전에 투여했고 다른 생쥐 군에 5 Gy(1.01 Gy/min)의 방사선량으로 전신조사를 했고 대조군에 생리적 식염수 (0.1 ml)를 투여 한 후 간 조직에서 환원형 글루타치온(GSH)과 산화형 글루타치온(GSSG)의 함량을 각각 검사하였다. 그 결과 방사선조사군(Rad)보다 동충하초투여군(EF+Rad)과 백삼투여군(WG+Rad)에서 환원형 글루타치온 (GSH)함량이 유의성 있게 증가했으나 산화형 글루타치온(GSSG)의 함량은 유의성 있게 감소하였다. 총 환원형 글루타치온(total GSH)과 산화형 글루타치온(GSSG)의 함량 비율은 방사선조사군(Rad)보다 동충하초투여군(EF+Rad)과 백삼투여군(WG+Rad)에서 유의성 있게 감소하였다.

백삼, 홍삼, 흑삼 추출물의 생리활성 비교 연구 (The comparative study on physiological activity of White ginseng, Red ginseng and Black ginseng extract)

  • 이 연구에서는 백삼(WG), 홍삼(RG) 흑삼(BG) 추출물의 효능차이를 비교하고자 항산화 및 항염 활성을 측정하였다. 세포독성 측정 결과 WG, RG, BG 모두 95% 이상의 세포 생존율을 나타내었고, 페놀함량, DPPH, ABTS 라디컬 소거능, ROS 생성량을 조사한 결과 WG와 RG에 비해 BG가 높은 항산화 활성을 보였다. NO생성 저해활성은 RG가, $PGE_2$생성 저해활성은 WG가 높았다. 염증성 사이토카인 생성량 감소에 미치는 영향을 측정한 결과, IL-$1{\beta}$ 생성량의 경우, WG와 RG가, IL-6 생성량의 경우 BG가 가장 큰 것으로 확인하였다. TNF-${\alpha}$ 생성량은 WG, RG, BG 모두 대조군과 비교하여 차이가 없었다. 이상의 분석 자료가 건강기능 식품과 기능성 화장품 원료 개발 등 전 임상단계의 기초적 자료로 가치가 있을 것으로 판단된다.

압출성형 백삼추출물의 화학적 조성 및 항산화 활성 (Chemical Compositions and Antioxidant Activity of Extract from a Extruded White Ginseng)

  • 본 연구는 압출성형 공정을 통한 인삼 시료의 성분 변화를 측정하기 위하여 일반성분 및 항산화활성을 측정하였다. 수분함량을 20%로 고정시키고, 배럴온도는 100, $140^{\ciirc}C$로 변화시켜 압출성형백삼을 제조하였다. 백삼, 홍삼, 압출성형백삼의 일반성분 실험에서는 압출성형 공정을 통한 성분의 증가를 확인할 수 없었다. 항산화활성 측정 결과 산성다당체 함량은 홍삼이 62.94 mg/mL로 높게 측정되었고 총 폴리페놀함량과 DPPH 측정에서는 $140^{\circ}C$ 압출성형백삼이 각각 14.99 mg/g, 80.21%로 높게 측정되어 압출성형 공정을 통한 항산화 물질의 증가를 확인할 수 있었다. $140^{\circ}C$ 압출성형백삼의 조성 분석을 위하여 BuOH 분획을 실시한 결과 총페놀성 화합물과 산성다당체 측정 결과가 각각 27.2 mg/g, 217.6 mg/mL로 측정되었으며 HPLC를 통해 알아본 사포닌 조성은 $140^{\circ}C$ 압출성형백삼에서 모든 성분의 증가를 확인할 수 있었고 특히 홍삼의 특유성분인 Rg3s, Rg3r의 증가로 압출성형 공정을 통한 백삼의 홍삼화 가능성을 확인할 수 있었다.

Anti-obesity effects of black ginseng extract in high fat diet-fed mice

  • Black ginseng is produced by a repeated steaming process. The aim of this study was to investigate the anti-obesity effects of black ginseng ethanol extract (BG-EE) in high fat (HF) diet-fed mice. Two groups were fed either a normal control (NC) diet or a HF diet (45% kcal fat). The other three groups were given a HF diet supplemented with 1% BG-EE, 3% BG-EE, and 5% BG-EE for 12 wk. The anti-obesity effects of the BG-EE supplement on body weight, the development of fat mass, and lipid mechanisms were assessed in obese mice. HF-induced hyperlipidemia, fat accumulation in the liver, and white adipose tissues were reduced after BG-EE supplementation. Total fecal weight and the amount of fecal fat excretion also were increased after BG-EE supplementation. These results suggest that BG-EE may be useful to ameliorate HF-induced obesity through the strong inhibition of fat digestion.

인지능 개선 효과 증진을 위한 백삼 추출물 조제 연구 (Study on White Ginseng Extract Preparation for Cognition Improvement)

  • Background: The study was conducted to elucidate the extraction conditions under which white ginseng has cognition-improving efficacy. Methods and Results: Extracts from white ginseng under different solvent and temperature conditions were analyzed for ginsenoside content and inhibitory effect on N-methyl-D-aspartate (NMDA) receptor and acetylcholinesterase. The total ginsenoside contents and amounts of ginsenoside Rb1 plus ginsenoside Rg1 from the 1st extracts (prepared with EtOH/$H_2O$ as solvent) were higher than those from the 2nd extracts (extracted with $H_2O$ after the 1st EtOH/$H_2O$ extraction). The contents in the 1st and 2nd extracts produced at $80^{\circ}C$ were also higher than those obtained at $50^{\circ}C$. Samples from the 1st extraction at $80^{\circ}C$ indicated higher inhibitory activities on NMDA receptors-whose excessive activation is thought to mediate the calcium-dependent neurotoxicity associated with several neurodegenerative diseases-than those from the 2nd extraction. Among the samples prepared at varying temperatures, the extract prepared at $50^{\circ}C$ showed the highest suppression activity on NMDA receptors. Note, however, that the extracts from the 2nd extraction at $50^{\circ}C$ inhibited acetylcholinesterase-whose inhibition could be a therapeutic strategy for neurodegenerative diseases with cognitive deficits and memory malfunction-more effectively than those from the 1st extraction. Conclusions: To enhance the cognition-improving activity of white ginseng extract, it is suggested that the extracts be utilized after being combined the 1st extracts (made with EtOH/$H_2O$ solvent) and the 2nd extracts (prepared with $H_2O$) at low temperature.
