The Geometry of the Space of Symmetric Bilinear Forms on ℝ2 with Octagonal Norm

  • Let $d_*(1,w)^2 ={\mathbb{R}}^2$ with the octagonal norm of weight w. It is the two dimensional real predual of Lorentz sequence space. In this paper we classify the smooth points of the unit ball of the space of symmetric bilinear forms on $d_*(1,w)^2$. Physiological Effects of Different Underwear Materials Thermoregulatory Response during Exercise with Sweating at Cold Environments (한랭환경하에서 운동발한시 인체의 체온조절반응에 대한 내의소재의 생리학적 의의)

  • This study conducted 4 different kinds of underwear materials, which were A (Cotton 100%), B (Wool 100%), C (Cotton/Wool, 50/50%) and D (Acrylic/Cotton, 50/50%) and were done in a climate chamber under cold ambient $10{\pm}1^{\circ}C$, $40{\pm}5%RH$ by 6 male subjects who were in good health. Physiological parameters such as rectal and local skin temperature(forehead, forearm, hand, trunk, thigh, leg, foot, back and chest), heart rate, body weight loss, clothing microclimate, blood lactic acid concentration, and wearing sensation were measured. Started with a 15-min rest period, 15-min of exercise 1 (the condition of 4.5 mile/hr walking speed equivalent to with 8.5 Kcal energy consumption on the treadmill) period, 15-min rest period, exercise 2 (after 3minutes warming-up at 3.0. 3.7, 4.5. 5.2. 6.0, 6.7 mile/hr) until exhaustion period, and final 15-min of recovery period were performed. The results were as follows: The lowest mean skin temperature was acrylic/cotton in order of wool > cotton/wool > cotton > acrylic/cotton (F=13. 79. p<0.00l). Most of all skin temperature by parts of body had turned out in sequence of temperature wool > cotton/wool > acrylic/cotton > cotton. Fore arm part showed highest temperature about $32.43^{\circ}C$ on wool and had a tendency approximately $1.8^{\circ}C$ higher than cotton which had the lowest temperature, and had the biggest difference among garments in terms of skin temperature. The back temperature within clothing showed about $2^{\circ}C$ higher than the chest temperature within clothing. but the back humidity within clothing showed about 4~12% higher than the chest humidity within clothing. Body weight loss by each garment was this sequence; cotton > acrylic/cotton > wool > cotton/wool.

  • Welding deformation causes critical problems under construction and in use of steel structures by varying the magnitude of the steel structures and deteriorating mechanic strength. Existing method to construct steel structures in civil engineering needs preassembly process for a part of or the whole structures on a broad space to examine the size of structures inevitably varied in the process of welding (assembly process). It leads to waste of time, space and human efforts, worry of safety accidents with the characteristic of the work to be performed on a high place, and non-efficiency and non-economy by using such supplementary equipments as crane. This paper, to remove the needless preassembly process by pre-estimating welding deformation produced under construction of large steel structures, devises a method modeling welded part for applying the equivalence load method and examines the effects of welding sequence and self weight on welding deformation by the method.

  • The secretory overexpression of Clostridium thermocellum endoglucanase A gene (celA) was examined in Saccharomyces cerevisiae using Kluyveromyces marxianus exoinulinase (INU1) signal sequence and GAL10 promoter. The two plasmids, pYEG-CT1 with its own signal sequence, and pYInu-CT1 with INU1 signal sequence were introduced to S. cerevisiae SEY2102 and S. cerevisiae 2805 host strains, respectively, and then each transformant was selected on the synthetic defined media lacking uracil. The expression level and secretion efficiency of endoglucanase A was increased by $18{\sim}22%$ and 11%, respectively, by INU1 signal sequence over celA signal sequence. By considering the high level of expression (361 unit/I), plasmid stability (89%), and secretion efficiency (70%), S. cerevisiae 2805 harboring plasmid pYInu-CT1 was selected as the opti-mal host vector system for the production of cellulose-degrading enzyme and recombinant yeast probiotic. The total expression and secretion efficiency of endoglucanase A was 418 unit/l and 73%, respectively, in the batch fermentation of S. cerevisiae 2805/pYlnu-CT1 on galactose medium. The mo-lecular weight of secreted endoglucanase A was found to be greater than 100 kDa, presumably due to the N-linked glycosylation.

Molecular Cloning and Analysis of Nucleotide Sequence of Xylanase Gene (xynk) from Bacillus pumilus TX703 (Bacillus pumilus TX703 유래 Xylanase 유전자(xynK)의 Cloning과 염기서열 분석)

  • A gene coding for xylanase from thermo-tolerant Bacillus pumilus TX703 was cloned into Escherichia coli DH5 $\alpha$ using pUC19. Among 7,400 transformants, four transformants showed clear zones on the detection agar plates containing oat-spells xylan. One of them which showed highest xylanase activity was selected and its recombinant plasmid, named pXES106, was found to carry 2.24 kb insert DNA fragment. When the nucleotide sequence of the cloned xylanase gene (xynK) was determined, xynK gene was found to consist of 1,227 base-pair open reading frame coding for a polypeptide of 409 amino acids with a deduced molecular weight of 48 kDa. The coding sequence was preceded by a putative ribosome binding site, the transcription initiation signals, and cia-acting catabolite responsive element. The deduced amino acids sequence of xylanase is similar to those of the xylanases from Hordeum vulgare (barley) and Clostridium thermocellum, with 39 and 31% identical residues, respectively. The amino acids sequence of this xylanase was quite different from those of the xylanases from other Bacillus species.

Gene Cloning, Nucleotide Sequence and Efficent Expression of Peptidyl proryl cis-trans Isomerase from Bacillus stearothermophilus (Bacillus stearothermophilus의 Peptidyl Prolyl cis-trans Isomerase 유전자 분리 염기배열 및 발현)

  • A PPIase gene of Bacillus stearothermophilus was screened from a genomic library by plaque hybridization using the A-1 primer as a probe. A PPIase positive plaque contained a 3.0kb insert of the chromosomal DNA. A 3.0kb fragment was subcloned into pUC18, resulting pPI1-40. A DNA fragment encoding the N-terminal portion of the PPIase in pPi-40 was amplified by polymerase chain reaction(PCR) method using the A-1 and B-2 primers. The amplified fragment was cloned into the Sma I site of pUC18 and recombinant plasmid was designated as pSN-18. The nucleotide sequence of 167bp fragment was determined. The deduced amino acid sequence of PPIase was completely matched with the determined N-terminal amino acid sequence of PPIase B. stearothermophilus. The translated protein sequence of PPIase B. stearothermophilus was compared with sequence from periplasmic PPIase from Escherichina coil ; homogies of 16 and 58%, respectively, were found. The clond PPIase gene was over-expressed in E. coil cell using pUC19 as an expression vector. The enzyme was partially purified by heat treatment and colum chromatochraphy on DEAE-Sepharose CL-6B. The molecular weight of the enzyme was dermined to be about 18.0 kDal by SDS-PAGE.

The Molecular Weight Dependance of Paramagnetic Gd-chelates on T1 and T2 Relaxation Times (상자성 복합체의 분자량에 따른 T1 및 T2 자기이완시간에 관한 연구)

  • To evaluate the T1, T2 magnetic relaxation properties of water molecule according to molecular weight of paramagnetic complex. 4-aminomethyicyclohexane carboxylic acid (0.63 g, 4 mmol) was mixed with the suspension solution of DMF (15 ml) and DTPA-bis-anhydride (0.71 g, 2 mmol) to synthesize the ligand. The ligand was then mixed with $Gd_2O_3$ (0.18 g, 0.5 mmol) to synthesize Gd-chelate. For the measurement of magnetic relaxivity of paramagnetic compounds, the compounds were diluted to 1 mM and then the relaxation times were measured at 1.57 (64 MHz). Inversion-recovery pulse sequence was employed for T1 relaxation measurement and CPMG (Carr-Purcell-Meiboon-Gill) pulse sequence was employed for T2 relaxation measurement. In case of inversion recovery sequence, total 35 images with different inversion time(T1)s ranging from 50 msec to 1,750 msec. To estimate the relaxation times, the signal intensity of each sample was measured using region of Interest (ROI) and then fitted by non-linear least square method to yield T1, T2 relaxation times and also R1 and R2. Compared to T1=($205.1{\pm}2.57$) msec and T2=($209.4{\pm}4.28$) msec of Omniscan (Gadodiamide), which is commercially available paramagnetic MR agent, T1 and T2 values of new paramagnetic complexes were reduced along with their molecular weight. That is, T1 value was ranged from $(96.35{\pm}2.04)\;to\;(79.38{\pm}1.55)$ msec and T2 value was ranged from $(91.02{\pm}2.08)\;to\;(76.66{\pm}1.84)$ msec. Among new paramagnetic complexes, there is a tendency that the R1 and R2 increase as the molecular weight is increases. As molecular weight of paramagnetic complex increases, T1 and T2 relaxation times reduce and thus the increase of relaxivity (R1 and R2) Is proportional to molecular weight.

Growth and Yield Characteristics by Tillering Sequence in Coix lachryma-jobi L. var. mayuen Stapf (율무의 분얼순서에 따른 생육 및 수량 기여도)

  • This study was conducted to evaluate growth and yield characteristics and the contribution to the yield according to the emergent sequence of tiller under two planting density $(60{\times}15\;cm,\;60{\times}25\;cm)$ in Coix lachryma-jobi L. var. mayuen Stapf. The average culm length and number of rachillas of $60{\times}25\;cm$ planting density were 20 cm shorter and 4.7 higher, respectively, whereas seed setting position of $60{\times}25\;cm$ planting density was 1.7 nodes lower compared with $60{\times}15\;cm$ planting density. Culm length and culm diameter of each tiller became shorter and thinner as the tiller emerged later. Length of rachillas was longer, number of grains was higher and ripening rate was also higher in $60{\times}25\;cm$ planting density than those of $60{\times}15\;cm$ planting density, whereas 100 grain weight in $60{\times}25\;cm$ planting density was lower by 0.5 g compared with $60{\times}15\;cm$ planting density. Length of rachillas and number of grains according to the emergent sequence of tiller were shown shorter and lower as the tiller emerged later. The highest number of bract was obtained from the main culm in both planting densities. 100 grain weight according to the emergent sequence of tiller became lower as the tiller emerged later in $60{\times}15\;cm$ planting density, but there was no clear tendency in $60{\times}25\;cm$ planting density. Among 8 tillers, 5th tiller and 4th tiller of main culm contributed 84.4% to the yield in $60{\times}15\;cm$ planting density, while it was 84.1% in $60{\times}25\;cm$ planting density. Analysis of path coefficient to grain yield showed that direct effect was the highest in the number of grain per tiller followed by in the 100 grain weight and in ripening rate.

Mapping QTL for Grain Quality Traits Using an Introgression Line Population from a Cross between Ilpumbyeo and Moroberekan in Rice (일품벼/모로베레칸 이입계통을 이용한 미질특성 관련 QTL 분석)

  • We conducted a QTL analysis of grain quality traits using 117 $BC_3F_4$ and $BC_3F_5$ lines developed from a cross between Ilpumbyeo and Moroberekan. Genotypes of 117 $BC_3F_5$ lines were determined using 134 simple sequence repeat (SSR) markers. A linkage map constructed using 134 SSR markers was employed to characterize quantitative trait loci (QTL). The 117 $BC_3F_4$ and $BC_3F_5$ lines were evaluated for eleven grain quality traits in 2005 and 2006. A total of 18 QTLs were identified for eleven traits, and the phenotypic variance explained by each QTL ranged from 9.9% to 35.2%. Moroberekan alleles contributed positive effects in the Ilpumbyeo background at two QTL loci for 1,000 grain weight. Four QTLs, two for chalky rice and one each for 1,000 grain weight and head rice were consistently detected in two consecutive years indicating that these QTLs are stable. Clusters of QTLs were observed in three chromosome regions. One cluster harboring five QTLs including head rice and brown rice ratio near SSR markers RM190 and RM314 was detected on chromosome 6. Another cluster harboring grain weight and white belly was detected on chromosome 2. Increase in white belly at this locus might be due to the increase in grain weight due to the presence of the Moroberekan allele. The Moroberekan alleles at two QTL loci, gw3 and gw4 associated with increased grain weight might be useful in breeding programs to develop high-yielding cultivars.

Molocular Cloning and Sequence Analysis of the Immediate Early Viral Gene, IE1, from Bombyx mori Nuclear Polyhedrosis Virus K1

  • We have cloned and characterized an immediate early-1 gene, iel, which is activated immediately upon entrance of the viral genome into the cell nucleus, from Bombyx mori nuclear polyhedrosis virus (BmNPV) K1 strain. This gene encodes a protein 584 amino acids with a predicted molecular weight of 67 kDa. The promoter and coding regions of BmNPV-K1 ie1 showed high homology with Autographa californica nuclear polyhedrosis virus and BmNPV T3 strain. The BmNPV-K1 ie1 was different from amino acid sequence at 4 positions in BmNPV T3. The location of ie1 gene in the BmNPV-K1 genome was confirmed by Southern blot analysis and its expression patterns at the transcriptional level in the infected cells were confirmed by Nerthern hybridization analysis.
