인간의 질병을 치료하기 위한 여러 가지 의료 시스템의 개발이 생명공학의 발전과 함께 많은 연구가 이루어지고 있다. 또한 약물이나 유전자와 같은 생리활성물질을 체내에 안전하게 전달할 수 있는 시스템의 개발과 함께 이루어지고 있다. 이러한 시스템에 있어서 가장 중요한 것은 생체적합성 및 생체분해성 그리고 무독성의 특성을 가진 생체의료용 고분자를 개발하는 것이다. 천연고분자물질인 키토산은 좋은 생체적합성과 생체활성의 특성을 가지고 있어서 생체의료용 재료로 심도 있게 고려되어지고 있다. 키토산의 물성은 키틴의 결정성 구조에 따라 다르게 설명되므로 키틴의 구조적 분석에 대한 연구가 생체재료로써의 응용을 위해서 선행되어야 한다. 이러한 관점에서 본 총설에서는 키틴의 결정성 구조 분석, 키토산의 일반적인 물성 그리고 생체의료용 재료로써 저분자량 수용성 키토산의 가능성을 소개하였다. 또한 다양한 기능성기를 이용한 저분자량 수용성 키토산의 화학적인 개질을 약물전달체로써의 가능성을 강조하고 생체이용율의 향상을 위해 수행하였다.
A coagulation process for RO (reverse osmosis) membrane pretreatment system was an effective technology to remove colloidal and particulate matters. However, coagulant residuals from the pretreatment process may negatively affect RO membrane performance. The bench-scale coagulant exposure study was performed to investigate the effect of their residual on adsorbed mass which related to the membrane performance. Coagulant addition in this study ranged from 0 to 5mg/L ferric chloride, alum, and 2mg/L cationic polymer(poly-di-methyldiallyl ammonium chloride) as coagulant aids. This results showed that adsorbed mass is not significantly increased during short-time period, however, accumulated mass of coagulants on the membrane surface is significantly increased during long-time experimental period. The effect of pH on coagulants adsorption characteristics was significantly differed due to the electrostatic repulsive interactions between soluble coagulants and membrane surface charge. This data suggest that the RO membrane performance of drinking water treatment plant could be decreased by adsorption of residual coagulants when applied for the coagulant pretreatment process.
아세클로페낙은 높은 결정성을 갖는 난용성 약물이다. 이러한 난용성 약물의 용해도를 증진시키기 위해서 고체분산법을 바탕으로 한 분무건조기를 이용하여 미립구를 제조하였다. PVP-K30을 수용성 담체로 사용하였고 폴록사머는 계면활성화제로 사용하였다. 제조된 아세클로페낙 고체분산체의 특성을 SEM, DSC, XRD 그리고 FT-IR을 이용하여 확인하였다. SEM, DSC, XRD을 통하여 아세클로페낙 고체분산체가 무정형임을 알 수 있었고 FT-IR을 통하여 아세클로페낙과 PVP-K30간에 수소결합을 통해 염을 형성하고 있다는 것을 확인할 수 있었다. 제조된 미립구는 pH 6.8에서 방출을 실시하였으며 시판제인 $Airtal^{(R)}$과 용출률을 비교하였으며 분무건조를 통해 제조한 미립구가 시판제인 $Airtal^{(R)}$ 보다 용출률이 크다는 것을 확인하였다.
This experiment was conducted to examine the effects of a short-term treatment of high pressure $CO_2$ on shelf-life of strawberry fruit. A short-term treatment (12 hr) of 100% $CO_2$ resulted in the increase of fruit firmness up to 71.9% compared to that at harvest. The firmness of $CO_2$ treated fruit remained a significantly higher than that of control (air) up to 15 days. The alteration of pectic polymers was observed by $CO_2$ treatment such as an increase of EDTA soluble pectins and decrease of water soluble ones. The $CO_2$ treatment resulted in the increase of total amount of wall bound calcium. Pectate lyase activity, an important agent of strawberry fruit softening, was also significantly reduced by $CO_2$ treatment. Contents of soluble solids and acids of $CO_2$ treated fruit were higher than those of control fruit. Short-term treatment of high pressure $CO_2$ affected shelf-life through firmness increase whereas the visual quality and decay incidence of strawberry fruit were not affected.
The luminescence spectra of $Ru(bpy)_3^{2+}$ in poly(methacrylic acid) (PMA) solutions varied sensitively with pH. At pH < 5.5, the emission intensity increased with pH up to 4 times, while it decreased with pH beyond the pH. The enhanced emission intensity was accompanied by blue-shift of the emission maxima as much as 15 nm. The enhancement of emission intensity was attributed to the restricted rotational mobility of ligand of the cation bound to densely coiled PMA molecules at pH < 5.5. The sharp decrease in emission intensity with increasing pH near pH 5.6 was accounted for conformational transition of the polymer to more extended structure, which was also revealed in viscosity measurement. The enhancement of emission intensity became higher as NaCl concentration of the solution increased. The binding constant of $Ru(bpy)_3^{2+}$ with two carboxylate groups of PMA was calculated as $2{\times}10^5\;M^{-1}$ in 0.1 M NaCl at pH 5.2. The pH dependence of luminescence quenching rate of $Ru(bpy)_3^{2+}$ by $Cu^{++}$ also showed maximum near pH 5, and the rate was more than $10^3$ times higher than that in water, whereas the maximum enhancement of quenching rate (about 20 times) in poly(acrylic acid) (PAA) solution occurred at pH 4.5. On the other hand, the pH dependence for neutral water soluble nitrobenzene (NB) exhibited opposite trend to that of $Cu^{++}$. The quenching constant vs pH curve for $MV^{++}$ was composite of those for $Cu^{++}$ and NB. The anomalous high quenching rate for $Cu^{++}$ in PMA solution at pH < 5.5 was attributed to the binding of $Ru(bpy)_3^{2+}$ and $Cu^{++}$ to the same region of PMA, when it conforms densely coiled structure in the pH range. The observation of mininium quenching rate for NB near pH 5.5 indicated that the $Ru(bpy)_3^{2+}$ bound to the densely coiled PMA is not accessible by NB, which is in bulk water phase. The composite nature of the pH dependence of quenching rate for $MV^{++}$ in PMA solution was attributed to the smaller binding affinity of the cation to PMA, compared to that of $Cu^{++}$. The sharp, cooperative conformational transition with pH observed in PMA was not revealed in PAA. But, the pH dependence of quenching rates in this polymer reflected increased charge density and, thus, binding of cations to the polymer, and expansion of the polymer chain with pH.
The release of heparin from monolithic devices composed of different ratios of polythylene oxide (PEO MW 20,000) and hydrophobic polydimethylsiloxane or polyurethane was investigated. Water soluble PEO blended into the polymers provided a controlled release of heparin. The release rate of heparin could be controlled by varying the content of PEO. The heparin release rate from the devices increased as the content of PEO in the devices increased. The release mechanism may be associated with the creation of pore of domain through the devices following the swelling and the change in the physical structure of the polymer network. Hydrophobic polydimethylsiloxanes and polyurethanes containing PEO can provide an antith rombogenic material for prologed release of heparin from blended system.

Sodium hyaluronate (NaHA), water soluble polymer having ultra-high molecular weight, is characterized by using on-line frit inlet asymmetrical flow field-flow fractionation (FI-AFlFFF) and multiangle light scattering (MALS). This study demonstrates the capability of power programming FI-AFlFFF for the separation of NaHA and the applicability of FI-AFlFFF with MALS for the characterization of molecular weight distribution and their structural information. Since sample injection and relaxation in FI-AFlFFF are achieved by using hydrodynamic relaxation, separation of high molecular weight polymers can be achieved smoothly without halting the separation flow. Experiments are carried out with the two different NaHA products (a raw NaHA sample and a thermally degraded NaHA product) and molecular weight distribution and conformations in solution are determined. Influence of sample filtration on the change of molecular weight distribution is also discussed.
Long-circulating liposomes have prompted interests in using them as a drug delivery system. This improvements in delivery has been thought to be the results from sustained action of liposomes in plasma without RES uptake. Although methotrexate(MTX) has been one of the most widely used antineoplastc drug, its use was limited by prompt RES uptake. The purpose of this study was to prepare long-circulating liposomes for MTX using highly water-soluble polymer (PEG-PE). In vitro, release of MTX from liposomes in phosphate buffer (pH 7.4), rat liver homogenate, and rat plasma was investigated. The pharmacokinetics and organ distribution of fee drug, conventional and long-circulating liposomes were also compared with one another after intravenous administration to rats.
The release rate of heparin from monolithic devices composed of raffinose, ${\beta}-cyclodextrin$, polyethylene oxide (Mw 20,000, PEO), and hydrophobic polyether urethane (biomer) was investigated. Water soluble raffinose, ${\beta}-cyclodextrin$, and PEO blended into the biomer provided a controlled release of heparin. The release rate of heparin could be controlled by the content of raffinose, ${\beta}-cyclodextrin$, and PEO in the devices. The mechanism of release rate increased by the raffinose, ${\beta}-cyclodextrin$, and PEO may result from the formation of channels and pores in the biomer matrices following the swelling and the change in the physical structure of polymer net work. Hydrophobic polyurethane containing raffinose, ${\beta}-cyclodextrin$, and PEO can provide a hydrophilic antithrombogenic material for prolonged release of heparin.
