Phenylpropanoid Glycosides of Paulownia coreana Uyeki Leaves

  • The leaves of Paulownia coreana Uyeki were collected, extracted with acetone-$H_2O$ (7:3, v/v), concentrated under reduced pressure and successively fractionated using n-hexane, methylene chloride, ethyl acetate and water on a separatory funnel. A portion of the ethyl acetate soluble powder was chromatographed on a Sephadex LH-20 column using aqueous methanol and ethanol-hexane as washing solvents. Two isomeric phenylpropanoid glycosides were isolated and elucidated as verbascoside and isoverbascoside by NMR and MS spectrometers.

  • A multi-layer of the dye, natural colorant Berberine, was successfully developed by the self-assembly deposition from water-soluble cationic dye(Berberine chloride) and anionic polyelectrolyte PSS(Polysodium 4-styrenesulfonate) in aqueous solution via electrostatic attraction. The corresponding results on multi-layer were characterized by UV-Vis absorbance measurements. The growth of multi-layer formed by the sequential interaction was also determined. The findings measured by UV-Vis spectrophotometer showed that the bilayer deposition characteristic was linear and highly reproducible from layer to layer.

  • An efficient process for the solution-phase synthesis of biaryl amides has been developed. Girard's reagent T. an inexpensive scanvenger. was found to be very efficient in trapping excess aromatic acid chlorides. resulting in water soluble by-products. which were easily removed from the products by liquid-liquid extraction. The ease of use. and the excellent purity of the amide libraries obtained are important features of this protocol. (omitted)

Osmose Process에 의한 목재방부(木材防腐) 처리(處理)에 관(關)한 연구(硏究) (A Study on the Preservative Treatment of Wood by Osmose Process)

  • In order to investigate the effectiveness of Osmose process for the practical treatment of wood this study has been made using water soluble preservatives such as Malenit and chromated zinc chloride. The results obtained in this investigation are as follows: 1. The penetration of Malenit in sapwood has been observed deeper than that of chromated zinc chloride for all species tested in this investigation. 2. The penetration of preservatives applied in soft wood, ie. Pinus densiflora and Larix leptolepis has been observed better results than that of hard wood, i.e., Quercus accutissima and Carpinus laxiflora. 3. The longer stack covering, despite of preservatives applied and size of wood tested, has given better penetration for all species tested, and the fastest diffusion has been occured in 15 days from they day started. Following after 15 days diffusion had gradualy become slower. 4. The length of time needed for effective penetration has taken 45 days for all species tested, reaching twenty millimeters (20mm) in depth in case of Malnit, that means also more than 50% of penetration into sapwood portion. However it has taken 45 days fer Pinus densiflora and Larix leptolepis, reaching fifteen millimeters (15mm) and 60 days for Quercus accutissima and Carpinus laxiflora, reaching same fifteen millimeters in case of chromated zinc chloride, that means also less than 50% (except 50% for Larix) of penetration into sapwood portion. 5. Deeper penetration of preservatives from the wood surface has been observed in the larger wood than the smaller wood for all species tested, although the penetration ratio between the width of sapwood and the length penetrated has been observed smaller in larger wood than smaller wood. 6. The relation between moisture content of wood and the penetration of preservatives into wood tested has shown the linear regression, that is, the more moisture content brought the deeper penetration. 7. Following the result obtained at this investigation osmose process with Malenit applied has indicated as a useable process for the none pressure treatment of wood.

Monascus anka의 균주선발 및 색소생성 조건 (Pigment production in Monascus anka)

  • 유기 용매에 추출되는 유리형의 홍국 색소를 보다 많이 얻기 위하여 Monascus anka의 균주를 자연 및 자외선 변이균주에서 선발하고 배양 조건을 탐색하였다. 균주 N3를 Lin's medium에서 진탕 배양, Nishikawa's medium에서 정치 배양 및 진탕 배양하여 균체성장과 색소생성에 소요되는 시간, 유기용매에 추출되는 색소의 양을 비교하여 보았다. Lin's medium에서의 액침 배양에서는 수용성 색소가 우세하게 생성되는 반면 Nishikawa's medium에서의 정치 배양에서는 보다 많은 유기용매 가용성의 색소를 얻을 수 있었다. 한편 탄소원으로는 자당외에 에탄올이 가장 효과적이었다. Sterol 합성저해제를 처리하였을 때는 상당한 색소 형성의 증가를 볼 수 있었다. 이차원 TLC로써 아직 보고된 바 없는 색소들이 존재함을 알 수 있었다. 이 균주의 색소중 가장 다량으로 존재하는 황색소의 하나를 분리하였고 분광학적 방법으로 ankaflavin으로 동정하였다.

탈삽조건에 따른 대봉감의 품질 변화 (Effect of Astringency Removal Conditions on the Quality of Daebong Persimmon)

  • The application of persimmon for making new products was investigated by analyzing the degree of astringency removal, softness of fruit, and sensory properties of Daebong persimmons (Diospyros kaki L) treated with three different astringency removal methods of steeping in water, steeping in 1% salt solution, and vacuum packing at $40^{\circ}C$. The weight of persimmon fruit increased with increasing steeping time, but decreased with increasing vacuum packing time. The fruit steeped in $40^{\circ}C$ salt solution (1%, w/w) was not detected with soluble tannin and not developed color with ferric chloride after steeping for 9 h. A total of 12 h of steeping time was required for removing soluble tannin using other methods. The lightness (L) in color and hardness in texture decreased when storage time increased, regardless of astringency removal methods. In overall, the preference was shown with the highest score in persimmon steeped in salt solution for 9 h and 12 h that were vacuum packed at $40^{\circ}C$. In conclusion, the astringency removal condition of Daebong persimmon was good after being steeped on 1% salt solution for 9 h at $40^{\circ}C$.

Enhanced stability of NADH/dehydrogenase mixture system by water-soluble phospholipid polymers

  • To maintain activity in a coenzyme/enzyme mixture system, such as ${\beta}$-nicotinamide adenine dinucleotide (NADH)/dehydrogenase, the water-soluble 2-methacryloyloxyethyl phosphorylcholine (MPC) polymers as an additive were synthesized and investigated for their stabilizing function. The inhibitor for the NADH/dehydrogenase reaction was spontaneously formed when the NADH was stored in the dehydrogenase solution. Therefore, we hypothesized that if the additive polymer could interact with an inhibitor without any adverse effect on the dehydrogenase, the activity in the NADH/dehydrogenase mixture could be maintained. We selected lactose dehydrogenase (LDH) as the enzyme, and the NADH was dissolved and incubated at $37^{\circ}C$ in the LDH solution containing the polymers. The phospholipid polymers used in this study were poly(MPC) (PMPC), poly(MPC-co-3-trimethylammonium-2-hydroxypropyl methacrylate chloride) (PMQ) and poly[MPC-co-potassium 3-methacryloyloxypropyl sulfonate ($MSO_3$)] ($PMMSO_3$). The poly($MSO_3$) was used as a reference. For the PMQ and $PMSO_3$ aqueous solutions, the activity of the NADH/LDH mixture system decreased with incubation time as the same level or lower than that in the Tris buffered solution in the absence of the polymers. However, for the poly($MPC-co-MSO_3$) ($PMMSO_3$) aqueous solution, the activity of the NADH/LDH mixed system was six times higher than that in the buffered solution even after a 3-days incubation. The LDH activity was 1.5-1.8 times higher in the presence of the $PMMSO_3$ compared with that in the $PMSO_3$ solution. The mixture of two polymers, poly(MPC) and poly($MSO_3$), did not produce any stabilization. Thus, both the MPC and $MSO_3$ units in the polymer chain had important and cooperative effects for stabilizing the NADH/LDH mixture.

주요 시판 두부의 물리화학적 특성 평가 (Evaluation of Physico-Chemical Characteristics of Commercial Tofu Products in Korea Market)

  • This study was conducted to compare the quality characteristics of commercial tofu products from the market in Korea. Seventeen types of commercial tofu samples were taken and their physicochemical properties, including soluble solid contents, salinity, pH, total acidity and moisture (total solid contents), were analyzed. The hardness of tofu was negatively correlated with the moisture contents of tofu (r=-0.667**). The commercial tofu showed pH 5.80~6.24, total acidity of 0.016~0.034%, soluble solids of 1.50~3.45°Brix, salinity of 1.20~2.30%, and moisture content of 79.91~87.57%, respectively. All 17 tofu samples sold in the Korean market were prepared using crude MgCl2 and sea water as a coagulant. The quality characteristics vary depending on the constituent's of soybeans, and the ratio and amount of coagulants of tofu used. The origin of soybean seeds affected the yellowness of tofu; tofu made from imported soybean showed a higher b value than domestic soybean. These results are expected to be useful for understanding trends in the domestic tofu industry.

생물공정 처리수의 PAC (Poly Aluminum Chloride) 응집에 의한 고효율 인 제거 특성 (High-Rate Phosphorous Removal by PAC (Poly Aluminum Chloride) Coagulation of A2O Effluent)

  • 2011년부터 시행될 2단계 수질 오염 총량제에 대비하여 고효율 인 제거 공정 개발이 필요하며 본 연구에서는 생물학적 처리와 화학적 응집처리를 조합한 공정이 대안으로 검토되었다. 2세대 응집제로서 최근 많이 사용되고 있는 PAC(poly aluminum chloride)를 이용하고 실제 하수종말처리장 A2O 공정의 호기조 유출수를 시료로 하여 인 응집 특성 및 적정 조건, 효율 향상 방안 등에 대해 실험연구를 수행하였다. 먼저 적정 PAC 투입 농도는 부피기준으로 30 ppm, mol비로는 2.81 mol Al/mol P인 것으로 나타났다. 단순침전에서 17.2%에 불과하던 용존성 인의 제거율이 PAC 10 ppm 투입으로 30.3%까지 증가하였으며, 20 ppm에서 30 ppm으로 PAC 투입량을 증가시킬 때 49.3%에서 88.4%로 제거율을 크게 향상시킬 수 있었다. 이때 총 인의 제거율은 92.4%, 유출수 총 인 농도는 0.3 mg/L로 총량제에 부합하는 방류수 수질을 달성할 수 있었다. 적정한 응집시간은 4분, 침전시간은 20분, 초기 pH는 7로 나타났고 원시료 (pH 7.0)는 별도의 pH 조절 없이 응집 가능한 것으로 판단되었다. pH 7 조건에서 용존성 인의 제거가 크게 향상되었으며, 추가적인 확인 연구가 필요하나, 이는 인의 경우 $Al(OH)_3$에 의한 sweep floc formation이 주요 제거 기작임을 의미하였다. 2차침전조 유입부에 PAC를 투입하여 2차침전조를 응집침전조로 활용하는 방안과 별도의 응집침전조를 2차침전조 후단에 설치하여 PAC를 투입하는 방안을 비교한 결과 PAC 투입량 대비 인 제거효율 증진 관점에서 후자가 바람직한 것으로 조사되었다. 이때 처리수의 인 농도는 0.18 mg/L, 제거율은 95.4%로 향상시킬 수 있었다. 끝으로 응집 처리수를 반송할 경우 인 제거 효율을 높일 수 있었으며 적정 반송율은 0.3인 것으로 분석되었다.

수용성 단백질에 대한 새우껍질 Chitosan의 흡착 특성 (Adsorption Property of Shrimp Shell Chitosan to Water Soluble Proteins)

  • Chitosan의 탈아세틸화도와 분자량 등을 달리하여 chitosan의 제조조건에 따른 수용성 단백질에 대한 흡착율 및 흡착에 영향을 미치는 인자에 대해 검토하였다. Chitosan의 탈아세틸화 조건을 $60\%,\;70\%,\;80\%$로 달리 하여 albumin, hemoglobin, albumin-myoglobin 혼합용액에 적용했을 때 chitosan의 탈아세틸화도가 높을 수록 chitosan과 수용성 단백질 사이의 흡착율은 높게 나타났다. 초음파 처리에 의하여 chitosan의 분자량을 변화시켰을 때 chitosan의 분자량이 작을수록 chitosan과 수용성 단백질 사이의 홉착율은 높게 나타났다 pH 변화에 따른 chitosan과 수용성 단백질의 흡착율은 albumin 및 albumin-myoglobin 혼합용액에서는 pH 4에서, hemoglobin 용액에서는 pH 7에서 홉착율이 높게 나타났다. Chitosan과 수용성단백질과의 흡착에서 반응시간은 albumin 및 albumin-myoglobin 혼합용액에서는 4시간, hemoglobin 용액에서는 3시간까지 흡착을이 증가하였고, 그 이상의 시간을 반응시켜도 흡착율의 증가는 거의 나타나지 않았다. 수용성 단백질 용액에 NaCl 농도를 0.1에서 1.0M로 증가시켜 첨가했을 때 염의 농도가 높을수록 chitosan과 수용성 단백질 사이의 흡착이 잘 일어나지 않았다.
