This study was conducted to assess effects of soil moisture condition and ventilation on development of fermented fruit of oriental melon. In higher soil moisture condition (-10 kPa), roots absorbed more water and transpiration was decreased under low temperature and high humidity conditions. Development of fermented fruit may be come from absorbed water concentrated placenta with higher soluble solid. Fermented fruit was developed with higher level at Shintozoa and Elite, was developed with lower level at Hongtozoa, and was not developed at self-rooting of oriental melon. Ventilation on fruit did not affect development of fermented fruit at Shintozoa, Elite, Hongtozoa and self-rooting seedling. Ca contents of flesh and placenta of fruit under ventilation treatment were higher than control but contents of Acetaldehyde, Ethanol and Ethylacetate were lower.
Sugars in Korean jujube fruit and jujube fruit drinks were studied. The sugars in Korean jujube fruit were extracted by boiling with water for 30min and stirring for 1 hour at 7$0^{\circ}C$ after soaking in the water for 24 hours at 4$^{\circ}C$ followed by crushing. Korean jujube fruit was found to contain 25% of sucrose, 21.2% of fructose and 20.7% of glucose. Korean jujube fruit drinks of V company and H company were found sugar composition of 5.9% and 6.9% of sucrose, 2.2% and 2.1% of fructose and 2.4% and 2.4% of glucose, respectively. No other mono- and oligosaccharides were detected in the test of TLC and HPLC. A viscous material in jujube fruit and jujube fruit drinks was separated by ethanol fractionation, and identified as pectin by 1H-NMR and carbazole analysis. Pectin of Korean jujube fruit, jujube drink of V company and jujube drink of H company was found to contain 61, 58 and 55% of galacturonic acid, respectively.
The seeds of Elaeocarpus serratus, a tropical underutilized fruit tree are characterized by hard seed coat and consequent poor water uptake and low germination. To improve the regeneration through seeds, various parameters such as viability of seeds, water uptake, and effect of seed mass on germination and pretreatments were performed using a completely randomized design (CRD). Tetrazolium (TZ) test was conducted using fresh, mature seeds revealed $50{\pm}2.56%$ mean viability. Seeds of different weight classes showed similar pattern of water uptake and the saturation level was achieved at 60 hrs of soaking. Seeds belong to weight class 2.6-3.5g were germinated ($12.5{\pm}1.26%$) with $175{\pm}1.75days$ (d) of mean time taken for germination (MTG). Germination capacity of seeds varied significantly among different populations and Varkala population gave $12.5{\pm}1.1%$ germination with $174.6{\pm}2.5d$ MTG. Among various seed treatments, mechanical scarification was superior in germination and significant reduction in MTG ($p{\leq}0.05$). The mechanical scarification by complete removal of seed coat resulted in $49.2{\pm}1.52%$ germination within a short period of time ($9.52{\pm}0.89d$ MTG). However, the complete removal of seed coat without damaging to embryo is a difficult task. An alternate treatment (Mechanical scarification II) by making cracks on nut faces vertically followed by soaking in distilled water for 24 hrs gave $48.4{\pm}1.73%$ germination with significantly reduced MTG ($12.14{\pm}0.56d$) over unsoaked, untreated control ($6.5{\pm}1.84%$ germination and $197.18{\pm}1.79d$ MTG; $p{\leq}0.05$). This treatment (Mechanical scarification II) is therefore recommended for E. serratus seeds as it can adopt easily and can achieve 7 fold increases in germination over control. The recorded germination through mechanical scarification is in tune with realized viability percentage of the seeds.
This study examined the anti-microbiological effects of chlorine treatment on the surface of fresh fruits, in order to improve microbiological safety in school foodservice operations. Non-peeled fruit(strawberries) and peeled fruit(bananas) were treated with different concentrations of chlorinated water and rinsing numbers, followed by microbiological testing. The fruits were immersed at different concentrations of chlorinated water(0 ppm, 50 ppm, and 100 ppm) and durations(3 min and 5 min), and were then rinsed with tap water(one time, two times, or three times). The total viable cell counts of both the strawberries and bananas ranged from $10^3$ CFU/g to $10^4$ CFU/g, and coliform levels ranged from $10^2$ CFU/g to $10^3$ CFU/g. As the chlorine concentration, immersion time, and rinsing number increased, anti-microbiological activity increased. The largest microbial reduction was shown with immersion for 5 min in 100 ppm chlorinated water and three rinsings. In the strawberries, this treatment reduced the initial population of total viable cells and coliforms by 3.29 log CFU/g and to an undetectable level, respectively, no total viable cells or coliforms were detected on the banana surface following this treatment. However, after a sterilization treatment with immersion for 5 min in 50 ppm chlorinated water and three rinsings, the total viable cell counts and coliform counts of the strawberries and bananas decreased to acceptable levels, based on the microbiological standards for ready-to-eat foods. Overall, it was shown that the sterilization treatment of 50 ppm chlorinated water, soaking for 5 min, and three rinsings provided an effective reduction in surface microbes, and enhanced the microbiological safety of the fruit.
A severe brown rot on peach fruit caused by a Phytophthora sp. has occurred at peach orchards in Taegu of Korea from late June to early August in 1997. Infected fruits showed irregularly round or circular water soaking brown regions. In the severe case, fruits were entirely rotten and surface of the fruits were wrinkled. Occasionally, white mycelia and abundant sporangia were developed on the surface of fruit. Inner tissues of the fruits were also discolored to brown. The causal fungus was identified as Phytophthora cactorum based on following characteristics. Sporangia were ovoid, conspicuously papillate, caducous and measured as 28.4~48.1$\times$21.9~37.2 (av. 39.9$\times$30.4) ${\mu}{\textrm}{m}$. Sexuality of the fungus was homothallic. Oogonia were 25.0~34.0 (av. 29.9) ${\mu}{\textrm}{m}$ in size. Most antheridia were paragynous and measured av. 10.5$\times$13.0 ${\mu}{\textrm}{m}$. Optimum temperature for mycelia growth was around 25~3$0^{\circ}C$. However none of the isolates grew under 7$^{\circ}C$ and over 35$^{\circ}C$. The fungus revealed high pathogenicity to fruits, shoots and leaves of peach, apple and pear with different degrees. Phytophthora fruit rot of peach caused by Phytophthora cactorum has not been reported in Korea previously.
Objective: The study was carried out to evaluate the hepatoprotective and antioxidant potential of Ziziphus mucronata (ZM) fruit extract. Methods: The different types of fruit extract were prepared by soaking the dry powdered fruit in different solvents followed by rotary evaporation. Each extract was tested for its phenol content and antioxidant activities. An in vivo study was performed in Sprague-Dawley (SD) rats. Thirty adult male SD rats (aged 21 weeks) were divided into six groups of five rats each and treated as follows: The normal control (NC) received distilled water while the dimethoate control (DC) received 6 mg/kg.bw.day-1 dimethoate dissolved in distilled water. The experimental groups E1, E2, E3, and E0 received dimethoate (6 mg/kg.bw) + ZMFM (100 mg/kg.bw-1), dimethoate (6 mg/kg.bw) + ZMFM (200 mg/kg.bw-1), dimethoate (6 mg/kg.bw) + ZMFM (300 mg/kg.bw-1), and ZMFM (300 mg/kg.bw-1) only. Both the normal control and the dimethoate control groups were used to compare the results. After 90 days, rats were sacrificed, blood was collected for biochemical assays, and livers were harvested for histological study. Results: High phenol content was estimated, and 2, 2-diphenyl-1-picryl hydrazyl radical (DPPH) spectrophotometric, thin layer chromatography (TLC) and 2, 2-Azobis-3-ethyl benzothiazoline-6-sulphonic acid (ABTS) assays showed a high antioxidant activity among the extracts. The preventive effects observed in the E1, E2 and E3 groups proved that the extract could prevent dimethoate toxicity by maintaining normal reduced glutathione (GSH), vitamin C and E, superoxide dismutase, catalase, cholineasterase and lipid profiles. The preventive effect was observed to be dose dependent. The EO group showed no extract-induced toxicity. Histological observations agreed with the results obtained in the biochemical studies. Conclusion: The study demonstrated that ZM methanol fruit extract is capable of attenuating dimethoate-induced toxicity because of its high antioxidant activity.

The microcrystalline cellulose (MCC) was prepared from oil palm biomass, empty fruit bunch (EFB) for increasing the usability of EFB. The morphological, physical and chemical properties of MCC made from EFB were evaluated by comparing with those of the commercial MCC obtained from AVICEL. The EFB-MCC had the wider distribution in particle size and there were many small particles around $10{\mu}m$. There were no significant differences in the cellulose crytallinity and the chemical composition between EFB-MCC and AVICEL-MCC. The properties of tablet samples made by the common direct compression process were evaluated depending on the types of MCC and the compression pressure during tablet making process. The tablet made of EFB MCC showed the higher compressed structure, which resulted in the less disintegration by the water soaking treatment than those made of Avicel-MCC. The results of this study showed that the EFB-MCC could be utilized as one of the commercial MCC.
A blue mold on tomato fruit caused by Penicillium oxalicum occurred sporadically in a greenhouse at Gyeongsangnam-do Agricultural Research and Extension Services. Infection usually occurred through wounds or cracks on the fruits. Symptoms began with water soaking lesions, then became watery and softened eventually. Colony of the causal fungus was white at the early growing stage, turned green on Czapek yeast extract agar and malt extract agar. Conidia were ellipsoidal in shape and $2-6{\times}2-4{\mu}m$ in size. Stipes were septate, smooth, thin walled, and $90-280{\times}3-4{\mu}m$ in size. Penicilli were mostly biverticillate. Ramuli were 1-3 groups, smooth, and $10-16{\times}2-3{\mu}m$ in size. Rami were 1-2 groups and $6-30{\times}2-4{\mu}m$ in size. Metulae were 2-3(5) verticils, smooth, and $12-20{\times}3-4{\mu}m$ in size. Phialides were 5-7 verticilate, ampulliform to cylindroidal, smooth, and $8-12{\times}2-3{\mu}m$ in size. Optimum temperature for growth was about $25^{\circ}C$. Pathogenicity of the fungus was proved on tomato fruit according to Koch's postulation. On the basis of mycological and pathological characteristics, the fungus was identified as P. oxalicum Currie & Thom. This is the first report of the blue mold on tomato fruit caused by P. oxalicum in Korea.

Recently sawdust cultivation of Shiitake mushroom (Lentinula edodes ) is getting increased because log cultivation is getting difficult to get oak logs. It is important to make mycelia browning on the substrate surface in sawdust cultivation. This browned surface plays an important role like as artificial bark of the oak log, which protects the other pests and suppresses water evaporation in the substrate. The period for mycelia browning is so long that the sawdust cultivation of Shiitake mushroom can not spread well into the mushroom farms. In this article we would like to discuss about the effect of environmental condition to the mycelial browning during sawdust bag cultivation for the To reduce the period required for browning of substrates, sawdust substrates was illuminated light with difference intensity. One hundred Lux light illumination was needed for producing normal yield of fruit body but fruit body yield was low and abnormally shaped fruit body was produced when cultured under the dark condition of incubation. Illumination over 200lux is necessary for the successful browning of substrates during incubation. Optimum incubation temperature for browning of substrates and fruiting was $25^{\circ}C$. The treatment of cotton plug with different size to identify the effect of aeration on the browning of substrates and fruiting showed rapid mycelial growth and reduced the periods for browning as the size of cotton plug was bigger. However, yield of fruit body was the highest at 16mm diameter cotton plug as compared to 20mm of that. $CO_2$ content in vessel of substrates was low as the size of cotton plug was bigger during incubation. $CO_2$ content during incubation of substrate was highest in periods between 8 week and 14 week after inoculation of shiitake when substrate was changed color into brown. $C_2H_4$ content in vessel with substrates was highest at 8mm diameter cotton plug and it was increased by order of 12, 16, 20, 0, 4 mm diameter cotton plug during substrate incubation. Sawdust substrate was soaked in cold water for different time to identify soaking effect of sawdust substrate on fruit body yield and activities of enzymes in these substrates were investigated. The fruit body yield was increased up to 40% by soaking substrates in comparison with unsoaked substrates. The soaked substrates showed 165, 175g/1,000ml at treatment of 4 and 15 hours, respectively. Cellulose activities in soaked substrates were not changed with soaking time, but activities of laccase, lignin degradation enzyme, were drastically increased up to 4 times in comparison with unsoaked substrates.
The fruit rot of pumpkin caused by Sclerotium rolfsii occurred sporadically at Gyeongsangnam-do Agricultural Research and Extension Services, Korea in 2004 and 2005. The typical symptom of the disease included water-soaking, fruit rotting and wilting. White mycelial mats were spread over infected fruits and then sclerotia were formed on the lesions and near soil surface. The sclerotia were globoid in shape, 1$\sim$3 mm in size and white to brown in color. The optimum temperature for mycelial growth and sclerotia formation was at $30^{\circ}C$ on PDA and the hyphal width ranged from 4 to 8 ${\mu}m$. The typical clamp connections were observed in the hyphae of the fungus grown on PDA. On the basis of mycological characteristics and pathogenicity to host plants, this fungus was identified as Sclerotium rolfsii Saccardo. This is the first report on the fruit rot of Cucurbita spp. caused by S. rolfsii in Korea.
