Effect of Water Extracts from Root of Taraxacum officinale on Innate and Adaptive Immune Responses in Mice (민들레 뿌리 물 추출물의 마우스 선천 및 획득 면역계에 미치는 효과)

  • Hot-water($100^{\circ}C$) and cold-water($4^{\circ}C$) extracts of Taraxacum officinale root were assessed for the effects of innate and adaptive immune responses in mice. Hot water extracts(TO-100) and cold water extracts(TO-4) did not affect the viability of macrophages at concentrations below to 18 mg/ml and 8 mg/ml, respectively. The thioglycollate-induced macrophages cultured with TO-100 and TO-4 produced a significantly higher quantity of various cytokines, such as IL-6 and IL-12, than those treated with medium. This shows that the extracts potently stimulated the innate immune response. When mice were subcutaneously immunized(sc) with OVA+FIA(Freund's incomplete adjuvant)-emulsified TO-100, TO-100 did not affect the production of IgE, but enhanced the production of IgG1, IgG2a and IgG2b. The culture supernatant obtained from the splenocytes of mice treated with OVA+FIA-emulsified TO-100 also evidenced elevated levels of both OVA-specific Th1-type(IFN-$\gamma$) and Th2-type cytokines(IL-4, IL-6 and IL-10). These results suggested that TO-100 can modulate the immune responses to allergens in mice.

Effect of Water Addition Ratio, Stirring Time and Ca Salts on Textural Properties of Soygel (콩묵 제조시 가수량, 교반시간 및 Ca염의 양이 텍스쳐 특성에 미치는 영향)

  • Rheological properties of whole soybean gel(soygel) were investigated as affected by the water addition ratio, stirring time and Ca salts. The soygel was prepared by suspension of whole soy flour(WSF, 300 mesh) in boiling water, addition of sodium alginate and Ca salts followed by thorough mixing and gel formation at $4^{\circ}C$. The texture properties of hardness, adhesiveness and cohesiveness of the gel were increased as the stirring time prolonged from 5 to 30 minutes. From the results of the rheological and sensory properties, 20 minutes of stirring time was selected for whole soybean gel preparation. Eventhough increase in water addition ratio from 8 to 12 times(water/WSF, v/w) resulted a decrease in hardness and adhesiveness, 10 times ratio was chosen as proper the water addition based on textural uniformity. Among the Ca salts, $CaSO_4$ produced the highest hardness followed by Ca $gluconate-CaSO_4$ mixture(413g) and Ca gluconate at the water addition level of 10 times. In order to determine the amounts of Ca salts, and 0.125g of Ca gluconate or $CaSO_4$ per g WSF were found to be optimum in terms of textural and sensory properties. The proper mixing ratio of Ca gluconate and $CaSO_4$ was found to be 50 : 50, 25 : 75 and 0 : 100.

  • In order to investigate the pathogenic genes and genetic relationships of Y. pseudotuberculosis, we isolated 9 strains of Y. pseudotuberculosis from about 380 spring water sites in Seoul and carried out antibiotic susceptibility test, biological test and molecular typing. All isolated strains were distributed throughout the northeast area in Seoul (Mt. Bookhan, Mt. Soorak, Mt. Boolam and etc...).Antibiotic susceptibility test revealed that all the strains were susceptible to chloramphenicol, gentamicin, neomycin and amoxicillin/clavulanic acid, but were resistant to novobiocin and vancomycin. Effect of Food Waste Mixing on Hydrogen Gas Production in Anaerobic Digestion of Brown Water from Urine Diversion Toilet (소변분리변기오수(Brown water)의 혐기성 처리 시 음식물 쓰레기 혼합에 따른 수소생산 특성)

  • The study was conducted to evaluate the effect of addition of food waste in brown water for anaerobic hydrogen production. Batch experiment was carried out to determine appropriate food waste to brown water mixing ratio. Maximum hydrogen yield of $6.92mmol\;H_2/g\;COD_{removed}$ was obtained at 70% food waste and 30% brown water. Semi-pilot scale reactor was operated based on result of batch experiment. Semi-pilot reactor operated, mixing 70% food waste and 30% brown water showed significant increment in butyric acid concentration. B/P (Butyric to propionic acid ratio) which is considered as governing factor for hydrogen production was found high (52.64). Maximum hydrogen yield of $25.03mmol\;H_2/g\;COD_{removed}$ was obtained. Result of this study concluded that mixing of food waste to brown water at appropriate ratio assists in enhanced hydrogen fermentation.

  • Physiological activities of the 70% ethanol extract from boiled-water of Enteroctopus dofleini (BE) upon a gamma irradiation were determined to examine the possible use of the food processing by-product as a functional material for food or cosmetic composition. Electron donating ability and the content of total phenolic compounds increased from 65% to 72% and from 406 ppm to 476 ppm, respectively, by 10 kGy gamma irradiation. A Salmonella mutagenicity assay indicated that the gamma-irradiated BE did not show any mutagenicity. Tyrosinase inhibition activity of the irradiated BE also increased from 57% to 86% with irradiation doses. These results show that irradiation can be used to increase the physiological activities of BE for its potential use as food or cosmetic materials.

  • Fibrionolytic activity(FA) of kimchi in rat and FAs of water and methanol extracts of kimchi ingredient were determined using fibrin plate method to see where the active principles for FA are present in kimchi. Nine Sprague-Dawley per each group were fed diet containing 3, 5 or 10% of freeze dried kimchi for 6 weeks. The FA of plasma obtained from 10% kimchi group showed stronger FA than that of control. FAs of 3 and 5% kimchi group were not noticeable. FA from methanol extract of kimchi was approximately 6 times higher than that from water extract assuming that the active material may be present in a fat soluble fraction. From water extract of kimchi ingredients, dropwort, green onion, radish and baechu showed FA in descending order. And from methanol extracts, red pepper powder, radish, green onion and baechu showed FA in decending order. The Fa of methanol extract of red pepper powder was the most noticeable among samples.

Suitability of Hot Water Extract from Panax ginseng Sprout Powder as a Dairy Additive (새싹인삼 분말 열수추출물의 유식품 적용 연구)

  • This study aimed to determine the effect of extraction temperature and time on the antioxidant activity of hot water extract from Panax ginseng sprout powder and to evaluate the suitability of this extract for use in dairy products. Water-soluble fractions of commercial Panax ginseng sprout powder were obtained by hot water extraction at 25, 60, or 80℃ for 0.5, 2, 12, or 24 h. The antioxidant activity of each extract was evaluated by measuring its free radical scavenging activity against 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS). DPPH and ABTS free radical scavenging activity increased with extraction temperature from 25 to 80℃. At 80℃, increasing the extraction time from 0.5 to 2 h led to increases in DPPH and ABTS free radical scavenging activity. Thus, the extract obtained under 2 h at 80℃ was selected for addition to milk and yogurt. After 16 days of storage, there were no significant changes in the pH of the milk or the antioxidant activity of the extract. With regard to yogurt fermentation, adding the extract did not affect the pH or the number of viable lactic acid bacteria. In conclusion, hot water extract from Panax ginseng sprout powder can be added to dairy products to enhance antioxidant activity.

  • Antioxidant activity of Akebia quinata Decne was evaluated. Water extract (0.5 g/50 mL) of flowers and leaves of A. quinata were prepared and total phenol contents and radical scavenging activity of the extracts was determined for antioxidant activity. The total phenol contents of extracts from A. quinata flowers (FAQ) and leaves (LAQ) were $30.05{\mu}M\;and\;20.23{\mu}M$, while the radical scavenging activity of FAQ and LAQ were 60.51 % and 52.97%, respectively. In addition, the effect of FAQ and LAQ extract on DNA damage induced by $H_2O_2$ in human lymphocytes was evaluated by comet assay. The FAQ and LAQ showed strong inhibitory effect against DNA damage induced by $200{\mu}M$ of $H_2O_2$. These results suggest that water extracts of A. quinata Decne flowers and leaves showed significant (p<0.05) antioxidant activity and protective effect against oxidative DNA damage.

Effects of Pretreatment Time and pH low set value on Continuous Mesophilic Hydrogen Fermentation of Food Waste (열처리 시간과 pH 하한값이 음식물쓰레기 연속 중온 수소 발효에 미치는 영향)

  • Since 2005, food waste has been separately collected and recycled to animal feed or aerobic compost in South Korea. However, the conventional recycling methods discharge process wastewater, which contain pollutant equivalent to more than 50% of food waste. Therefore, anaerobic digestion is considered as an alternative recycling method of food waste to reduce pollutant and recover renewable energy. Recent studies showed that hydrogen can be produced at acidogenic stage in two-stage anaerobic digestion. In this study, the authors investigated the effects of pretreatment time and pH low set value on continuous mesophilic hydrogen fermentation of food waste. Food waste was successfully converted to $H_2$ when heat-treated at $70^{\circ}C$ for 60 min, which was milder than previous studies using pH 12 for 1 day or $90^{\circ}C$. Organic acid production dropped operational pH below 5.0 and caused a metabolic shift from $H_2/butyrate$ fermentation to lactate fermentation. Therefore, alkaline addition for operational pH at or over 5.0 was necessary. At pH 5.3, the result showed that the maximum hydrogen productivity and yield of 1.32 $m^3/m^3$.d and 0.71 mol/mol $carbohydrate_{added}$. Hydrogen production from food waste would be an effective technology for resource recovery as well as waste treatment.

Biological activity of functional foods and anti-microbial activity of phenolics from Sambucus sieboldiana var. pendula leaves

  • This study aimed to investigate the biological and antibacterial activities of phenolics from Sambucus sieboldiana var. pendula leaf extract. The phenolic contents of the S. sieboldiana var. pendula extracted with water and ethanol was 11.60 and 12.39 mg/g, respectively. The inhibitory effects of the water and ethanol extracts on the angiotensin converting enzyme (ACE) were 71.94-92.08 and 48.42-78.33%, respectively, and on xanthine oxidase (XOase) were 42.11-100 and 35.61-100%, respectively, at a phenolic concentration of 50-200 ㎍/mL. All these effects were found to be concentration-dependent. Additionally, the leaf extracts exhibited excellent antibacterial activities against Streptococcus mutans, Staphylococcus aureus, and Propionibacterium acnes. Hence, S. sieboldiana var. pendula was confirmed to have excellent antihypertensive, antigout, and antimicrobial properties.
